Porcine epidemic diarrhea virus (PEDV) remains a major threat to the global swine industry, with current inactivated vaccines offering limited cross-protection due to imperfect antigen design and weak immune activation. Here, we engineered a full-structural-protein virus-like particle (VLP) co-assembled with PEDV S, M, N, and E proteins into uniform nanoscale particles, further formulated with a dual-adjuvant system of AS03 and CpG ODN. The VLP + AS03 + CpG nanovaccine elicited 10-12-fold higher PEDV-specific IgG than unadjuvanted VLPs, outperformed a commercial inactivated vaccine in mice, an

Current cancer vaccines mainly focusing on inducing cancer cells-specific immune responses showed very limited efficacy because of the extremely complicated tumor microenvironments. Herein, we facilely construct Se nanoadjuvant-based bivalent DNA nanovaccines targeting both melanoma and cancer-associated fibroblast (CAF), based on the crucial roles of CAF in desmoplastic tumors and Se element in immune regulations. Intriguingly, the bivalent Se nanovaccines significantly induce robust bi-specific immune responses such as more activated macrophages, higher population of OVA+FAP+CD8+ T lymphocyt

Tumor vaccines represent a promising strategy for the prevention and treatment of cancer. However, suboptimal targeting efficiency to dendritic cells (DCs) and inadequate antigen presentation by metabolically dysfunctional DCs significantly hinder vaccination efficacy. In vitro drug screening reveals that lovastatin (Lov) effectively inhibits antigen degradation in DCs by modulating the mevalonate (MVA) pathway, thereby enhancing antigen presentation. Additionally, tumor-derived water-insoluble neoantigens contain abundant immunogenic mutated components but suffer from limited vaccination effi

Nanoparticles (NPs) have been widely utilized in vaccine design. Although numerous NPs have been explored, NPs with adjuvant effects on their own have rarely been reported. We produce a promising self-assembled NP by integrating the pentameric Escherichia coli heat-labile enterotoxin B subunit (LTB) (studied as a vaccine adjuvant) with a trimer-forming peptide. This fusion protein can self-assemble into the NP during expression, and polysaccharide antigens (OPS) are then loaded in vivo using glycosylation. We initially produced two Salmonella paratyphi A conjugate nanovaccines using two LTB su

An alternative strategy of choosing photothermal and weak-immunostimulatory porous silicon@Au nanocomposites as particulate cores to prepare a biomimetic nanovaccine is reported to improve its biosafety and immunotherapeutic efficacy for solid tumors. A quantitative analysis method is used to calculate the loading amount of cancer cell membranes onto porous silicon@Au nanocomposites. Assisted with foreign-body responses, these exogenous nanoparticulate cores with weak immunostimulatory effect can still efficiently deliver cancer cell membranes into dendritic cells to activate them and the down

BACKGROUND: Tumor cell-derived extracellular vesicles (tEVs) have garnered significant attention as promising antigen delivery vehicles for the development of cancer vaccines. However, their practical applications are hindered by weak immunogenicity and inadequate lymph node targeting. In this study, we engineered tEVs into "self-adjuvant" multiantigenic nanovaccines that simultaneously accumulate in tumors and lymph nodes (LNs), effectively triggering innate and adaptive immunity capable of recognizing both tumor cells and virus antigen-modified tumor cells to inhibit tumor progression.

Despite the availability of licensed vaccines, influenza causes considerable morbidity and mortality worldwide. Current influenza vaccines elicit an immune response that primarily targets the head domain of the viral glycoprotein hemagglutinin (HA). Influenza viruses, however, readily evade this response by acquiring mutations in the head domain. While vaccines that target the more conserved HA stalk may circumvent this problem, low levels of antistalk antibodies are elicited by vaccination, possibly due to the poor accessibility of the stalk domain to B cell receptors. In this work, it is dem

The virus responsible for the coronavirus viral pandemic is the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Emerging SARS-CoV-2 variants caused by distinctive mutations within the viral spike glycoprotein of SARS-CoV-2 are considered the cause for the rapid spread of the disease and make it challenging to treat SARS-CoV-2. The manufacturing of appropriate efficient vaccines and therapeutics is the only option to combat this pandemic. Nanomedicine has enabled the delivery of nucleic acids and protein-based vaccines to antigen-presenting cells to produce protective immunity aga

Nanotechnology has developed rapidly, giving rise to "nanovaccinology". In particular, protein-based nanocarriers have gained widespread attention because of their excellent biocompatibility. As the development of flexible and rapid vaccines is challenging, modular extensible nanoparticles are urgently needed. In this study, a multifunctional nanocarrier capable of delivering various biomolecules (including polysaccharides, proteins, and nucleic acids) was designed by fusing the cholera toxin B subunit with streptavidin. Then, the nanocarrier was used to prepare a bioconjugate nanovaccine agai

