[mRNA expression of metastasis-suppressor gene nm23 in carcinoma of buccal mucosa. II. Quantitative reverse transcription PCR amplification].

The nm23 gene is a conspicuous metastasis-suppressor gene. The authors detected both nm23-H1 and nm23-H2 mRNA levels in 47 tissues samples of patients with carcinoma of buccal mucosa (CBM) by quantitative reverse transcription PCR amplification. The results showed that expression levels of both nm23-H1 and nm23-H2 mRNA varied in normal buccal mucosa, leukoplakia, adjacent nontumorous mucosa, submandibular gland, CBM and lymph nodes with or without metastasis. The nm23-H1 mRNA expression levels in CBM with lymph nodes metastases were lower than those in CBM without metastases (P < 0.05), while

Isolation and characterization of the human genomic locus coding for the putative metastasis control gene nm23-H1.

Nm23-H1 gene expression is inversely correlated with tumor metastatic potential in certain tumors, including melanomas, breast carcinomas, and hepatocellular carcinomas. Using nm23-H1 c-DNA primer and genomic polymerase chain reaction (PCR) amplification, we purified three PCR fragments (one of 4kb and two of 2 kb) covering the whole human genomic locus of the gene (8.460bp). We recombined the PCR products into pUC18 and produced a restriction map to perform subcloning. Complete sequencing of genomic PCR fragments, including the whole coding region of nm23-H1, revealed that the gene consists o

Isolation and characterization of the promoter region of human nm23-H1, a metastasis suppressor gene.

Allelic deletions in the nm23, a metastasis suppressor gene, are known to occur in neuroblastomas, breast and colorectal carcinomas. Down-regulation of nm23 expression has been reported in various rodent and human tumor cells with high metastasis phenotype. Colorectal tumors showed overexpression of nm23. To elucidate the regulatory mechanisms of nm23, we isolated, cloned and sequenced the presumptive regulatory DNA fragment spanning the 5' region of the human nm23-H1 gene. The region's nucleotide sequence shows the presence of motifs typical for transcriptional elements such as TFIID, AP-1 an

NME4 mediates metabolic reprogramming and promotes nonalcoholic fatty liver disease progression.

Nonalcoholic fatty liver disease (NAFLD) is mainly characterized by excessive fat accumulation in the liver, and it is associated with liver-related complications and adverse systemic diseases. NAFLD has become the most prevalent liver disease; however, effective therapeutic agents for NAFLD are still lacking. We combined clinical data with proteomics and metabolomics data, and found that the mitochondrial nucleoside diphosphate kinase NME4 plays a central role in mitochondrial lipid metabolism. Nme4 is markedly upregulated in mice fed with high-fat diet, and its expression is positively corre

Identification of key metabolic enzymes involved in the activation of obeldesivir and remdesivir to the active triphosphate metabolite.

Obeldesivir (GS-5245, ODV), an orally administered 5'-isobutyryl ester prodrug of GS-441524, has demonstrated anti-SARS-CoV-2 activity in preclinical and clinical settings. ODV is hydrolyzed in plasma to provide high systemic exposures of GS-441524, enabling the efficient intracellular formation of the bioactive 5'-triphosphate GS-443902, an ATP analog inhibiting the SARS-CoV-2 RNA-dependent RNA polymerases. Remdesivir (RDV), the first US FDA-approved antiviral treatment for COVID-19, is also activated to GS-443902. In this paper, we systematically profiled the human enzymes potentially involv

Exploring Lamiaceae diterpenoids as potential multitarget therapeutics for leishmaniasis and chagas disease.

Neglected tropical diseases such as Leishmaniasis and Chagas disease remain critical public health challenges. This study applied ligand-based virtual screening to a dataset of 4,150 secondary metabolites from the Lamiaceae family, aiming to identify multitarget molecules against four Leishmania species (L. infantum, L. donovani, L. amazonensis, and L. braziliensis) and Trypanosoma cruzi forms. Random forest models exhibited high accuracy (over 72%), leading to the identification of 82 molecules with potential multitarget activity across five of six predictive models. Nineteen prioritized mole

[NDPK/nm23 expression and its correlation with lymph node metastasis in human lung cancer].

