Driving mechanism of different nutrient conditions on microbial mediated nitrate reduction in magnetite-present river infiltration zone.

Significant research is focused on the ability of riparian zones to reduce groundwater nitrate contamination. Owing to the extremely high redox activity of nitrate, naturally existing electron donors, such as organic matter and iron minerals, are crucial in facilitating nitrate reduction in the riparian zone. Here, we examined the coexistence of magnetite, an iron mineral, and nitrate, a frequently observed coexisting system in sediments, to investigate nitrate reduction features at various C/N ratios and evaluate the response of microbial communities to these settings. Additionally, we aimed

Complete genome sequence of Nguyenibacter sp. L1, a phosphate solubilizing bacterium isolated from Lespedeza bicolor rhizosphere.

Phosphorus (P) deficiency is a predominant constraint on plant growth in acidified soils, largely due to the sequestration of P by toxic aluminum (Al) compounds. Indigenous phosphorus-solubilizing bacteria (PSBs) capable of mobilizing Al-P in these soils hold significant promise. A novel Al-P-solubilizing strain, Al-P Nguyenibacter sp. L1, was isolated from the rhizosphere soil of healthy Lespedeza bicolor plants indigenous to acidic terrains. However, our understanding of the genomic landscape of bacterial species within the genus Nguyenibacter remains in its infancy. To further explore its b

Identification of Sinorhizobium (Ensifer) medicae based on a specific genomic sequence unveiled by M13-PCR fingerprinting.

A collection of nodule isolates from Medicago polymorpha obtained from southern and central Portugal was evaluated by M13-PCR fingerprinting and hierarchical cluster analysis. Several genomic clusters were obtained which, by 16S rRNA gene sequencing of selected representatives, were shown to be associated with particular taxonomic groups of rhizobia and other soil bacteria. The method provided a clear separation between rhizobia and co-isolated non-symbiotic soil contaminants. Ten M13-PCR groups were assigned to Sinorhizobium (Ensifer) medicae and included all isolates responsible for the form

Beneath the surface: expanding the known repertoire of methylotrophic metabolism.

Although the metabolic pathways that allow the utilization of one-carbon compounds as sole sources of carbon and energy (methylotrophy) are well characterized, this understanding has been substantially refined and expanded in recent years. The paradigm-shifting discovery of the lanthanide-dependent methanol dehydrogenase, XoxF, established the biological relevance of rare-earth metals and revealed that methylotrophy required reassessment. We now know that XoxF is broadly distributed among bacteria and may actually constitute an ancestral form by which methylotrophy initially evolved, as well a

Microbial community dynamics mediate rapid biomass loss and phosphorus recycling in pelagic Sargassum.

Sargassum blooms in the tropical North Atlantic have surged to unprecedented levels, causing massive algal tides that severely impact coastal ecosystems. To improve the current understanding of Sargassum biomass dynamics and the contribution of this productive algal bloom to carbon and nutrient fluxes between the Atlantic and the Caribbean, we characterized the growth and decomposition rates of this pelagic alga, nutrient release rates during litter decay, and the bacterial microbiome at several stages of this decay. The analysis revealed no exceptional growth under increased nutrient availabi

Effects of vegetation type on microbial biomass carbon and nitrogen in subalpine mountain forest soils.

BACKGROUND/PURPOSE: Microbial biomass plays an important role in nutrient transformation and conservation of forest and grassland ecosystems. The objective of this study was to determine the microbial biomass among three vegetation types in subalpine mountain forest soils of Taiwan. METHODS: Tatachia is a typical high-altitude subalpine temperate forest ecosystem in Taiwan with an elevation of 1800-3952 m and consists of three vegetation types: spruce, hemlock, and grassland. Three plots were selected in each vegetation type. Soil samples were collected from the organic layer, topsoil, and su

The genome of Rhizobiales bacteria in predatory ants reveals urease gene functions but no genes for nitrogen fixation.

Gut-associated microbiota of ants include Rhizobiales bacteria with affiliation to the genus Bartonella. These bacteria may enable the ants to fix atmospheric nitrogen, but no genomes have been sequenced yet to test the hypothesis. Sequence reads from a member of the Rhizobiales were identified in the data collected in a genome project of the ant Harpegnathos saltator. We present an analysis of the closed 1.86 Mb genome of the ant-associated bacterium, for which we suggest the species name Candidatus Tokpelaia hoelldoblerii. A phylogenetic analysis reveals a relationship to Bartonella and Bruc

Small but big player: the important role of microRNAs in legume crops.

Legumes are essential components of global cropping systems due to their nutritional value and contribution to sustainable agriculture. Among the regulatory molecules, small RNAs (sRNAs), particularly microRNAs (miRNAs), play crucial roles in plant development and in responses to biotic and abiotic stresses. miRNAs regulate genes involved in diverse developmental processes, including nodule formation, which is fundamental for the nitrogen-fixing symbiosis that characterizes legumes. Functional studies have demonstrated that miRNAs are key modulators of plant defense, contributing to resistance

Chromium(VI) Toxicity in Legume Plants: Modulation Effects of Rhizobial Symbiosis.

Most legume species have the ability to establish a symbiotic relationship with soil nitrogen-fixing rhizobacteria that promote plant growth and productivity. There is an increasing evidence of reactive oxygen species (ROS) important role in formation of legume-rhizobium symbiosis and nodule functioning. Environmental pollutants such as chromium compounds can cause damage to rhizobia, legumes, and their symbiosis. In plants, toxic effects of chromium(VI) compounds are associated with the increased production of ROS and oxidative stress development as well as with inhibition of pigment synthesi

Effects of Vegetation Restoration Type on Soil Greenhouse Gas Emissions and Associated Microbial Regulation on the Loess Plateau.

