Extracorporeal membrane oxygenation combined with continuous renal replacement therapy in cutaneous burn and inhalation injury caused by hydrofluoric acid and nitric acid.

RATIONALE: Hydrofluoric acid (HF) is a highly corrosive agent and can cause corrosive burns. HF can penetrate deeply into tissues through intact skin and the lipid barrier, leading to painful liquefactive necrosis, and inducing hypocalcemia and hypomagnesemia. In this study, we hypothesize that continuous renal replacement therapy (CRRT) may be beneficial in addressing hemodynamic instability in cases of HF poisoning. PATIENT CONCERNS: A 25-year-old man fell into an electroplating pool containing 10% HF and 50% nitric acid. DIAGNOSES: He had severe cutaneous injuries involving approximately

Computational Foretelling and Experimental Implementation of the Performance of Polyacrylic Acid and Polyacrylamide Polymers as Eco-Friendly Corrosion Inhibitors for Copper in Nitric Acid.

Copper is primarily used in many industrial processes, but like many other metals, it suffers from corrosion damage. Polymers are not only one of the effective corrosion inhibitors but also are environmentally friendly agents in doing so. Hence, in this paper, the efficacy of two polyelectrolyte polymers, namely poly(acrylic acid) (PAA) and polyacrylamide (PAM), as corrosion inhibitors for copper in molar nitric acid medium was explored. Chemical, electrochemical, and microscopic tools were employed in this investigation. The weight-loss study revealed that the computed inhibition efficiencies

Retinoic acid-mediated enhancement of the cholinergic/neuronal nitric oxide synthase phenotype of the medial septal SN56 clone: establishment of a nitric oxide-sensitive proapoptotic state.

It is unclear what mechanisms lead to the degeneration of basal forebrain cholinergic neurons in Alzheimer's or other human brain diseases. Some brain cholinergic neurons express neuronal nitric oxide (NO) synthase (nNOS), which produces a free radical that has been implicated in some forms of neurodegeneration. We investigated nNOS expression and NO toxicity in SN56 cells, a clonal cholinergic model derived from the medial septum of the mouse basal forebrain. We show here that, in addition to expressing choline acetyltransferase (ChAT), SN56 cells express nNOS. Treatment of SN56 cells with re

Chemically functionalized diglycolamide resin for rapid and efficient recovery of Am(III) from nitric acid medium.

A diglycolamide (DGA)-functionalized silica-based extraction chromatography resin was synthesized and evaluated for the separation of trivalent actinides from nitric acid medium. Batch sorption studies showed strong affinity of the resin toward Am3+, with distribution coefficients (Kd) exceeding 5000 mL/g above 3 M HNO3. Sorption equilibrium was achieved within 20 min, indicating rapid uptake kinetics suitable for separation applications. Equilibrium sorption studies were performed using Eu3+ as a non-radioactive surrogate for Am3+. The resin exhibited a maximum Eu3+ uptake capacity of 9.5±0.5

Nitric oxide donor-modified extracellular matrix hydrogel loaded with salvianolic acid B for diabetic wound healing.

Nitric oxide (NO) donor-based gas therapy has been widely explored to promote angiogenesis in diabetic wound treatment, particularly to compensate for insufficient endogenous NO levels in damaged tissues. Here, we present a skin-derived extracellular matrix (ECM)-based platform that leverages its inherent biocompatibility and cell affinity for efficient NO delivery. An NO donor was chemically grafted onto the ECM scaffold, which was subsequently processed into a hydrogel (H@ECM-SNO/Sal-B) capable of encapsulating salvianolic acid B (Sal B), a bioactive phytochemical with pro-healing potential.

Carnosic Acid Activates the STING/IRF3 Pathway to Induce Nitric Oxide-Mediated Apoptosis in Osteosarcoma Cells.

Osteosarcoma (OS) is a highly aggressive bone cancer with limited therapeutic options. Carnosic acid (CA), a phenolic diterpene with well-established antioxidant properties, has shown anticancer activity, yet its mechanisms in OS remain unclear. In this study, we found that CA suppressed proliferation and induced apoptosis in human osteosarcoma cells in a dose-dependent manner. Mechanistically, CA activated the STING/IRF3 signaling pathway and enhanced nitric oxide (NO) production, factors closely linked to redox modulation and mitochondrial apoptotic signaling. Pharmacological inhibition or s

Jasmonic acid improves cadmium tolerance in rice (Oryza sativa) by reducing the production of nitric oxide.

