Periostin Is Required for the Maintenance of Muscle Fibers during Muscle Regeneration.

Skeletal muscle regeneration is a well-organized process that requires remodeling of the extracellular matrix (ECM). In this study, we revealed the protective role of periostin, a matricellular protein that binds to several ECM proteins during muscle regeneration. In intact muscle, periostin was localized at the neuromuscular junction, muscle spindle, and myotendinous junction, which are connection sites between muscle fibers and nerves or tendons. During muscle regeneration, periostin exhibited robustly increased expression and localization at the interstitial space. Periostin-null mice showe

Reliability of an ultrasound-motion capture approach to measure the lengthening behavior of the gastrocnemius medialis muscle-tendon unit, muscle belly, and Achilles tendon in vivo during gait.

BACKGROUND: Human muscle-tendon units (MTUs) exhibit significant plasticity and are subject to structural and functional alterations in response to various pathological conditions, such as diseases or disorders. The assessment of these pathological modifications, as well as the evaluation of therapeutic interventions, necessitates the application of robust and scientifically validated examination methodologies. In this study, a simple approach combining two-dimensional (2D) ultrasound and three-dimensional (3D) motion capture was developed and evaluated with regard to the reliability of dynami

Severe Flexor Digitorum Profundus Muscular Adhesion by Pseudo-Volkmann's Contracture without Fracture: A Case Report and Literature Review.

Volkmann's ischemic contracture is a condition characterized by permanent ischemic damage to muscles and nerves due to vascular insufficiency, resulting in flexion contractures of the affected limb. In contrast, pseudo-Volkmann's contracture presents with similar clinical features but lacks ischemic damage and has the potential for complete recovery. We report a case of a 39-year-old man who developed failure of extension in the middle and ring fingers of the left hand following blunt forearm trauma from a rolling machine. Despite no skin breakage or fracture, his symptoms progressively worsen

MIF versus SIF Motoneurons, What Are Their Respective Contribution in the Oculomotor Medial Rectus Pool?

Multiply-innervated muscle fibers (MIFs) are peculiar to the extraocular muscles as they are non-twitch but produce a slow build up in tension on repetitive stimulation. The motoneurons innervating MIFs establish en grappe terminals along the entire length of the fiber, instead of the typical en plaque terminals that singly-innervated muscle fibers (SIFs) motoneurons establish around the muscle belly. MIF motoneurons have been proposed to participate only in gaze holding and slow eye movements. We aimed to discern the function of MIF motoneurons by recording medial rectus motoneurons of the oc

Regulation of myonuclear positioning and muscle function by the skeletal muscle-specific CIP protein.

The appropriate arrangement of myonuclei within skeletal muscle myofibers is of critical importance for normal muscle function, and improper myonuclear localization has been linked to a variety of skeletal muscle diseases, such as centronuclear myopathy and muscular dystrophies. However, the molecules that govern myonuclear positioning remain elusive. Here, we report that skeletal muscle-specific CIP (sk-CIP) is a regulator of nuclear positioning. Genetic deletion of sk-CIP in mice results in misalignment of myonuclei along the myofibers and at specialized structures such as neuromuscular junc

Isolated pectoralis minor tear at the costal origin: a case-based review of the literature.

BACKGROUND: Isolated tears of the pectoralis minor (PMi) are rare and often misdiagnosed as pectoralis major or costochondral injuries. Since the first report in 2004, only eleven cases have been described in the English-language literature, all involving the myotendinous junction or coracoid insertion. A tear at the costal origin has not previously been reported. CASE PRESENTATION: A 46-year-old male developed acute anterior chest pain after pulling a heavy hospital bed. Physical examination revealed focal tenderness over the third to fifth ribs and pain with resisted internal rotation. Magn

Potential cross-talk between muscle and tendon in Duchenne muscular dystrophy.

PURPOSE: To describe potential signaling (cross-talk) between dystrophic skeletal muscle and tendon in Duchenne muscular dystrophy. MATERIALS AND METHODS: Review of Duchenne muscular dystrophy and associated literature relevant to muscle-tendon cross-talk. RESULTS AND CONCLUSIONS: Duchenne muscular dystrophy results from the absence of the protein dystrophin and the associated dystrophin - glycoprotein complex, which are thought to provide both structural support and signaling functions for the muscle fiber. In addition, there are other potential signal pathways that could represent cross-ta

Division Tenorrhaphy: A Novel Technique for Chronic or Failed Nonoperatively Treated Achilles Tendon Rupture.

UNLABELLED: Here we describe a modified open technique for the repair of a ruptured Achilles tendon using multiple looped sutures with the creation of interdigitating tendon stumps maximizing surface area for suture application as well as allowing for significant tissue overlay. This technique produces a high strength repair that is useful in cases of extensive degeneration or poor-quality tissue. Degenerative tissue may be encountered with chronic ruptures or failed nonoperative treatment, as well as those ruptures that occur at the proximal myotendinous junction. We present 2 cases in which

Rupture of the lateral head of the gastrocnemius muscle at the musculotendinous junction mimicking a compartment syndrome.

This case report documents an acute rupture of the lateral head of the gastrocnemius muscle at the myotendinous junction and a lateral dislocation of the tibialis anterior tendon with a clinical presentation consistent with a compartment syndrome, despite the intraoperative finding of a rupture of the posterior and lateral compartments. Extensive hematoma formation led to marked edema, paresthesias, muscle weakness, and severe pain in the involved leg. Surgical repair of the torn muscle and dislocated tendon and evacuation of the dissecting hematoma resulted in a well-functioning extremity. Th

Partial tear at the musculotendinous junction of the teres major after anterior latissimus dorsi and teres major tendon transfer: associated factors and clinical impact in 256 patients.

