A tetrahedral framework nucleic acid-based co-delivery platform enhances the antibacterial activity of nisin and quercetin against MRSA.

The emergence of antibiotic-resistant bacteria necessitates alternative antimicrobial strategies. Here, tetrahedral framework nucleic acids (tFNAs) were engineered as a nanoplatform to co-deliver nisin and quercetin (tFNQ) against a methicillin-resistant Staphylococcus aureus (MRSA) strain isolated from retail pork. tFNAs enhanced bacterial association and enabled nuclease-responsive release, significantly improving antibacterial activity at sub-minimum inhibitory concentrations (sub-MIC) compared with the free combination. tFNQ induced membrane disruption and depolarization, accompanied by re

d-Methionine and d-Phenylalanine Improve Lactococcus lactis F44 Acid Resistance and Nisin Yield by Governing Cell Wall Remodeling.

Lactococcus lactis encounters various environmental challenges, especially acid stress, during its growth. The cell wall can maintain the integrity and shape of the cell under environmental stress, and d-amino acids play an important role in cell wall synthesis. Here, by analyzing the effects of 19 different d-amino acids on the physiology of L. lactis F44, we found that exogenously supplied d-methionine and d-phenylalanine increased the nisin yield by 93.22% and 101.29%, respectively, as well as significantly increasing the acid resistance of L. lactis F44. The composition of the cell wall in

Decontamination of seeds destined for edible sprout production from Listeria by using chitosan coating with synergetic lysozyme-nisin mixture.

This study aimed at decontamination of seeds destined for edible sprout production from Listeria using chitosan (CS) coatings incorporated with synergetic lysozyme-nisin (LYS-NIS) mixtures. Low molecular weight (LMW) CS coating showed the highest potency against Listeria innocua, followed by medium molecular weight (MMW) and high molecular weight (HMW) CSs. The LMW CS film with LYS-NIS also caused almost 1.5-fold greater log reduction (∼5 log) in initial L. innocua load of broth culture than MMW and HMW CS films with LYS-NIS within 6 days. Moreover, LMW CS coating with LYS-NIS reduced the init

Nisin, a Probiotic Bacteriocin, Modulates the Inflammatory and Microbiome Changes in Female Reproductive Organs Mediated by Polymicrobial Periodontal Infection.

Periodontitis-related oral microbial dysbiosis is thought to contribute to adverse pregnancy outcomes (APOs), infertility, and female reproductive inflammation. Since probiotics can modulate periodontitis and oral microbiome dysbiosis, this study examined the effects of a probiotic bacteriocin, nisin, in modulating the reproductive microbiome and inflammation triggered by periodontitis. A total of 24 eight-week-old BALB/cByJ female mice were randomly divided into four treatment groups (control, infection, nisin, and infection+nisin group), with 6 mice per group. A polymicrobial (Porphyromonas

Nisin, a food preservative produced by Lactococcus lactis, affects the localization pattern of intermediate filament protein in HaCaT cells.

Nisin is a food preservative produced by Lactococcus lactis subsp. lactis. Previous blood biochemical research revealed that nisin has physiological effects in mammals; although the site of action has yet to be identified, keratinocytes have been proposed as a possible target. In this study, we investigated whether nisin affects keratinocytes by examining the effects on eukaryotic intermediate filaments in HaCaT human keratinocytes. Treatment with 93 μg/ml nisin for 24 h decreased the localization of the intermediate filament proteins cytokeratin (CK)5 and CK17 at the cell periphery, which wer

Dynamic encapsulation of hydrophilic nisin in hydrophobic poly (lactic acid) particles with controlled morphology by a single emulsion process.

Hydrophilic nisin-loaded hydrophobic poly (lactic acid) (PLA) particles with controlled size and shape were successfully produced utilizing a one-step single emulsification method. Preliminary shear stress and temperature tests showed that there was no significant loss in the nisin inhibition activity during this process. PLA/nisin composite particles were prepared into solid nanocomposite spheres (50-200 nm) or hollow microcomposite spheres (1-5 μm) under the operative conditions developed in our previous study, in which the hydrophilic nisin in the aqueous phase solution could be entrapped i

Synergistic antimicrobial effect of nisin and p-cymene on Salmonella enterica serovar Typhi in vitro and on ready-to-eat food.

Foods contaminated with Salmonella enterica serovar Typhi are a major cause of typhoid fever, leading to public health problems and economic losses worldwide. Nisin and rho-cymene were tested in this study for their antimicrobial activity against S. Typhi at 4 degrees C and 37 degrees C. Nisin and rho-cymene, when used separately, did not inhibit the bacterium at either temperature. A synergistic antimicrobial effect between both compounds was observed when they were used simultaneously. This synergism was greater at 37 degrees C than at 4 degrees C. The lowest concentrations of nisin and rho-

Inactivation of Listeria monocytogenes in skim milk and liquid egg white by antimicrobial bottle coating with polylactic acid and nisin.