Using labelled streptavidin-biotin (LSAB) method, we examined the expression of nucleoside diphosphate kinase(NDPK), the product of metastasis suppressor gene nm23, in human lung cancer. Of 88 patients tested, 48 (54.5%) showed positive staining. The positive staining rate was higher in adenocarcinoma (28/42, 66.7%) than in squamous cell carcinoma (20/46, 43.5%; P < 0.05). Higher incidence of positive staining was also found in squamous cell carcinoma without hilar or mediastinal lymph node metastasis (16/27, 59.3%) than in that with hilar or mediastinal lymph node involvement (4/19, 21.1%; P

The NM23-H1/H2 homolog NDK-1 is required for full activation of Ras signaling in C. elegans.

The group I members of the Nm23 (non-metastatic) gene family encode nucleoside diphosphate kinases (NDPKs) that have been implicated in the regulation of cell migration, proliferation and differentiation. Despite their developmental and medical significance, the molecular functions of these NDPKs remain ill defined. To minimize confounding effects of functional compensation between closely related Nm23 family members, we studied ndk-1, the sole Caenorhabditis elegans ortholog of group I NDPKs, and focused on its role in Ras/mitogen-activated protein kinase (MAPK)-mediated signaling events duri

Uridine diphosphate N-acetylglucosamine homeostasis via CgNgk1 kinase modulates chitin levels and cell-wall integrity in Candida glabrata.

Candida glabrata is a clinically significant cause of candidemia, yet how it supplies uridine diphosphate-N-acetylglucosamine (UDP-GlcNAc) for cell-wall chitin synthesis remains unclear. Here, we identify an N-acetylglucosamine (GlcNAc) kinase encoded by CAGL0M00682g and designated CgNgk1. In vitro, purified CgNgk1 preferentially phosphorylated GlcNAc with markedly higher catalytic efficiency than other sugars and accepted multiple nucleoside triphosphates, including ATP, as phosphoryl donors. Site-directed mutagenesis indicated that Asp197 and Lys155 are required for catalysis and ATP binding

Proteomic analysis of skeletal deformity in diploid and triploid rainbow trout (Oncorhynchus mykiss) larvae.

A proteomic screening approach was employed to achieve a better understanding of the changes that occur in protein expression patterns associated with skeletal deformities in both diploid and triploid rainbow trout larvae. Triploidy was induced through the application of heat shock of 28°C for 10min to eggs 10-min post fertilization in an aquarium equipped with a heater. Percentage of skeletal deformity in heat-shocked larvae (2.88±0.30, mean±S.E.) was significantly (P<0.05) greater than that of the diploids (0.55±0.24). At five days after hatching, proteins of normal and deformed specimens of

NME3 promotes clear cell renal cell carcinoma progression and modulates TKI response through MFN1/2-dependent mitochondrial fusion and mitophagy suppression.

Renal cell carcinoma (RCC), particularly the clear cell subtype (ccRCC), is a prevalent malignancy characterized by aggressive progression and heterogeneous therapeutic responses. Although mitochondrial dynamics are increasingly recognized as critical regulators of cancer metabolism and survival, the role of nucleoside diphosphate kinase 3 (NME3) in this process remains poorly understood. In this study, we integrated bioinformatic analyses of public datasets with validation in patient-derived tissues, in vitro functional assays, and in vivo xenograft models to elucidate the role of NME3 in ccR

Dietary Nucleotides as Potential Modulators of Inflammatory and Metabolic Pathways with Implications for Insulin Resistance.

Dietary nucleotides and nucleic-acid-derived compounds are considered bioactive nutrients, but their metabolic relevance remains uncertain. This narrative review, supported by a structured literature search, evaluates intestinal handling and evidence on glucose and lipid metabolism, redox homeostasis, immune function, gut barrier regulation, and microbiota. Direct supplementation studies are distinguished from nucleoside/nucleobase evidence, dietary nucleic acids or purine-rich foods, and endogenous extracellular purinergic signaling. Preclinical studies suggest defined nucleotide preparations

The let-7f-5p-Nme4 pathway mediates tumor necrosis factor α-induced impairment in osteogenesis of bone marrow-derived mesenchymal stem cells.