BACKGROUND: Anterior latissimus dorsi and teres major (aLDTM) tendon transfer is an established procedure for isolated irreparable subscapularis tendon tear and anterosuperior irreparable rotator cuff tears. However, partial tear at the musculotendinous junction (MTJ) of the teres major (TM) has been observed on post-operative magnetic resonance imaging (MRI). The associated factors and clinical significance of these lesions remain unclear. This study aimed to identify factors associated with these lesions and to evaluate their impact on clinical outcomes. METHODS: We retrospectively reviewed

Isolated avulsion of the short head of the biceps tendon from the coracoid insertion: A case report and narrative review of the literature.

Isolated injuries to the short head of the biceps (SHB) are uncommon, and the existing literature mainly includes case reports documenting tears at the muscle belly or myotendinous junction. This case report presents the first reported SHB tendon injury at the coracoid insertion in a 34-year-old healthy male patient after trauma while playing with his daughter. The injury occurred with the arm forced into hyperextension, abduction, and external rotation, with the elbow fully extended. Diagnosis was confirmed by MRI, showing an isolated insertional tear of the SHB tendon with intact coracobrach

Overuse Injuries in the Muscle-Tendon-Enthesis Unit.

Overuse injuries of the bone-tendon-muscle unit are a common yet underrecognized source of pain and dysfunction in athletes and the general population. They arise from repetitive mechanical loading and age-related degenerative changes that exceed the adaptive capacity of musculoskeletal tissues. Imaging is central to early detection, accurate characterization, and treatment planning. This review presents a practical, imaging-based framework using magnetic resonance imaging and radiography, emphasizing key diagnostic features, anatomical considerations, and classification. Content is organized

Endoscopic repair of proximal hamstring avulsion.

Hamstring muscle injuries are common in athletes and mostly consist of sprains at the myotendinous junction, which often respond well to conservative treatment. Proximal hamstring avulsion injuries, though less common, can be severely debilitating. This injury is often seen in water skiers but has been described in many other sports and in middle-aged patients. Complete avulsions in young and active individuals do not respond well to conservative treatment and may require surgical repair. In contrast, many partial tears may be treated nonoperatively. However, when symptoms continue despite a t

Soluble Heparin Binding Epidermal Growth Factor-Like Growth Factor Is a Regulator of GALGT2 Expression and GALGT2-Dependent Muscle and Neuromuscular Phenotypes.

GALGT2 (also B4GALNT2) encodes a glycosyltransferase that is normally confined to the neuromuscular and myotendinous junction in adult skeletal muscle. GALGT2 overexpression in muscle can inhibit muscular dystrophy in mouse models of the disease by inducing the overexpression of surrogate muscle proteins, including utrophin, agrin, laminins, and integrins. Despite its well-documented biological properties, little is known about the endogenous regulation of muscle GALGT2 expression. Here, we demonstrate that epidermal growth factor receptor (EGFR) ligands can activate the human GALGT2 promoter.

Plakins in striated muscle.

Striated muscle cells contain numerous architectural proteins that contribute to the function of muscle as generators of mechanical force. Among these proteins are crosslinkers belonging to the plakin family, namely plectin, microtubule-actin crosslinking factor (ACF7/MACF1), bullous pemphigoid antigen 1 (Bpag1/dystonin), and desmoplakin. These plakin family members, in particular plectin and Bpag1/dystonin, exist as several isoforms. The domain organization of these plakin variants dictates their subcellular location and the proteins with which they interact. Several studies suggest that plak

Latissimus dorsi. Case report of a rare lesion diagnosed by magnetic resonance imaging.

The latissimus dorsi is a flat muscle that covers most of the back. Its injury happens when there is avulsion of the tendon. There are currently few reported cases in the literature, but most sources agree that latissimus dorsi injuries are more common among professional athletes. The main injury sites should be highlighted: tendon, myotendinous junction, ventral muscle, and costal muscle. Patients with latissimus dorsi muscle injury describe a burning or popping sensation in the armpit followed by pain and weakness. Magnetic resonance imaging confirms the diagnosis of avulsion injury, definin

Congenital nystagmus: fine structure of human extraocular muscles.

The ultrastructure of the extraocular muscles of patients affected by congenital nystagmus is still to be defined. Specimens obtained from patients suffering from either oculomotor congenital nystagmus or nystagmus associated with strabismus were studied and compared with specimens obtained from patients enucleated for various pathologies but not affected by any oculomotor system disorder. The scleral myotendinous junction and the muscle body (venter) were examined. In both these areas, damaged muscle fibers were found. 'Mitochondrial cores' and concentric myofibrils characterized the extraocu

Review: myogenic and muscle toxicity targets of environmental methylmercury exposure.

A number of environmental toxicants are noted for their activity that leads to declined motor function. However, the role of muscle as a proximal toxicity target organ for environmental agents has received considerably less attention than the toxicity targets in the nervous system. Nonetheless, the effects of conventional neurotoxicants on processes of myogenesis and muscle maintenance are beginning to resolve a concerted role of muscle as a susceptible toxicity target. A large body of evidence from epidemiological, animal, and in vitro studies has established that methylmercury (MeHg) is a po

Is there any sense in the Palisade endings of eye muscles?

Palisade endings (PEs), which are unique to the eye muscles, are associated with multiply innervated muscle fibers. They lie at the myotendinous junctions and form a cap around the muscle fiber tip. They are found in all animals investigated so far, but their function is not known. Recently, we demonstrated that cell bodies of PEs and tendon organs lie around the periphery of the oculomotor nucleus in the C- and S-groups. A morphological analysis of these peripheral neurons revealed the existence of different populations within the C-group. We propose that a small group of round or spindle-sha
