Targeting the V-ATPase A Subunit in Phytophthora sojae: Discovery of Compound A15.

Phytophthora sojae poses a major threat to global soybean production. Here, CRISPR/Cas9-mediated promoter replacement revealed that the V-ATPase A subunit gene (PsVHA-A) plays an important role in the growth, development, and pathogenicity of P. sojae, highlighting its potential as a molecular target. Using this target, a multistage docking-based virtual screening identified compound A15 as a potent antioomycete inhibitor, with EC50 values of 4.082-8.974 μg/mL against six oomycetes. A15 markedly inhibited mycelial growth and key developmental stages in P. sojae in vitro and protected soybean l

Antifungal Activity of Plant-Derived Essential Oils on Pathogens of Pulse Crops.

Pulse crops such as chickpeas, lentils, and dry peas are grown widely for human and animal consumption. Major yield- and quality-limiting constraints include diseases caused by fungi and oomycetes. The environmental and health concerns of synthetic fungicides used for disease management, emergence of fungicide-resistant pathogens, and demand for organic pulse crop products necessitate the search for effective alternatives. Safe and environmentally friendly plant-derived essential oils (EOs) have been reported effective against some pathogenic fungi. Growth on EO-amended growth medium and an in

AtRTP5 negatively regulates plant resistance to Phytophthora pathogens by modulating the biosynthesis of endogenous jasmonic acid and salicylic acid.

Plants have evolved powerful immune systems to recognize pathogens and avoid invasions, but the genetic basis of plant susceptibility is less well-studied, especially to oomycetes, which cause disastrous diseases in many ornamental plants and food crops. In this research, we identified a negative regulator of plant immunity to the oomycete Phytophthora parasitica, AtRTP5 (Arabidopsis thaliana Resistant to Phytophthora 5), which encodes a WD40 repeat domain-containing protein. The AtRTP5 protein, which was tagged with green fluorescent protein (GFP), is localized in the nucleus and plasma membr

Gentisaldehyde Suppresses Phytophthora sojae by Combined Damage to Membrane Integrity and Energy Metabolism.

The soil-borne oomycete Phytophthora sojae causes severe Phytophthora root and stem rot (PRR) in soybean. Rhizosphere metabolites, integral to plant defense, represent an underexplored resource for novel control strategies. A metabolomic comparison of rhizospheres from resistant (Williams82) and susceptible (Williams) soybeans identified gentisaldehyde as a key differential metabolite. In vitro, gentisaldehyde demonstrated broad, concentration-dependent inhibition of P. sojae, suppressing mycelial growth, oospore formation, oospore survival, zoosporangium formation, zoospore encystment, and cy

Characterization of Aphanomyces invadans isolates using pyrolysis mass spectrometry (PyMS).

Twenty-one isolates of Aphanomyces invadans, the fungal pathogen associated with the Asian fish disease epizootic ulcerative syndrome (EUS), were compared with other Oomycete fungi in terms of their pyrolysis mass spectrometry (PvMS) profiles. Canonical variate analysis (CVA) of the pyrolysis mass spectra distinguished the Aphanomyces species from a wide scatter of Achlya and Saprolegnia isolates. Further CVA and hierarchal cluster analysis (HCA) separated the Aphanomyces species into two main groups. The first group clustered A. invadans isolates from EUS outbreaks in Thailand, Bangladesh, In

Trichoderma species isolated from grapevine with tolerance towards common copper fungicides used in viticulture for plant protection.

BACKGROUND: Copper-containing fungicides are applied broadly in organic viticulture against downy mildew caused by Plasmopara viticola. Although long-term application of copper-based fungicides is associated with ecotoxic effects on the environment, their use in viticulture is required until sustainable alternatives are available. Trichoderma spp. might be a promising approach to fungicide reduction while promoting plant growth and development and displaying biocontrol activity. This study aims to examine the tolerance and compatibility of Trichoderma spp. to copper fungicides. This work contr

Production of small cysteine-rich effector proteins in Escherichia coli for structural and functional studies.

Although the lifestyles and infection strategies of plant pathogens are diverse, a prevailing feature is the use of an arsenal of secreted proteins, known as effectors, which aid in microbial infection. In the case of eukaryotic filamentous pathogens, such as fungi and oomycetes, effector proteins are typically dissimilar, at the protein sequence level, to known protein families and functional domains. Consequently, we currently have a limited understanding of how fungal and oomycete effectors promote disease. Protein biochemistry and structural biology are two methods that can contribute grea

Predatory Mechanisms of Myxobacteria and Their Potential in the Development of Biocontrol Agents.

