No evidence for the involvement of the argasid tick Ornithodoros faini in the enzootic maintenance of marburgvirus within Egyptian rousette bats Rousettus aegyptiacus.

BACKGROUND: The cave-dwelling Egyptian rousette bat (ERB; Rousettus aegyptiacus) was recently identified as a natural reservoir host of marburgviruses. However, the mechanisms of transmission for the enzootic maintenance of marburgviruses within ERBs are unclear. Previous ecological investigations of large ERB colonies inhabiting Python Cave and Kitaka Mine, Uganda revealed that argasid ticks (Ornithodoros faini) are hematophagous ectoparasites of ERBs. Yet, their potential role as transmission vectors for marburgvirus has not been sufficiently assessed. FINDINGS: In the present study, 3,125

Use of a Bayesian network model to identify factors associated with the presence of the tick Ornithodoros erraticus on pig farms in southern Portugal.

The soft tick Ornithodoros erraticus occurs on pig farms in southern Portugal and Spain and transmits several important pathogens of humans and livestock. Its distribution is patchy and the determinants of its distribution are uncertain. Here, we use a Bayesian network model to explore possible associations between climate, farm management and the presence of O. erraticus. The resulting network confirms previous suggestions that the presence of O. erraticus is more likely in traditionally constructed pig housing, and indicates that carbon dioxide traps are highly effective for the detection of

Cloning, nucleotide sequence and expression of the gene encoding factor Xa inhibitor from the salivary glands of the tick, Ornithodoros savignyi.

The N-terminal sequence of the competitive and slow tight-binding factor Xa inhibitor (fXaI; Ki = 0.83 +/- 0.10 nM) isolated from the salivary glands of Ornithodoros savignyi ticks (Acari: Argasidae) was employed to design a degenerate gene-specific primer (GSP) for 3'-rapid amplification of cDNA ends (3'-RACE). The primer consisted of a sequence encoding for amino acid residues 5-11. A full-length gene was next constructed from the 3'-RACE product in a two-step PCR procedure and successfully expressed by the BAC-TO-BAC baculovirus expression system. The deduced amino acid sequence of the gene

Detection of pig farms with Ornithodoros erraticus by pig serology. Elimination of non-specific reactions by carbohydrate epitopes of salivary antigens.

Ornithodoros erraticus is the European vector of African Swine Fever. It is therefore essential to know on which pig farms the tick is present in order to prevent contact with swine. Currently, studies are being made to ascertain this through the detection of anti-O. erraticus antibodies in the sera of swine, using three extracts from the salivary glands of the parasite (SGE): a complete extract (SGE-1), a soluble antigens extract (SGE-2), and a tissue antigens extract (SGE-3). The results of the present work show that SGE-2 gives the best differentiation between swine bitten by O. erraticus a

Vertical transmission of Borrelia turicatae (Spirochaetales: Borreliaceae ) by autogenously reproducing Ornithodoros turicata (Ixodida: Argasidae) female naturally infected with the spirochetes.

UNLABELLED: Ornithodoros turicata is a vector of relapsing fever (RF) spirochetes in North America and transmits Borrelia turicatae to a variety of vertebrate hosts. The remarkably long lifespan of O. turicata and its ability to maintain spirochetes horizontally (between life stages) and vertically to progeny promotes the perpetuation of B. turicatae in nature. Nevertheless, the reproductive biology of O. turicata is poorly understood. In this report, we collected ticks from a park within a neighborhood of Austin, Texas. They were reared to adulthood and male ticks were individually housed wit

Detection of Rickettsia sp. strain Itinguçú in Ornithodoros faccinii (Acari: Argasidae) parasitizing the toad Rhinella ornata (Anura: Bufonidae) in Brazil.

The pivotal role of amphibians in food webs and their value as indicators of disequilibrium in ecosystem health have long been recognized by wildlife biologists. However, massive pathogen-induced declines in global amphibian populations reported during the last 30 years served to alert the scientific community that knowledge of amphibian disease ecology, including parasitic and vector-borne conditions, was and remains incipient. Herein, we report the detection of a Rickettsia bacterium infecting larvae of the argasid tick Ornithodoros faccinii, collected from the toad Rhinella ornata, in South

Coxiella-like endosymbiont in argasid ticks (Ornithodoros muesebecki) from a Socotra Cormorant colony in Umm Al Quwain, United Arab Emirates.

Coxiella burnetii is a pathogen causing Q fever in domestic animals and humans. Seabirds have been implicated as possible reservoirs of this bacterium in the Arabian Gulf and in the Western Indian Ocean. Recently, Coxiella species closely related to C. burnetii was detected from ticks collected from oil rigs used as roosting areas by Socotra Cormorants (Phalacrocorax nigrogularis) in the western Arabian Gulf. We collected ticks from the largest breeding colony of Socotra Cormorants in the United Arab Emirates on the eastern extreme of the species' breeding range to determine the prevalence of

Integrating Systematic Surveys With Historical Data to Model the Distribution of Ornithodoros turicata americanus, a Vector of Epidemiological Concern in North America.

Globally, vector-borne diseases are increasing in distribution and frequency, affecting humans, domestic animals, and wildlife. Science-based management and prevention of these diseases requires a sound understanding of the distribution and environmental requirements of the vectors and hosts involved in disease transmission. Integrated Species Distribution Models (ISDM) account for diverse data types through hierarchical modeling and represent a significant advancement in species distribution modeling. We assessed the distribution of the soft tick subspecies Ornithodoros turicata americanus. T

[The effect of the causative agent of tick-borne relapsing fever on the respiration of the vector Ornithodoros papillipes (Birula, 1895) (Ixodoidea, Argasidae)].

