Oxalic acid supported Si-18F-radiofluorination: one-step radiosynthesis of N-succinimidyl 3-(di-tert-butyl[18F]fluorosilyl)benzoate ([18F]SiFB) for protein labeling.

N-Succinimidyl 3-(di-tert-butyl[(18)F]fluorosilyl)benzoate ([(18)F]SiFB), a novel synthon for one-step labeling of proteins, was synthesized via a simple (18)F-(19)F isotopic exchange. A new labeling technique that circumvents the cleavage of the highly reactive active ester moiety under regular basic (18)F-labeling conditions was established. In order to synthesize high radioactivity amounts of [(18)F]SiFB, it was crucial to partially neutralize the potassium oxalate/hydroxide that was used to elute (18)F(-) from the QMA cartridge with oxalic acid to prevent decomposition of the active ester

Insights into the 1O2 generation of activation of persulfate by zinc ferrite loaded on oxalic acid-modified activated carbon: Interface mechanism and molecular dynamics.

Developing carbonaceous composites with strong non-radical activity and recyclability is key for sustainable wastewater purification. Herein, a novel photocatalytic process was designed using zinc ferrite loaded on oxalic acid-modified powdered activated carbon (ZFO/OA/PAC) to activate persulfate (PDS) under visible light (VL) irradiation. ZFO was in-situ grown onto OA/PAC via C=O bridging, forming stable Zn/Fe-O-C bonds that suppressed magnetic agglomeration and enabled directional electron transfer. The optimized interfacial binding energy of 22.45 kcal/mol reduced the activation energy barr

Influence of water and sodium diuresis and furosemide on urinary excretion of vitamin B(6), oxalic acid and vitamin C in chronic renal failure.

INTRODUCTION: Several errors in mechanisms of specific antigen response are manifested as cutaneous inflammation. Some clinical features required to fulfil the diagnostic criteria are inapplicable to paediatric patients, as they develop over a prolonged period of time or in older age. Effective treatment requires identification of the defective mechanism. Biopsy of the lesions is considered a gold standard in diagnostics; however invasive methods should be avoided in children. Cytokine-producing T helper cells are important players in skin immune homeostasis. Distribution of selected subsets i

Inhibition of calcium oxalate crystal growth in vitro by uropontin: another member of the aspartic acid-rich protein superfamily.

The majority of human urinary stones are primarily composed of calcium salts. Although normal urine is frequently supersaturated with respect to calcium oxalate, most humans do not form stones. Inhibitors are among the multiple factors that may influence the complex process of urinary stone formation. We have isolated an inhibitor of calcium oxalate crystal growth from human urine by monoclonal antibody immunoaffinity chromatography. The N-terminal amino acid sequence and acidic amino acid content of this aspartic acid-rich protein, uropontin, are similar to those of other pontin proteins from

Selenium oxide nanostructure-based electrodes for rapid and simultaneous electrochemical determination of oxalic and ascorbic acids in food matrices.

The development of nanostructured electrochemical platforms for rapid and selective sensing of coexisting biomolecules remains a key challenge in food analysis. Herein, selenium oxide nanostructures (SeO2NSs) were synthesized and integrated into carbon-based electrodes to enable the simultaneous electrochemical detection of ascorbic acid (AA) and oxalic acid (OA) in complex food matrices. Comprehensive morphological, structural, and electrochemical characterization studies confirmed the uniform distribution, high surface area, and excellent redox activity of the SeO2NSs. Under optimized chrono

Short Chain Fatty Acids Prevent Glyoxylate-Induced Calcium Oxalate Stones by GPR43-Dependent Immunomodulatory Mechanism.

Calcium oxalate (CaOx) stones are the most common type of kidney stones and are associated with high recurrence, short chain fatty acids (SCFAs), and inflammation. However, it remains uncertain whether SCFAs affect the formation of CaOx stones through immunomodulation. We first performed mass cytometry (CyTOF) and RNA sequencing on kidney immune cells with glyoxylate-induced CaOx crystals (to elucidate the landscape of the associated immune cell population) and explored the role of SCFAs in renal CaOx stone formation through immunomodulation. We identified 29 distinct immune cell subtypes in k

Development of a bioresource-based adsorbent from activated coconut shell and chitosan-oxalic acid for safranin O dye removal: RSM optimization and circular resource management.

