Alz-50 immunoreactivity in the hypothalamus of the normal and Alzheimer human and the rat.

Alz-50 is a monoclonal antibody recognizing a 68 kilodalton protein that is abundant in Alzheimer's disease (AD) but not detectable by immunoblotting methods in normal brains. When used for immunohistochemistry in AD cortex, Alz-50 recognizes large numbers of neurofibrillary tangles (NFT), neuritic plaques, and some neurons that show no evidence of neurofibrillary degeneration by conventional histopathological staining methods. Alz-50 immunoreactivity is described at the light and electron microscopic levels in the hypothalamus of brains obtained at autopsy from normal and AD subjects. Alz-50

The natural history of dementia in Down's syndrome.

In a prospective longitudinal study with death as the end point in 17 middle-aged patients with Down's syndrome, dementia was clinically diagnosed in 15 patients, by means of careful observations in daily circumstances. Autopsies were performed in 10 cases: 8 demented patients and 2 nondemented patients. Neuropathologically, Alzheimer-type abnormalities were demonstrated in 9 patients, both demented and nondemented, and combined Alzheimer-type abnormalities with infarctions were demonstrated in 1 patient. In the 14 demented patients who did not show evidence of cerebrovascular or systemic vasc

Progressive dementia associated with Sheehan's syndrome--an autopsied case.

This is a report of the study of progressive dementia associated with Sheehan's syndrome in a 61-year-old woman who had a 13-year history of insidious progressive decline in global cognitive abilities. At the onset, there were only slight difficulties with abstract thoughts, and a shortened attention span, but the symptoms gradually increased in severity until death at the age of 61. The clinical deficits fluctuated in severity. During the last years of life she was at all times confused, apathetic, and incapable of self-care. A necropsy revealed a moderately severe neuronal loss in the centra

Animal models of brain ageing and dementia.

Six animal models of human brain ageing or of age-related diseases, especially Alzheimer's disease, are evaluated. Special emphasis is laid on comparing the neuropathological symptoms found in aged human brains or in Alzheimer patients, with the neuronal symptoms induced experimentally by the different treatments described here. For each model, the experimental methods for induction of the neuropathological symptoms (lipofuscin accumulation, senile plaques, neurofibrillary degeneration, etc.) and for their identification are discussed. In addition, the behavioral significance of each animal mo

Meningio-angiomatosis: a report of six cases with special reference to the occurrence of neurofibrillary tangles.

We report six cases of meningio-angiomatosis, a disorder of the cerebral cortex of probable malformative origin frequently associated with neurofibromatosis and either asymptomatic or associated with a seizure disorder. The patients, three males and three females, ranged from 10 to 70 years of age at diagnosis. The lesion was an incidental autopsy finding in two patients; four subjects had a seizure disorder which improved following surgical resection of the cortical lesion. In five instances the process was unifocal; one patient with neurofibromatosis had multifocal involvement. Grossly, the

Neuronal pathology in the nucleus basalis and associated cell groups in senile dementia of the Alzheimer's type: possible role in cell loss.

The loss of cortical cholinergic innervation in senile dementia of the Alzheimer's type (SDAT) is associated with cell loss in the nucleus basalis and related cell groups (magnocellular basal nucleus, MBN). We examined MBN in Nissl-, acetylcholinesterase- and thioflavin S-stained sections in two cases of SDAT and in four control brains. Using these sensitive methods, senile plaques were easily demonstrated in MBN, and most MBN neurons showed neurofibrillary degeneration as an early change. Cell loss appeared to be due to maturation of neurofibrillary tangles, displacing normal cellular content

Neurofibrillary tangles, granulovacuolar degeneration, and neuron loss in Down Syndrome: quantitative comparison with Alzheimer dementia.

The degree of neurofibrillary tangle formation, granulovascuolar degeneration, and nerve cell loss was quantified in serial sections of the hippocampal formation from the brains of 5 adults dying with Down syndrome (monogolism). These morphometric results were compared with values obtained from a series of mentally normal elderly patients reported previously. Granulovacuolar change appeared to be related as much to patient age as to the existence of monogolism. By contrast, the number of Alzheimer neurofibrillary tangles and loss of pyramidal neurons from the hippocampus exceeded levels found

Oxidative stress, mutant SOD1, and neurofilament pathology in transgenic mouse models of human motor neuron disease.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease that primarily affects motor neurons in the spinal cord and brain stem. About 10% of all ALS cases are familial (FALS), inherited in an autosomal dominant manner. One fifth of FALS patients carry mutations in the Cu/Zn superoxide dismutase (SOD1) gene, and several lines of transgenic mice have been engineered to express mutant forms of the SOD1 gene that are linked to FALS. Significantly, many of these transgenic lines of mice develop a motor neuron disease (MND) that resembles human FALS. Oxidative stress induced by human SOD1

Early-onset Alzheimer's disease in 2 large Belgian families.

