TDP-43 pathology, cognitive decline, and dementia in old age.

IMPORTANCE: Cognitive decline is a leading cause of disability and death in old age but its neurobiological bases are not well understood. OBJECTIVE: To test the hypothesis that transactive response DNA-binding protein 43 (TDP-43) is related to late-life cognitive decline. DESIGN, SETTING, AND PARTICIPANTS: Longitudinal clinical-pathologic cohort study involving more than 40 Catholic groups across the United States. A total of 130 older Catholic nuns, priests, and monks underwent annual clinical evaluations, including detailed cognitive testing, for a mean of 10.1 years prior to death. On ne

What the world's religions teach, applied to vaccines and immune globulins.

For millennia, humans have sought and found purpose, solace, values, understanding, and fellowship in religious practices. Buddhist nuns performed variolation against smallpox over 1000 years ago. Since Jenner developed vaccination against smallpox in 1796, some people have objected to and declined vaccination, citing various religious reasons. This paper reviews the scriptural, canonical basis for such interpretations, as well as passages that support immunization. Populous faith traditions are considered, including Hinduism, Buddhism, Jainism, Judaism, Christianity, and Islam. Subjects of co

Activation and control of pathogenic T cells in OSP/claudin-11-induced EAE in SJL/J mice are dominated by their focused recognition of a single epitopic residue (OSP58M).

Oligodendrocyte-specific protein (OSP)/claudin-11 has been recently implicated in multiple sclerosis pathophysiology. Yet, the pathogenic autoimmunity against OSP has been poorly investigated. We previously showed that OSP-induced experimental autoimmune encephalomyelitis (EAE) and optic neuritis in SJL/J mice are primarily associated with CD4+ T cells reactive against OSP55-80. Dissecting the fine epitope specificity to the level of epitopic residues recognized by OSP-specific encephalitogenic T cells revealed their focused recognition of OSP58M. Accordingly, OSP58M predicted by computer mode

Long-term survival of chosen atrial-based pacing modalities.

Atrial-based cardiac pacing modalities were chosen in 341 of 684 (50%) patients selected for permanent cardiac pacing at the Grey Nuns Community Health Center between 1989 and 1995. There were 183 males and 158 females. Mean age was 70 years (range 8-97 years). The indications for atrial-based pacing were: advanced atrioventricular (AV) block (n = 166) 49%; sick sinus syndrome (n = 153) 45%; hypersensitive carotid sinus syndrome (n = 11) 3%; neurocardiac syncope (n = 10) 2.9%; and hypertrophic cardiomyopathy (n = 1) 0.3%. Forty-eight patients had a known history of paroxysmal atrial arrhythmia

Determinants of oxidative stress in orthodox Christian women: A comparative analysis of monastic and non-monastic lifestyles.

BACKGROUND: Oxidative stress has been implicated in aging and metabolic dysfunction, modulated by diet, lifestyle, and body composition. While structured dietary practices such as intermittent fasting and plant-based regimens have demonstrated antioxidative benefits, the biological impact of long-term religious fasting remains underexplored. Orthodox Christian fasting, characterized by periodic abstention from animal products and alignment with circadian rhythms, offers a unique naturalistic model for examining redox adaptation in humans. OBJECTIVES: To identify the physiological and biologic

Multi-omic subtypes of Alzheimer's dementia are differentially associated with psychological traits.

IMPORTANCE: Psychological traits reflecting neuroticism, depressive symptoms, loneliness, and purpose in life are risk factors of AD dementia; however, the underlying biologic mechanisms of these associations remain largely unknown. OBJECTIVE: To examine whether one or more multi-omic brain molecular subtypes of AD is associated with neuroticism, depressive symptoms, loneliness, and/or purpose in life. DESIGN: Two cohort-based studies; Religious Orders Study (ROS) and Rush Memory and Aging Project (MAP), both ongoing longitudinal clinical pathological studies that began enrollment in 1994 an

Urinary sodium excretion and blood pressure in chronic psychiatric in-patients.

