Development and Application of a 1,8-Naphthalimide-Based Fluorescent Probe for Sensitive Detection of Hydrogen Sulfide in Human Blood Serum.

This work implies the preparation, structural and fluorescent characterization, and application of the 1,8-naphthalimide-based fluorescent probe for H2S detection in human blood serum. The mechanism of fluorescent detection of H2S is based on the reduction of the azide group, as the chemical reactive site, in the fluorescent probe structure, to the amino group. Compound 2 possesses another important structural motif, ethanolamine moiety, which modulates physicochemical properties. The synthetic pathway for the preparation of compound 2 is consisted of two reaction steps, and the target compoun

A Serinol-Modified 1,8-Naphthalimide Fluorescent Probe for Selective Detection of H₂S in Human Serum and Hypertensive Patients.

This research involved the design, preparation, and both structural and fluorescent characterization of a novel fluorescent probe (probe 2) for H2S detection in human blood serum as well as in samples from patients with hypertension. The fluorophore 1,8-naphthalimide was selected, while an azide group served as the chemical reactive site for H2S. To increase the bioavailability of the prepared fluorescent probe, the basic 1,8-naphthalimide scaffold was modified by introducing a 2-aminopropane-1,3-diol (serinol) moiety. Fluorescence spectra were recorded to evaluate the effects of pH, time, sel

A naphthalimide-aminal-based pH-sensitive fluorescent donor for lysosome-targeted formaldehyde release and fluorescence turn-on readout.

A naphthalimide-aminal-based fluorescent donor (NAP-FAD-1) is a pH-sensitive smart donor, which features accelerated FA release at pH 5.0 compared to that at pH 7.4 and fluorescence turn-on readout for facile tracking and quantification of FA release. Confocal imaging studies of HeLa cells co-incubated with Lyso-Tracker Red suggested that its FA release is lysosome-targeted.

Nature and Dynamics of the Excited States of Ethynyl-Functionalized Naphthalimide-Phenanthroline Re(I) Complexes Studied by TA and TRIR Spectroscopies.

The synthesis and photophysical properties of four [Re(R-NI-phen)(CO)3Cl] complexes (Re-R), where NI = 1,8-naphthalimide and phen = 1,10-phenanthroline, are reported. The R-NI chromophores were substituted at their 4-positions with R = H (Re-H), ethynyl benzene (Re-EB), ethynyl thiophene (Re-ES), ethynyl trimethylsilane (Re-ET), yielding a series of metal-organic bichromophores. The R-NI-phen organic chromophores were investigated in parallel to better understand the photophysical properties exhibited in the Re-R complexes. The excited-state processes of these molecules were studied by transie

In Situ Real-Time Imaging of Benzyl-Naphthalimide Dyes Revealing Diverse Structural Evolution Dynamics at the Single-Assembly Level.

Fluorescence imaging has emerged as a powerful technique for in situ monitoring of self-assembly dynamics providing critical insights into structural evolution and functional potential. However, the inherent complexity of supramolecular systems presents significant challenges for real-time visualization, particularly in capturing the growth kinetics of individual assemblies and the hierarchical formation of network structures at the single-assembly level. Here, we visualized the dynamic growth and network formation of benzyl-naphthalimide dye assemblies in situ by confocal laser scanning micro

A bio-friendly biotin-coupled and azide-functionalized naphthalimide for real-time endogenous hydrogen sulfide analysis in living cells.

Hydrogen sulfide (H2S) is involved in various biological processes. Thereby, abnormal levels of H2S are reported to be related to various human diseases including cancer. Currently, many fluorescent probes are pioneered to detect H2S by taking advantages of naphthalimides' unique internal charge transfer (ICT) property. However, most probes often require a high content of organic solvents or surfactants, and are limited to the analysis of exogenous H2S treated externally in live cell studies, and have difficulties in analyzing endogenous H2S, thus limiting their practical use. In this study, w

Naphthalimide-Based Template for Inhibitor Screening via Cross-Linking and In-Gel Fluorescence: A Case Study against HCA II.