The involvement of jasmonic acid (JA) in the rice's response to cadmium (Cd) stress is well recognized, though the underlying mechanisms remain unclear. In this study, exposure to Cd stress rapidly elevated endogenous JA concentrations in rice roots, meanwhile, a mutant coleoptile photomorphogenesis 2 (cpm2) which produces less JA, was more sensitive to Cd stress than its wild type (WT). JA mitigated Cd toxicity by decreasing Cd absorption in root cell wall and shoot, which was achieved by up-regulating the expression of the Cd-chelation and efflux-related genes such as OsHMA3, OsABCG36 and Os

Lanthanum supplementation with abscisic acid modulates rhizosphere dynamics through changes in nitric oxide synthesis in wheat.

Lanthanum (La), a rare earth element (REE) found its wide applications in the agricultural land practices. Utilization of La as a fertilizer is based on its concentration for staple crops, including wheat (Triticum aestivum). We have investigated the role of a beneficial dose of La (60 μM) along with ABA (10 μM) in improving wheat root dynamics and defense systems (ascorbate-glutathione pathway, secondary metabolites, and nitric oxide (NO) biosynthesis). Co-application of La60 and ABA significantly enhanced all the root dynamic attributes along with defense systems and reduced production of re

Jasmonic acid and nitric oxide orchestrate a hierarchical melatonin cascade for Botrytis cinerea resistance in tomato.

Tomato gray mold, caused by Botrytis cinerea (B. cinerea), poses a major challenge to tomato production. In plants, B. cinerea resistance is positively regulated by melatonin; however, the regulatory mechanism of melatonin biosynthesis during B. cinerea infection is not known. Here, we established the working model of jasmonic acid (JA) and nitric oxide (NO) on melatonin biosynthesis in the state of B. cinerea infection. NO responded to B. cinerea infection earlier than JA. In the early stage of infection, tomato caffeic acid O-methyltransferase 2 (SlCOMT2) was S-nitrosylated by NO at Cys344,

β-Carotene Suppresses Lipopolysaccharide-induced Nitric Oxide Production in Microglia via Retinoic Acid Receptor-dependent Mechanisms.

Carotenoids are one of the common food ingredients with potential anti-inflammatory effects. Screening of carotenoids for their anti-inflammatory effects has predominantly been conducted using macrophages, whereas comparative analyses of their effects on excessive activation of microglia remain limited. This study aimed to evaluate the anti-inflammatory effects of eight different carotenoids in lipopolysaccharide (LPS)-stimulated microglia, focusing on nitric oxide (NO) production suppression and underlying molecular mechanisms. Mouse microglia cell line MG6 was pretreated with carotenoids, fo

Enhanced detection of aromatic oxidation products using NO3 - chemical ionization mass spectrometry with limited nitric acid.

Nitrate ion-based chemical ionization mass spectrometry (NO3 --CIMS) is widely used for detection of highly oxygenated organic molecules (HOMs). HOMs are known to participate in molecular clustering and new particle formation and growth, and hence understanding the formation pathways and amounts of these compounds in the atmosphere is essential. However, the absence of analytical standards prevents robust quantification of HOM concentrations. In addition, nitrate-based ionization is usually very selective towards the most oxygenated molecules and blind to less oxygenated compounds hindering th

Why so blue? A novel presentation of methaemoglobinaemia secondary to an inhaled occupational nitric acid exposure.

INTRODUCTION: Nitric and hydrofluoric acids are commonly used in the commercial cleaning industry. We are unaware of reports of nitric acid inhalation forming methaemoglobin. Additionally, methaemoglobinaemia and treatment with methylthioninium chloride (methylene blue) may precipitate clinical uncertainty due to similar wavelengths of absorbance in pulse oximetry. CASES: We report two patients with respiratory distress from symptomatic methaemoglobinaemia following a prolonged, inhaled occupational exposure to nitric acid in the context of industrial cleaning. Their methaemoglobinaemia was s

Successful DTPA therapy in the case of 239Pu penetration via injured skin exposed to nitric acid.

