Slow and fast synaptic inhibition evoked by pattern-generating neurons of the gastric mill network in spiny lobsters.

1. In this paper we begin an assessment of the role of synaptic properties, especially synaptic time course, in the function of the central pattern generator circuit (CPG) that controls rhythmic movements of the gastric mill in the foregut of spiny lobster (Panulirus interruptus). 2. The majority of neurons in the gastric CPG are motor neurons (MNs) that innervate striated muscles of the gastric mill but that also make electrical and inhibitory chemical interconnections within the neuropil of the stomatogastric ganglion. We studied the ionic dependence, pharmacology, and time course of inhibit

Presynaptic glutamate receptor--possible involvement of a K+ channel.

Intra-axonal recording was made from the excitatory axon of the lobster walking leg near the nerve terminal and the effects of L-glutamate were studied. Topical application of glutamate to the synapse produced hyperpolarization in the presynaptic membrane with increase in conductance. The glutamate-induced hyperpolarization was reversed to a depolarization at about -100 mV. The reversal potential was not significantly changed by altering the external Cl- but was shifted to more positive values by increasing the external K+. A spider toxin (JSTX), which specifically blocks the postsynaptic glut

Graded synaptic transmission between identified spiking neurons.

Graded synaptic transmission between spiking motoneurons of the pyloric group was studied in the stomatogastric ganglion of the spiny lobster, Panulirus interruptus. Intracellular microelectrodes were placed in the cell bodies of both pre- and postsynaptic neurons. Graded synaptic transmission was found between all tested cell pairs that were known to display spike-evoked synaptic transmission, including PD to LP, PD to PE, PD to PL, PL to LP, and LP to PD. Graded synaptic transmission was effective below the threshold for spikes. Thus, it was possible to study the influence of graded synaptic

The physiological properties of amine-containing neurones in the lobster nervous system.

1. Our previous studies have shown that octopamine and serotonin are found associated with a system of neurones in the connective tissue sheath of the second roots of lobster thoracic ganglia. To try to understand the mechanism of activation of these neurones, we undertook an examination of their general physiological properties. 2. All of the neurones receive excitatory synaptic input that has a cholinergic pharmacology, which suggests that it may be from sensory neurones. A very limited number of cells, possibly one, provdes the total synaptic input to all the cells in the roots of the secon

Porous scaffold hydroxyapatite from sand lobster shells (Panulirus homarus) using polyethylene oxide/chitosan as polymeric porogen for bone tissue engineering.

The hydroxyapatite (HAp; Ca10 (PO4 )6 (OH)2 )) has good biocompatibility, bioactivity, and osteoconductivity as a bone implant because the main inorganic mineral of human bone is HAp. The use of scaffold HAp from biogenic resources that contain high calcium and polymer as a pore forming agent to support bone growth is a longstanding area of interest. In this study, porous scaffolds based on HAp were synthesized from sand lobster (SL; Panulirus homarus) shells as a source of calcium using the porogen leaching method with polyethylene oxide (PEO) and chitosan (Chs) as polymeric porogen. The pres

Modulation of a neural network by physiological levels of oxygen in lobster stomatogastric ganglion.

Although a large body of literature has been devoted to the role of O2 in the CNS, how neural networks function during long-term exposures to low but physiological O2 partial pressure (PO2) has never been studied. We addressed this issue in crustaceans, where arterial blood PO2 is set in the 1-3 kPa range, a level that is similar to the most frequently measured tissue PO2 in the vertebrate CNS. We demonstrate that over its physiological range, O2 can reversibly modify the activity of the pyloric network in the lobster Homarus gammarus. This network is composed of 12 identified neurons that spo

A new method for labelling saxitoxin and its binding to non-myelinated fibres of the rabbit vagus, lobster walking leg, and garfish olfactory nerves.

1. A new method of labelling saxitoxin (STX) is described, based on transfer of tritium from tritiated water to the toxin. 2. The radiochemical purity of the labelled toxin has been directly determined, rather than being based on indirect biochemical means, as in previous experiments with Wilzbach-labelled STX and TTX. 3. The specific activity of the labelled toxin, 66 d..m.f-mole-1, corresponds with one tritium atom per molecule STX, an improvement of about 300-fold over other means of labelling TTX and STX. 4. The binding of this toxin to rabbit, lobster and garfish olfactory nerve fibres ha

A physiological analysis of walking in the American lobster (Homarus americanus).

The normal, unrestrained, forward walking of the lobster was studied with a closed-circuit television system and a video-tape recorder. A frame-by-frame analysis was undertaken and measurements made of unilateral stepping sequences, contralateral and ipsilateral phase relations between pairs of legs, the movements at the leg joints primarily involved in stepping and their differences in each of the four pereiopods. The order of stepping was expressed in terms of the probability of any leg following any other leg and it was found that while there is a preferred order, there is considerable vari

Role of Ih in differentiating the dynamics of the gastric and pyloric neurons in the stomatogastric ganglion of the lobster, Homarus americanus.

The hyperpolarization-activated inward cationic current (Ih) is known to regulate the rhythmicity, excitability, and synaptic transmission in heart cells and many types of neurons across a variety of species, including some pyloric and gastric mill neurons in the stomatogastric ganglion (STG) in Cancer borealis and Panulirus interruptus However, little is known about the role of Ih in regulating the gastric mill dynamics and its contribution to the dynamical bifurcation of the gastric mill and pyloric networks. We investigated the role of Ih in the rhythmic activity and cellular excitability o

Methyl mercury inhibits short-circuit current and Cl- influx across isolated epipodite of European lobster (Homarus gammarus).

