Divergent tau pathology profiles induced by distinct alpha-synuclein aggregates in non-human primates.

Proteinopathies are frequently accompanied by co-pathologies, which complicate understanding the pathophysiological mechanisms of neurodegenerative diseases. In this study, we employed a well-characterised baboon cohort to investigate the occurrence of tau pathology following intrastriatal injections of two distinct α-synuclein extracts (small aggregates: SA and large aggregates: LA) derived from the brains of Parkinson's disease (PD) patients. Biochemical and histopathological analyses (6, 12, or 24 months after injections) revealed significantly higher phospho-tau expression and greater freq

Effects of in vivo and in vitro administration of neuro-Behcet's disease IgG.

Antibodies directed against membrane antigens of neuronal axonal processes (neuropil) have been recently identified in neuro-Behcet's disease (NBD) patients. To delineate the potential pathogenic action of these antibodies, pooled sera from seven NBD patients with neuropil antibodies and seven healthy controls were divided into purified IgG and IgG-depleted serum (IgG-DS) fractions and each fraction was administered into lateral ventricles of rats. NBD IgG-injected rats showed reduced locomotor activity in the open field test as compared to NBD IgG-DS, healthy control IgG, healthy control IgG-

Immunohistochemical quantification of the synapse-related protein synaptophysin in Alzheimer disease.

Alzheimer disease (AD) as well as other dementing disorders are characterized by a continuous loss of neurons in cortical and subcortical areas and probably by an extensive synaptic loss. In order to substantiate and localize the loss of synapses in AD, we quantified by microdensitometry the neuropil immunoreactivity to an antibody that labels the protein synaptophysin (p38), which is localized in the presynaptic terminals. We found in the AD cases an average 50% decrease in the density of the granular neuropil immunoreaction in parietal, temporal and midfrontal cortex. In contrast, Pick disea

A comparative study of MAP2 immunostaining in areas 9 and 17 in schizophrenia and Huntington chorea.

Increasing evidence suggests that there may be significant morphological changes in the neuropil of the dorsolateral prefrontal cortex in schizophrenia. A controversial issue surrounding these deficits in the cortical neuropil is the confounding effects of antipsychotic (neuroleptic) medication as well as the question of generality to psychiatric disorders. To begin to address these issues we examined brains from Huntington's patients matched to a cohort of schizophrenics and controls. Many Huntington's patients take neuroleptics similar to schizophrenics; therefore, by comparing the two group

Central projections of photoreceptor axons originating from ectopic eyes in Drosophila.

Ectopic expression of the retinal determination gene eyeless (ey) induces the formation of supernumerary eyes on antennae, legs, wings, and halteres. These ectopic eyes form ommatidia that contain photoreceptors and accessory cells and respond to light. Here, we demonstrate that ectopic eyes on antennae and legs extend axonal projections to the central nervous system. Furthermore, electroretinograms and morphological evidence indicate that the photoreceptor axons of at least the antennal ectopic eyes can form completely constituted ectopic synapses with foreign postsynaptic elements and sugges

Architecture of the basal region of the subcommissural organ in the brush-tailed possum (Trichosurus vulpecula).

The architecture of the basal region of the subcommissural organ (SCO) and the subjacent neuropil was studied in the brush-tailed possum, Trichosurus vulpecula (Marsupialia). Several structural features suggest that the basal mode of SCO-secretion may be as prominent as the well-established apical secretion. Some of the features that speak in favour of basal secretion are: (1) the existence of deep processes of secretion-laden SCO cells which reach and surround the capillaries in the hypendyma and the subjacent neuropil; (2) the presence of perivascular spaces, some of which may contain a mate

An immunohistochemical study of serotoninergic nerves in the colon and rectum of children with Hirschsprung's disease.

Serotonin (5-HT)-like immunoreactive nerve fibers were investigated in gut tissue obtained from seven Japanese children with Hirschsprung's disease. In the control untreated tissues, 5-HT-like immunoreactive fibers were observed neither in the normoganglionic nor in the aganglionic regions. After pargyline treatment, 5-HT-positive neuropils were consistently detected in association with the myenteric plexus in the normoganglionic segment, while in the aganglionic segment immunoreactive fibers could not be demonstrated through the entire layer of the bowel tissue. The occurrence of 5-HT-like im

On the Larval Sensory Organs and Central Nervous System of the Zoea 1 of the Asian Shore Crab Hemigrapsus sanguineus (Decapoda, Brachyura).