Although tumor necrosis factor α (TNF-α)-mediated inflammation significantly impacts osteoporosis, the mechanisms underlying the osteogenic differentiation defects of bone marrow-derived mesenchymal stem cells (BM-MSCs) caused by TNF-α remain poorly understood. We found that TNF-α stimulation of murine BM-MSCs significantly upregulated the expression levels of several microRNAs (miRNAs), including let-7f-5p, but this increase was significantly reversed by treatment with the kinase inhibitor BAY 11-7082. To study gain- or loss of function, we transfected cells with an miRNA inhibitor or miRNA m

The distribution of the components of the cyclic GMP cycle in retina.

Frozen sections of retinas from rabbit (mostly rods), ground squirrel (mostly cones), and monkey (mixed rods and cones) were freeze dried, and samples from all the discrete layers analyzed for the enzymes which form cyclic GMP and subsequently convert it back to GTP. The distribution of cyclic GMP was also measured in monkey retina, and the retinal layers of both monkey and rabbit were analyzed for GTP, GTP plus GDP, ATP, ATP plus ADP, and UTP plus CTP. The ratio of guanylates to adenylates was found to be about 1:1 in photoreceptor cell layers, but only 1:4 or 5 in deeper layers. In all speci

nm23-H1 protein immunoreactivity in laryngeal carcinoma.

BACKGROUND: The nm23-H1 gene encodes a 17-kilodalton cytoplasmic and nuclear protein that has recently been shown to be reduced in a number of human carcinomas including breast, colorectal, lung, gallbladder, and biliary tract carcinomas. This study examines the immunohistochemical staining characteristics of the nm23-H1 protein in human laryngeal carcinomas and nonneoplastic laryngeal polyps, and attempts to determine if there is any relationship between reduction of nm23-H1 protein immunoreactivity and prognosis of patients with laryngeal carcinoma. METHODS: Routine streptavidin-biotin immu

On the role of phosphatase in regulation of cardiac L-type calcium current by cyclic GMP.

Does cGMP, via protein kinase G, inhibit cAMP-stimulated Ca(2+) current (I(Ca(L))) in mammalian ventricular myocytes by phosphorylating the calcium channel at a site different from that acted on by cAMP or by dephosphorylating the calcium channel through phosphatase(s)? We tested these possibilities in guinea pig ventricular myocytes superfused with Tyrode's solution (35 degrees C) and dialyzed with adenosine 5'-O-(3-thiotriphosphate) ([ATPgammaS](pip)). ATPgammaS is a kinase substrate but thiophosphorylated proteins are not phosphatase substrates. With 5 mM [ATPgammaS](pip), I(Ca(L)) increase

Metastasis suppressor NME1 in exosomes or liposomes conveys motility and migration inhibition in breast cancer model systems.

Tumor-derived exosomes have documented roles in accelerating the initiation and outgrowth of metastases, as well as in therapy resistance. Little information supports the converse, that exosomes or similar vesicles can suppress metastasis. We investigated the NME1 (Nm23-H1) metastasis suppressor as a candidate for metastasis suppression by extracellular vesicles. Exosomes derived from two cancer cell lines (MDA-MB-231T and MDA-MB-435), when transfected with the NME1 (Nm23-H1) metastasis suppressor, secreted exosomes with NME1 as the predominant constituent. These exosomes entered recipient tum

Label-Free Mass Spectrometry Proteomics Reveals Different Pathways Modulated in THP-1 Cells Infected with Therapeutic Failure and Drug Resistance Leishmania infantum Clinical Isolates.

As the world is facing increasing difficulties to treat leishmaniasis with current therapies, deeper investigation into the molecular mechanisms responsible for both drug resistance and treatment failure (TF) is essential in drug discovery and development. So far, few available drugs cause severe side effects and have developed several resistance mechanisms. Drug resistance and TF parasite strains from clinical isolates may have acquired altered expression of proteins that characterize specific mechanisms leading to therapy inefficacy. This work aims to identify the biochemical pathways of THP
