Self-Propelled Magnesium Microrobots With Enzyme-Encapsulated Metal-Organic Framework for Accelerated Chemical Warfare Agent Degradation in Harsh Environments.

The fast and efficient degradation of chemical warfare agents (CWA) is an unmet challenge due to the vulnerability of catalytic and biocatalytic reactions to various environmental conditions and their slow mass transfer conditions. While the phosphotriesterase (PTE) enzyme is highly efficient for CWA neutralization, its practical utility is greatly limited by its poor stability under variable environmental conditions. Furthermore, the efficiency of traditional PTE platforms relies solely on passive diffusion, resulting in slow mass transport and degradation kinetics. Here, we describe PTE-enca

Turning Color Confusion to Clarity: A Layered Ce-Based Nanozyme Drives Rapid Wavelength Transition for Discriminative Detection of Nerve Agent during Decontamination.

Organophosphate (OP)-based nerve agents (NAs) pose serious risks to human life, demanding materials that facilitate rapid decontamination and reliable detection with selectivity. Ce-based metal organic frameworks (MOFs) are considered as promising candidates due to their high reactivity. However, their intrinsic yellow color misleads detection of NAs such as paraoxon, which releases yellow p-nitrophenol. Here, we report a bioinspired 2D metal-organic layer (MOL) construct, Ce-BTB-MOL, a phosphotriesterase nanozyme with abundant binding sites that not only facilitate rapid degradation of paraox

Polyionic complexes of butyrylcholinesterase and poly-l-lysine-g-poly(ethylene glycol): Comparative kinetics of catalysis and inhibition and in vitro inactivation by proteases and heat.

We previously reported that recombinant human butyrylcholinesterase (rhBChE) complexed with a series of copolymers of poly-l-lysine (PLL) with grafted (polyethylene) glycol (PEG) (i.e., PLL-g-PEG) showed reduced catalytic activity but relatively similar concentration-dependent inactivation of the organophosphorus inhibitor paraoxon. Herein, we compared the kinetics of catalysis (using butyrylthiocholine as the substrate) and inhibition (using four different inhibitors) of free and copolymer-complexed rhBChE. Using scanning electron microscopy, polyionic complexes of rhBChE with three different

Bioremediation of organophosphorus compounds using highly stable packed bed microreactor developed with OPH (organophosphorus hydrolase) coated nanofiber.

Cellulose nanofibers (CNF) provide a large surface area for enzyme immobilization and facilitate easy recovery from solution, enabling repeated use. The crosslinked coating of organophosphorus hydrolase (OPH) on these nanofibers significantly enhanced enzyme stability compared to the free form. Among the tested systems, diethylaminoethyl cellulose/magnetic particle-OPH (DEAE/MP-OPH) exhibited the highest activity retention, maintaining approximately 90% of its initial activity after 30 days. Similarly, carboxymethyl cellulose/margnetic nanopaticle-OPH (CMC/MP-OPH) and cellulose nanofiber-OPH (

Cholinergic Activation Enhances Resistance to Oral Salmonella Infection by Modulating Innate Immune Defense Mechanisms at the Intestinal Barrier.

Inflammation is a crucial defense mechanism that protects the body from the devastating effects of invading pathogens. However, an unrestrained inflammatory reaction may result in systemic manifestations with dire consequences to the host. The extent of activation of the inflammatory response is tightly regulated through immunological and neural pathways. Previously, we demonstrated that cholinergic stimulation confers enhanced protection in experimental animals orally infected with virulent Salmonella enterica serovar Typhimurium. In this study, we investigated the mechanism by which this enh

Role of metabolism in parathion-induced hepatotoxicity and immunotoxicity.

The objective of this study was to investigate whether metabolic activation of parathion by cytochrome P-450s (CYPs) was responsible for pesticide-induced hepatotoxicity and immunotoxicity. Initially, to investigate parathion metabolism in vitro, the production of paraoxon and p-nitrophenol, major metabolites of parathion, was determined by high-performance liquid chromatography (HPLC). Subsequently, metabolic fate and CYP enzymes involved in the metabolism of parathion were partially monitored in rat liver microsomes in the presence of the NADPH-generating system. Among others, phenobarbital

[Decrease of paraoxonase activity in the first day after abdominal operations in women].

INTRODUCTION: Paraoxonase (PON1) is one of enzymes participating in antioxidative defence mechanism, which protects the organism from the results of action of reactive oxygen species. PON1 is an esterase contained in plasma high density lipoproteins (HDL) which protects HDL from peroxidation and plasma membranes from free radical injury. PON1 serum activity is diminished in some diseases (atherosclerosis, diabetes, coronary artery disease) and after cardiosurgery operations. But there is no information about changes in PON1 activity after abdominal operations. THE AIM: Of this study was the a

Acetylcholinesterase in zebrafish embryos as a tool to identify neurotoxic effects in sediments.