Many decapod crustaceans occupy different habitats during their ontogeny and undergo a niche shift during the transition from the larval to the juvenile phase. Their life cycle comprises pelagic larvae and, in most species, benthic juvenile-adult stages. Already at hatching, decapod larvae possess a wealth of organ systems, necessary to autonomously survive and develop in the plankton. They also exhibit a rich behavioral repertoire to appropriately respond to environmental stimuli such as light, gravity, hydrostatic pressure, tidal currents, temperature, salinity, chemical cues, and food conce

Morphologically distinct plaque types differentially affect dendritic structure and organisation in the early and late stages of Alzheimer's disease.

We have investigated the effects of the deposition of insoluble beta-amyloid plaques on dendritic morphology within the neocortex. Labelling for beta-amyloid identified three morphologically distinct plaque types present both within the brains of preclinical Alzheimer's disease (AD) and end-stage AD cases. In both preclinical and end-stage AD, the percentage area occupied by diffuse plaques contained a greater density of labelling for microtubule-associated protein-2 (MAP2) relative to the surrounding neuropil (case type, ratio of MAP2 labelling in plaque to MAP2 labelling in surrounding neuro

Pigmentary macular degeneration with multifocal necrotizing encephalopathy.

A previously healthy 10-year-old girl suffered sudden, binocular visual deterioration. During the next few years her neurologic and visual condition progressively worsened and she developed hypertension, seizures, ataxia, and lactic acidemia, leading to death at the age of 16 years. Bilateral optic disk pallor was followed by the loss of the foveal reflex and pigmentary maculopathy, manifested as disorganization of the retinal layers, loss of ganglion cells, degeneration of the photoreceptors and nuclei, and irregular infiltration of the retina by pigment epithelial cells. The optic nerves and

Impairment of the GABAergic system in the anterior midcingulate cortex of heroin-addicted males.

Opioid addiction is a global concern and the largest health burden among drug use disorders. The multifunctional anterior midcingulate cortex (aMCC) is critical for processing nociceptive input and negative emotions, which play a prominent role in the maintenance of opioid addiction. GABAergic interneurons regulate the output of the aMCC, whose dysfunction has been linked to the behavioural abnormalities observed in addiction. In these neurons, glutamate decarboxylase (GAD), with its isoforms GAD 65 and 67, is a key enzyme in the synthesis of GABA. However, there is a lack of research investig

Widespread spinal cord involvement in progressive supranuclear palsy.

We describe the histopathologic features of spinal cord lesions in 10 cases of progressive supranuclear palsy (PSP) and review the literature. Histologic examination revealed atrophy with myelin pallor in the anterior funiculus and anterolateral funiculus in the cervical and thoracic segments in eight of the 10 cases, whereas the posterior funiculus was well preserved. The degrees of atrophy of the anterior funiculus and the anterolateral funiculus correlated with that of the tegmentum of the medulla oblongata. Myelin pallor of the lateral corticospinal tract was observed in two of the 10 case

A GFP-based genetic screen reveals mutations that disrupt the architecture of the zebrafish retinotectal projection.

The retinotectal projection is a premier model system for the investigation of molecular mechanisms that underlie axon pathfinding and map formation. Other important features, such as the laminar targeting of retinal axons, the control of axon fasciculation and the intrinsic organization of the tectal neuropil, have been less accessible to investigation. In order to visualize these processes in vivo, we generated a transgenic zebrafish line expressing membrane-targeted GFP under control of the brn3c promoter/enhancer. The GFP reporter labels a distinct subset of retinal ganglion cells (RGCs),

Optical recording of odor-evoked responses in the olfactory brain of the naïve and aversively trained terrestrial snails.

