Targeting CXCL12/CXCR4 signaling with oncolytic virotherapy disrupts tumor vasculature and inhibits breast cancer metastases.

Oncolytic viruses hold promise for the treatment of cancer, but their interaction with the tumor microenvironment needs to be elucidated for optimal tumor cell killing. Because the CXCR4 receptor for the stromal cell-derived factor-1 (SDF-1/CXCL12) chemokine is one of the key stimuli involved in signaling interactions between tumor cells and their stromal microenvironment, we used oncolytic virotherapy with a CXCR4 antagonist to target the CXCL12/CXCR4 signaling axis in a triple-negative 4T1 breast carcinoma in syngeneic mice. We show here that CXCR4 antagonist expression from an oncolytic vac

Varicella zoster virus infection of malignant glioma cell cultures: a new candidate for oncolytic virotherapy?

BACKGROUND: Glioblastoma multiforme is a highly aggressive tumor with a median survival of 14 months despite all standard therapies. Focusing on alternative treatment strategies, we evaluated the oncolytic potential of varicella zoster virus (VZV) in malignant glioma cell cultures. MATERIALS AND METHODS: Replication of wildtype and mutant VZV was comparatively analyzed in glioma cell lines (U87, U251 and U373) and in primary malignant glioma cells (n=10) in vitro by infectious foci assay, immunofluorescence microscopy and western blot analysis. Additionally, the tumor-targeting potential of V

A Decade of Oncolytic Virotherapy in Pediatric Cancers: A Systematic Review of Safety, Immune Awakening, and Emerging Efficacy.

Oncolytic virotherapy employs genetically modified viruses to selectively lyse tumor cells while activating antitumor immune responses. In pediatric oncology, where outcomes for high-grade gliomas and refractory solid tumors remain poor, oncolytic viruses represent a promising therapeutic strategy. A systematic review was conducted in accordance with Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) 2020 guidelines, including searches of PubMed, Embase, Scopus, Web of Science, and ClinicalTrials.gov for studies published between 2015 and 2025 that evaluated oncolytic

An in silico approach uncovering the competency of oncolytic human adenovirus 52 for targeted breast cancer virotherapy.

Breast cancer remains a major health threat throughout the world specifically in women above 30 years of age however, it is rarely known to affect men as well. It is characterized by the abnormal division of cells in the breast tissue resulting in the development of breast malignancies. Various risk factors contributing to breast cancer include age, family history, genetic mutations (chiefly in BRCA1 and BRCA2 genes) along with hormonal imbalances (oestrogen, progesterone, HER2). Early detection which can be obtained through frequent rounds of self-examination, mammographic scanning, and clini

Liver-resident cellular determinants of oncolytic virotherapy in hepatocellular carcinoma: viral susceptibility and spatial immune remodeling.

Hepatocellular carcinoma (HCC) develops in a chronically injured and immunologically specialized organ that is both the treatment target and a major determinant of viral clearance and toxicity. Oncolytic viruses (OVs) can couple tumor-selective infection with immunogenic lysis, innate sensing, antigen presentation, and adaptive immune recruitment, but the clinical consequences of these processes depend on viral platform, delivery route, liver-resident cell handling, spatial disease distribution, and hepatic reserve. This review synthesizes HCC OV biology across four linked levels: platform-spe

Oncolytic virotherapy with chimeric rVSV-NDV causes tumor-specific immunity in a DC-activation-dependent manner.

In recent years, activation of the immune system has been recognized as a key mechanism in supporting the therapeutic outcome of oncolytic virotherapy for cancer. Most studies have focused on T cells in the characterization of immune responses to therapy; however, the role of dendritic cells (DCs) in priming these cytotoxic T cell responses has been far less explored. rVSV-NDV is a chimeric oncolytic virus that causes a highly immunogenic cell death in infected tumor cells. In tumor-bearing mice, this leads to increased levels of circulating and tumor-infiltrating tumor-specific CD8+ T cells u

Oncolytic Virotherapy for Relapsed, IDH-Mutant, Grade 3 Astrocytoma: A New Promising Approach - A Case Report.

INTRODUCTION: IDH-mutant astrocytomas are high-grade gliomas with a poor prognosis. Transformation to glioblastoma multiforme is common, which further shortens overall survival and frequently renders the tumor inoperable. Oncolytic viruses (OVs) have been shown to be safe and effective agents for the treatment of some malignant brain tumors. Intra-tumoral application may further enhance their therapeutic potential. CASE PRESENTATION: This report presents a case of a 37-year-old female patient with advanced relapsed grade 3 astrocytoma, with multiple foci, including one along the rim of the le

Pevonedistat, a first-in-class NEDD8-activating enzyme inhibitor, sensitizes cancer cells to VSVΔ51 oncolytic virotherapy.

The clinical efficacy of VSVΔ51 oncolytic virotherapy has been limited by tumor resistance to viral infection, so strategies to transiently repress antiviral defenses are warranted. Pevonedistat is a first-in-class NEDD8-activating enzyme (NAE) inhibitor currently being tested in clinical trials for its antitumor potential. In this study, we demonstrate that pevonedistat sensitizes human and murine cancer cells to increase oncolytic VSVΔ51 infection, increase tumor cell death, and improve therapeutic outcomes in resistant syngeneic murine cancer models. Increased VSVΔ51 infectivity was also ob

Pre-surgical neoadjuvant oncolytic virotherapy confers protection against rechallenge in a murine model of breast cancer.

