This study compared aggressiveness between two distinct wild populations of Japanese medaka: a Northern population, Oryzias sakaizumii, and a Southern population, O. latipes. When four males competed in intra-population contests, the social hierarchy was determined based on aggressive acts in both populations. Dominants of the Southern population showed higher aggressive acts than did dominants of the Northern population. Increased aggressiveness of Southern males compared with Northern males was also observed when a pair of Northern and Southern males were compared in inter-population contest

Radiation-induced bystander effects are established consequences of exposure to ionizing radiation. The operation of this mechanism has been seen in vitro and also between fish, mammals, and plants in vive where stress signals from treated organisms induce responses in neighbors. In vitro research shows that DNA repair deficient cells produce more toxic bystander responses. To test this in vivo two strains of Japanese medaka were tested. One is a mutant, repair deficient strain (ric2) and the other, the wildtype repair proficient strain (CAB). Irradiated fish swam with unirradiated partners in

Japanese medaka (Oryzias latipes) embryos exposed to ethanol have developed craniofacial, cardiovascular and skeletal defects which can be compared with the phenotypic features of fetal alcohol spectrum disorder (FASD) observed in human. The present experiment was designed to show that the disruption in circulation by ethanol during embryogenesis is a potential cause of FASD. Fertilized eggs were exposed to ethanol (0, 100 and/or 400 mM) for 24 or 48 h at various developmental stages (Iwamatsu stages 4-30) and were analyzed at 6 day post fertilization (dpf). It was observed that controls and t

Increasing evidence suggests that sublethal effects of natural or xenobiotic chemicals in the environment may be mediated via the stimulation of apoptosis. To investigate whether apoptosis can be induced in fish by weakly estrogenic and androgenic chemicals, adult male Japanese medaka (Oryzias latipes) were exposed to 100 ppb of the estrogenic alkylphenol, 4-nonylphenol, and adult female medaka were exposed to 100 ppb of the aromatase-inhibiting bioflavonoid, quercetin, for 6 weeks. Exposure to nonylphenol and quercetin had no significant effect on the length, weight or condition factors compa

Atrazine (ATZ), a widely used triazine herbicide frequently detected in aquatic environments, poses ecological risks, yet its toxicity mechanisms in estuarine/marine organisms remain unclear. The present study employed juvenile Oryzias melastigma (marine medaka) as the model organism, and the 96-h median lethal concentration (LC50) of ATZ was determined to be 31.65 mg/L. Juvenile marine medaka were exposed to 0, 0.09, 0.36, 1.07, and 3.28 mg/L ATZ for 30 days. Histopathology revealed severe gill alterations (hyperplasia, apical enlargement, disintegration of the lamellae and filament congestio

The marine medaka is emerging as a potential behavioral model organism for ocean studies, namely on marine ecotoxicology. However, not much is known on the behavior of the species and behavioral assays lack standardization. This study assesses the marine medaka as a potential model for chemical communication. We investigated how short exposure to visual and chemical cues mediated the stress response to social isolation with the light/dark preference test (LDPT) and the open field test (OFT). After a 5-day isolation period, and 1 h before testing, isolated fish were randomly assigned to one of

The COVID-19 pandemic has resulted in huge amounts of face masks worldwide. However, there is a lack of awareness on the additives and their potential risk to aquatic ecosystems of face masks. To address this issue, the additives and their toxicity in 13 face masks (e.g., polypropylene, polyethylene, and polylactic acid) were determined using nontarget analysis and bioassays. A total of 826 organic additives including intermediates (14.8%), surfactants (9.3%), plasticizers (8.2%), and antioxidants (6.1%) were tentatively identified, with 213 compounds being assigned confidence levels of 1 and

PURPOSE: Chronic exposure to ionizing radiation (IR) at low doses (<100 mGy) has been insufficiently studied to understand fully the risk to health. Relatively little knowledge exists regarding how species and healthy tissues respond at the protein level to chronic exposure to low doses of IR, and mass spectrometric-based profiling of protein expression is a powerful tool for studying changes in protein abundance.
MATERIALS AND METHODS: SDS gel electrophoresis, LC-MS/MS mass spectrometry-based approaches and bioinformatic data analytics were used to detect proteomic changes following chronic

Shoaling can be considered a simple form of affective behavior displayed by social fish in which a single fish exhibits an tendency to approach others. In the present study, we adopted a dual approach to investigate shoaling behavior in the medaka fish (Oryzias latipes): a behavioral pharmacological approach to assess mirror approaching behavior and an immunohistochemical approach to examine the neurotransmitter distribution in the medaka telencephalon. In order to gain an insight into shoaling activity, we examined the pharmacological effects of the positive allosteric modulator of the gamma-

Proliferation of stromal cells is a common occurrence in experimentally induced hepatocarcinogenesis in fish. However, the role of these cells in fish hepatic injury and neoplasia is unknown. To better understand the biology of the cells comprising the hepatic stroma, livers from medaka (Oryzias latipes) experimentally induced by diethylnitrosamine or methylazoxymethanol acetate were labeled with keratin, actin, and desmin antibodies. The distribution of these proteins in hepatocellular carcinoma, cholangiocarcinoma, spindle cell proliferative lesions, and spongiosis hepatis was assessed, incl