Regular spontaneous oscillations were recorded both electro- and optophysiologically using a voltage-sensitive absorption dye in the olfactory part of the brain (procerebral lobe of the cerebral ganglia) of the gastropod mollusk Helix lucorum. Odor application caused transient changes in procerebral oscillations, and an odor-evoked potential was recorded in the procerebrum (PC). The wave of evoked potential originated near the place of olfactory nerve entrance into the PC and propagated via the procerebral neuropile toward the cell body layer. The spread of the odor-evoked potential correspond

In vivo toxicity of prion protein in murine scrapie: ultrastructural and immunogold studies.

Prion protein (PrP) is a cell surface, host coded, sialoglycoprotein which accumulates in excess in scrapie, Creutzfeldt-Jakob disease, bovine spongiform encephalopathy and other transmissible spongiform encephalopathies. Infection of mice with the 87 V or ME7 scrapie strains results in distinctive and very different light microscopical patterns of vacuolation and disease specific PrP accumulation. In both of these scrapie strains immunogold electron microscopy was used to locate PrP to the plasmalemma of neurons from where it was released into the neuropil. Initial PrP accumulation around neu

[Optical recording of the odor-evoked responses in olfactory structures of the brain of the terrestrial mollusk Helix].

Regular oscillations were recorded in olfactory part of the brain (procerebrum) of gastropod mollusk Helix both electrographically and optically. In general, oscillations resembled those in slugs reported earlier. Odor application caused a transient change in the procerebral oscillations followed by appearance of a special pattern. For the first time the evoked potential was recorded in procerebrum and mapped in reference to the area of oscillations. The area of spreading of evoked potential roughly corresponded to the neuropil projection, while the oscillations were recorded in the projection

[Distribution of GABA-immunoreactive elements in the reptile amygdaloid complex].

GABA-immunoreactive (GABA-I) elements (neuronal somata and neuropile) are detected in turtle Emys orbicularis and lizard Ophysaurus spodus in all structures of ventral and dorsal parts of amygdaloid complex (AC) considered as phylogenetic more ancient and younger, respectively by means of the immunohistochemical method. Their maximal quantity in the ventral section of AC is found in the lateral region, lesser--in the ventral, central and medial regions. Besides in lizards a specialized laminar distribution of GABA-I elements in n. sphaericus is observed. GABA-I neurons are also detected in str

Degeneration of Pick bodies visualized by methenamine-silver staining and immunohistochemistry.

The degeneration process of Pick's bodies (PB) was investigated using methenamine-silver (MS) staining and immunohistochemistry. Methenamine-silver staining sensitively detected extracellular PB as well as intracellular PB in the granular cell layer of the dentate gyrus. Extracellular PB appeared as granular masses with ill-defined boundaries in the neuropil. For MS electron microscopy, the extracellular PB were composed of randomly oriented fibrillary components measuring 9-12 nm in diameter with astroglial processes and degenerated organelles. Tau immunoreactivity of extracellular PB was red

Immunohistochemical study of advanced glycation end products in aging and Alzheimer's disease brain.

Advanced glycation end products (AGEs) in the brain were immunohistochemically examined in Alzheimer's disease (AD) and aging using anti-AGE antibody recognizing mainly carboxymethyllysine. AGE positive staining diffusely located in the neuronal perikarya of hippocampus and parahippocampus in AD and aged brains without dementia, but not in young brains less than 17 years of age. Extra-neuroperikaryal AGE deposits were also detected in the neuropil of AD and aged brains. The extra-neuroperikaryal AGE deposits markedly increased in AD brains as compared to aged brains. These AGE-positive deposit

Argyrophilic grains in late-onset Creutzfeldt-Jakob diseased brain.

Braak's argyrophilic grains (ArG) are spindle-shaped structures originally described in patients with dementia. Herein, a unique case of sporadic Creutzfeldt-Jakob disease (CJD) accompanied by numerous ArG is presented. The pathological picture was typical of CJD based on the findings of routine hematoxylin-eosin staining. The highest density of ArG was observed throughout the parahippocampal gyrus and the temporal gyri; however, the sector CA1 of the hippocampus showed less ArG. An immunohistochemical analysis for prion protein (PrP) revealed diffuse fine neuropil staining in the cerebral cor
