Strong expression of interleukin-1 receptor type I in the rat carotid body.

One of the unsolved key questions in neuroimmunomodulation is how peripheral immune signals are transmitted to the brain. It has been reported that the vagus might play a role in this regard. The underlying mechanism for this immune system-to-brain communication route is related to the binding of cytokines, such as interleukin (IL)-1beta originating from activated immune cells, to their receptors in glomus cells of the vagal paraganglia. The existence of IL-1 receptor type I (IL-1RI) in vagal paraganglia has been proved. On the basis of these studies, a hypothesis is raised that the carotid bo

Neuroimmune interactions in experimental colitis. An immunoelectron microscopic study.

With its abundance of neurons and immunocytes, the gut is a potentially important site for the study of the interaction between the nervous and immune systems. In this electron microscopic study we have investigated the distribution of substance P (SP)- and vasoactive intestinal polypeptide (VIP)-immunoreactive (IR) nerve terminals and the immunocytes during experimental colitis in the rat. A mild colitis was induced by a luminal enema containing trinitrobenzene sulfonic acid. The most severe inflammation was detected after 2 days and the density and the distribution of the SP- and VIP-IR nerv

Zika Virus as a Possible Risk Factor for Autism Spectrum Disorder: Neuroimmunological Aspects.

The recent outbreak of the Zika virus (ZIKV) and the discovery that perinatal Zika exposure can lead to the Congenital Zika Syndrome has promoted a call for prevention measures. Due to the increased number of babies born with microcephaly, structural brain abnormalities, and neurological alterations in regions affected by ZIKV, investigations were carried out in order to better understand this process. The maternal immune system directly influences the fetal central nervous system, and complications during pregnancy have been associated with neurodevelopmental disorders. Autism spectrum disord

The role of melatonin in the cells of the innate immunity: a review.

Melatonin is the major secretory product synthesized and secreted by the pineal gland and shows both a wide distribution within phylogenetically distant organisms from bacteria to humans and a great functional versatility. In recent years, a considerable amount of experimental evidence has accumulated showing a relationship between the nervous, endocrine, and immune systems. The molecular basis of the communication between these systems is the use of a common chemical language. In this framework, currently melatonin is considered one of the members of the neuroendocrine-immunological network.

A Review of the Prospective Effects of Methadone on Peripheral Neuropathic Pain in Diabetic Patients.

Peripheral neuropathic pain (PNP) is a significant complication for diabetic patients, often linked to poor glycemic control and elevated levels of glycosylated hemoglobin (HbA1c). High serum levels of cytokines, such as interleukin (IL)-6, and an increase in T-lymphocytes are crucial factors in developing neuropathic complications. Research suggests that substances like opiates and methadone can provide pain relief for these patients. This literature review is aimed at exploring the advantages and disadvantages of prescribing methadone to individuals with diabetes. We conducted a search of se

Vagal-immune interactions involved in cholinergic anti-inflammatory pathway.

Inflammation and other immune responses are involved in the variety of diseases and disorders. The acute response to endotoxemia includes activation of innate immune mechanisms as well as changes in autonomic nervous activity. The autonomic nervous system and the inflammatory response are intimately linked and sympathetic and vagal nerves are thought to have anti-inflammation functions. The basic functional circuit between vagus nerve and inflammatory response was identified and the neuroimmunomodulation loop was called cholinergic anti-inflammatory pathway. Unique function of vagus nerve in t

Amphotericin B Increases Transglutaminase 2 Expression Associated with Upregulation of Endocytotic Activity in Mouse Microglial Cell Line BV-2.

Amphotericin B (AmB), a polyene antibiotic, is reported to cause the microglial activation to induce nitric oxide (NO) production and proinflammatory cytokines expression, and change neurotrophic factors expression in cultured microglia (Motoyoshi et al. in Neurochem Int 52:1290-1296, 2008). On the other hand, tissue-type transglutaminase (TG2) is involved in connection to phagocytes with apoptotic cells. Engulfment of neurons by activated microglia is thought to cause neurodegenerative diseases but detail is unclear, and involvement of TG2 in phagocytosis has been reported in our previous stu

Neuroimmune interactions in fascia and myofiber regeneration: a narrative review.

When not treated adequately, neuromusculoskeletal, tendinous, and joint tissue injuries may become chronic, leading to impaired tissue function due to fibrosis, extracellular matrix densification, and fatty connective tissue accumulation, ultimately resulting in reduced joint and muscle mobility. Timely treatment involving the mobilization of fascia and targeted muscle exercise has been shown to enhance and promote tissue regeneration. Key phases in tissue regeneration after injury include the activation of the innate immune system, followed by its resolution. Although several treatment modali

Chronic pain in Alzheimer disease and the role of neuroimmune interactions.

