GABA regulates excitatory synapse formation in the neocortex via NMDA receptor activation.

The development of a balance between excitatory and inhibitory synapses is a critical process in the generation and maturation of functional circuits. Accumulating evidence suggests that neuronal activity plays an important role in achieving such a balance in the developing cortex, but the mechanism that regulates this process is unknown. During development, GABA, the primary inhibitory neurotransmitter in adults, excites neurons as a result of high expression of the Na(+)-K(+)-2Cl(-) cotransporter (NKCC1). Using NKCC1 RNA interference knockdown in vivo, we show that GABA-induced depolarizatio

Fossil evidence favors a role for vision in the modular evolution of the primate neocortex.

Neocortical expansion is a key innovation of crown primates, although fossil evidence for how the neocortex has evolved remains elusive. We studied neocortical evolution using virtual brain endocasts from extant and Eocene-Miocene fossil primates. We found that neocortex size increased in parallel across crown primate clades, with rapid increases in tarsiers and anthropoids. Although the frontal lobe is commonly described as having expanded disproportionately in primates, we show that it evolved gradually, following a common pattern for allometric scaling. By contrast, nonfrontal neocortical r

Primary cilia as architects of the neocortex: Roles in brain development, function, and microcephaly.

The etiology of primary hereditary microcephaly (MCPH), a condition closely linked to neocortex development, remains poorly understood. Initially, MCPH genes were thought to regulate a limited set of cellular processes, but recent studies reveal that many encode multifunctional proteins, often converging on primary cilia, organelles that orchestrate the development of most vertebrate tissues and organs. In this perspective article, we examine the role of primary cilia in brain development and explore how disruptions in MCPH and microcephaly-associated proteins compromise ciliary dynamics and f

Molecular and cellular dynamics of the developing human neocortex at single-cell resolution.

The development of the human neocortex is a highly dynamic process and involves complex cellular trajectories controlled by cell-type-specific gene regulation1. Here, we collected paired single-nucleus chromatin accessibility and transcriptome data from 38 human neocortical samples encompassing both the prefrontal cortex and primary visual cortex. These samples span five main developmental stages, ranging from the first trimester to adolescence. In parallel, we performed spatial transcriptomic analysis on a subset of the samples to illustrate spatial organization and intercellular communicatio

To Do or Not to Do: The cerebellum and neocortex contribute to predicting sequences of social intentions.

Humans read the minds of others to predict their actions and efficiently navigate social environments, a capacity called mentalizing. Accumulating evidence suggests that the cerebellum, especially Crus 1 and 2, and lobule IX are involved in identifying the sequence of others' actions. In the current study, we investigated the neural correlates that underly predicting others' intentions and how this plays out in the sequence of their actions. We developed a novel intention prediction task, which required participants to put protagonists' behaviors in the correct chronological order based on the

Periventricular nodular heterotopia is coupled with the neocortex during resting and task states.

Periventricular nodular heterotopia (PVNH) is a well-defined developmental disorder characterized by failed neuronal migration, which forms ectopic neuronal nodules along the ventricular walls. Previous studies mainly focus on clinical symptoms caused by the PVNH tissue, such as seizures. However, little is known about whether and how neurons in the PVNH tissue functionally communicate with neurons in the neocortex. To probe this, we applied magnetoencephalography (MEG) and stereo-electroencephalography (sEEG) recordings to patients with PVNH during resting and task states. By estimating frequ

Three-Dimensional Synaptic Organization of Layer III of the Human Temporal Neocortex.

In the present study, we have used focused ion beam/scanning electron microscopy (FIB/SEM) to perform a study of the synaptic organization of layer III of Brodmann's area 21 in human tissue samples obtained from autopsies and biopsies. We analyzed the synaptic density, 3D spatial distribution, and type (asymmetric/symmetric), as well as the size and shape of each synaptic junction of 4945 synapses that were fully reconstructed in 3D. Significant differences in the mean synaptic density between autopsy and biopsy samples were found (0.49 and 0.66 synapses/μm3, respectively). However, in both ty

Regional Specialization of Pyramidal Neuron Morphology and Physiology in the Tree Shrew Neocortex.

The mammalian cerebral cortex is divided into different areas according to their function and pattern of connections. Studies comparing primary visual (V1) and prefrontal cortex (PFC) of primates have demonstrated striking pyramidal neuron (PN) specialization not present in comparable areas of the mouse neocortex. To better understand PFC evolution and regional PN specialization, we studied the tree shrew, a species with a close phylogenetic relationship to primates. We defined the tree shrew PFC based on cytoarchitectonic borders, thalamic connectivity and characterized the morphology and ele

Apoptosis of glutamatergic neurons fails to trigger a neurogenic response in the adult neocortex.