Magnetic hyperthermia therapy (MHT) is an emerging thermotherapeutic approach that has shown promising potential in cancer treatment. However, conventional MHT strategies exhibit limited capacity to activate anti-tumor immune responses, an essential factor for preventing cancer recurrence and metastasis. Therefore, improving the immunomodulatory efficacy of MHT remains a critical challenge. This study designed and developed a novel magneto-thermal probe, PSRNP, by conjugating anti-PD-L1 and RS09 peptides onto the surface of superparamagnetic iron oxide nanoparticles. The resulting PSRNP nanopr

Tumor nanovaccines have attracted great interest recently, due to a variety of advantages including high stability, efficient antigen packing and delivery, and the capacity to elicit sustained anti-tumor immune responses. Despite these advantages, existing nanovaccines are constrained by intricate manufacturing procedures and high production costs, prompting a need to develop simpler and more cost-effective solutions. In this study, we introduce a novel STING-activating tumor nanovaccine, designated OVA/Tp@Mn-DNA. The preparation of OVA/Tp@Mn-DNA nanovaccine is a straightforward process involv

Food allergy is an escalating global health challenge. Although oral immunotherapy is clinically promising, its efficacy is limited by gastrointestinal adverse effects, allergen instability, and transient desensitization. We developed mannan-decorated allergen nanoparticles (Mann/Allergen NPs) that protected allergen integrity, prolonged intestinal residence (>8 h), and selectively targeted tolerogenic antigen-presenting cells in the small intestinal lamina propria. Mann/Allergen NPs exhibited uniform size, excellent stability, and enhanced dendritic cell uptake compared with free allergen. In

Cancer immunotherapy based on nanovaccines aims to elicit specific anti-tumor immune responses, demonstrating significant therapeutic potential. However, its clinical translation remains hindered by two major challenges: inadequate immunogenicity of the vaccine itself and limited antigen presentation efficiency in dendritic cells (DCs). To overcome these bottlenecks, we have developed a biomimetic nanovaccine for cervical cancer treatment. This vaccine employs a sophisticated biomimetic design: its core consists of iron oxide nanoparticles that serve as both an immune adjuvant and a carrier fo

Brucellosis is a widespread zoonotic disease caused by Brucella, which is a facultative intracellular pathogen. Brucellosis poses a significant challenge to vaccine development due to the ability of Brucella to evade innate immunity, primarily through its atypical and low-toxicity lipopolysaccharide (LPS). To improve the suboptimal immunogenicity of subunit vaccines, a pathogen-mimicking nanovaccine was engineered to deliver the antigen and monophosphoryl lipid A (MPLA, a detoxified LPS analogue) in the present study. Chronic hepatitis B virus (HBV) infection is a global health problem that substantially increases the risk of developing liver disease. The development of a novel strategy to induce anti-HB seroconversion and achieve a long-lasting immune response against chronic HBV infection remains challenging. Here, we found that chronic HBV infection affected the signaling pathway involved in STING-mediated induction of host immune responses in dendritic cells (DCs) and then generated a lymph node-targeted nanovaccine that co-delivered hepatitis B surface antigen (HBsAg) and cyclic diguanylate monophospha

BACKGROUND: Engineered strains of Escherichia coli have been used to produce bioconjugate vaccines using Protein Glycan Coupling Technology (PGCT). Nanovaccines have also entered the vaccine development arena with advances in nanotechnology and have been significantly developed, but chassis cells for conjugate nanovaccines have not been reported.
RESULTS: To facilitate nanovaccine preparation, a generic recombinant protein (SpyCather4573) was used as the acceptor protein for O-linked glycosyltransferase PglL, and a glycol-engineered Escherichia coli strain with these two key components (SC457

Streptococcus pneumoniae is the main cause of diseases such as meningitis, pneumoniae and sepsis, especially in children and old people. Due to costly antibiotic treatment, and increasing resistance of pneumococcus, developing high-efficient protective vaccine against this pathogen is an urgent need. Although the pneumoniae polysaccharide vaccine (PPV) and pneumonia conjugate vaccines (PCV) are the efficient pneumococcal vaccine in children and adult groups, but the serotype replacement of S. pneumoniae strains causes the reduction in efficacy of such vaccines. For overcoming the aforesaid dra

In situ generation of autologous tumor vaccines from dying tumor cells offers a promising route for personalized cancer immunotherapy, yet is often limited by phosphatidylserine (PS)-mediated immunosuppressive clearance that can restrict downstream immune activation and favor M2-like macrophage polarization. Here, we report a sequentially orchestrated nanovaccine (NT@ANX5-P) that couples photodynamic therapy (PDT)-induced immunogenic tumor-cell damage with on-demand PS masking to enhance in situ tumor vaccination. The nanovaccine consists of a photosensitizer core cloaked with genetically engi
