Long-term nitrogen deposition reduces the diversity of nitrogen-fixing plants.

Biological nitrogen fixation is a fundamental part of ecosystem functioning. Anthropogenic nitrogen deposition and climate change may, however, limit the competitive advantage of nitrogen-fixing plants, leading to reduced relative diversity of nitrogen-fixing plants. Yet, assessments of changes of nitrogen-fixing plant long-term community diversity are rare. Here, we examine temporal trends in the diversity of nitrogen-fixing plants and their relationships with anthropogenic nitrogen deposition while accounting for changes in temperature and aridity. We used forest-floor vegetation resurveys o

Harnessing Rhizobial Inoculation for Sustainable Nitrogen Management in Mung Bean (Vigna radiata L.).

As a pulse crop, mung beans are associated with nitrogen-fixing bacteria, which can improve soil fertility, lower the need for nitrogen fertilizers, and increase yield and soil quality for subsequent harvests. This study aimed to identify effective rhizobial inoculants for mung beans (Vigna radiata L.) by evaluating selected strains for cowpea (Vigna unguiculata L.), soybean (Glycine max L.), and common bean (Phaseolus vulgaris L.) under controlled (axenic) conditions. Cowpea, soybean, and common bean strains were tested as mung beans inoculants under axenic conditions. Promising strains were

Analysis of the nitrogen-fixing ability of the diazotrophic community living in the rice rhizosphere using a novel test tube chemostat culture.

The rice rhizosphere is a hot spot for nitrogen fixation using carbon sources from root exudates. We herein investigated the intensity of the nitrogen-fixing ability of each diazotroph interacting with other bacteria within the rhizosphere microbiota. We established the Test Tube Chemostat replicating Rice Rhizosphere (TCRR) based on four characteristics of the rice rhizosphere: root exudates, percolation, radial oxygen loss, and soil. The TCRR flora constructed by culturing paddy soil using TCRR showed the pH and high nitrogen-fixing ability of the rice rhizosphere. Twenty-five species of dia

Hacking heterocysts: advances in the genetic regulation of heterocyst differentiation.

Heterocyst differentiation in certain filamentous cyanobacteria is a multifaceted process essential for nitrogen fixation, orchestrated by a sophisticated regulatory network that encompasses several key stages. These include induction, pattern differentiation, commitment, extracellular layer formation, cell-cell communication, and ultimately, nitrogen fixation and metabolism. Key regulators like NtcA and HetR control heterocyst development, while proteins such as PatS, HetN, and PatA modulate pattern formation. Certain non-coding RNAs, such as NsiR1, Yfr1, and NsiR4, also regulate gene express

Same core just with the paradoxical condition: A review of biogeochemical mechanisms of oxic methane production in lakes.

The discovery of oxic methane production has significantly expanded the conceptual boundaries of traditional methane production theory, extending research from marine to lake environments. This review systematically synthesizes current research progress on oxic methane production mechanisms in lakes, with particular emphasis on the processes by which methylated nitrogen (N)-, sulfur (S)-, and phosphorus (P)-compounds serve as key precursors to generate methane through both enzymatic biological reactions and chemical transformation pathways, and oxic methane production mechanisms driven by fund

Engineering nitrogen-fixing symbiosis: bridging signaling and intracellular entry.

Nitrogen availability is a major constraint on plant growth and agricultural productivity. Legumes and several other plant lineages circumvent this limitation by establishing symbiotic associations with nitrogen-fixing microorganisms, which convert atmospheric dinitrogen into ammonia that the host assimilates. Since root nodules were first recognized as sites of biological nitrogen fixation in the late 19th century, engineering this capacity in nonlegume crops has remained a long-standing goal. However, two key challenges remain: enabling intracellular rhizobial infection and establishing a mo

Carnivorous Nepenthes Pitchers with Less Acidic Fluid House Nitrogen-Fixing Bacteria.

Carnivorous pitcher plants are uniquely adapted to nitrogen limitation, using pitfall traps to acquire nutrients from insect prey. Pitcher plants in the genus Sarracenia may also use nitrogen fixed by bacteria inhabiting the aquatic microcosms of their pitchers. Here, we investigated whether species of a convergently evolved pitcher plant genus, Nepenthes, might also use bacterial nitrogen fixation as an alternative strategy for nitrogen capture. First, we constructed predicted metagenomes of pitcher organisms from three species of Singaporean Nepenthes using 16S rRNA sequence data and correla

The nitrogen cycle.

