Based on clinical observation of a flock of approximately 5000 sheep (breed Merino) from southern Brazil, the failure of anthelmintic treatments was suspected. The sensitivity of the gastrointestinal nematodes that infected these sheep to nine drugs (Levamisole, Moxidectin, Albendazole, Ivermectin, Nitroxynil, Disophenol, Trichlorphon, Closantel, and a combination of Ivermectin, Levamisole and Albendazole) was evaluated. None of the drugs reduced the count of nematode eggs per gram of feces (EPG) in the sheep to an adequate extent (defined as a 95% reduction in EPG). Therefore, we found multip

The influence of route of administration on the pharmacokinetics and efficacy of macrocyclic lactone anthelmintics has been a subject of interest due to its potential to influence the development of anthelmintic resistance. For most parasite species studied so far, oral administration results in the highest concentrations of drug in the parasites and the highest efficacy against resistant genotypes. However, a recent study in cattle measured the highest levels of ivermectin in the abomasal Ostertagia ostertagi following subcutaneous injection, but it was not possible to correlate these elevate

Anthelmintic resistance (AR) in gastrointestinal nematodes (GIN) is a worldwide problem in all livestock production systems. Several prevalence studies performed internationally demonstrate that AR in cattle parasites is a growing problem; however, there are no published studies on prevalence in the United States (USA). In this study, we performed fecal egg count reduction tests (FECRT) on twelve cow-calf farms in Georgia, USA. On all farms the efficacy of eprinomectin pour-on (Eprinex®) and doramectin injectable (Dectomax®) was tested individually, as well as a combination treatment of dorame

The goal of the current work was to perform an integrated evaluation of monepantel (MNP) pharmacokinetics (PK) and pharmacodynamics, measured as anthelmintic efficacy, after its oral administration to calves naturally infected with GI nematodes resistant to ivermectin (IVM) and ricobendazole (RBZ) on three commercial farms. On each farm, forty-five calves were randomly allocated into three groups (n = 15): MNP oral administration (2.5 mg/kg); IVM subcutaneous (SC) administration (0.2 mg/kg); and RBZ SC administration (3.75 mg/kg). Eight animals from the MNP treated group (Farm 1) were selected

We describe a new fixed-dose combination injectable (FDCI) formulated with doramectin and levamisole hydrochloride (HCl) to target broad and overlapping spectra of gastrointestinal nematodes (GINs) through two distinct modes of action. Here, we demonstrate the superior efficacy of the FDCI against mixed populations of cattle GINs in two dose confirmation studies conducted in Australia using artificially induced adult (Study 1) and immature (Study 2) GIN infections. Artificial infections consisted of Cooperia spp., Haemonchus placei, Ostertagia ostertagi, and Trichostrongylus axei. In both stud

The synlophe (longitudinal, surface cuticular ridges) of Sarwaria bubalis is described for the first time. It is a tapering lateral synlophe of about 40 ridges. The synlophe of S. bubalis is similar to that of Ostertagia ostertagi but markedly different from that of species of Spiculopteragia and Mazamastrongylus. New information is provided also on the structure of the esophagus and perivulval pores. The esophageal valve is more than twice as long as wide. The bilateral perivulval pores were located 192-267 microns posterior to the vulva and dorsal to the lateral lines. The new information wi

A series of alpha-cyano-beta-hydroxypropenamides was prepared and tested for anthelmintic activity. alpha-Cyano-beta-hydroxy-N-[4- (trifluoromethyl)phenyl]-3-[4-(trifluoromethyl)phenyl]propenamide (1) showed good activity against the nematode Nematospirodes dubius in a mixed parasite infection in mice; several of the analogues were also effective against the cestode Hymenolepis nana. In sheep trials, 1 caused 100% reduction of the hematophagous nematode Haemonchus contortus after a single dose of 20 mg/kg but did not show satisfactory control of Trichostrongylus colubriformis or Ostertagia cir

The lungs and gastrointestinal tracts from 18 hunter-killed bighorn rams (Ovis canadensis californiana) were examined in total or in part for helminth parasites during a two-year study of three separate herds in Eastern Oregon. Prevalence was 100% with the lungworm Protostrongylus stilesi. The gastrointestinal fauna from 11 rams comprised Cooperia oncophora, Marshallagia marshalli, Nematodirus oiratianus, Oesophagostomum spp., Ostertagia occidentalis, O. ostertagi, Skrjabinema ovis, Trichostrongylus axei and Trichuris spp. Adult Wyominia tetoni and cysticerci of Taenia hydatigena were recovere

The efficacy of albendazole in removing gastrointestinal nematodes and tapeworms was studied in 16 treated and 16 untreated (control) beef calves. The anthelmintic was administered as an oral drench at a dose level of 5 ml/45.5 kg of body weight (5 mg/kg). Percentage of efficacy against the various genera was: Trichostrongylus axei, Oesophagostomum radiatum, and Moniezia-100.0; Haemonchus placei, Cooperia spp, and Trichostrongylus colubriformis - greater than 99.0; Ostertagia ostertagi -98.3; and Bunostomum phlebotomum -96.2. Efficacy against Trichuris was 20.2. Efficacy against immature nemat