This work demonstrates the design of a bioresource-based adsorbent for the effective removal of organic dyes, embodying the principles of circular resource management by transforming waste into a water-treatment resource. Thus, the current study aims to valorize activated lignocellulosic biomass (coconut shell) combined with chitosan-oxalic acid to develop an eco-friendly adsorbent of cross-linked chitosan-oxalic acid/activated coconut shell (CTS-OX/CS-H) for the uptake of safranin O (SFO) dye from water systems. Several methods, including pHpzc, XRD, CHNS-O, FTIR, BET, and FESEM, were used to

PRMT1-mediated methylation of UBE2m promoting calcium oxalate crystal-induced kidney injury by inhibiting fatty acid metabolism.

Calcium oxalate (CaOx) is the most common type of kidney stone, and its crystal deposition can induce oxidative stress, inflammatory responses, and cell death. This further aggravates kidney structural and functional damage, which in turn, promotes kidney stone recurrence, forming a vicious cycle of repeated stone formation and renal injury. Therefore, identifying precise and effective therapeutic targets is crucial to prevent the damage and inflammation caused by kidney stones. Protein arginine methyltransferase 1 (PRMT1) is a well-known epigenetic regulatory enzyme involved in renal metaboli

Gut Microbiota-Isoallolithocholic Acid Crosstalk Promotes Calcium Oxalate Kidney Stone Formation via PARP1-Mediated Parthanatos.

Bile acids have been implicated in calcium oxalate (CaOx) nephrolithiasis. Here, we employ multi-omics approaches to identify isoallolithocholic acid (isoalloLCA) as the key bile acid elevated in the feces, serum, kidney, and urine of CaOx rats, confirmed by spatial metabolomics. 16S sequencing and metagenomic analyses indicate that elevated isoalloLCA levels in CaOx rats or individuals are likely of microbial origin. Mechanistically, isoalloLCA binds to PARP1 via specific molecular interactions validated by surface plasmon resonance and cellular thermal shift assays. Knockdown of PARP1 by ade

An Ultrasmall Self-Assembled Gallic-Acid-Based Natural Multifunctional Defense Networks for Therapeutic Application in Calcium Oxalate Nephropathies.

Kidney stones, which have high prevalence and recurrence rates, often cause severe oxidative damage and inflammation. The ultrasmall hydrodynamic diameter of nanoparticles is crucial for their enrichment in the kidneys to exert biological activity. However, integrating crystallization inhibition and therapeutic functions into a single ultrasmall nanoparticle is challenging. A novel ultrasmall iron (Fe)-gallic acid (Ga) metal-phenolic networks (Fe-Ga MPNs) is developed for treating calcium oxalate (CaOx) nephropathies. These MPNs can specifically adsorb on the high-energy ( 1 ¯ 01 $\bar{1}01$

Untargeted and targeted metabolomics reveal bile acid profile changes in rats with ethylene glycol-induced calcium oxalate nephrolithiasis.

Calcium oxalate (CaOx) nephrolithiasis is a prevalent disorder linked to metabolism. Examining metabolic alterations could potentially give an initial understanding of the origins of CaOx nephrolithiasis. This study aims to determine gut metabolic biomarkers differentiating CaOx nephrolithiasis utilizing untargeted and targeted metabolomics. CaOx nephrolithiasis model rats were built by 1% ethylene glycol administration. Histologic staining and renal function measurement revealed the presence of crystals in the lumen of the renal tubules, the renal injury and interstitial fibrosis in CaOx rats

New salicylic acid derivatives, double inhibitors of glycolate oxidase and lactate dehydrogenase, as effective agents decreasing oxalate production.

The synthesis and biological evaluation of double glycolate oxidase/lactate dehydrogenase inhibitors containing a salicylic acid moiety is described. The target compounds are obtained in an easily scalable two-step synthetic procedure. These compounds showed low micromolar IC50 values against the two key enzymes in the metabolism of glyoxylate. Mechanistically they behave as noncompetitive inhibitors against both enzymes and this fact is supported by docking studies. The biological evaluation also includes in vitro and in vivo assays in hyperoxaluric mice. The compounds are active against the

Mitigating cadmium stress in rice (Oryza sativa L.) using succinic and oxalic acids with focus on cellular integrity and antioxidant responses.