Benzotriazole ultraviolet stabilizers (BUVs) are widespread and bioaccumulative aquatic contaminants. However, their immunotoxicity and underlying molecular mechanisms remain largely unexplored. In this study, we measured the concentrations of ten BUVs in the East China Sea and selected UV-P for further study based on its highest detected concentration. Consequently, marine medaka (Oryzias melastigma) were exposed to an environmentally relevant concentration (3.5 μg/L) and two elevated concentrations (35 and 350 μg/L) of UV-P for 28 days. Immunological parameters, histopathological alterations

Visual perception plays a crucial role in foraging, avoiding predators, mate selection, and communication. The regulation of color vision is largely dependent on opsin, which is the first step in the formation of the visual transduction cascade in photoreceptor cells. Short-wave-sensitive 1 (sws1) is a visual pigment that mediates short-wavelength light transduction in vertebrates. The depletion of sws1 resulted in increased M-opsin in mice. However, there is still no report on the visual function of sws1 in teleost fish. Here, we constructed the sws1 knockout medaka using CRISPR/Cas9 technolo

The synthetic estrogen 17α-ethinylestradiol (EE2) is a common component of hormone therapy and oral contraceptives and has been widely used for nearly 60 years. Numerous studies have shown that exposure to EE2 can affect embryonic development in a number of fish species. The effects of parental and embryonic EE2 exposure on embryo developmental toxicity and the underlying molecular mechanisms, however, have rarely been examined. In this study, embryos collected from parental EE2-exposed adult fish were examined to assess EE2-induecd toxicity during embryo development. The rate of embryo develo

Toxicity tests were performed to investigate possible harmful effects on medaka exposed to nano-iron. Dose-dependent decreases of superoxide dismutase (SOD) and increases of malondialdehyde (MDA) were induced in the medaka embryo, suggesting that oxidative damage was induced by nano-iron. For adult medaka, the disturbance of antioxidative balance was observed during the early exposure period based on the monitoring of the hepatic and cerebral SOD and reduced glutathione (GSH). No terminal oxidative damage occurred during the whole exposure period, probably due to the high self-recovering capab

Tralopyril, a novel antifouling agent widely used in hull coatings, has raised ecological concerns due to its increasing application. This study employed marine medaka as a model to systematically evaluate the behavioral toxicity and underlying mechanisms of tralopyril exposure. Tralopyril significantly reduced locomotor activity, enhanced phototaxis, and impaired olfactory function. Histopathological analysis revealed damage in multiple organs, including retinal thinning, olfactory epithelial disruption, brain vacuolization, cardiac atrophy, muscle disorganization, and cranial cartilage contr

Neuropeptide Y receptor Y2 (npy2r) is an important receptor gene involved in anxiety and feeding regulation in mammals. Since NPY receptors have different receptor gene deletions in mammals and teleost fish, it is not clear whether npy2r has the similar function in fish as in mammals. In this study, we used the CRISPR/Cas9 system to establish npy2r-deficient medaka (Oryzias latipes). Unexpectedly, the deletion of npy2r resulted in the npy2r +/- medaka were all-male, therefore, npy2r homozygous mutant lines could not be established. The deletion of npy2r increased the food intake in medaka, and

Japanese medaka embryos were exposed to water accommodated fractions (WAF) and chemically-enhanced WAF of two types of diluted bitumen (dilbit) at concentrations bracketing the EC50s for developmental malformations. Within these treatments, fish were grouped based on the presence or absence of developmental malformations (e.g., blue sac disease (BSD)), and analyzed for novel transcriptomic responses. Microarray analyses identified novel biomarkers and gene networks in dilbit-exposed malformed embryos that were not evident in dilbit-exposed fish without BSD or in controls without dilbit. The to

Ciguatoxins are produced by toxic benthic dinoflagellates and cause ciguatera fish poisoning worldwide, but the toxic effects on developing marine fish have not been well investigated. The Pacific ciguatoxin (P-CTX-1), is a potent sodium channel agonist, which is one of the most toxic members among all CTXs. This study evaluated the toxic effects of microinjecting purified Pacific ciguatoxin-1 (P-CTX-1) on embryonic development of marine medaka Oryzias melastigma. A lower 96h-LD50 value was estimated for eleuthero-embryos (1.32ngg-1) than that for embryos (1.71ngg-1), indicating that P-CTX-1 i

Heat shock protein 70 (Hsp70) is one of the most widely used biomarker for monitoring environment perturbations in biological systems. To facilitate the analysis of hsp70 expression as a biomarker, we generated a Tg(hsp70:gfp) transgenic medaka line in which green fluorescence protein (GFP) reporter gene was driven by the medaka hsp70 promoter. Here, we characterized Tg(hsp70:gfp) medaka for inducible GFP expression by seven environment-relevant heavy metals, including mercury, arsenic, lead, cadmium, copper, chromium, and zinc. We found that four of them (mercury, arsenic, lead, and cadmium)

DNA methylation is a common epigenetic modification of DNA levels in the genome of eukaryotic cells, and an aberrant elevation of DNA methylation in gene promoter regions can inhibit gene expression. DNA methyltransferases (DNMTs) are involved in genomic DNA methylation, divided into maintenance DNA methyltransferases and de novo methylases, which are expressed to different degrees in the testis and ovaries. 5-aza-2'-deoxycytidine (5-aza-dC) is a cytidine analog with a strong methylation inhibition. In this experiment, medaka fish fries were treated with 5-aza-dC at 0 μg/L, 50 μg/L, and 100 μg
