Assaying Nociception Behaviors in Drosophila Larvae During Parasitoid Wasp Attacks.

Nociception in fruit fly (Drosophila melanogaster) larvae is characterized by a stereotyped escape behavior. When a larva encounters a noxious (potentially harmful) stimulus, it responds by curving its body into a c-shape and rolling in a corkscrew-like manner around its long-body axis. This rolling behavior may serve to quickly remove the larva from the source of the noxious stimulus, and is particularly adaptive to escape from a common natural predator of fruit fly larvae: parasitoid wasps (Leptopilina boulardi). L. boulardi completes its life cycle by using fruit fly larvae as hosts for its

Cortical astrocyte activation triggers meningeal nociception and migraine-like pain.

Cortical astrocytes were implicated in migraine attacks involving cortical spreading depolarizations and related aura symptoms. However, whether the activation of cortical astrocytes is sufficient to generate migraine headaches is unknown. We investigated the role of cortical astrocytes in migraine pain triggering using a DREADD-based chemogenetic approach to activate visual cortex astrocytes, assessing its effect on the responses of meningeal nociceptors, the development of migraine-like behaviors, and their dependence on peripheral CGRP signaling. In a rat model, activating cortical astrocyt

Self-assembled innervated vasculature-on-a-chip to study nociception.

Nociceptor sensory neurons play a key role in eliciting pain. An active crosstalk between nociceptor neurons and the vascular system at the molecular and cellular level is required to sense and respond to noxious stimuli. Besides nociception, interaction between nociceptor neurons and vasculature also contributes to neurogenesis and angiogenesis. In vitromodels of innervated vasculature can greatly help delineate these roles while facilitating disease modeling and drug screening. Herein, we report the development of a microfluidic-assisted tissue model of nociception in the presence of microvas

Neuropathic corneal pain and dry eye: a continuum of nociception.

Throughout the body, damage to peripheral nerves normally involved in nociception may produce a constellation of symptoms-including irritation, itchiness and pain. The neurobiological processes involved in corneal symptoms of dry eye (DE) and neuropathic corneal pain (NCP) have not been clearly considered in terms of nociceptive processing. The conventional underlying presumption is that a labelled line principle is responsible; that these distinct perceptions are hard coded by primary afferent inputs to the central nervous system. This presumption oversimplifies the neurobiological mechanisms

Early Life Nociception is Influenced by Peripheral Growth Hormone Signaling.

A number of cellular systems work in concert to modulate nociceptive processing in the periphery, but the mechanisms that regulate neonatal nociception may be distinct compared with adults. Our previous work indicated a relationship between neonatal hypersensitivity and growth hormone (GH) signaling. Here, we explored the peripheral mechanisms by which GH modulated neonatal nociception under normal and injury conditions (incision) in male and female mice. We found that GH receptor (GHr) signaling in primary afferents maintains a tonic inhibition of peripheral hypersensitivity. After injury, a

CGRPα within the Trpv1-Cre population contributes to visceral nociception.

The role of calcitonin gene-related peptide (CGRP) in visceral and somatic nociception is incompletely understood. CGRPα is highly expressed in sensory neurons of dorsal root ganglia and particularly in neurons that also express the transient receptor potential cation channel subfamily V member 1 (Trpv1). Therefore, we investigated changes in visceral and somatic nociception following deletion of CGRPα from the Trpv1-Cre population using the Cre/lox system. In control mice, acetic acid injection (0.6%, ip) caused significant immobility (time stationary), an established indicator of visceral pa

Serotonergic mechanisms of trigeminal meningeal nociception: Implications for migraine pain.

Serotonergic mechanisms play a central role in migraine pathology. However, the region-specific effects of serotonin (5-HT) mediated via multiple types of receptors in the nociceptive system are poorly understood. Using extracellular and patch-clamp recordings, we studied the action of 5-HT on the excitability of peripheral and central terminals of trigeminal afferents. 5-HT evoked long-lasting TTX-sensitive firing in the peripheral terminals of meningeal afferents, the origin site of migraine pain. Cluster analysis revealed that in majority of nociceptive fibers 5-HT induced either transient

Swimming training and functional assessment of sciatic nociception in rats.

INTRODUCTION: The protective effect of exercise on individuals with peripheral neuropathy is controversial. OBJECTIVE: To assess the influence of physical training of moderate and progressive intensity on nociception of rats in an experimental sciatica model. MATERIALS AND METHODS: The study assessed 18 rats divided into the following three groups: control group (CG); low-intensity swimming group (LISG); progressive-time swimming group (PTSG). To meet the physical training protocol, the animals were placed in a water tank for six weeks, three alternate days per week. In LISG group, the anima

The influence of gender and sex steroids on craniofacial nociception.

Several pain conditions localized to the craniofacial region show a remarkable sex-related difference in their prevalence. These conditions include temporomandibular disorders and burning mouth syndrome as well as tension-type, migraine, and cluster headaches. The mechanisms that underlie sex-related differences in the prevalence of these craniofacial pain conditions remain obscure and likely involve both physiological and psychosocial factors. In terms of physiological factors relevant to the development of headache, direct evidence of sex-related differences in the properties of dural affere

Ontogeny of mu opioid agonist anti-nociception in postnatal rats.

