Opposite changes in adenosine A1 and A2A receptor mRNA in the rat following sleep deprivation.

Extracellular levels of adenosine increase in basal forebrain following prolonged wakefulness. Moreover, perfusion of adenosine into basal forebrain increases sleep. In this study we have examined the adenosine receptor subtypes, A1 and A2A, for changes in the levels of mRNA using RT-PCR and in situ hybridization and the receptor ligand binding efficiency using autoradiography following 3 and 6 h of sleep deprivation. We observed that A1 receptor mRNA levels increased in basal forebrain with no changes in other forebrain areas examined. A1 receptor binding was not affected. A2A receptor mRNA a

Ascending projections of the mammillary region in the pigeon: emphasis on telencephalic connections.

The ascending projections of the mammillary region of the hypothalamus and adjacent posterolateral hypothalamus were investigated by autoradiographic and horseradish peroxidase histochemical techniques. Analysis of the anterograde data revealed that the main contingent of mammillary fibers ascends in the medial forebrain bundle (MFB) in the lateral hypothalamic area. These fibers distribute to a number of telencephalic regions via three pathways. (1) Fibers course dorsomedially into the medial and lateral parts of the septum and continue into the hippocampus with a dense terminal field in the

The distribution of dopamine immunoreactivity in the forebrain and midbrain of the lizard Gekko gecko: an immunohistochemical study with antibodies against dopamine.

The distribution of dopamine (DA) immunoreactivity in the forebrain and the midbrain of the lizard Gekko gecko was studied by using recently developed antibodies against DA. Dopamine-containing cells were found around the glomeruli of the olfactory bulb, in several parts of the periventricular hypothalamic nucleus, in the periventricular organ, the ependymal wall of the infundibular recess, the lateral hypothalamic area and the pretectal posterodorsal nucleus of the diencephalon, and in the ventral tegmental area, the substantia nigra, and the presumed reptilian equivalent of the mammalian A8

Quantitative studies on the supraoptic nucleus in the rat. II. Afferent fiber connections.

A quantitative electron microscopic study of synaptic terminal degeneration was performed in the supraoptic nucleus (NSO) after a variety of major transections or ablations, destroying or interrupting in different combinations the afferent pathways known from earlier and own light microscopic degeneration studies. Solutions of a set of equations, expressing the percentage degenerations in synaptic profiles after different combinations in which the several pathways are interrupted by the various interferences, enabled the authors to give the following percentage numbers for afferent synapses fr

Effects of diazepam and ketamine administered individually or in combination on regional rates of glucose utilization in rat brain.

The effects of diazepam, which acts at GABAA receptors to enhance the effects of GABA, and ketamine, a non-competitive N-methyl-D-aspartate receptor antagonist, on local rates of cerebral glucose utilization (ICMRglc) were examined in unrestrained rats. Four groups were studied: vehicle-injected controls; and ketamine-treated, diazepam-treated and combined ketamine- and diazepam-treated animals. Ketamine alone produced a heterogeneous pattern of changes in ICMRglc (e.g. significant increases in the corpus callosum, olfactory tubercle and the entire Papez circuit, in addition to other limbic ar

Selective MPP+ uptake into synaptic dopamine vesicles: possible involvement in MPTP neurotoxicity.

1. In the present study we provide evidence for a saturable, Mg2+/ATP- and temperature-dependent, tetrabenazine-, dopamine-, and amphetamine-sensitive uptake of 1-methyl-4-phenylpyridinium ion (MPP+) in synaptic vesicles from mouse striatum. 2. Similarity in the properties of the vesicular uptake suggests that in the striatum dopamine and MPP+ share the vesicular carrier. 3. The presence of MPP+ vesicular uptake in dopamine-rich regions such as striatum, olfactory, tubercles and hypothalamus, as well as its absence in cerebellum, cortex and pons-medulla, suggest that monoamine vesicular carrie

Short-term effects of ether, equithesin and droperidol/fentanyl on catecholamine and indolamine metabolism in the brain of the rat.

Levels of biogenic amines and their major metabolites were measured in the frontal cortex, dorsal striatum (caudate nucleus) and ventral striatum (nucleus accumbens and olfactory tubercle) of rats which had received anaesthetic or analgesic drugs or no treatment, shortly before decapitation. Apart from a reduced level of homovanillic acid in the ventral striatum after ether, no differences were observed between ether- or Equithesin-treated and untreated animals in the levels of dopamine, dihydroxyphenylacetic acid, homovanillic acid, 3-methoxytyramine, noradrenaline, serotonin or 5-hydroxyindo

The distribution of GAP-43 immunoreactivity in the central nervous system of adult opossums (Didelphis virginiana) with notes on their development.

The distribution of the growth associated protein GAP-43 has been described in the brain and spinal cord of rats and other placental mammals but not marsupials. In order to provide such information, we employed a monoclonal antibody to immunostain for GAP-43 in the central nervous system of adult and developing opossums, Didelphis virginiana. The GAP-43 immunoreactivity was widely distributed in the brain of adult opossums, but it was particularly dense within specific layers of the olfactory bulb and hippocampal formation, layer I of the cerebral cortex, the bed nucleus of the stria terminali

Effects of in utero ethanol exposure on the development of LHRH neurons in the mouse.

Prenatal ethanol exposure has been shown to result in craniofacial malformations as well as alterations of central nervous system morphology and function. Previous studies have demonstrated that acute ethanol exposure on gestational day 7 in the developing C57BL/6J mouse resulted in craniofacial abnormalities similar to that of children with fetal alcohol syndrome. We investigated the effect of ethanol on the migration and number of immunoreactive LHRH (irLHRH) neurons in this strain of mouse. Pregnant mice were intubated with 2 doses of a 25% solution of ethanol 4 h apart on gestational day 7

Molecular cloning and expression of a D1 dopamine receptor linked to adenylyl cyclase activation.