The use of oncolytic viruses (OVs) for cancer treatment is emerging as a successful strategy that combines the direct, targeted killing of the cancer with the induction of a long-lasting anti-tumor immune response. Using multiple aggressive murine models of triple-negative breast cancer, we have recently demonstrated that the early administration of oncolytic Maraba virus (MRB) prior to surgical resection of the primary tumor is sufficient to minimize the metastatic burden, protect against tumor rechallenge, cure a fraction of the mice and sensitize refractory tumors to immune checkpoint block

Intraperitoneal oncolytic virotherapy for patients with malignant ascites: Characterization of clinical efficacy and antitumor immune response.

Oncolytic viruses mediate antitumor responses through direct tumor cell lysis and induction of host antitumor immunity. However, the therapeutic efficacy of oncolytic viruses against malignant ascites has rarely been explored. This study aimed to evaluate the efficacy, safety, and immunomodulatory effect of an intraperitoneal injection of human type 5 recombinant adenovirus (called H101) against malignant ascites. Forty patients with malignant ascites were recruited and treated with intraperitoneal H101 in the Fudan University Shanghai Cancer Center. The 4-week clinical responses were determin

Oncolytic virotherapy mobilizes tumor-resident, granzyme B-producing bystander CD4+ T cells to inhibit systemic microbial infection.

Patients with cancer are at elevated risk of life-threatening infections due to immunosuppression caused by both malignancy and its treatments, which compromises pre-existing anti-microbial immune memory established by prior infection or vaccination. Despite this systemic suppression, pathogen-specific memory T (TMEM) cells can infiltrate the tumor microenvironment in large numbers as tumor antigen-irrelevant bystander TMEM cells. We previously developed an engineered oncolytic virus encoding bystander TMEM cell epitopes (OV-BYTE) that redirected tumor-infiltrating bystander TMEM cells against

Cross-species oncolytic virotherapy in vitro: BoHV4EGFPΔTK as a novel candidate against canine osteosarcoma and histiocytic sarcoma.

Canine osteosarcoma (cOSA) and histiocytic sarcoma (cHS) are aggressive tumors characterized by poor prognoses and limited therapeutic options. Oncolytic viruses represent a promising treatment option for these tumors. Based on this hypothesis, this study aimed to evaluate direct and indirect oncolytic effects of Bovine Herpes virus type 1 (BoHV1) and a recombinant type 4 (BoHV4EGFPΔTK). Here we used cOSA, cHS and cutaneous fibroblast (FDOG) cell lines. BoHV1 and BoHV4EGFPΔTK can successfully infect cOSA and cHS cell lines with 1 multiplicity of infection (MOI). Notably, BoHV4EGFPΔTK displayed

Clinical activity of single-dose systemic oncolytic VSV virotherapy in patients with relapsed refractory T-cell lymphoma.

Clinical success with intravenous (IV) oncolytic virotherapy (OV) has to-date been anecdotal. We conducted a phase 1 clinical trial of systemic OV and investigated the mechanisms of action in responding patients. A single IV dose of vesicular stomatitis virus (VSV) interferon-β (IFN-β) with sodium iodide symporter (NIS) was administered to patients with relapsed/refractory hematologic malignancies to determine safety and efficacy across 4 dose levels (DLs). Correlative studies were undertaken to evaluate viremia, virus shedding, virus replication, and immune responses. Fifteen patients receive

KLS-3021: Innovative oncolytic virotherapy for the treatment of advanced primary and metastatic cutaneous squamous cell carcinoma.

Cutaneous squamous cell carcinoma (cSCC) is a highly prevalent malignancy with increasing incidence and limited therapeutic options in advanced or metastatic stages. Patients with high-risk or metastatic cSCC often experience poor outcomes, underscoring the need for novel therapeutic strategies. We investigated the potential of KLS-3021, a recombinant oncolytic vaccinia virus encoding hyaluronidase, interleukin-12, and soluble programmed cell death 1 fused to an Fc domain. KLS-3021 exhibited tumor-selective cytotoxicity and robust replication in cSCC cell lines while sparing normal keratinocyt

MicroRNA-124-targeted recombinant Zika virus: a dual-functional and safe candidate for vaccination and oncolytic virotherapy.

UNLABELLED: Zika virus (ZIKV) remains a significant public health threat due to its pronounced neurotropism linked to Congenital Zika Syndrome (CZS). Paradoxically, the same neurotropism also provides a unique opportunity for exploiting ZIKV as an oncolytic agent against malignant brain tumors such as glioma. However, achieving precise control of ZIKV replication in specific cell types or tissues remains a major challenge. Here, we show that the brain-specific and enriched microRNA, miR-124, is highly expressed in neural progenitor cells (NPCs) and neurons, but minimally expressed in glioma st

State-of-the-art in oncolytic virotherapy using adenoviruses other than the commonly applied adenovirus type 5.

In most clinical trials, oncolytic viruses (OAds) based on human adenovirus type 5 (HAdV-C5) were explored, and only rarely a complete switch to another adenovirus type was evaluated. This review highlights the broader diversity of human adenoviruses and discusses advances in engineering non-HAdV-C5 OAds. We will discuss ongoing research to refine adenoviral constructs derived from alternative adenovirus species, including chimeric viruses, to optimize viral delivery and enhance antitumor immune responses. We summarize translational and clinical studies using these alternative OAds, emphasizin
