Dietary antioxidant for disease prevention corroborated by the Nrf2 pathway.

Dietary antioxidants are widely distributed in various types of our food. They are strongly associated with reduced risk of many chronic diseases such as atherosclerosis, cancer, and Alzheimer's diseases. They include vitamins such as vitamins A, E, C, and carotenoids. Also, some minerals like; zinc, manganese, copper, iron, and selenium are essential for the activity of antioxidant enzymes. Furthermore, dietary polyphenols and flavonoids are considered as potent antioxidant compounds. Vegetables, fruits, and edible herbs are the richest sources of such antioxidants. Antioxidants reduce oxidat

Biogenic Nanoselenium Particles Effectively Attenuate Oxidative Stress-Induced Intestinal Epithelial Barrier Injury by Activating the Nrf2 Antioxidant Pathway.

In the present study, a new form of selenium nanoparticle (biogenic nanoselenium (BNS) particles) was synthesized using bacteria. The protection of BNS particles against oxidative stress-induced intestinal barrier dysfunction and the inherent mechanisms of this process were investigated, and selenomethionine (SeMet) and chemically synthesized nanoselenium (Nano-Se) particles were used for comparison. Characterization of BNS particles revealed that they were monodispersed and homogeneous spheres, with an average size of 139.43 ± 7.44 nm. In the mouse model of intestinal oxidative stress, BNS pa

Roles nrf2 plays in myeloid cells and related disorders.

The Keap1-Nrf2 system protects animals from oxidative and electrophilic stresses. Nrf2 is a transcription factor that induces the expression of genes essential for detoxifying reactive oxygen species (ROS) and cytotoxic electrophiles. Keap1 is a stress sensor protein that binds to and ubiquitinates Nrf2 under unstressed conditions, leading to the rapid proteasomal degradation of Nrf2. Upon exposure to stress, Keap1 is modified and inactivated, which allows Nrf2 to accumulate and activate the transcription of a battery of cytoprotective genes. Antioxidative and detoxification activities are imp

Inhibitory effects of benzaldehyde derivatives from the marine fungus Eurotium sp. SF-5989 on inflammatory mediators via the induction of heme oxygenase-1 in lipopolysaccharide-stimulated RAW264.7 macrophages.

Two benzaldehyde derivatives, flavoglaucin (1) and isotetrahydro-auroglaucin (2), were isolated from the marine fungus Eurotium sp. SF-5989 through bioassay- and 1H NMR-guided investigation. In this study, we evaluated the anti-inflammatory effects of these compounds in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. We demonstrated that compounds 1 and 2 markedly inhibited LPS-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production by suppressing inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression without affecting cell viability. We also

4-amino-3-(phenylselanyl) benzenesulfonamide attenuates intermittent cold stress-induced fibromyalgia in mice: Targeting to the Nrf2-NFκB axis.

Stress is widely recognized as the primary environmental factor associated with chronic pain conditions, including fibromyalgia. A recent study demonstrated the potential antinociceptive effects of 4-amino-3-(phenylselanyl) benzenesulfonamide (4-APSB) in acute nociceptive animal models due to its antioxidant and anti-inflammatory properties. However, the efficacy of 4-APSB in managing chronic painful conditions, such as fibromyalgia, has not been explored so far. This study investigated the pharmacological effects of 4-APSB in an experimental model of fibromyalgia induced by intermittent cold

Polystyrene microplastics mediate inflammatory responses in the chicken thymus by Nrf2/NF-κB pathway and trigger autophagy and apoptosis.

Microplastics (MPs) are a hot environmental contaminant now. However, researchers paid little attention to their effects on immune organs such as the thymus. Here, we exposed chickens to a concentration gradient of polystyrene microplastics (PS-MPs) and then followed the decrease in the thymus index. HE staining showed cellular infiltration in the thymus. The assay kit corroborated that PS-MPs impelled oxidative stress in the thymus: increased MDA levels, downregulated antioxidants such as SOD, CAT, and GSH, and significantly undermined total antioxidant capacity. Western blotting and qRT-PCR

Geniposide suppressed OX-LDL-induced osteoblast apoptosis by regulating the NRF2/NF-κB signaling pathway.