We describe a rapid electrophoresis-based method for profiling of carbonic anhydrase inhibitors. In addition to the pharmacophore moiety intended for reversible interaction with a target enzyme, a fluorescent template with a built-in azide group for photoaffinity labeling is also included as a part of the inhibitor design. Following incubation and irradiation, gel electrophoresis with visualization under UV allows assessment of the efficiency of cross-linking. The relative efficiency of cross-linking of various probes can be regarded as a reflection of their inhibition potencies, an assumption

A reusable cyanide sensor via activation of C-H group: trifluoromethylcarbinol-directed meta-C-H cyanomethylation of naphthalimide.

A fluorescent and colorimetric anion sensor based on the activated C-H group has been developed, and its reusability and ability to selectively detect cyanides have been demonstrated. The system utilizes trifluoromethylcarbinol-directed C-H regioselective cyanomethylation for direct C-C bond formation between the fluorophore and acetonitrile. The present paper also describes a novel strategy for the enhancement of carbanion stability by a potential intramolecular C[double bond, length as m-dash]OH-C hydrogen bonding system, thus providing a new way to develop activated CH-based anion receptors

Antibacterial profiling, mechanistic characterization and in silico studies of pyrrolo[3,4-d]isoxazolidine-naphthalimide hybrids targeting Staphylococcus aureus.

In an effort to diverge from the molecular framework of conventional drug molecules, a series of pyrrolo[3,4-d]isoxazolidine-naphthalimide hybrids have been synthesized via a 1,3-dipolar cycloaddition reaction between azomethine N-oxides and N-substituted maleimide, with the aim of exploring moieties capable of lessening bacterial infections that continue to burden human health and the livestock industry. Several of the synthesized analogues demonstrate potent antibacterial activity at low concentrations. Notably, 9k and 9l, featuring indole and N,N-dimethylaniline moieties, respectively, outp

Fluorescent RET-Based Chemosensor Bearing 1,8-Naphthalimide and Styrylpyridine Chromophores for Ratiometric Detection of Hg2+ and Its Bio-Application.

Dyad compound NI-SP bearing 1,8-naphthalimide (NI) and styrylpyridine (SP) photoactive units, in which the N-phenylazadithia-15-crown-5 ether receptor is linked with the energy donor naphthalimide chromophore, has been evaluated as a ratiometric fluorescent chemosensor for mercury (II) ions in living cells. In an aqueous solution, NI-SP selectively responds to the presence of Hg2+ via the enhancement in the emission intensity of NI due to the inhibition of the photoinduced electron transfer from the receptor to the NI fragment. At the same time, the long wavelength fluorescence band of SP, ari

A Naphthalimide-Based Cd(2+) Fluorescent Probe with Carbamoylmethyl Groups Working as Chelators and PET-Promoters under Neutral Conditions.

We have developed a novel naphthalimide-based Cd(2+) fluorescent probe (1), featuring almost no background response, high sensitivity and selectivity toward Cd(2+) through its high association constant [K=(2.10±0.423)×10(6) ], and a practical working pH range. Membrane-permeability was conferred on 1 by replacing the imide and amide substituents with n-butyl groups, and hence the derivative (4) has found practical utility on fluorescent imaging of Cd(2+) in HeLa cells. Comparison of fluorescent properties between various compounds derived from 1 has demonstrated that the carbamoylmethyl groups

Effects of the 3- and 4-methoxy and acetamide substituents and solvent environment on the electronic properties of N-substituted 1,8-naphthalimide derivatives.

The photophysical properties of polar molecules in solution with an intramolecular charge-transfer effect in the excited state depend strongly on the polarity and proticity of the solvents. UV-visible spectra of 1,8-naphthalimide and some N-substituted derivatives in acetic acid, acetonitrile, dichloromethane, and p-dioxane were carried out. Several molecular cluster geometries formed with N-substituted 1,8-naphthalimide derivatives and a large set of random positioning of some solvent molecules in their environment were optimized by a semiempirical method. It provided a complete screening of

Design, synthesis, and biological evaluation of naphthalimide derivatives as dual monoamine oxidase-B and amyloid-β aggregation inhibitors for Alzheimer's disease.

