Molybdenum doping enhances backbonding at neighboring iron centers in iron-sulfur clusters.

Nitrogenases employ Fe-S co-factors to reduce the weakly π-acidic ligand N2 to NH3. The Mo nitrogenase is more efficient than the V and Fe-only nitrogenases, but how the Mo ion enhances N2 reduction is not fully understood. In this study, we undertake a comparative structural and spectroscopic analysis of synthetic [MoFe3S4] and [Fe4S4] clusters that ultimately reveals how the presence of Mo impacts the ability of Fe-S clusters to interact with π-acids. Specifically, we find that compared with the Fe centers in the [Fe4S4] clusters, those in the [MoFe3S4] clusters are more adept at backbonding

Dinitrogen Coordination to a High-Spin Diiron(I/II) Species.

Dinitrogen coordination to iron centers underpins industrial and biological fixation in the Haber-Bosch process and by the FeM cofactors in the nitrogenase enzymes. The latter employ local high-spin metal centers; however, iron-dinitrogen coordination chemistry remains dominated by low-valent states, contrasting the enzyme systems. Here, we report a high-spin mixed-valent cis-(μ-1,2-dinitrogen)diiron(I/II) complex [(FeBr)2 (μ-N2 )Lbis ]- (2), where [Lbis ]- is a bis(β-diketiminate) cyclophane. Biotransformation of cyanide to methane and ammonia by Klebsiella oxytoca.

Klebsiella oxytoca, isolated from cyanide-containing industrial wastewater, was shown to be able to biodegrade cyanide to non-toxic endproducts using cyanide as the sole nitrogen source. In this study, ammonia was one of the detected endproduct of cyanide biodegradation by the concentrated resting cells of K. oxytoca. Moreover, cyanide has been shown to be biotransformed to methane through the actions of concentrated resting cells. Biodegradation of cyanide by cell-free extracts was not observed, which might be due to the inactivation of nitrogenase (an oxygen-labial enzyme) caused by the oxyg

Toxicity of chromium and tin to Anabaena doliolum. Interaction with sulphur-containing amino acids and thiols.

Toxicity of chromium and tin on growth, heterocyst differentiation, nitrogenase activity and 14CO2 uptake of Anabaena doliolum and its amelioration by sulphur-containing amino acids and thiols has been studied. The final growth yield was found to be approximately 51% and 58% of control at sublethal concentration of chromium and tin respectively. Among various amino acids tested, cysteine (0.05 mM) significantly restored growth, heterocyst differentiation, nitrogenase and 14CO2 uptake of test alga. Dithiothreitol (1 mM) restored all the parameters and processes better than monothiol, mercaptoet

Growth and cyanide degradation of Azotobacter vinelandii in cyanide-containing wastewater system.

Azotobacter vinelandii, a strict aerobic nitrogen-fixing bacterium, has been extensively studied with regard to the ability of N2-fixation due to its high expression of nitrogenase and fast growth. Because nitrogenase can also reduce cyanide to ammonia and methane, cyanide degradation by A. vinelandii has been studied for the application in the bioremediation of cyanide-contaminated wastewater. Cyanide degradation by A. vinelandii in NFS (nitrogen-free sucrose) medium was examined in terms of cell growth and cyanide reduction, and the results were applied for cyanide-contaminated cassava mill

Diazotrophic community restructuring and soil metabolome reprogramming drive enhanced nitrogen fixation in pecan plantations of different ages.

Biological nitrogen fixation (BNF) is a key ecological process underpinning sustainable agroforestry by reducing dependence on synthetic nitrogen fertilizers. We used integrated high-throughput sequencing and untargeted metabolomics to examine how stand age affects BNF activity, diazotrophic community structure, and soil metabolomic profiles in pecan (Carya illinoinensis) orchards at two developmental stages (7year and 12-year stands). Pecan age significantly increased diazotrophic α-diversity (by 23.52%, p < 0.01) and nitrogenase activity (p < 0.001). Pseudomonadota dominated across all sampl

Functional methylthio-alkane reductases are widely distributed for bacterial organic sulfur acquisition.