Mu opiate agonists morphine, fentanyl and meperidine are administered short-term to pediatric patients, from the neonatal period through adolescence. However, there has been no assessment of the effect of age on the analgesic efficacy or the concentration-response relationship for these opioids in human pediatric patients. Few studies in animals have correlated opioid anti-nociception and tissue levels of these opioids commonly administered to pediatric patients. The present study was conducted to examined the role of age on opioid anti-nociceptive potency and efficacy and brain and plasma opi

[Clinical exploration of nociception with the use of reflexologic techniques].

In the first part of this report a methodology is described which allows an objective and specific exploration of experimental pain in man by using some electrophysiological features of cutaneous reflexes. This method can be summarized as follows: in normal and trained volunteers, we studied simultaneously the recruitment curves of the nociceptive flexion reflex of a knee-flexor muscle (biceps femoris muscle) and that of pain sensation elicited by electrical stimulation of the ipsilateral sural nerve at the ankle. In this procedure, we found that the reflex threshold (Tr) was closely related t

Daily variation of nociceptive responses in Drosophila.

The circadian clock regulates a wide range of physiological and behavioral rhythms. Daily variation in nociceptive responsiveness to harmful stimuli has been demonstrated in mammalian models; however, whether such variation is present in non-mammalian species including insects has remained unclear. Here, we found that behavioral responses to noxious heat stimuli in the fruit fly Drosophila melanogaster exhibit significant variation throughout the day. In contrast, null mutants for the clock genes per and tim failed to exhibit daily variation in nociceptive responses. Consistently, we found dai

Nrxn3 reduces myofascial nociceptive pain.

Neurexin 3 (Nrxn3) has a role in neuronal signaling. Previous reports indicated that reducing Nrxn3 expression in the central amygdala increased orofacial neuropathic pain. A common temporomandibular disorder is myofascial pain. Thus, we hypothesized that Nrxn3 would reduce myofascial hypersensitivity. To test this hypothesis Nrxn3 shRNA was infused into the central amygdala of male rats. Then a ligature of the tendon attachment of the anterior superficial portion of the masseter muscle was performed to induce inflammatory orofacial pain. Dark phase meal duration was measured continuously, and

A nociceptive signaling role for neuromedin B.

Here we used an array-based differential screen to uncover the expression of the neuropeptide neuromedin B (NMB) in the trigeminal ganglia of mice. Double-labeling experiments reveal NMB is expressed in a subset of sensory neurons that colabel with calcitonin gene-related peptide and TRPV1 suggestive of a role for NMB in nociception. Indeed, administration of NMB antagonist greatly attenuates edema and nerve sensitization following stimulation of peripheral nerves with mustard oil, demonstrating that NMB contributes to neurogenic inflammation. Moreover, direct injection of NMB causes local swe

Nociceptive and inflammatory effects of subcutaneous TNFalpha.

Tumor necrosis factor alpha (TNF) is a potent pro-inflammatory cytokine that produces pain and hyperalgesia following injection. Its algesic effects are due to sensitizing actions on nociceptive primary afferents and to the upregulation of other pro-inflammatory and algesic proteins. In anesthetized rats, we investigated the effect of subcutaneously injected TNF on background activity and mechanical sensitivity of C nociceptors of the sural nerve, as well as its effects on cutaneous plasma extravasation. TNF sensitized C nociceptors dose-dependently; the optimal dose (5 ng) lowered threshold i

[Nociceptive modulation during intravenous anesthesia].

The surgical stress can be defined as a complex array of neuroendocrine, haemodynamic, metabolic and inflammatory changes following surgical injury. These responses, when excessive, may contribute to increased postoperative morbidity and mortality. Thus a major endpoint of general anaesthesia is to protect the patient against the noxious stress response components. The different classes of anaesthetic or associated drugs are reviewed in this respect: sedative/hypnotics (barbiturates, etomidate, propofol, benzodiazepines), opioids, alpha-2 adrenergic agonists (clonidine), non-steroidal antiinfl

Expression of nociceptive ligands in canine osteosarcoma.

BACKGROUND: Canine osteosarcoma (OS) is associated with localized pain as a result of tissue injury from tumor infiltration and peritumoral inflammation. Malignant bone pain is caused by stimulation of peripheral pain receptors, termed nociceptors, which reside in the localized tumor microenvironment, including the periosteal and intramedullary bone cavities. Several nociceptive ligands have been determined to participate directly or indirectly in generating bone pain associated with diverse skeletal abnormalities. HYPOTHESIS: Canine OS cells actively produce nociceptive ligands with the capa

Substance P in nociceptive sensory neurons.

Substance P is contained within a subpopulation of nociceptive primary sensory neurons that project to the superficial dorsal horn of the spinal cord. Stimulation of the peripheral processes of primary afferent fibres at intensities that activate A delta and C fibres elicits a pronounced release of substance P from the cat spinal cord in vivo. Experiments with the neurotoxins capsaicin and 5,6-dihydroxytryptamine have shown that substance P release from the spinal cord in vivo derives largely from afferent fibres. Intrathecal perfusion of the cat spinal cord with morphine abolishes the nerve-e

Intraoperative Nociception Monitoring Using the NoL Index: Phase-Specific Assessment of Nociceptive Responses During Spinal Surgery.

Background: Quantifying intraoperative nociceptive responses under general anesthesia remains challenging, particularly during complex procedures such as spinal surgery. The Nociception Level (NoL) index is a multiparametric tool designed to reflect the dynamic balance between nociception and analgesia in anesthetized patients. This study aimed to evaluate NoL fluctuations during predefined phases of spinal surgery and assess their relationship to anesthetic administration. Methods: This prospective observational study enrolled 44 adult patients undergoing lumbar discectomy, laminectomy, or sp