BACKGROUND: Gastrointestinal nematode (GIN) control is traditionally achieved with the use of anthelmintic drugs, however due to regulations in organic farming and the rise in anthelmintic resistance, alternatives are sought after. A promising alternative is the use of bioactive plant feeding due to the presence of plant secondary metabolites (PSMs) such as proanthocyanidins (PAs). This study focussed on the perennial shrub heather (Ericaceae family), a plant rich in PAs, highly abundant across Europe and with previously demonstrated anthelmintic potential.
METHODS: In vitro assays were used

When sheep grazed alone, there was no significant increase in the number of worm species normally found in sheep, namely Ostertagia circumcincta and Nematodirus spathiger, with an increase in stocking rate. BACKGROUND: Benzimidazole (BZ) resistance in gastrointestinal nematodes is associated with a single nucleotide polymorphism (SNP) at codons 167, 198 and 200 in the isotype 1 of beta-tubulin gene although in some species these SNPs have also been associated with resistance to macrocyclic lactones. In the present study we compared the levels of resistance in Teladorsagia circumcincta and Trichostrongylus colubriformis by means of the faecal egg reduction test (FECRT) and the percentage of resistant alleles obtained after pyrosequencing. The study was conducted in 10 naturally infected sheep floc

Anthelmintic resistance (AR) is a well-recognized challenge in farmed ruminants. The use of anthelmintics in combination is one of the strategies recommended to slow the rate of AR development. Two studies were undertaken in 2017 and 2019 to assess the efficacy of single-dose macrocyclic lactone (ML) anthelmintic and ML combination drenches. In total, 11 Faecal Egg Count Reduction Trials (FECRTs) were set up in 10 different beef herds, with results available from 10 of those FECRTs (9 herds). AR to a single ML anthelmintic was detected in all 9 herds, with resistance to Cooperia and Haemonchus

In early 1989, two-thirds of the Soay sheep population on St Kilda died over 12 weeks. Post-mortem examinations revealed emaciated carcasses and considerable nematode burdens, with protein-energy malnutrition as the probable cause of death. Haematological and blood biochemical changes in the sheep, as well as fecundity of gastrointestinal nematodes, suggested the hosts were immunosuppressed. In parallel, laboratory experiments in which Soay sheep on a high plane of nutrition were artificially infected with Ostertagia circumcincta, showed no clinical signs or mortality when supporting worm burd

Resistance to the benzimidazole and macrocyclic lactone anthelmintics is widespread in Cooperia spp. on cattle farms in New Zealand. Since this was first documented in 2006 little has changed in cattle farming systems except for the widespread use of levamisole to control Cooperia spp. in young cattle (i.e., parasite control has maintained an almost total reliance on use of anthelmintics). Here we report the emergence of simultaneous resistance to the benzimidazole, macrocyclic lactone and levamisole anthelmintics in Cooperia spp. and in Ostertagia spp. Anthelmintic efficacy against nematode p

AIMS: To investigate the association between gastrointestinal parasites (GIP) and animal behaviour in dairy calves under New Zealand pastoral conditions, using animal-mounted, accelerometer-based sensors.
METHODS: Thirty-six, 5-6-month-old, Friesian-Jersey, heifer calves fitted with animal activity sensors to track behaviour were randomly allocated to one of two treatment groups. Half the animals were challenged with an oral dose of 20,000 larvae of Ostertagia ostertagi and Cooperia oncophera once a week for 3 weeks and half were unchallenged. Five weeks after the last dose, seven infected an

Control of helminth infections is a major task in livestock production to prevent health constraints and economic losses. However, resistance to established anthelmintic substances already impedes effective anthelmintic treatment in many regions worldwide. Thus, there is an obvious need for sensitive and reliable methods to assess the resistance status of at least the most important nematode populations. Several single nucleotide polymorphisms (SNPs) in the β-tubulin isotype 1 gene of various nematodes correlate with resistance to benzimidazoles (BZ), a major anthelmintic class. Here we descri

Five groups of 20 weaned beef calves were injected subcutaneously with either an ivermectin, a doramectin, an abamectin long-acting formulation, an ivermectin long-acting formulation or a saline control, at turnout and 60 and 120 days later. The animals grazed the same pasture and were sampled and weighed at turnout and 30, 60, 90, 120, 150 and 180 days later. At turnout the mean bodyweights of all the groups were similar and faecal culture showed that they had a mixed strongyle infection of Cooperia, Haemonchus, Ostertagia, Oesophagostomum and Trichostrongylus species. We analyzed the content of the abomasum (n = 79) and small intestine (n = 83) of Spanish ibex from Sierra Nevada Natural Park, southern Spain. Fifteen species of trichostrongylid nematodes were identified, 4 of which were found for the first time in this host, i.e., Nematodirus fillicollis, N. oiratianus, Ostertagia lyrata, and O. ostertagi. Teladorsagia circumcincta and Marshallagia marshalli were the most abundant abomasal species, whereas N. abnormalis, N. davtiani, and N. oiratianus were dominant in the small intestine. Counts of both abomasal and intestinal nematodes were generally low (y
