Single-dose live-attenuated vesicular stomatitis virus-based vaccine protects African green monkeys from Nipah virus disease.

Nipah virus is a zoonotic paramyxovirus that causes severe respiratory and/or encephalitic disease in humans, often resulting in death. It is transmitted from pteropus fruit bats, which serve as the natural reservoir of the virus, and outbreaks occur on an almost annual basis in Bangladesh or India. Outbreaks are small and sporadic, and several cases of human-to-human transmission have been documented as an important feature of the epidemiology of Nipah virus disease. There are no approved countermeasures to combat infection and medical intervention is supportive. We recently generated a recom

A systematic review of Nipah virus disease epidemiological parameters, outbreaks, and mathematical models.

Our systematic review, based on PRISMA guidelines (PROSPERO CRD42023393345), characterised the epidemiology, outbreaks, and mathematical models of Nipah virus, an important public health threat in south and southeast Asia. We searched PubMed and Web of Science from database inception to March 14, 2025, and extracted 243 parameters, 89 risk factors, 39 models, and 23 distinct outbreaks from 119 papers. IgG seroprevalence estimates in the general population ranged from 0% to 12·5%. Nipah virus causes severe disease, with pooled case-fatality ratio estimates ranging widely from 9·1% (95% CI 0·2-4

Post-acute sequelae after Nipah virus infection: a systematic review and meta-analysis.

BACKGROUND: Nipah virus was first detected in the 1998-1999 Malaysia outbreak and remains a significant public health concern due to its high epidemic potential and recurrent outbreaks in South Asia. Incidence patterns of post-acute sequelae, characterised by persistence or delayed onset after the acute phase of an infection, are not well documented after infectious disease outbreaks. We aimed to address this knowledge gap. METHODS: We conducted a systematic review and meta-analysis on the prevalence, incidence, duration, and characteristics of post-acute sequelae in survivors of Nipah virus

One Health Insights From Pteropus medius: Nipah Virus Spillover, Microbiota, and Antimicrobial Resistance.

Pteropus medius is a major reservoir of Nipah virus (NiV), a zoonotic pathogen responsible for recurrent fatal encephalitis outbreaks in Bangladesh. Human infections are primarily associated with consuming raw date palm sap contaminated with bat excreta. Beyond viral spillover, growing evidence suggests that bat-associated microbiota and guano are potential reservoirs of antimicrobial resistance (AMR), contributing to the environmental dissemination of resistant bacteria and resistance genes. This narrative literature review examined the relationship among NiV spillover, bat microbiota, and AM

Seroprevalence of Nipah virus and related paramyxoviruses in native frugivorous bats, Luzon, Philippines.

Nipah virus (NiV) is a highly virulent zoonotic virus classified as a priority pathogen and biohazard. In 2014, an outbreak of NiV-like disease in the Province of Sultan Kudarat, Mindanao, Philippines resulted in a 53% case fatality rate. Here, we identified wildlife bat hosts of NiV by conducting monthly serological surveillance of flying foxes and other native frugivorous bat species across Luzon. We estimated 13.92% NiV seroprevalence in native flying foxes. We also detected NiV neutralizing activity in some flying fox sera and identified factors such as age and seasonality as drivers of hi

Immune correlates of protection for SARS-CoV-2, Ebola and Nipah virus infection.

Correlates of protection (CoP) are biological parameters that predict a certain level of protection against an infectious disease. Well-established correlates of protection facilitate the development and licensing of vaccines by assessing protective efficacy without the need to expose clinical trial participants to the infectious agent against which the vaccine aims to protect. Despite the fact that viruses have many features in common, correlates of protection can vary considerably amongst the same virus family and even amongst a same virus depending on the infection phase that is under consi

Development of Nipah virus-specific IgM & IgG ELISA for screening human serum samples.

BACKGROUND & OBJECTIVES: Nipah virus (NiV) is a zoonotic paramyxovirus that causes fatal encephalitis in humans. Enzyme Linked Immunosorbent Assay (ELISA) is a safe, sensitive, specific, and affordable diagnostic tool that can be used during screening of large-scale epidemiological investigations. Development and evaluation of IgM and IgG ELISA for screening serum samples of NiV suspected cases would also help in planning public health interventions. METHODS: An IgM capture (MAC) ELISA and an indirect IgG ELISA were developed using NiV antigen to detect IgM and IgG antibodies against NiV in h

rVSV-ΔG-EBOV-GP-Nipah-G (also designated PHV02) replication-competent, recombinant vesicular stomatitis virus vaccine for prevention of Nipah virus disease: Standardized template with key considerations for a risk/benefit assessment.

A recombinant vesicular stomatitis virus (rVSV) expressing the glycoprotein (GP) of Ebola virus (rVSV-EBOV GP) has been approved for preventive immunization against Ebola (Ervebo®, Merck & Co.). The rVSV-EBOV vector was modified to express an additional glycoprotein (G) of Nipah virus (NiV), a bat-borne henipavirus endemic/epidemic in South and Southeast Asia that causes fatal encephalitis and pneumonia. The rVSV-EBOV-GP-NiV G vaccine candidate ("PHV02") is a unique chimeric virus with tropism and immunological properties derived from the wild-type (wt) vesicular stomatitis virus (VSV) parent,

Nipah virus infection as a putative risk factor for Alzheimer's disease: A testable hypothesis.

Nipah virus is a neurotropic paramyxovirus known to cause severe acute encephalitis and delayed or relapsing neurological syndromes. Increasing evidence suggests that both systemic infections and central nervous system inflammation may act as contributors to, or accelerators of, Alzheimer's disease pathology. In this context, we propose that Nipah virus infection may confer long-term susceptibility to Alzheimer's disease through several converging mechanisms, including direct neuronal injury, sustained microglial activation mediated by Ephrin-B2/B3 receptor engagement, and persistent disruptio

Nipah virus infects the brain and triggers neuronal and glial damage in a hamster model.