In order to clarify the suitability of zebrafish (Danio rerio) embryos for the detection of neurotoxic compounds, the acetylcholinesterase assay was adapted and validated with a series of priority pollutants listed as relevant for the European water policy (Aroclor 1254, 2,3-benzofuran, bisphenol A, chlorpyrifos, paraoxon-methyl, quinoline, and methyl mercury chloride) as well as acetonic extracts from three sediments of known contamination. The acute toxicities of the model substances and the sediment extracts were determined by means of the fish embryo test as specified in OECD TG 236, and c

Micronuclei, Pesticides, and Element Mixtures in Mining Contexts: The Hormetic Effect of Selenium.

The contexts where there are mining and agriculture activities are potential sources of risk to human health due to contamination by chemical mixtures. These contexts are frequent in several Colombian regions. This study explored the potential association between the frequency of micronuclei and pesticides and elements in regions with ferronickel (Montelibano, Córdoba) and gold (Nechí, Antioquia) mining, and a closed native mercury mine (Aranzazu, Caldas), with an emphasis in the potential effect of selenium as a potential chelator. A cross-sectional study was carried out with 247 individuals.

Enzyme Nanoreactor for In Vivo Detoxification of Organophosphates.

A nanoreactor containing an evolved mutant of Saccharolobus solfataricus phosphotriesterase (L72C/Y97F/Y99F/W263V/I280T) as a catalytic bioscavenger was made for detoxification of organophosphates. This nanoreactor intended for treatment of organophosphate poisoning was studied against paraoxon (POX). Nanoreactors were low polydispersity polymersomes containing a high concentration of enzyme (20 μM). The polyethylene glycol-polypropylene sulfide membrane allowed for penetration of POX and exit of hydrolysis products. In vitro simulations under second order conditions showed that 1 μM enzyme in

Unprecedented Alkylation of the Catalytic Histidine in the Aging of Cholinesterases after Inhibition by Organophosphorus Pesticides.

Organophosphorus (OP) compounds pose a serious risk to human health by covalently modifying acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Currently approved oxime therapeutics can reactivate OP-inhibited AChE and BChE, despite significant limitations. The OP-inhibited enzymes undergo a secondary O-dealkylation event, known as aging, for which no currently approved therapeutics are effective as treatments. Many decades of research have studied the aging mechanism in AChE and BChE. It has previously been accepted that aging occurs via a spontaneous O-dealkylation event, by loss o

Identification and characterization of a promiscuous metallohydrolase in metallo-β-lactamase superfamily from a locally isolated organophosphate-degrading Bacillus sp. strain S3wahi.

In this present study, characteristics and structure-function relationship of an organophosphate-degrading enzyme from Bacillus sp. S3wahi were described. S3wahi metallohydrolase, designated as S3wahi-MH (probable metallohydrolase YqjP), featured the conserved αβ/βα metallo-β-lactamase-fold (MBL-fold) domain and a zinc bimetal at its catalytic site. The metal binding site of S3wahi-MH also preserves the H-X-H-X-D-H motif, consisting of specific amino acids at Zn1 (Asp69, His70, Asp182, and His230) and Zn2 (His65, His67, and His137). The multifunctionality of S3wahi-MH was demonstrated through

Brain catecholamine metabolism changes and hypothermia in intoxication by anticholinesterase agents.

Sublethal doses of physostigmine, paraoxon and soman induce a short-lasting fall in rat core temperature potentiated by alpha-methyl-para-tyrosine (alpha-MT) (early effects). When the own hypothermic effect of the anticholinesterase agent has disappeared (late effects), alpha-MT induces a new decrease in temperature. Parallel biochemical studies of catecholamine levels and turnover were performed in several brain areas. The norepinephrine (NE) turnover is generally increased particularly in the hypothalamus, suggesting that NE hypothalamic changes might be linked to a latent perturbation of th

Paraoxonase and susceptibility to organophosphorus poisoning in farmers dipping sheep.

OBJECTIVES: Human serum paraoxonase (PON1) hydrolyses organophosphate pesticides (OPs) entering the blood circulation and tissue fluid thus limiting toxicity. The PON1 coding region has two polymorphisms involving the amino acids at position 55 (Lt<--M) and 192 (Qt<--R), giving rise to isoenzymes which differ in their catalytic rate for the hydrolysis of OPs. We therefore hypothesized that individuals inheriting low activity isoforms of PON1 would be more liable to report symptoms of OP toxicity. METHODS: We have therefore investigated the relationship between PON1 genetic polymorphisms and P