Climate change, environmental degradation, and the depletion of finite resources are intensifying the search for sustainable, bio-based solutions across agriculture, environmental management, and chemical production. Against this backdrop, myxobacteria have emerged as a valuable but still underused microbial resource, distinguished by their ecological versatility, multicellular behavior, predatory lifestyle, and biosynthetic capacity. This review integrates the mechanistic basis of myxobacterial predation with its translational potential in biocontrol, linking microbial ecology to applied biot

Fabrication of highly and poorly oxidized silver oxide/silver/tin(IV) oxide nanocomposites and their comparative anti-pathogenic properties towards hazardous food pathogens.

Herein, we report the fabrication of highly oxidized silver oxide/silver/tin(IV) oxide (HOSBTO or Ag3+-enriched AgO/Ag/SnO2) nanocomposite under a robust oxidative environment created with the use of concentrated nitric acid. Tin(IV) hydroxide nanofluid is added to the reaction mixture as a stabilizer for the Ag3+-enriched silver oxide in the nanocomposite. The formation of Ag nanoparticles in this nanocomposite originates from the decomposition of silver oxides during calcination at 600 °C. For comparison, poorly oxidized silver oxide/silver/tin(IV) oxide (POSBTO with formula AgO/Ag/SnO2) nan

Control of litchi downy blight by zeamines produced by Dickeya zeae.

Zeamines (ZMS), a class of polyamine-polyketide-nonribosomal peptide produced by bacterial isolate Dickeya zeae, were shown recently to be potent antibiotics against some bacterial pathogens. In this study, the results indicated that ZMS showed antifungal activity against Peronophythora litchii and other fungal pathogens. The activity of ZMS against the oomycete pathogen P. litchi, which causes the devastating litchi downy blight, was further investigated under in vitro and in vivo conditions. ZMS displayed potent inhibitory activity against the mycelial growth and sporangia germination of P.

Lysobacter capsici AZ78 can be combined with copper to effectively control Plasmopara viticola on grapevine.

The bacterial genus Lysobacter represents a still underdeveloped source of biocontrol agents able to protect plants against pathogenic oomycetes. In this work the L. capsici strain AZ78 was evaluated with regard to the biological control of Plasmopara viticola, the causal agent of grapevine downy mildew. L. capsici AZ78 is able to resist copper ions and its resistance to this metal is probably due to the presence of genes coding for copper oxidase (copA) and copper exporting PIB-type ATPases (ctpA). The presence of both genes was also detected in other members of the Lysobacter genus. Resistan

Interactive effect of chromium compounds and a fungal parasite on carp eggs.

A fungal parasite (Saprolegniales) of carp eggs was exposed to hexavalent and trivalent chromium compounds at concentrations identical to those used in a study of the effects of the same compounds on carp eggs hatchability. Comparison of data from both experiments revealed that increased egg mortality and increased fungal growth were cooupled at low concentrations of the compounds. A mechanism for the interaction of a chemical stress and a biological stress on carp egg survival in natural systems is presented.

Alcoholic extracts of Russian sage (Salvia yangii) contain bioactive terpenoids with inhibitory activity against grapevine downy mildew (Plasmopara viticola).

BACKGROUND: Plant extracts are promising eco-friendly alternatives to synthetic fungicides for developing sustainable plant protection strategies. Grapevine downy mildew, caused by Plasmopara viticola, is a devastating disease that requires frequent fungicide applications, making it an ideal target for plant-based products. Salvia officinalis extracts are known to reduce downy mildew severity, but little is known about the efficacy and chemical composition of other fast-growing Salvia species with low economic value. This study aimed to evaluate the efficacy of alcoholic extracts of Russian sa

New discoveries and applications of mosquito fungal pathogens.

Mosquitoes are a major threat to human health globally because they transmit infectious diseases, such as malaria, lymphatic filariasis, and arboviruses. The conventional mosquito control efforts, based on synthetic insecticides, have been compromised owing to the eventual development of insecticide resistance and the adverse environmental impacts of insecticides. Alternative eco-friendly approaches using entomopathogenic fungi to alleviate vector-borne disease burden have gained an increasing interest because of their selective specificity and environmental safety. Existing literature reveale

Salicylate-independent lesion formation in Arabidopsis lsd mutants.

In many interactions of plants with pathogens, the primary host defense reaction is accompanied by plant cell death at the site of infection. The resulting lesions are correlated with the establishment of an inducible resistance in plants called systemic acquired resistance (SAR), for which salicylic acid (SA) accumulation is a critical signaling event in Arabidopsis and tobacco. In Arabidopsis, the lesions simulating disease (lsd) mutants spontaneously develop lesions in the absence of pathogen infection. Furthermore, lsd mutants express SAR marker genes when lesions are present and are resis