In order to clone the D1 dopamine receptor linked to adenylyl cyclase activation, the polymerase chain reaction was used with highly degenerate primers to selectively amplify a cDNA sequence from NS20Y neuroblastoma cell mRNA. This amplification produced a cDNA fragment exhibiting considerable sequence homology to guanine nucleotide-binding (G)-protein-coupled receptors that have been cloned previously. To characterize this cDNA further, a full-length clone was isolated from a rat striatal library by using the cDNA fragment as a probe. Sequence analysis of this cDNA clone indicated that it is

Early life stress exacerbates behavioural and neuronal alterations in adolescent male mice lacking methyl-CpG binding protein 2 (Mecp2).

The methyl-CpG binding protein 2 gene (MECP2) encodes an epigenetic transcriptional regulator implicated in neuronal plasticity. Loss-of-function mutations in this gene are the primary cause of Rett syndrome and, to a lesser degree, of other neurodevelopmental disorders. Recently, we demonstrated that both Mecp2 haploinsuficiency and mild early life stress decrease anxiety-like behaviours and neuronal activation in brain areas controlling these responses in adolescent female mice. Here, we extend this work to males by using Mecp2-null and wild type adolescent mice subjected to maternal separat

Immunocytochemical localization and description of neurons expressing serotonin2 receptors in the rat brain.

Serotonin2 receptors have been implicated in a variety of behavioral and physiological processes, as well as a number of neuropsychiatric disorders. To specify the brain regions and specific cell types possessing serotonin2 receptors, we conducted an immunocytochemical study of the rat brain using a polyclonal serotonin2 receptor antibody. Perfusion-fixed rat brain sections were processed for immunocytochemistry and reactivity was visualized using an immunoperoxidase reaction. Numerous small, round neurons were heavily labeled in the granular and periglomerular regions of the olfactory bulb. H

[Pineal gland glutathione peroxidase activity in rats and its age-associated change].

Glutathione peroxidase activity has been studied in the pineal gland (epiphysis) of young and aging female Wistar rats (2-4 and 17-19 month old). For comparison the same activity was studied in the pyramids of medulla oblongata and in the olfactory tubercle. These two brain structures represent white and gray matter respectively. The determination of the activity was performed with H2O2 as a substrate and with 5,5'-dithio-bis-(2-nitrobenzoic acid) for estimation of the decrease of restored form of glutathione concentration. The glutathione peroxidase activity was higher in the pineal gland tha

The development of the human brain, including the longitudinal zoning in the diencephalon at stage 15.

Twenty-six embryos (6-11 mm) of stage 15 (approximately 33 days) were studied in detail and graphic reconstructions of three of them were prepared. Characteristic features of this stage include closed lens vesicles, presence of nasal pits, and retinal pigment. The neuromeric pattern is still visible. Each cerebral hemisphere is limited by the torus hemisphericus internally and by the di-telencephalic sulcus externally. The medial (diencephalic) eminence of the basal nuclei (previously misinterpreted by others as the lateral) had appeared in stage 14, and the lateral eminence, which is telencep

Development of a modified weight-drop apparatus for closed-skull, repetitive mild traumatic brain injuries in a mouse model.

Repetitive mild traumatic brain injury (rmTBI) is a major contributor to long-term neurological dysfunction, yet many preclinical models lack precise control and quantification of biomechanical forces across impacts. We developed a reproducible, closed-skull mouse model of rmTBI using a custom-built weight-drop apparatus featuring a solenoid-based rebound arrest system, integrated high-speed videography, and accelerometry to track head kinematics during impact. Adult male and female mice received either a single impact or nine daily impacts. Linear and angular acceleration data were analyzed a

Oxytocin and vasopressin immunoreactivity within the forebrain and limbic-related areas in the mustached bat, Pteronotus parnellii.

The nonapeptides, oxytocin and arginine vasopressin, play an important role in mammalian social and reproductive behavior. Using immunocytochemical procedures, we identified oxytocin-immunoreactive neurons in the frontal and auditory cortices, limbic areas such as the medial septal nucleus, horizontal limb of the diagonal band and the amygdala. Only arginine vasopressin neurons were present in the bed nucleus of the stria terminalis. In limbic-related areas, the hypothalamic paraventricular and supraoptic nuclei and the nucleus centralis contained both oxytocin and arginine vasopressin neurons

An immunohistochemical study of the telencephalon of the senegal bichir (Polypterus senegalus).

The telencephalon in ray-finned fish (actinopterygians) is everted, in contrast to the evaginated telencephalic hemispheres in all other vertebrates. In the more derived ray-finned fish, the teleosts, proliferation of neurons and their migration from the ependymal zone of the pallium renders comparisons between telencephalic cell groups of the teleosts and members of other vertebrate groups extremely difficult. The telencephalon of Polypterus (a primitive living ray-finned fish), although everted, is cytoarchitecturally much simpler than that of teleosts. We have thus applied immunohistochemic

VGF expression in the brain.

VGF is a neuronal polypeptide first identified as a cDNA clone in a gene library from nerve growth factor-stimulated PC12 cells. In the present paper, the expression of VGF is examined for the first time throughout the adult rat central nervous system with immunocytochemistry and Northern blot analysis. VGF RNA was found in all brain regions studied, including hypothalamus, hippocampus, cerebellum, olfactory bulb, and cortex. In contrast to the relatively strong immunostaining of hypothalamic neurons, the level of VGF RNA expression in the hypothalamus was relatively low in comparison with oth