This study was to develop an antimicrobial bottle coating method to reduce the risk of outbreaks of human listeriosis caused by contaminated liquid foods. Liquid egg white and skim milk were inoculated with Listeria monocytogenes Scott A and stored in glass jars that were coated with a mixture of polylactic acid (PLA) polymer and nisin. The efficacy of PLA per nisin coating in inactivating L. monocytogenes was investigated at 10 and 4 degrees C. The pathogen grew well in skim milk without PLA/nisin coating treatments, reaching 8 log CFU/mL after 10 d and then remained constant up to 42 d at 10

Synthesis, characterization, and evaluation of antimicrobial double-layer mat incorporating nisin and thyme essential oil to enhance food safety.

Antimicrobial active packaging is a promising strategy to control foodborne contamination, yet the instability and rapid inactivation of natural preservatives in complex food matrices limit their practical use. Here, we fabricate and evaluate an electrospun double-layer nanobiocomposite mat engineered to protect bioactives and enable their slow release. The inner layer comprises chitosan/poly (vinyl alcohol) loaded with nisin and Shirazi thyme essential oil (CS/PVA/N/EO), while the outer layer is cellulose acetate containing thyme essential oil (CA/EO). Solutions were electrospun and character

AcrR1, a novel TetR/AcrR family repressor, mediates acid and antibiotic resistance and nisin biosynthesis in Lactococcus lactis F44.

Lactococcus lactis, widely used in the manufacture of dairy products, encounters various environmental stresses both in natural habitats and during industrial processes. It has evolved intricate machinery of stress sensing and defense to survive harsh stress conditions. Here, we identified a novel TetR/AcrR family transcription regulator, designated AcrR1, to be a repressor for acid and antibiotic tolerance that was derepressed in the presence of vancomycin or under acid stress. The survival rates of acrR1 deletion strain ΔAcrR1 under acid and vancomycin stresses were about 28.7-fold (pH 3.0,

A nisin-inducible chromosomal gene expression system based on ICE Tn5253 of Streptococcus pneumoniae, transferable among streptococci and enterococci.

The present work reports the development and validation of a chromosomal expression system in Streptococcus pneumoniae which permits gene expression under the control of Lactococcus lactis lantibiotic nisin. The system is based on the integrative and conjugative element (ICE) Tn5253 of S. pneumoniae capable of site-specific chromosomal integration and conjugal transfer to a variety of bacterial species. We constructed an insertion vector that integrates in Tn5251, an ICE contained in Tn5253, which carries the tetracycline resistance tet(M) gene. The vector contains the nisRK regulatory system

Nisin lantibiotic prevents NAFLD liver steatosis and mitochondrial oxidative stress following periodontal disease by abrogating oral, gut and liver dysbiosis.

Oral microbiome dysbiosis mediates chronic periodontal disease, gut microbial dysbiosis, and mucosal barrier disfunction that leads to steatohepatitis via the enterohepatic circulation. Improving this dysbiosis towards health may improve liver disease. Treatment with antibiotics and probiotics have been used to modulate the microbial, immunological, and clinical landscape of periodontal disease with some success. The aim of the present investigation was to evaluate the potential for nisin, an antimicrobial peptide produced by Lactococcus lactis, to counteract the periodontitis-associated gut d

Nisin resistance is increased through GtcA mutation induced loss of cell wall teichoic acid N-acetylglucosamine modifications in Listeria monocytogenes.

Nisin resistance development is one of food safety challenges posed by Listeria monocytogenes, an important foodborne pathogen that causes human listeriosis. The GtcA flippase enzyme is functionally crucial in two separate pathways that glycosylate cell envelope wall teichoic acids (WTA) with N-acetylglucosamine (NAG) and lipoteichoic acids (LTA) with galactose, respectively. This study investigated phenotypic roles and molecular mechanisms underlying GtcA involvement in L. monocytogenes nisin resistance. A GtcAA65V mutation was linked with increased nisin resistance in a food processing envir

The "One-Two Punch": a conceptual analysis of nisin and lysozyme synergy against Listeria monocytogenes biofilms and their dependence on environmental factors.

AIMS: The persistence of Listeria monocytogenes biofilms in food processing environments poses a significant challenge to food safety, as these resilient communities exhibit high tolerance to conventional sanitation. This conceptual review synthesizes current literature on two natural antimicrobials: nisin and lysozyme, to evaluate their combined potential against L. monocytogenes biofilms and to propose a novel "One-Two Punch" model describing their sequential synergistic action. It further examines how environmental factors, including surface physicochemical properties, biofilm maturity, and

Thermosonication Combined with Natural Antimicrobial Nisin: A Potential Technique Ensuring Microbiological Safety and Improving the Quality Parameters of Orange Juice.

Currently, thermal pasteurisation (TP) remains the most widely applied technique for commercial orange juice preservation; however, a high temperature causes adverse effects on the quality attributes of orange juice. In order to explore a novel non-thermal sterilization method for orange juice, the impacts of thermosonication combined with nisin (TSN) and TP treatments on the quality attributes including microbial and enzyme inactivation and the physicochemical, nutritional, functional, and sensory qualities of orange juice were studied. Both TP and TSN treatments achieved desirable bactericid
