A potential target for organophosphate insecticides leading to spermatotoxicity.

Organophosphate (OP) insecticides as an anticholinesterase also act on the diverse serine hydrolase targets, thereby revealing secondary or unexpected toxic effects including male reproductive toxicity. The present investigation detects a possible target molecule(s) for OP-induced spermatotoxicity (sperm deformity, underdevelopment, and reduced motility) from a chemical standpoint. The activity-based protein profiling (ABPP) approach with a phosphonofluoridate fluorescent probe pinpointed the molecular target for fenitrothion (FNT, a major OP insecticide) oxon (bioactive metabolite of FNT) in

Intermediate syndrome following organophosphate insecticide poisoning.

Acute organophosphate insecticide poisoning can manifest 3 different phases of toxic effects, namely, acute cholinergic crisis, intermediate syndrome (IMS), and delayed neuropathy. Among them, IMS has been considered as a major contributing factor of organophosphate-related morbidity and mortality because of its frequent occurrence and probable consequence of respiratory failure. Despite a high incidence, the pathophysiology that underlies IMS remains unclear. Previously proposed mechanisms of IMS include different susceptibility of various cholinergic receptors, muscle necrosis, prolonged ace

[Clinical and electrophysiological aspects of acute organophosphate intoxication].

INTRODUCTION: Acute organophosphate (OP) intoxications, accidental or voluntary, are responsible for a high mortality. They cause extensive muscular paralysis by acetyl cholinesterase activity inhibition at the neuromuscular junction level. AIM: To underline the rarity and the characteristic electrophysiological pattern during cholinergic crisis. OBSERVATION: A 28-year-old woman was admitted to the medical intensive care unit for Malathion acute intoxication with signs of glandular hypersecretion, complicated tetraparesis, and respiratory distress. The cholinesterase activity was 17%. The el

A case report and overview of organophosphate (OP) poisoning.

A case of organophosphate (OP) poisoning who recovered after requiring almost 1000 mg of atropine, 10 gm of PAM and ventilatory support for 7 days is presented here. The overview of organophosphate poisoning and its management is given. With the approach adopted, the mortality reported in the general medicine unit in the central hospital in Nepal is 7.4%. The two important aspects of the management are vigilance of the atropine drip, especially at night, and other physical and psychological support care of the patients.

Suicide and exposure to organophosphate insecticides: cause or effect?

BACKGROUND: Suicide using pesticides as agent is recognized as a major cause of pesticide poisoning. METHODS: A literature review of mortality and morbidity studies related to suicide among pesticide-exposed populations, and of human and animal studies of central nervous system toxicity related to organophosphate (OP) pesticides was performed. RESULTS: Suicide rates are high in farming populations. Animal studies link OP exposure to serotonin disturbances in the central nervous system, which are implicated in depression and suicide in humans. Epidemiological studies conclude that acute and c

Mechanisms of organophosphate insecticide-induced airway hyperreactivity.

It has been suggested that pesticide exposure may be a contributing factor underlying the increased incidence of asthma in the United States and other industrialized nations. To test this hypothesis, airway hyperreactivity was measured in guinea pigs exposed to chlorpyrifos, a widely used organophosphate pesticide. Electrical stimulation of the vagus nerves caused frequency-dependent bronchoconstriction that was significantly potentiated in animals 24 h or 7 days after a single subcutaneous injection of either 390 mg/kg or 70 mg/kg of chlorpyrifos, respectively. Mechanisms by which chlorpyrifo

Intensive care management of organophosphate insecticide poisoning.

INTRODUCTION: Organophosphate (OP) insecticides inhibit both cholinesterase and pseudo-cholinesterase activities. The inhibition of acetylcholinesterase causes accumulation of acetylcholine at synapses, and overstimulation of neurotransmission occurs as a result of this accumulation. The mortality rate of OP poisoning is high. Early diagnosis and appropriate treatment is often life saving. Treatment of OP poisoning consists of intravenous atropine and oximes. The clinical course of OP poisoning may be quite severe and may need intensive care management. We report our experience with the intens

Genotoxicity from domestic use of organophosphate pesticides.

This report describes the types of chromosome damage in peripheral blood found in patients exposed to domestic application of organophosphate pesticides. These changes serve as a biomarker of cumulative toxic exposure. Susceptible individuals show DNA damage as chromosome alterations. The importance of these findings is that the apparent genotoxic changes occurred from domestic application of two of the commonly used organophosphate pesticides in America and adds one more adverse potential effect from these hazardous chemicals. Peripheral blood from eight patients exposed to the domestic spray

Risk factors for nonoccupational organophosphate pesticide poisoning.

The 535 reports of nonoccupational/nonagricultural poisoning with malathion in the Pesticide Incident Monitoring System (PIMS), a data base maintained by the Environmental Protection Agency, were coded and classified to describe the occurrence and to determine risk factors for poisoning. Between 1966 and 1980, 335 (63%) of reported incidents were associated with at least one symptomatic person, with a total of 670 persons exposed. Home exposures accounted for 92% of reports and arose from improper use and labeling problems in 61% and 3% of reports, respectively. Seventeen reports (3%) were ass

Organophosphate pesticide poisoning in 50 Ethiopian patients.

Organophosphate pesticide poisoning in 50 patients treated at the Tikur Anbessa Hospital over a period of 6 years is described. Most of the cases were attempted suicide (94%). The majority of the patients presented with the typical clinical features of Parasympathetic overactivity. Atropine was the mainstay of treatment. Mortality from organophosphate pesticide poisoning was 20%. There is a strong need to device a national system of control of pesticides and educate the public about the dangers of pesticides.

Respiratory failure of acute organophosphate and carbamate poisoning.

Respiratory failure (RF) developed in 43 (40.2 percent) of 107 patients with acute organophosphate or carbamate poisoning; 22 (51.2 percent) died. The 64 patients who did not develop RF survived. All cases of RF developed within 96 hours after poisoning: within 24 hours in 35 patients (acute onset) and between 24 and 96 hours in eight patients (subacute onset). Severity of poisoning was the primary determinating factor for RF. Cardiovascular collapse and pneumonia were also associated with RF. In 19 patients with cardiovascular collapse, 17 had acute onset of RF and two had subacute onset. In

End-plate dysfunction in acute organophosphate intoxication.

Acute organophosphate intoxication resulting from suicide attempts in 14 patients produced a series of electrophysiologic abnormalities that correlated with the clinical course. Spontaneous repetitive firing of single evoked compound muscle action potentials (CMAP) was the earliest and most sensitive indicator of the acetylcholinesterase inhibition. A decrement of evoked CMAP following repetitive nerve stimulation was the most severe abnormality. At the height of the intoxication no CMAP was evoked after the first few stimuli. The decrement-increment phenomenon occurred only at milder stages o