UNLABELLED: Methylthio-alkane reductase (MAR) is a relative of nitrogenase that cleaves methylthio-alkanes for methanethiol liberation and subsequent methionine biogenesis. For example, dimethylsulfide, the most abundant volatile organic sulfur compound, is cleaved to methanethiol and methane. To date, the only verified MAR enzymes are from Alphaproteobacteria like Rhodospirillum rubrum, despite the long-time observation of gene homologs with potential MAR function present across diverse taxa. Synthesis of MAR genes from uncultivated organisms and expression in R. rubrum reveals that functiona

Light-driven Transformation of Carbon Monoxide into Hydrocarbons using CdS@ZnS : VFe Protein Biohybrids.

Enzymatic Fisher-Tropsch (FT) process catalyzed by vanadium (V)-nitrogenase can convert carbon monoxide (CO) to longer-chain hydrocarbons (>C2) under ambient conditions, although this process requires high-cost reducing agent(s) and/or the ATP-dependent reductase as electron and energy sources. Using visible light-activated CdS@ZnS (CZS) core-shell quantum dots (QDs) as alternative reducing equivalent for the catalytic component (VFe protein) of V-nitrogenase, we first report a CZS : VFe biohybrid system that enables effective photo-enzymatic C-C coupling reactions, hydrogenating CO into hydro

Substrate specificity and evolutionary implications of a NifDK enzyme carrying NifB-co at its active site.

The in vitro reconstitution of molybdenum nitrogenase was manipulated to generate a chimeric enzyme in which the active site iron-molybdenum cofactor (FeMo-co) is replaced by NifB-co. The NifDK/NifB-co enzyme was unable to reduce N(2) to NH(3), while exhibiting residual C(2)H(4) and considerable H(2) production activities. Production of H(2) by NifDK/NifB-co was stimulated by N(2) and was dependent on NifH and ATP hydrolysis. Thus, NifDK/NifB-co is a useful tool to gain insights into the catalytic mechanism of nitrogenase. Furthermore, phylogenetic analysis of D and K homologs indicates that s

Genome-wide identification of MtTCTPs and functional analysis of MtTCTP3 as a positive regulator of nodulation in Medicago truncatula.

The Translationally Controlled Tumor Protein (TCTP) is a conserved eukaryotic regulator of cell proliferation and stress responses, but its function in legume-rhizobium symbiosis remains unclear. Here, we identified five MtTCTP genes in the model legume Medicago truncatula and characterized the role of MtTCTP3 in symbiotic nodulation. MtTCTP3 is highly expressed in root nodules, particularly in the nodule apical meristem. Overexpression of MtTCTP3 significantly enhanced nodule formation, nitrogenase activity, and biomass accumulation, while silencing of MtTCTP3 caused reduced nodule number and

Rhizobia block Cd absorption in peanut via rhizosphere microbiome assembly and N-regulated host defense.

Cadmium (Cd) contamination severely threatens peanut (Arachis hypogaea L.) production and symbiotic nitrogen fixation (SNF). Although rhizobia inoculation can alleviate heavy metal toxicity in plants, how nitrate (NO3-) regulates Cd translocation and the defense of the "rhizobia-root-nodule" system remains unclear. Here, we investigated the mechanisms by which the peanut rhizobium, Rhizobium sp. HM13, mitigates Cd toxicity and sustains SNF under varying NO3- levels (N- and N+) via field and pot trials. Field trials showed that HM13 reduced rhizosphere bioavailable Cd and decreased seed Cd accu

Simultaneous bioprecipitation of cadmium to cadmium sulfide nanoparticles and nitrogen fixation by Rhodopseudomonas palustris TN110.

This study investigated the abilities of a purple non-sulfur bacterium, Rhodopseudomonas palustris TN110 to bioremediate cadmium through the biosynthesis of CdS nanoparticles and to fix nitrogen simultaneously. Under microaerobic-light conditions, R. palustris TN110 synthesized CdS nanoparticles. The produced CdS nanoparticles had a spherical shape and an average size of 4.85 nm. The Fourier transform infrared spectrum of the nanoparticles reveals the carbonyl groups, bending vibrations of the amide I and II bands of proteins, and CN stretching vibrations of aromatic and aliphatic amines. New insights on glyphosate mode of action in nodular metabolism: Role of shikimate accumulation.