The effect of methyl mercuric chloride (MeHg) on short-circuit current (I(SC)) was studied in the isolated perfused epipodite preparation from the branchial chamber of European lobster (Homarus gammarus) acclimated to dilute seawater. When applied at the apical surface, 0.2, 1.0 and 3.0 microM MeHg depressed I(SC) by a 26%, 81% and 98%, respectively. The half-maximal inhibitory concentration (IC(50)) of apically applied MeHg was 0.6 microM. Basolaterally added MeHg (3.0 microM) had no effect on I(SC), whereas addition of the specific Na(+),K(+)-ATPase inhibitor ouabain (1.5 mM) reduced I(SC) b

Crustacean hyperglycemic hormone in the lobster nervous system: localization and release from cells in the subesophageal ganglion and thoracic second roots.

Crustacean hyperglycemic hormones (CHHs) are neuropeptides involved in the regulation of hemolymph glucose. The primary source of CHHs has been identified as the neurosecretory neurons of the eyestalk X-organ and its associated neurohemal organ, the sinus gland. We have identified another source of CHH-like peptides in the nervous system. With the use of immunocytochemistry, cells in the second roots of the thoracic ganglia have been observed to stain positively for CHH-reactive material. We also identified a pair of cells in the subesophageal ganglion that contain large amounts of CHH-reactiv

Sensory-induced plasticity of motor pattern selection in the lobster stomatogastric nervous system.

In a previous study, a bilateral sensory input pathway to the crustacean stomatogastric nervous system was reported to induce the functional switching of an identified motor neuron (VD) from one rhythm generating neural network (the pyloric circuit) to another (the cardiac sac network). In the present in vitro study on the spiny lobster, Palinurus vulgaris, we have shown that under certain conditions, repetitive trains of phasic stimulation (1 s, 40 Hz) of one of these sensory nerves elicits either an increase or a decrease in efficacy of the VD switching response. In preparations showing no p

Nickel induces oscillatory behavior and enhanced synaptic and electrotonic transmission between stomatogastric neurons of Panulirus interruptus.

The pyloric pattern generator network of the stomatogastric ganglion uses a mixture of burst-inducing plateau potentials, synaptic transmission, and electrical coupling to produce its patterned output. This study examines the effects of two divalent, calcium channel blockers, nickel and cadmium, on voltage oscillations, synaptic transmission, and electrical coupling between the two pyloric dilator (PD) neurons and lateral pyloric (LP) neuron of Panulirus interruptus. The in vitro stomatogastric ganglion was bathed in saline containing tetrodotoxin (TTX) to eliminate Na-spikes and the spontaneo

Prawn Shell Chitosan Has Anti-Obesogenic Properties, Influencing Both Nutrient Digestibility and Microbial Populations in a Pig Model.

The potential of natural products to prevent obesity have been investigated, with evidence to suggest that chitosan has anti-obesity effects. Short-term synaptic plasticity compensates for variability in number of motor neurons at a neuromuscular junction.

We studied how similar postsynaptic responses are maintained in the face of interindividual variability in the number of presynaptic neurons. In the stomatogastric ganglion of the lobster, Homarus americanus, the pyloric (PY) neurons exist in variable numbers across animals. We show that each individual fiber of the stomach muscles innervated by PY neurons received synaptic input from all neurons present. We performed intracellular recordings of excitatory junction potentials (EJPs) in the muscle fibers to determine the consequences of differences in the number of motor neurons. Despite the va

The precursors of governance in the Maine lobster fishery.

Collective action is more likely to occur and to be effective when it is consistent with the self-interest of the affected individuals. The Maine lobster fishery is an instructive example of biological and technological circumstances combining with individual self-interest to create conditions favorable to collective action. The model describes the way social structure emerges from the adaptive behavior of competing fishers. Fishers compete in two ways: in a scramble to find the lobsters first and by directly interfering in other fishers' ability to compete, i.e., by cutting their traps. Both

Comparing neuronal spike trains with inhomogeneous Poisson distribution: evaluation procedure and experimental application in cases of cyclic activity.

A procedure is proposed to compare single-unit spiking activity elicited in repetitive cycles with an inhomogeneous Poisson process (IPP). Each spike sequence in a cycle is discretized and represented as a point process on a circle. The interspike interval probability density predicted for an IPP is computed on the basis of the experimental firing probability density; differences from the experimental interval distribution are assessed. This procedure was applied to spike trains which were repetitively induced by opening-closing movements of the distal article of a lobster leg. As expected, th

Interacting biological and electronic neurons generate realistic oscillatory rhythms.

Small assemblies of neurons such as central pattern generators (CPG) are known to express regular oscillatory firing patterns comprising bursts of action potentials. In contrast, individual CPG neurons isolated from the remainder of the network can generate irregular firing patterns. In our study of cooperative behavior in CPGs we developed an analog electronic neuron (EN) that reproduces firing patterns observed in lobster pyloric CPG neurons. Using a tuneable artificial synapse we connected the EN bidirectionally to neurons of this CPG. We found that the periodic bursting oscillation of this

Dopamine modulates graded and spike-evoked synaptic inhibition independently at single synapses in pyloric network of lobster.

Bath application of dopamine (DA) modifies the rhythmic motor pattern generated by the pyloric network in the stomatogastric ganglion of the spiny lobster, Panulirus interruptus. Synaptic transmission between network members is an important target of DA action. All pyloric neurons employ both graded transmitter release and action-potential-mediated synaptic inhibition. DA was previously shown to alter the graded synaptic strength of every pyloric synapse. In this study, we compared DA's effects on action-potential-mediated and graded synaptic inhibition at output synapses of the lateral pylori