Nitrogen is the fourth most abundant element in cellular biomass, and it comprises the majority of Earth's atmosphere. The interchange between inert dinitrogen gas (N2) in the extant atmosphere and 'reactive nitrogen' (those nitrogen compounds that support, or are products of, cellular metabolism and growth) is entirely controlled by microbial activities. This was not the case, however, in the primordial atmosphere, when abiotic reactions likely played a significant role in the inter-transformation of nitrogen oxides. Although such abiotic reactions are still important, the extant nitrogen cyc

Spatiotemporal Regulation of N2 Fixation and Photosynthesis in Trichodesmium: New Insights From mRNA CARD-FISH and nanoSIMS.

To protect the O2-sensitive nitrogenase enzyme, diazotrophic cyanobacteria separate N2 fixation from photosynthesis in space or time. However, in the filamentous Trichodesmium, technical limitations and conflicting results have so far prevented a consensus on the separation mechanism. Here, we specifically adapted single-cell methods for simultaneously capturing both photosynthesis and N2 fixation at the levels of gene expression, fixation and transport of carbon and nitrogen. Testing expression of nifH (nitrogenase) and psbA (photosystem II), we found that about half of the cells exclusively

Cyanophage lysis reshapes nitrogen cycling and microbiome composition in diazotrophic cyanobacterium Aphanizomenon flos-aquae.

Although it is presumed that viruses play a significant role in nutrient cycling and bacterial community dynamics, this has only rarely been addressed in studies of diazotrophic cyanobacteria. In this study, we therefore examined how cyanophage infection affects the expression of nitrogen (N) cycling genes, N₂ fixation rates, and population structure in the diazotrophic cyanobacterium Aphanizomenon flos-aquae, a prevalent bloom-forming species in temperate brackish and freshwater ecosystems. We also assessed the influence of A. flos-aquae lysis on co-occurring bacterial assemblages throughout

Active Methanotrophs in Suboxic Alpine Swamp Soils of the Qinghai-Tibetan Plateau.

Methanotrophs are the only biofilters for reducing the flux of global methane (CH4) emissions in water-logged wetlands. However, adaptation of aerobic methanotrophs to low concentrations of oxygen and nitrogen in typical swamps, such as that of the Qinghai-Tibetan Plateau, is poorly understood. In this study, we show that Methylobacter-like methanotrophs dominate methane oxidation and nitrogen fixation under suboxic conditions in alpine swamp soils. Following incubation with 13C-CH4 and 15N-N2 for 90 days under suboxic conditions with repeated flushing using an inert gas (i.e., argon), microbi

A Ce-UiO-66 Metal-Organic Framework-Based Graphene-Embedded Photocatalyst with Controllable Activation for Solar Ammonia Fertilizer Production.

Currently, nitrogen fertilizers feed half of the global population, but their use is limited by energy consumption and transportation. Therefore, it is important to study photocatalysts for use in solar nitrogen fertilizers. Herein, a new type of graphene-embedded Ce-based UiO-66 (Ce-UiO-66) photocatalyst (GSCe) is investigated. Ce-UiO-66 is activated by the breakage of benzene-C bonds and the formation of active sites by ultraviolet light in water. Moreover, embedding graphene effectively controls activation and improves nitrogen fixation. GSCe exhibited a remarkable apparent quantum efficien

Iron oxide minerals: promising materials for sustainable rice production via stimulating iron-reducing diazotrophs.

This mini-review highlights the emerging agricultural applications of iron oxides, the primary form of iron and one of the most abundant elements on Earth. Rice yield is strongly influenced by soil nitrogen fertility, which is supported by biological nitrogen fixation. Using soil metatranscriptomic analysis and isolation-cultivation experiments, we recently discovered iron-reducing Deltaproteobacteria as the predominant but previously overlooked drivers of nitrogen fixation in paddy soil. As these bacteria utilize Fe3+ as an electron acceptor during anaerobic respiration, we hypothesized that

Engineering a Photoautotrophic Microbial Coculture toward Enhanced Biohydrogen Production.