Investigating responses of soil greenhouse gas (GHG) emissions to vegetation restoration is important for global warming mitigation. On the Loess Plateau, a wide range of vegetation restoration strategies have been implemented to control land degradation. However, the thorough quantification of soil GHG emissions triggered by different modes of vegetation restoration is insufficient. There is still a knowledge gap regarding the regulation of soil biochemical and microbial processing on soil GHG emissions. To do so, we compared responses of soil GHG emissions to various types of vegetation rest

Type III effectors of symbiotic Rhizobia include diverse predicted transcriptional and post-transcriptional modulators.

Rhizobia are soil bacteria that establish nitrogen-fixing symbioses with legumes. While many rhizobia use a Type III Secretion System to deliver "Nodulation Outer Protein" (Nop) effectors, some uniquely use these proteins to initiate nodule organogenesis, bypassing classical signalling. The molecular functions of these effectors remain largely unknown due to extreme sequence divergence. Using AlphaFold2-mediated structural proteomics, we identified a modular architecture in rhizobial effectors composed of 22 distinct structural units. We reveal that many Nop effectors are cryptic transcription

Beyond elongation: The multifaceted roles of gibberellins in symbiosis and root development.

Plants regulate root development in response to fluctuating environmental conditions, including establishing symbiotic relationships with arbuscular mycorrhizal fungi and nitrogen-fixing bacteria under nutrient limitation. These processes are orchestrated by plant hormones, particularly gibberellins, and the repressors of gibberellin signalling, DELLA proteins. Gibberellin and DELLAs serve as critical regulators in symbiotic signalling and root organogenesis, integrating hormonal and environmental cues with cellular patterning to direct plant development. This review explores the current under

Rhizobium leguminosarum bv. trifolii PssP protein is required for exopolysaccharide biosynthesis and polymerization.

Rhizobium leguminosarum bv. trifolii produces an acidic exopolysaccharide (EPS) that is important for the induction of nitrogen-fixing nodules on clover. Recently, three genes, pssN, pssO, and pssP, possibly involved in EPS biosynthesis and polymerization were identified. The predicted protein product of the pssP gene shows a significant sequence similarity to other proteins belonging to the PCP2a family that are involved in the synthesis of high-molecular-weight EPS. An R. leguminosarum bv. trifolii TA1 mutant with the entire coding region of pssP deleted did not produce the EPS. A pssP mutan

Chronic intracellular infection of alfalfa nodules by Sinorhizobium meliloti requires correct lipopolysaccharide core.

Our analyses of lipopolysaccharide mutants of Sinorhizobium meliloti offer insights into how this bacterium establishes the chronic intracellular infection of plant cells that is necessary for its nitrogen-fixing symbiosis with alfalfa. Derivatives of S. meliloti strain Rm1021 carrying an lpsB mutation are capable of colonizing curled root hairs and forming infection threads in alfalfa in a manner similar to a wild-type strain. However, developmental abnormalities occur in the bacterium and the plant at the stage when the bacteria invade the plant nodule cells. Loss-of-function lpsB mutations,

Railroad derived nitrogen and heavy metal pollution does not affect nitrogen fixation associated with mosses and lichens at a tundra site in Northern Sweden.

Traffic derived nitrogen (N) and heavy metal pollution is a well-known phenomenon, but little explored in otherwise pristine ecosystems such as subarctic tundra. Here, the main source of N input to the ecosystem is via N2 fixation by moss- and lichen-associated bacteria. While inhibitory effects of N deposition on moss-associated N2 fixation have been reported, we still lack understanding of the effects of traffic derived N and heavy metal deposition on this ecosystem function in an otherwise pristine setting. To test this, we established a distance gradient (0-1280 m) away from a metal pollut

A diazotrophy-ammoniotrophy dual growth model for the sulfate reducing bacterium Desulfovibrio vulgaris var. Hildenborough.

Sulfate reducing bacteria (SRB) comprise one of the few prokaryotic groups in which biological nitrogen fixation (BNF) is common. Recent studies have highlighted SRB roles in N cycling, particularly in oligotrophic coastal and benthic environments where they could contribute significantly to N input. Most studies of SRB have focused on sulfur cycling and SRB growth models have primarily aimed at understanding the effects of electron sources, with N usually provided as fixed-N (nitrate, ammonium). Mechanistic links between SRB nitrogen-fixing metabolism and growth are not well understood, parti

Metaproteomics reveals metabolic transitions between healthy and diseased stony coral Mussismilia braziliensis.

Infectious diseases such as white plague syndrome (WPS) and black band disease (BBD) have caused massive coral loss worldwide. We performed a metaproteomic study on the Abrolhos coral Mussismilia braziliensis to define the types of proteins expressed in healthy corals compared to WPS- and BBD-affected corals. A total of 6363 MS/MS spectra were identified as 361 different proteins. Healthy corals had a set of proteins that may be considered markers of holobiont homoeostasis, including tubulin, histone, Rab family, ribosomal, peridinin-chlorophyll a-binding protein, F0F1-type ATP synthase, alpha

The anammox coupled partial-denitrification process in an integrated granular sludge and fixed-biofilm reactor developed for mainstream wastewater treatment: Performance and community structure.

This study describes an integrated granular sludge and fixed-biofilm (iGB) reactor innovatively designed to carry out the anammox/partial-denitrification (A/PD) process for nitrogen removal with mainstream municipal wastewater. The iGB-A/PD reactor consists of anammox granules inoculated in the lower region of reactor and an acclimated fixed-biofilm positioned in the upper region. Compared to the other reported A/PD systems for mainstream wastewater treatment, this iGB-A/PD reactor is notable due to its higher quality effluent with a total inorganic nitrogen (TIN) of ∼3 mg•L-1 and operation at