The short-term effects of the herbicide glyphosate (1.25-10 mM) on the growth, nitrogen fixation, carbohydrate metabolism, and shikimate pathway were investigated in leaves and nodules of nodulated lupine plants. All glyphosate treatments decreased nitrogenase activity rapidly (24 h) after application, even at the lowest and sublethal dose used (1.25 mM). This early effect on nitrogenase could not be related to either damage to nitrogenase components (I and II) or limitation of carbohydrates supplied by the host plant. In fact, further exposure to increasing glyphosate concentrations (5 mM) an

The effects of pesticides on bacterial nitrogen fixers in peanut-growing area.

In the peanut production, the applications of herbicides and fungicides are a common practice. In this work, studies done under field conditions demonstrated that pesticides affected negatively the number and nitrogenase activity of diazotrophic populations of soil. Agrochemical effects were not transient, since these parameters were not recovered to pre-treatment levels even 1 year after pesticides application. Results obtained from greenhouse experiments revealed that the addition of herbicide or fungicides diminished the free-living diazotrophs number reaching levels found in soil amended w

Regulatory mechanisms of CH4 : air volume ratios on metabolic flux partitioning in methane-oxidizing bacteria and their impact on single cell protein biosynthetic efficiency.

INTRODUCTION: C1 gas bioconversion for single-cell protein (SCP) production offers dual environmental benefits by mitigating greenhouse gases and generating protein resources. This study systematically determined optimal methane-to-air ratios (CH4:air, v/v) for enhancing methane-oxidizing bacteria (MOB) growth and SCP yield under three distinct nitrogen assimilation modes: nitrate-driven pMMO expression, ammonium-driven sMMO expression, and nitrogen-fixing sMMO expression. METHODS: Experiments were conducted under three nitrogen assimilation regimes: nitrate-fed pMMO expression, ammonium-fed

Effects of the co-application of biochar and trace elements on cadmium passivation and nitrogen biogeochemical processes in contaminated soil cultivated with soybean.

Cadmium (Cd) contamination of soil poses food safety concerns through its bioaccumulation and by disrupting biological nitrogen (N) fixation; therefore, reducing productivity and altering microbial community structure. This study evaluated the co-application of biochar (BC) with molybdenum (Mo), iron (Fe), or sulfur (S) to regulate Cd accumulation, N fixation, nitrification, rhizosphere microbial communities, and plant physiological responses in Cd-contaminated soybean cultivation. The effects of BC co-application differed depending on the trace element supplied. Indeed, BC-Mo treatment increa

Mechanistic Insights into Dinitrogen Reduction to Ammonia in Light-Controlled Nanocrystal:Nitrogenase Complexes.

Developing systems that can efficiently capture photon energy and convert this energy into fuels and chemicals requires understanding how to assemble molecular components with diverse functions into complete systems possessing selectivity and efficiency in directing charge carriers to catalytic reactions. There are many challenges to achieving this goal. One promising approach is the development of hybrid systems that combine semiconductor nanocrystals (NCs) for light capture and enzymes as efficient catalysts. Such biohybrid systems capitalize on the tunable electronic and optical properties

Impact of chromium and lead on Nostoc muscorum: regulation of toxicity by ascorbic acid, glutathione, and sulfur-containing amino acids.

Nostoc muscorum can be protected from Cr and Pb toxicity by supplementing 10 micrograms/ml of ascorbic acid, reduced glutathione, and 5 micrograms/ml of sulfur-containing amino acids (L-methionine and L-cysteine). Appreciable stimulation of different parameters of Nostoc by ascorbic acid and glutathione was noticed in the untreated culture. These substances when used in the presence of metals were found to restore ionic loss, growth, 14CO2 uptake, heterocyst production, and nitrogenase activity thus acting as reducing agents. Sulfur-containing amino acids (at 5 micrograms/ml) exert little stim

Introduction: Transition Metals and Sulfur.

The number of transition metal ions which are essential to life - also often called trace elements - increased steadily over the years. In parallel, the list of biological functions in which transition metals are involved, has grown, and is still growing tremendously. Significant progress has been made in understanding the chemistry operating at the biological sites where metal ions have been discovered. Early on, based on the application of physical, chemical, and biological techniques, it became likely that numerous of these metal centers carry sulfur ligands in their coordination sphere, su