Adult neurogenesis is actively studied in part because of the potential to manipulate endogenous neural stem and progenitor cells for tissue repair. Although constitutive generation of neurons in the adult rodent olfactory bulb and hippocampal dentate gyrus is widely accepted and stroke-induced generation of striatal inhibitory neurons consistently observed, evidence supporting the generation of neurons in the neocortex after neuronal loss remains slim. Nevertheless, a few studies suggested that targeted apoptosis of neocortical glutamatergic neurons could trigger the generation of new ones in

Cdk5rap2 interacts with pericentrin to maintain the neural progenitor pool in the developing neocortex.

Primary autosomal-recessive microcephaly (MCPH) and Majewski osteodysplastic primordial dwarfism type II (MOPDII) are both genetic diseases that result in decreased brain size at birth. MCPH is thought to arise from alterations in the size of the neural progenitor pool, but the cause of this defect has not been thoroughly explored. We find that one of the genes associated with MCPH, Cdk5rap2, is highly expressed in the neural progenitor pool and that its loss results in a depletion of apical progenitors and increased cell-cycle exit leading to premature neuronal differentiation. We link Cdk5ra

Opioids modulate cell division in the germinal zone of the late embryonic neocortex.

Opioid effects on cell division in the embryonic cerebral cortex were examined using two experimental approaches: (i) the presence of opioid receptors in the embryonic day 16 mouse neocortex was tested using immunohistochemical techniques; (ii) the values of the indices of [3H]thymidine pulse labelled cells and mitotic indices were estimated in the ventricular zone of the embryonic day 16 mouse neocortex 2.5, 4.5 and 8.5 h after administration to pregnant females of selected opioid receptor agonists or the opioid antagonist naloxone. Multiple olfactory activity in the human neocortex identified by magnetic source imaging.

To date, cortical regions activated by olfactory stimulation have not been identified precisely in humans. In this study we used magnetic source imaging to localize neuronal activity following olfactory stimulation with two odorants, hydrogen sulphide and vanillin. Peak latencies of the olfactory event-related magnetic fields corresponded to the ascending and descending slopes of the major deflections of the olfactory event-related potentials (OERP). At these latencies we obtained consistent activation of the anterior-central parts of the insula (agranular-periallocortical and dysgranular regi

Localization of superoxide dismutases in Alzheimer's disease and Down's syndrome neocortex and hippocampus.

Abnormalities in the cellular regulation and expression of antioxidant enzymes may have a role in mechanisms of central nervous system aging and neurodegeneration. We therefore examined, using isozyme-specific antibodies and immunohistochemistry, the localization of copper, zinc-superoxide dismutase and manganese-superoxide dismutase in the frontal and temporal neocortices and hippocampi of aged controls and individuals with Alzheimer's disease or Down's syndrome. Two different antibodies to copper, zinc-superoxide dismutase and one antibody to manganese-superoxide dismutase were evaluated by

Radial glia in the neocortex of adult rats: effects of neonatal brain injury.

Microgyria can be induced in otherwise normal rat neocortex by a freezing injury to the cortical plate before the completion of neuronal migration. We had previously reported radial glial like-immunoreactive fibers in the area of the microgyria in 32-day-old rats. Here we demonstrate that these glial fibers, which are immunoreactive to Rat-401, vimentin, and glial fibrillary acidic protein (GFAP) antibodies, are seen in adult rats. The appearance of these fibers is hypothesized to result from the release of a trophic factor during the recovery from neonatal injury which acts to either (1) halt

Topography of interhemispheric connections in neocortex of mice with congenital deficiencies of the callosal commissure.

Normally, axons within the corpus callosum are ordered according to the cortical regions from which they originate, and callosal cells and terminations form elaborate cortical patterns related to the underlying topographic representations of the sensory periphery. About 30% of mice of the BALB/c strain show congenital deficiencies of the callosal commissure which range from total absence of the corpus callosum to a moderate reduction in the size of this commissure. In the light of current theories about the origin of these callosal deficiencies, it seems likely that fibers crossing the midplan

[Interhemispheric asymmetry of the motor representation of the facial musculature in the neocortex of the white mouse].

In anesthetized white mice, the motor representation of the face muscles was determined with the aid of intracortical microstimulation in the frontal neocortex (fields 4, 6, 8). Significant difference was found in the motor responses to low-threshold stimulation in left and right hemispheres. Stimulation of the left hemisphere induced mostly movements of the upper lip and vibrissae, whereas stimulation of symmetrical area in the right hemisphere induced more variable movements. The area of the lip movement was much smaller in the right hemisphere.

[Organization of motor representation in the neocortex of white rats: results of macro- and microstimulation].

Motor and EMG responses were studied in thirty rats craniotomized under thiopental, in conditions of intracortical microstimulation and surface anode macrostimulation of the parietal and frontal neocortical areas. Strict contralaterality and distinct separation was found in motor representation (MRs) of the fore and hind limbs, of vibrissae fields and even of some joints and functional muscle groups. On the territory of the hind leg MR which was studied in greater details, centrally lies the compact MR zone of toes, mediocaudally from it - a vast MR of the foot, and further still more caudally