Familial Alzheimer's disease (FAD) is a dominantly inherited condition that may present with an early onset, and myoclonus occurs frequently in the course of the disease. We report clinical and neuropathologic data on 2 large Belgian families with FAD in which we obtained 17 autopsies of the CNS. In family A, each of 11 autopsies had the typical neuropathologic features of Alzheimer's disease (AD), and there were a few cerebellar plaques in the molecular layer. In family B, in addition to the typical characteristics of AD in 6 autopsies, there were numerous amyloid plaques in the cortical cere

[Neuropathologic principles of senile dementia].

Several groups of senile dementias with differing causes and neurohistological substrates can be distinguished. Genetically determined Pick's disease produces severe neuronal atrophy; Alzheimer's disease is characterized by pathological neurofibrillary changes. Jakob-Creutzfeldt disease and Gerstmann-Sträussler-Scheinker disease are slow virus diseases induced by unconventional agents. These diseases, after an incubation time of several years, take the course of slowly progressive degenerative diseases of the central nervous system without inflammatory and specific immune reactions of the host

Topography of neurofibrillary tangles and granulovacuoles in hippocampi of patients with Down's syndrome: quantitative comparison with normal ageing and Alzheimer's disease.

An XY pen recorder linked potentiometrically to a sampling stage microscope permitted the plotting of topographic "scattergrams" to determine the precise cytoarchitectonic localization of neurofibrillary tangles and granulovacuolar degeneration in hippocampal neurones of Down's syndrome adult brains. The area (and hence volume) of six cortical "zones" surveyed was measured with a digitizer and programmable calculator. In decreasing magnitude of affliction, the rank order for neurofibrillary tangles was: entorhinal greater than subiculum greater than H1 greater than endplate greater than H2 gre

[Neuropsychology and senile dementia].

The study of senile plaque dementias and neurofibrillary degenerations, represented by Alzheimer's disease, presbyophrenia, simple senile dementia, and senile dementia of the Alzheimer type, enables, by means of its specificity, a privileged method of approach to be taken in the comprehension of cerebral mechanisms, with a resulting different conception of neuropsychology. That which is described in this paper has evolved from the application of Genetic Psychology methods developed by J. Piaget. It demonstrates the value of a synchronic qualitative analysis of dementia, even for determination

Clinicopathologic studies of primary cerebral amyloid angiopathy.

Primary cerebrovascular amyloidosis resulting in significant cerebral parenchymal damage was encountered in 23 autopsied cases at the Mayo Clinic over the past 10 years. Patients were 60 to 97 years old and both sexes were equally represented. Large- and medium-sized leptomeningeal and cortical arteries showed the characteristic pattern of medial and intimal involvement, with luminal stenosis. The walls of smaller arteries were often diffusely infiltrated, with fibrinoid degeneration and miliary aneurysm formation. The amyloid nature of the infiltrate was confirmed by electron microscopic exam

Brain aging and Alzheimer's disease.

The most common cause of senile dementia appears to be a pathological process indistinguishable from that found in presenile dementia of the Alzheimer type. Consideration of the neuropathological changes suggest that this disease may involve in interaction of at least three processes: a viral-like infection, a disorder in the immune system and the neurotoxic effect of an environmental agent. The evidence in support of this hypothesis is reviewed.

Aluminium, neurofibrillary degeneration and Alzheimer's disease.

Aluminium is neurotoxic and results in the proliferation of 100 A diameter filaments in the cytoplasm of certain neurons. The aluminium concentration for 7 normal human brains was 1-9 +/- 0-7 SD mug/g dry weight (n = 208 samples). The aluminium content of 585 areas sampled in 10 post-mortem cases of Alzheimer's disease ranging in age from 50 to 88 years, in which the diagnosis was based on the specific histological appearances, revealed an elevated aluminium content in some regions. A range of 0-4 - 107-0 mug/g was encountered and 28 per cent of all regions sampled had concentrations in excess

Rational Design and Optimisation of CRISPR-Cas9 Delivery Systems for Targeted Genomic Transformation.

The CRISPR-Cas9 genome-editing technique offers a promising therapeutic strategy for genetic disorders, including neurodegenerative diseases like Alzheimer's disease (AD), characterised by inherited susceptibility and progressive cognitive decline, as well as other hallmarks such as amyloid beta (Aβ1-42) plaques and neurofibril tangles (NFTs). However, the blood-brain barrier (BBB) poses a significant challenge to the effectiveness of gene editing components in the affected brain region and impedes clinical translation. This comprehensive review compares various CRISPR-Cas9 delivery vectors, v

Topographical anatomy of the subthalamic region with special interest in the human medial forebrain bundle.

OBJECTIVE: The medial forebrain bundle (MFB) is a novel promising deep brain stimulation (DBS) target in severe affective disorders that courses through the subthalamic region according to tractography studies. Its potential therapeutic role arose in connection with the development of hypomania during stimulation of the subthalamic nucleus (STN) in Parkinson's disease, offering an alternative explanation for the occurrence of this side effect. However, until now its course exclusively described by tractography had not yet been confirmed by any anatomical method. The aim of this study was to fi