Studies of populations or communities with no rise in blood pressure (BP) with advancing age and low prevalence of hypertension, may provide aetiological clues on the cause of hypertension. Within westernised societies, low blood pressures have been reported amongst chronic psychiatric in-patients and closed order secluded nuns. To investigate factors associated with BP in chronic psychiatric in-patients, we surveyed the BP and lifestyle factors in 89 such subjects in low security wards in three psychiatric hospitals. The average age of examines was 48.1 years (s.d. 15.8) and the patients had

A DESCRIPTION OF ADDISON'S DISEASE, AND ITS NEUROPSYCHIATRIC MANIFESTATIONS COMPARING THE DISEASE AS IT IS NOW TO THE DISEASE AS EXPERIENCED BY SAINT ELIZABETH OF THE TRINITY IN 1906.

We describe Addison's disease, which is the disease caused by Adrenal Gland Insufficiency. Addison's disease is an uncommon endocrine disorder characterised by inadequate production of hormones, predominantly aldosterone and cortisol, by the adrenal glands. This condition occurs due to damage to the adrenal cortex, the region responsible for hormone synthesis. Clinical manifestations of Addison's disease are often insidious and nonspecific, including symptoms such as myasthenia, persistent fatigue, unintentional weight loss, hypotension, and hyperpigmentation of the skin, especially in areas s

Arteriolosclerosis that affects multiple brain regions is linked to hippocampal sclerosis of ageing.

Hippocampal sclerosis of ageing is a prevalent brain disease that afflicts older persons and has been linked with cerebrovascular pathology. Arteriolosclerosis is a subtype of cerebrovascular pathology characterized by concentrically thickened arterioles. Here we report data from multiple large autopsy series (University of Kentucky Alzheimer's Disease Centre, Nun Study, and National Alzheimer's Coordinating Centre) showing a specific association between hippocampal sclerosis of ageing pathology and arteriolosclerosis. The present analyses incorporate 226 cases of autopsy-proven hippocampal sc

The Neurostimulation of the Brain in Depression Trial: Protocol for a Randomized Controlled Trial of Transcranial Direct Current Stimulation in Treatment-Resistant Depression.

BACKGROUND: Major depressive disorder (MDD) is the second highest cause of disability worldwide. Standard treatments for MDD include medicine and talk therapy; however, approximately 1 in 5 Canadians fail to respond to these approaches and must consider alternatives. Transcranial direct current stimulation (tDCS) is a safe, noninvasive method that uses electrical stimulation to change the activation pattern of different brain regions. By targeting those regions known to be affected in MDD, tDCS may be useful in ameliorating treatment-resistant depression. OBJECTIVE: The objective of the Neuro

[Assessment of 24 years of infant growth monitoring in a rural area of Senegal (1969-1992)].

UNLABELLED: Undernutrition in young children in developing countries is associated with an increased risk of death. But in several studies, a decrease in mortality was not associated with any decrease in the prevalence of undernutrition. STUDY AREA: A rural population of Casamance (Senegal) has been under yearly demographic surveillance by The French National Institute of Demographic Studies (INED) since 1985. Between 1960-1964 and 1990-1994, under-5 and child (1-4 years) mortality rates dropped from 312 to 127 and from 201 to 68 per 1,000, respectively. Since 1961, French Catholic nuns who a

CAR T cells and T cell engagers for autoimmunity-lessons from hematology.

Autoimmune diseases are driven by dysregulated immune responses against self-antigens, resulting in chronic inflammation and progressive tissue damage. Chimeric antigen receptor (CAR) T cell therapy and T cell engagers, initially developed for the treatment of hematologic malignancies, have demonstrated that they can mediate profound and sustained depletion of pathogenic B cells, leading to long-term remissions in otherwise refractory diseases. In this review, we explore key insights gained from hematologic applications of CAR T cells and T cell engagers, and discuss how these lessons can guid