This paper presents results of the radiological study and DTPA therapy for a worker exposed to a plutonium nitrate solution. Plutonium levels were measured in excreta, blood, plasma and wound for several weeks. Plutonium renal clearance ranged from 110-190 ml min(-1) to 3-4 ml min(-1) at different stages of chelation therapy. Plutonium absorption into blood from the injured skin amounted to 4.3%. As a result of intensive therapy, 96% of absorbed plutonium was successfully excreted.

Luminal calcium in regulation of nitric oxide release and acid secretion in rat stomachs after damage.

We investigated the role of luminal Ca2+ in the regulation of nitric oxide (NO) release and acid secretion in the rat stomach following damage by sodium taurocholate (TC). Under urethane anesthesia, a rat stomach was mounted in an ex vivo chamber, perfused with saline, and transmucosal potential difference (PD), luminal pH, acid secretion, and luminal contents of Ca2+ and NO were measured before and after exposure to 20 mM TC for 30 min, with or without coapplication of EGTA (5 mM) and/or CaCl2 (10 mM). Mucosal exposure to TC caused a reduction in PD and a decrease of acid secretion, with a co

Nitrate tolerance is specific for nitric acid esters and its recovery requires an intact protein synthesis.

Using cultured LLC-PK1 cells the present study investigates mechanisms leading to nitrate tolerance and its reversal. A 5-h pretreatment with glyceryl trinitrate (GTN, 0.01-100 microM) resulted in desensitization of the intracellular cyclic GMP response to a subsequent 10-min challenge with GTN (1 microM). The spontaneous donor of nitric oxide (NO) spermine NONOate, which releases NO independently of enzymatic catalysis, did not induce tolerance to its own cyclic GMP stimulatory effect and remained fully effective in GTN-tolerant cells. Tolerant cells regained sensitivity to GTN after a 30-h i

Inducible nitric oxide synthase: identification of amino acid residues essential for dimerization and binding of tetrahydrobiopterin.

Nitric oxide synthases (NOSs) require tetrahydrobiopterin (BH4) for dimerization and NO production. Mutation analysis of mouse inducible NOS (iNOS; NOS2) identified Gly-450 and Ala-453 as critical for NO production, dimer formation, and BH4 binding. Substitutions at five neighboring positions were tolerated, and normal binding of heme, calmodulin, and NADPH militated against major distortions affecting the NH2-terminal portion, midzone, or COOH terminus of the inactive mutants. Direct involvement of residues 450 and 453 in the binding of BH4 is supported by the striking homology of residues 44

Carnosic acid ameliorates methotrexate-induced male infertility: targeting testicular redox imbalance and nitric oxide/CGMP signaling.

PURPOSE: Methotrexate (MTX), a folate antagonist used in the treatment of cancer and autoimmune disorders, unfortunately, induces testicular toxicity and impairs male fertility. Although the tissue-protective properties of Carnosic acid (CAR) are known, its effectiveness in mitigating MTX-induced reproductive toxicity has not been adequately studied. This study explores the protective role of CAR against the toxicity of MTX in rat testes. METHODS: Forty male Wistar rats were divided into four groups: control, MTX (20 mg/kg i.p., day 7), MTX + low-dose CAR (20 mg/kg p.o., 14 days), and MTX + h

Nitric oxide synthesis stimulated by arachidonic acid accumulation via PPARα in acetylcholine-stimulated gastric mucous cells.

NEW FINDINGS: What is the central question of this study? Arachidonic acid (AA) stimulates NO production in antral mucous cells without any increase in [Ca2+ ]i . Given that the intracellular AA concentration is too low to measure, the relationship between AA accumulation and NO production remains uncertain. Is AA accumulation a key step for NO production? What is the main finding and its importance? We demonstrated that AA accumulation is a key step for NO production. The amount of AA released could be measured using fluorescence-HPLC. The intracellular AA concentration was maintained at < 1

Cesium removal from a water system using a polysulfone carrier containing nitric acid-treated bamboo charcoal.

Laboratory scale sorption and desorption experiments were performed to investigate the cesium (Cs) removal efficiency of a bead-shaped polysulfone carrier containing HNO3-treated bamboo charcoal (BC). The average Cs removal efficiency of BC only and of polysulfone carrier without BC after 1 h sorption reaction was 53 and 18%, respectively. However, the Cs removal efficiency for the polysulfone carrier with 5% HNO3-treated BC (P-5N-BC) after 1 h and 24 h reaction was 66 and 98%, respectively. The Cs removal efficiency after 24 h reaction remained >85% over a wide range of pH and temperature con