BACKGROUND: Osteoporosis (OP), due to microarchitectural alterations, is associated with decreased bone mass, declined strength, and increased fracture risk. Increased osteoblast apoptosis contributes to the progression of OP. Natural compounds from herbs provide a rich resource for drug screening. Our previous investigation showed that geniposide (GEN), an effective compound from Eucommia ulmoides, could protect against the pathological development of OP induced by cholesterol accumulation. METHODS: The rat OP models were duplicated. Dual-energy X-ray absorptiometry, hematoxylin and eosin st

Chronic Exposure to Endocrine Disruptor Vinclozolin Leads to Lung Damage via Nrf2-Nf-kb Pathway Alterations.

Endocrine-disrupting substances (EDS) are common and pervasive in our environment and pose a serious risk to both human and animal health. Endocrine-disrupting compounds (EDCs) have been associated with a variety of detrimental human health effects, including respiratory issues, as a result of their ability to disrupt cell physiology. Vinclozolin ((RS)-3-(3,5-Dichlorophenyl)-5-methyl-5-vinyloxazolidine-2,4-dione) is a common dicarboximide fungicide used to treat plant diseases. Several studies have analyzed the effects of vinclozolin exposure on the reproductive system, but less is known about

The integrated stress response effector ATF4 is an obligatory metabolic activator of NRF2.

The redox regulator NRF2 becomes activated upon oxidative and electrophilic stress and orchestrates a response program associated with redox regulation, metabolism, tumor therapy resistance, and immune suppression. Here, we describe an unrecognized link between the integrated stress response (ISR) and NRF2 mediated by the ISR effector ATF4. The ISR is commonly activated after starvation or ER stress and plays a central role in tissue homeostasis and cancer plasticity. ATF4 increases NRF2 transcription and induces the glutathione-degrading enzyme CHAC1, which we now show to be critically import

Polycyclic Limonoids as Potential Nrf2 Inducers from Trichilia connaroides.

An investigation of Trichilia connaroides uncovered connarolides A-D (1-4, respectively), four limonoids with exceptional multiring systems. Compound 1 showcased a unique 5/8/6/6-fused ring system distinguished by a bicyclo[5.2.12,4]decane A/B ring framework. Compound 2 featured a 5/8/6/5/6 scaffold incorporating a triply fused furan ring, and a long-conjugated system resulted from 3-deoxygenation. X-ray crystallography established the rare 6/3/6/6/6 ring system of 3. Moreover, 4 was identified as an antioxidant, conferring cytoprotective activity through Nrf2 activation.

The Diet-Multiple Sclerosis Connection: Oxidative Stress and Emerging Mechanisms.

Multiple sclerosis (MS) is an autoimmune neuroinflammatory disease resulting in myelin degeneration and progressive disability. Oxidative stress plays a crucial role in MS pathogenesis and progression. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a master regulator of the antioxidant mechanisms and its upregulation is associated with beneficial effects in MS. Among the environmental factors influencing MS onset and progression, diet represents a promising non-pharmacological strategy to modulate Nrf2, potentially improving MS outcomes. Indeed, several natural compounds present in Medi

Targeting the Interplay Between Autophagy and the Nrf2 Pathway in Parkinson's Disease with Potential Therapeutic Implications.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder marked by the progressive degeneration of midbrain dopaminergic neurons and resultant locomotor dysfunction. Despite over two centuries of recognition as a chronic disease, the exact pathogenesis of PD remains elusive. The onset and progression of PD involve multiple complex pathological processes, with dysfunctional autophagy and elevated oxidative stress serving as critical contributors. Notably, emerging research has underscored the interplay between autophagy and oxidative stress in PD pathogenesis. Given the limited effica

Role of miR-128-3p/Nrf2 Axis in DPhP-Induced GC-2spd(ts) Apoptosis and Inhibition of Cellular Autophagy.

Organophosphorus flame retardants (OPFRs) are recognized as potential endocrine disruptors. Several studies have demonstrated that contamination with OPFRs increases the risk of infertility. Our prior research showed that inhibition of the nuclear factor erythroid-derived 2-related factor (Nrf2)/p53 signaling pathway is an essential regulator of DPhP-induced reproductive injury. In this study, we aimed to uncover potential epigenetic mechanisms involved in DPhP-induced GC-2 cell dysfunction. Our results established that DPhP promoted miR-128-3p expression in GC-2 cells, targeted the Nrf2 signa