Nipah virus (NiV) is distributed in South-East Asia and the Western Pacific, with currently active outbreak sites. NiV causes an acute febrile illness with high mortality (40-70%), encephalitis, and long-term disability among survivors. Currently, there are no effective therapeutic or preventive interventions, and its potential for explosive spread resulted in its designation as a pathogen of international epidemiological concern. However, the pathophysiology of NiV-induced encephalitis remains unclear. Using a hamster model of NiV infection, we demonstrated that NiV targets several brain area

Reframing early warning for Nipah virus in Bangladesh: A shift from detection to anticipation.

Nipah virus continues to pose a recurrent zoonotic threat in Bangladesh. The sporadic spillover events are associated with high case fatality and limited opportunities for clinical intervention. Despite nearly two decades of surveillance response, early warning systems remain reactive, relying heavily on laboratory confirmation and case accumulation. This Perspective argues that early warning has been conceptually misframed as outbreak detection, rather than risk anticipation. Drawing on the epidemiological characteristics of Nipah virus and Bangladesh's current surveillance architecture, this

Cross-protectivity of henipavirus soluble glycoprotein in an in vivo model of Nipah virus disease.

INTRODUCTION: Nipah virus (NiV) is one of a group of highly pathogenic viruses classified within the Henipavirus genus. Since 2012 at least 11 new henipa-like viruses have been identified, including from new locations and reservoir hosts; the pathogenicity of these new viruses has yet to be determined, but two of them have been associated with morbidity, including fatalities. METHODS: The efficacy and cross-reactivity of two vaccine candidates derived from the soluble glycoproteins of both NiV and Hendra virus (HeV) was evaluated in our recently established hamster model. RESULTS: Both vacci

Establishment of a novel minigenome system for the identification of drugs targeting Nipah virus replication.

Nipah virus (NiV) is a deadly zoonotic pathogen with high potential to cause another pandemic. Owing to biosafety concerns, studies on living NiV must be performed in biosafety level 4 (BSL-4) laboratories, which greatly hinders the development of anti-NiV drugs. To overcome this issue, minigenome systems have been developed to study viral replication and screen for antiviral drugs. This study aimed to develop two minigenome systems (transient and stable expression) based on a helper cell line expressing the NiV P, N and L proteins required to initiate NiV RNA replication. Stable minigenome ce

Advancements in Nipah virus treatment: Analysis of current progress in vaccines, antivirals, and therapeutics.

Nipah virus (NiV) causes severe encephalitis in humans. Three NiV strains NiV-Malaysia (NiVM ), NiV Bangladesh (NiVB ), and NiV India (NiVI reported in 2019) have been circulating in South-Asian nations. Sporadic outbreak observed in South-East Asian countries but human to human transmission raises the concern about its pandemic potential. The presence of the viral genome in reservoir bats has further confirmed that NiV has spread to the African and Australian continents. NiV research activities have gained momentum to achieve specific preparedness goals to meet any future emergency-as a resul

A highly potent, stable, and safe dePEGylated lipopeptide against Nipah virus and related henipaviruses.

Nipah virus (NiV) poses a significant threat owing to its high mortality and the lack of approved therapeutics. Targeting the conserved heptad repeat 1 (HR1) domain of the viral fusion (F) protein constitutes a promising antiviral strategy. Also, lipopeptides derived from the human parainfluenza virus 3 (HPIV3) heptad repeat 2 (HR2) regions inhibit NiV fusion by blocking formation of the critical six-helix bundle (6-HB); however, their efficacy has been impeded by the controversial use of PEGylation. To resolve these limitations, we employed our proprietary heptad repeat 2 C-terminal fragment

Assessing the pandemic potential of Nipah virus: From molecular pathogenesis to ecological spillover risks.

Nipah virus, a highly lethal zoonotic paramyxovirus with a case‑fatality rate of 40-75%, poses a formidable threat to global health security. This review synthesizes its evolving epidemiology, contrasting the Malaysian and Bangladesh‑India strains, and dissects molecular pathogenesis focusing on G/F glycoprotein recognition of ephrin receptors and the multi‑layered immune evasion mediated by P gene products. Phylogenetic analyses reveal 8-10% nucleotide divergence between lineages, with positive selection in the G protein that may compromise vaccine cross‑protection. Ecological risk amplifiers

Potent human neutralizing antibodies against Nipah virus derived from two ancestral antibody heavy chains.

Nipah virus (NiV) is a World Health Organization priority pathogen and there are currently no approved drugs for clinical immunotherapy. Through the use of a naïve human phage-displayed Fab library, two neutralizing antibodies (NiV41 and NiV42) targeting the NiV receptor binding protein (RBP) were identified. Following affinity maturation, antibodies derived from NiV41 display cross-reactivity against both NiV and Hendra virus (HeV), whereas the antibody based on NiV42 is only specific to NiV. Results of immunogenetic analysis reveal a correlation between the maturation of antibodies and their

Structure-Based Design of Nipah Virus Vaccines: A Generalizable Approach to Paramyxovirus Immunogen Development.

Licensed vaccines or therapeutics are rarely available for pathogens with epidemic or pandemic potential. Developing interventions for specific pathogens and defining generalizable approaches for related pathogens is a global priority and inherent to the UN Sustainable Development Goals. Nipah virus (NiV) poses a significant epidemic threat, and zoonotic transmission from bats-to-humans with high fatality rates occurs almost annually. Human-to-human transmission of NiV has been documented in recent outbreaks leading public health officials and government agencies to declare an urgent need for