The application of synthetic phototrophic microbial consortia holds promise for sustainable bioenergy production. Nevertheless, strategies for the efficient construction and regulation of such consortia remain challenging. Applying tools of genetic engineering, this study successfully constructed a synthetic community of phototrophs using Rhodopseudomonas palustris (R. palustris) and an engineered strain of Synechocystis sp PCC6803 for acetate production (Synechocystis_acs), enabling the production of biohydrogen and fatty acids during nitrogen and carbon dioxide fixation. Iron-enhanced rTCA cycle drives synergistic enhancement of inorganic carbon fixation and denitrification.

Iron-carbon autotrophic denitrification has shown potential for nitrogen removal from low-C/N wastewater. However, its application has been limited by slow Fe0 corrosion, increased alkalization, and insufficient inorganic carbon. CO2 induction represents a promising strategy for overcoming the above limitations. Here, a CO2 enhanced iron-carbon autotrophic denitrification process was established to elucidate how CO2 regulates iron corrosion, carbon metabolism, and denitrification. The results showed that the continuous CO2 supply maintained weakly acidic conditions (pH 6.0-6.5), accelerated Fe

Modulating plant growth-metabolism coordination for sustainable agriculture.

Enhancing global food security by increasing the productivity of green revolution varieties of cereals risks increasing the collateral environmental damage produced by inorganic nitrogen fertilizers. Improvements in the efficiency of nitrogen use of crops are therefore essential; however, they require an in-depth understanding of the co-regulatory mechanisms that integrate growth, nitrogen assimilation and carbon fixation. Here we show that the balanced opposing activities and physical interactions of the rice GROWTH-REGULATING FACTOR 4 (GRF4) transcription factor and the growth inhibitor DELL

Highly diverse diazotrophs drive high N2 fixation rates in a shallow submarine hydrothermal system.

Dinitrogen (N2) fixation provides nitrogen sources supporting active chemoautotrophic processes in submarine hydrothermal systems. Here, the diazotrophic phylogenetic diversity and N2 fixation profile were investigated in depth along a sharp biogeochemical gradient from the hydrothermal vents to ambient seawater in a shallow submarine hydrothermal system (SHS) off Kueishantao Islet, Taiwan. Diazotrophic community compositions in the SHS covered all four known nifH clusters (I, II, III, and IV) and contained both characteristics of diazotrophic communities in deep-sea hydrothermal systems and t

Composition of non-cyanobacterial diazotrophs in suspended and sinking particles is driven by productivity phase.

UNLABELLED: Biological nitrogen fixation is a key process impacting productivity in the Baltic Sea. Most studies of this process have focused on filamentous cyanobacterial diazotrophs, leaving the ecological roles of non-cyanobacterial diazotrophs (NCDs) poorly resolved. Here, we investigate the diazotrophic community across different primary productivity phases (productive and post-productive) and within distinct particle fractions (suspended and sinking) across the Northern Baltic Proper, Gulf of Finland, and Gulf of Bothnia. Using nifH gene amplicon sequencing, we show that NCDs dominate th

Nitrogenase cofactor biosynthesis using proteins produced in mitochondria of Saccharomyces cerevisiae.

Biological nitrogen fixation, the conversion of inert N2 to metabolically tractable NH3, is only performed by certain microorganisms called diazotrophs and is catalyzed by the nitrogenases. A [7Fe-9S-C-Mo-R-homocitrate]-cofactor, designated FeMo-co, provides the catalytic site for N2 reduction in the Mo-dependent nitrogenase. Thus, achieving FeMo-co formation in model eukaryotic organisms, such as Saccharomyces cerevisiae, represents an important milestone toward endowing them with a capacity for Mo-dependent biological nitrogen fixation. A central player in FeMo-co assembly is the scaffold pr

Nitrogen-fixing bacteria and Oxalis - evidence for a vertically inherited bacterial symbiosis.

BACKGROUND: Plant-endophyte symbioses often revolve around nitrogen metabolism, and involve varying degrees of intimacy. Although evidence for vertical inheritance of nitrogen-fixing endophytic bacteria is increasing, it is confined mostly to crop plants, and to date no such system has been reported for geophytes. METHODS: Bacterial endophytes associated with Oxalis, the most species-rich geophytic genus form the Cape Flora in southern Africa was studied. Culturable endophytes were isolated from surface-sterilized vegetative and reproductive plant organs for six host species at three location