Alzheimer's disease (AD) is a chronic and progressive neurodegenerative disorder with multifactorial pathogenesis. Amyloid beta (Aβ) and monoamine oxidase (MAO) are among the key therapeutic targets for its treatment. In this investigation, a series of 18 new naphthalimide-based derivatives were synthesized and assessed for multitarget activity against hAChE, esBuChE, MAO-A, MAO-B, Aβ, and paired helical fragment 6 (PHF6) aggregation. The screened compounds predominantly exhibited dual inhibition of MAO-B and Aβ aggregation to counteract oxidative stress and plaque formation. Among the tested

Water-soluble TREN-amino-1,8-naphthalimide fluorescent probes for selective, reversible Cu(II) detection and live-cell imaging.

Two water-soluble amino-1,8-naphthalimide-based fluorophores, TREN-Nap-1 and TREN-Nap-2, incorporating a tris(2-aminoethyl)amine (TREN) receptor unit, were synthesized through a straightforward single-step protocol and investigated as selective fluorescent chemosensors with high sensitivity for Cu(II) ions in aqueous media. Both probes exhibit strong intrinsic fluorescence that is significantly quenched upon coordination to Cu(II) and display negligible interference from competing metal ions. The sensing process is rapid (response time < 2 min) and highly sensitive, affording detection limits

Membrane-anchoring naphthalimide-based aggregation-induced emission photosensitizers for NIR fluorescence imaging and photodynamic elimination of multidrug-resistant bacteria.

Multidrug-resistant (MDR) bacterial infections demand antibacterial strategies that circumvent conventional resistance pathways and enable localized action with minimal host toxicity. Photodynamic therapy (PDT) represents a non-invasive approach; however, its effectiveness is constrained by insufficient bacterial targeting, aggregation-caused quenching, and limited reactive oxygen species (ROS) generation under physiological conditions. Herein, we develop two membrane-anchoring naphthalimide-based aggregation-induced emission (AIE) photosensitizers, TPAPV-NIM-mPy-M and TPAPV-NIM-Py-M, engineer

A naphthalimide based fluorescent liposomal system for sequential copper(II) and hydrogen sulfide detection and its application in hydrogel beads and bioimaging.

Hydrogen sulfide (H2S) is a key endogenous gaseous messenger, and its abnormal levels are closely linked to major physiological and pathological conditions. Developing selective and water-compatible H2S probes is therefore crucial. In this work, we report an amphiphilic naphthalimide-DPA-based fluorescent probe (Nap.Amp) capable of sequential Cu2+ and H2S detection via a fluorescence ON-OFF-ON mechanism. Nap.Amp exhibits strong green fluorescence that is quenched upon the formation of an in situ copper complex (Nap.Amp-Cu), which in turn displays excellent selectivity toward H2S, restoring the

From crystal packing to chemical sensing: substitution-guided design of naphthalimide schiff Bases for dual detection of carbonate and hydrogen sulfide.

A systematic substitution-based approach was used to probe how the molecular structure impacts crystal packing and anion-sensing in four Schiff bases derived from N-amino-1,8-naphthalimide. The derivatives, with 3-ethoxy (3E), 4-diethylamino (4D), 5-bromo (5B), and 2-hydroxynaphthyl (HN) substituents, were synthesized and characterized by NMR, FT-IR, HRMS, and single-crystal X-ray diffraction. Structural analysis discloses notable differences in molecular planarity and supramolecular organization based on substitutions, influenced by hydrogen bonding, π-π stacking, C-H···π, and halogen-bonding

Naphthalimide-Based Dyed Long Fluorescent Lifetime Probe for Lipid Droplet Viscosity Mapping in Non-Alcoholic Fatty Liver Disease.

Monitoring viscosity dynamics in lipid droplets is crucial for understanding the pathogenesis of non-alcoholic fatty liver disease (NAFLD), a condition affecting 25% of the global population. However, the application of fluorescence lifetime imaging microscopy (FLIM) for this purpose is hampered by the lack of probes with long fluorescence lifetimes and high signal-to-noise ratios. Herein, we developed two naphthalimide-based dyed fluorescent probes, NBC and NBA, which are viscosity-sensitive and feature donor-π-acceptor architectures. They operate via the restriction of intramolecular rotatio