Chronic pain conditions affect a large proportion of the global population and pain has tremendous personal and economic impact, affecting quality of life and ability to work while imposing a significant burden on families and health care systems. Neurological disorders and painful conditions are frequently accompanied by chronic pain, which is often jointly diagnosed with age-related neurodegenerative diseases, however, many patients do not receive adequate treatment. This review aims to create a working framework around the issue of under-recognized/undertreated chronic pain in Alzheimer dis

Targeted Intracranial Delivery of Secreted Immune Molecules to Investigate Neuroimmune Interactions.

Although secreted immune proteins have increasingly been recognized to influence nervous system function, technical barriers persist in defining their spatiotemporal dynamics within the brain's complex environment. For example, microglia, a population of key immune cells in the brain, release a variety of factors that interact with many diverse cellular targets. Microglia-derived proteins can signal to other cell types via mechanisms such as surface receptor binding without internalization, passive membrane transport, phagocytic uptake, or endocytosis-mediated uptake. Here, a method for locali

Neural state as a determinant of tumor immunity in lung cancer.

Immune checkpoint blockade has reshaped lung cancer therapy, yet durable benefit remains confined to a minority of patients, and resistance often emerges even in initially responsive disease. An increasingly coherent explanation is that neural state-encoded through hierarchical neuroimmune interactions-can profoundly recalibrate anti-tumor immunity and influence treatment responsiveness. In this Review, we synthesize evidence across three integrated levels of neuro-immune regulation in lung cancer. First, lung tumors engage in local neural-immune-tumor interactions, including synapse-like coup

The choroid plexus: both a gatekeeper and a conductor of neuroimmune communication.

The choroid plexus (ChP) is increasingly recognized as a dynamic neuroimmune interface that integrates peripheral and central signals to regulate cerebrospinal fluid (CSF) homeostasis, leukocyte trafficking, and inflammatory tone within the central nervous system (CNS). Recent studies reveal that beyond their classic roles in CSF production and barrier function, ChP epithelial cells are conductors of neuroimmune communication, actively remodeling the state of immune activity, immunosurveillance and responsiveness of immune cells in the CSF in response to infection, injury, malignancy, aging, a

Meningeal neuropeptide and neuroimmune interactions in the context of migraine.

Migraine is a highly prevalent, painful neurological disorder with increasing evidence to support the contribution of neuroimmune interactions to its pathophysiology. The cranial meninges are a key site of these interactions, and are characterized by rich innervation, vascular and lymphatic supply, connective tissue, and diverse immune cell populations. Migraine-related physiological events, such as cortical spreading depression and other external triggers, induce the release of neuropeptides, including calcitonin gene-related peptide (CGRP), substance P (SP), pituitary adenylate cyclase-activ

The cholinergic anti-inflammatory pathway in humans: State-of-the-art review and future directions.

The parasympathetic nervous system modulates inflammation through efferent vagus nerve signaling. Tracey (2002) termed this process as the cholinergic anti-inflammatory pathway (CAP). Interest in the potential practical use of this immune-modulatory process is increasing alongside increasing appreciation for the role of systemic inflammation in the etiology of somatic and psychological disease. A diverse literature exists providing expansive correlational evidence and some preliminary experimental evidence of the CAP in humans. However, so far this literature has not been well integrated and c

[The inflammatory reflex: the role of the vagus nerve in regulation of immune functions].

Experimental studies published in past years have shown an important role of the vagus nerve in regulating immune functions. Afferent pathways of this cranial nerve transmit signals related to tissue damage and immune reactions to the brain stem. After central processing of these signals, activated efferent vagal pathways modulate inflammatory reactions through inhibiting the synthesis and secretion of pro-inflammatory cytokines by immune cells. Therefore, pathways localized in the vagus nerve constitute the afferent and efferent arms of the so-called "inflammatory reflex" that participates in

Targeting the neuro-immune axis: immunological mechanisms and therapeutic strategies in male sexual dysfunction.

Male sexual dysfunction (MSD) is a multifactorial condition in which traditional models focusing on vascular, endocrine, and psychogenic factors are insufficient. Emerging evidence highlights dysregulated crosstalk between the nervous and immune systems-the neuroimmune axis-as a potential unifying pathological substrate. This review synthesizes fragmented yet compelling evidence linking neural injury, chronic inflammation (both systemic and neurogenic), and immune dysregulation to the pathogenesis of MSD. We propose a novel integrative framework in which disruption of the neuro-immune axis ser

Neuro-immune-vascular interactions in rosacea and hypertension: a mechanistic review.

Rosacea is a chronic inflammatory skin disorder characterized by recurrent flare-ups and significant variability in therapeutic responses. Its pathophysiology is complex and involves multilevel processes, including neurogenic dysregulation, immune activation, and alterations in vascular reactivity. Recent epidemiological studies suggest that rosacea may be associated with an increased risk of hypertension and other cardiometabolic diseases. However, it remains unclear whether this association merely reflects a comorbidity or indicates the presence of shared underlying pathophysiological mechan