Soil contamination with toxic heavy metals [such as cadmium (Cd)] is becoming a serious global problem due to the rapid development of the social economy. Organic chelating agents such as succinic acid (SA) and oxalic acid (OA) are more efficient, environmentally friendly, and biodegradable compared to inorganic chelating agents and they enhance the solubility, absorption, and stability of metals. To investigate this, we conducted a pot experiment to assess the impact of SA (0.25 and 0.5 mM) and OA (0.25 and 0.5 mM) on enhancing the phytoremediation of Cd under its toxic concentration of 0.1 m

Malic acid supplementation increases urinary citrate excretion and urinary pH: implications for the potential treatment of calcium oxalate stone disease.

BACKGROUND AND PURPOSE: Raising urinary pH and citrate excretion with alkali citrate therapy has been a widely used treatment in calcium nephrolithiasis. Citrate lowers ionized Ca(+2) concentrations and inhibits calcium salt precipitation. Conservative alternatives containing citrate such as fruit juices have been investigated and recommended. Any compound that induces systemic alkalosis will increase citraturia. Malate, a polycarboxylic anion like citrate, is a potential candidate for chelating Ca(+2) and for inducing systemic alkalinization. We undertook to investigate these possibilities.

Aluminium resistance requires resistance to acid stress: a case study with spinach that exudes oxalate rapidly when exposed to Al stress.

Spinach is a vegetable with a high oxalate concentration in its tissues. Oxalate efflux from spinach (Spinacia oleracea L. cv. Quanneng) roots was rapidly stimulated (within 30 min) by aluminium (Al) treatment. The efflux was constant within 6 h, but increased with increasing Al concentration. The efflux was confined to the root tip (0-5 mm), which showed a 5-fold greater efflux than the root zone distal to the tip (5-10 mm). Oxalate efflux could not be triggered by treatment with the trivalent cation lanthanum or by phosphorus deficiency, indicating that the efflux was specific to the Al trea

Phenylalanine primes renal tubular redox vulnerability and calcium oxalate crystal retention through a PPARδ-CPT1A fatty-acid oxidation axis.

Calcium oxalate (CaOx) nephrolithiasis is usually framed as a physicochemical disorder of urinary supersaturation, yet stable crystal retention also requires a permissive tubular epithelial state. Here, we tested whether circulating phenylalanine (Phe) functions as a systemic priming factor that sensitizes renal tubular epithelium to CaOx crystal retention under lithogenic stress. Targeted amino-acid metabolomics in patients with CaOx stones, two-sample Mendelian randomization, glyoxylate-induced mouse nephrolithiasis, HK-2 cell injury assays, renal transcriptomics, pathway-rescue experiments

STAT6 promoting oxalate crystal deposition-induced renal fibrosis by mediating macrophage-to-myofibroblast transition via inhibiting fatty acid oxidation.

OBJECTIVE AND DESIGN: Kidney stones commonly occur with a 50% recurrence rate within 5 years, and can elevate the risk of chronic kidney disease. Macrophage-to-myofibroblast transition (MMT) is a newly discovered mechanism that leads to progressive fibrosis in different forms of kidney disease. In this study, we aimed to investigate the role of MMT in renal fibrosis in glyoxylate-induced kidney stone mice and the mechanism by which signal transducer and activator of transcription 6 (STAT6) regulates MMT. METHODS: We collected non-functioning kidneys from patients with stones, established glyo

Ursolic acid treats renal tubular epithelial cell damage induced by calcium oxalate monohydrate via inhibiting oxidative stress and inflammation.

Ursolic acid (UA) has been proved to have antioxidant and anti-inflammatory effects. However, it is not clear whether it has a protective impact on kidney damage induced by crystals of calcium oxalate monohydrate (COM). This work aimed to make clear the potential mechanism of UA protecting COM-induced kidney damage. The results manifested that high- and low-dose UA reduced COM crystals in COM rats' kidney, down-regulated urea, creatinine, and neutrophil gelatinase-associated lipocalin (NGAL) levels in rat plasma, declined kidney tissue and HK-2 cell apoptosis, inhibited Bax expression but elev