A relatively low level of normal respiratory metabolism in O. papillipes has been observed as compared to most other invertebrates and even ixodid ticks. It is connected with biological peculiarities and habitat of this species. Changes in respiratory metabolism under the influence of Borrelia have been revealed. These changes vary at different stages of tick life cycle. In long-fasting infected individuals, respiration is further inhibited, and in nourished ones, is increased as compared to noninfected ticks.

Autogenous reproduction by Ornithodoros turicata (Ixodida: Argasidae) females and vertical transmission of the tick-borne pathogen Borrelia turicatae (Spirochaetales: Borreliaceae).

Previous research has implicated Ornithodoros ticks, including Ornithodoros turicata, as long-term reservoirs of relapsing fever (RF) spirochetes. Considering the tick's long lifespan and their efficiency in maintaining and transferring spirochetes within the population, the infection could persist in a given enzootic focus for decades. However, little is known about the relative importance of horizontal and vertical transmission routes in the persistence and evolution of RF Borrelia. Our observations on the reproductive biology of O. turicata in the absence of vertebrate hosts indicate an add

Borrelia nietonii sp. nov.: Relapsing Fever Spirochetes Transmitted By the Tick Ornithodoros hermsi Designated Previously as Borrelia hermsii Genomic Group II.

Background: The taxonomic status of the relapsing fever spirochete Borrelia hermsii in western North America was established in 1942 and based solely on its specific association with the soft tick vector Ornithodoros hermsi. Multilocus sequence typing (MLST) of the 16S rRNA, flaB, gyrB, glpQ, and 16S-23S rRNA intergenic spacer of B. hermsii isolates collected over many years from various geographic locations and biological sources identified two distinct clades designated previously as B. hermsii Genomic Group I (GGI) and Genomic Group II (GGII). Occurrence of the bat tick Ornithodoros (Alectorobius) Kelleyi Cooley & Kohls (Acari: Argasidae) in Costa Rica and its relation to human bites.

Several persons from two localities in Costa Rica (San Rafael de Coronado, 1510 m and Dulce Nombre de Tres Ríos, 1445 m) were bitten by ticks, subsequently identified as Ornitohodoros (A.) Kelleyi (Argasidae), commonly associated with bats. In the attics of their homes, live adults, larvae, nymphs and skins were found in the bat guano. Since there are no records of adult argasids in this country, a laboratory colony was established to observe the biology of these ticks and their association with bats. In this particular case, the bats were Molossidae and Vespertilionidae, both insectivorous. A

Quantitative proteomic analysis of salivary glands in female Ornithodoros lahorensis (syn. Alveonasus lahorensis) after blood feeding via DIA-coupled LC-MS/MS.

Ornithodoros lahorensis (syn. Alveonasus lahorensis) is a species of soft tick within the Argasidae family, known for causing debilitating symptoms, as well as transmitting pathogens that pose significant health risks and economic losses to affected livestock and potentially humans. The salivary gland is the key component in the pathogenic mechanisms of ticks, as it not only facilitates feeding but also serves as a medium for pathogen transmission. Here, data-independent acquisition (DIA) coupled to liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to quantitatively identify d

Chemical Synthesis and Structure-Activity Relationship Studies of the Coagulation Factor Xa Inhibitor Tick Anticoagulant Peptide from the Hematophagous Parasite Ornithodoros moubata.

Tick Anticoagulant Peptide (TAP), a 60-amino acid protein from the soft tick Ornithodoros moubata, inhibits activated coagulation factor X (fXa) with almost absolute specificity. Despite TAP and Bovine Pancreatic Trypsin Inhibitor (BPTI) (i.e., the prototype of the Kunitz-type protease inhibitors) sharing a similar 3D fold and disulphide bond topology, they have remarkably different amino acid sequence (only ~24% sequence identity), thermal stability, folding pathways, protease specificity, and even mechanism of protease inhibition. Here, fully active and correctly folded TAP was produced in r

Population structure of the soft tick Ornithodoros maritimus and its associated infectious agents within a colony of its seabird host Larus michahellis.

The epidemiology of vector-borne zoonoses depends on the movement of both hosts and vectors, which can differ greatly in intensity across spatial scales. Because of their life history traits and small size, vector dispersal may be frequent, but limited in distance. However, little information is available on vector movement patterns at local spatial scales, and particularly for ticks, transmitting the greatest diversity of recognized infectious agents. To test the degree to which ticks can disperse and disseminate pathogens at local scales, we investigated the temporal dynamics and population

Prevalence, distribution, and phylogenetics of the tick-borne relapsing fever spirochete Borrelia turicatae in the soft tick Ornithodoros turicata americanus in Florida.

In the southwestern United States, the occurrence and potential for disease spread of Tick-Borne Relapsing Fever (TBRF) has been studied. In Florida, Borrelia pathogens that cause TBRF have only been found in two domestic dogs (Canis lupus familiaris), yet the soft tick vector, Ornithodoros turicata americanus, is commonly found throughout the state. The goal of our study was to provide the first large-scale investigation of this disease system in the southeastern US. Our objectives were to: 1) describe the occurrence and prevalence of Borrelia spp. in ticks found throughout their distribution

Transcriptomic and Metabolomic Profiling Identifies a Core Gene-Metabolite Axis Driving African Swine Fever Virus Replication in the Soft Tick Ornithodoros lahorensis.

African swine fever virus (ASFV) causes an incurable swine disease with nearly 100% mortality, posing a catastrophic threat to global pig production. The soft tick Ornithodoros lahorensis acts as a critical biological vector that sustains persistent ASFV replication and mediates long-distance viral transmission, yet the molecular mechanisms governing ASFV-tick interplay remain poorly understood. Here, we integrated transcriptomics and metabolomics to systematically dissect molecular changes in O. lahorensis across three infection stages: Uninfected control, early infection (7 days post-infecti
