The superior colliculus contains a discrete region involved in the control of jaw movements: role of GABAA receptors.

The role of GABA(A) receptors in the superior colliculus in the production of rat repetitive jaw movements was examined, as this nucleus receives tonic GABAergic inhibitory inputs from the dorsolateral part of the substantia nigra pars reticulata and the entopeduncular nucleus. Both regions are also connected with the ventrolateral striatum where stimulation of either dopamine or acetylcholine receptors has been found to elicit distinct types of jaw movements in rats. The GABA(A) receptor antagonist bicuculline (50 and 150 ng/0.2 microl per side) dose-dependently produced repetitive jaw moveme

Effects of intranigral cannabinoids on rotational behavior in rats: interactions with the dopaminergic system.

The effect of unilateral intranigral cannabinoid receptor stimulation on rotational behavior was explored. The potent cannabinoid agonist CP 55,940 (5 and 10 micrograms/0.5 microliter) induced contralateral turning when microinjected unilaterally into the substantia nigra pars reticulata. In addition, the cannabinoid agonist markedly attenuated the contralateral rotation induced by the dopamine D1 agonist SKF 82958 and completely reversed the ipsilateral rotation induced by the dopamine D2 agonist quinpirole. In both cases, the coadministration of the cannabinoid agonist together with the D1 o

Chronic neuroleptic treatment in rats produces persisting changes in GABAA and dopamine D-2, but not dopamine D-1 receptors.

The effects of continuous treatment with haloperidol (HAL) or fluphenazine (FLU) for 10 months on dopamine and GABA receptors in the rat brain was examined using in vitro autoradiography. Rats treated with HAL, but not FLU, showed an increase in D-2 receptor binding in the caudate-putamen as revealed by [3H]spiperone. Labeling of D-1 receptors by [3H]SCH23390 revealed no changes in either drug-treated group. Both drug-treated groups, however, exhibited a significant increase in [3H]muscimol binding in substantia nigra, pars reticulata (SNR). These dopaminergic-GABAergic receptor alterations ma

Saccade responses to dopamine in human MPTP-induced parkinsonism.

Depletion of dopamine content in the substantia nigra resulting from 1-methyl-4-phenyl-1,2,5,6-tetrahydropyridine (MPTP) toxicity produces parkinsonism. Management of 3 patients with MPTP-induced parkinsonism required drug holidays during which there was a state of dopamine depletion followed by dopamine replacement. We used this opportunity to study the effect of the selective loss of pars compacta dopaminergic cells on vertical and horizontal saccade (fast) eye movements. During the drug holidays, visually guided saccades were hypometric and had long latencies but retained a normal saccade v

[A case of Hallervorden-Spatz disease with magnetic resonance imaging data].

Hallervorden-Spatz disease (HSD) is an uncommon disorder, progressive and degenerative of the basal ganglia. It begins in the first or second decade of life and is characterized by a dominant extrapiramidal signs, dystonia and progressive dementia. It is autosomic recessive, although sporadic cases are seen in 15%. There is no biological marker for the disease. The post-mortem findings include iron deposits in the globus pallidum and pars reticulata of the substantia nigra. Magnetic resonance imaging (MRI) in T2 shows symmetric hypointense lesions in both globus pallidum with a hyperintense ce

Tolerance to diazepam and methyl-beta-carboline-3-carboxylate measured in substantia nigra of benzodiazepine tolerant rats.

The spontaneous activity of neurons in the pars reticulata of substantia nigra (SNpr) was studied in chloral hydrate anesthetized rats. As a function of dose, intravenous diazepam decreased, and methyl-beta-carboline-3-carboxylate (beta CCM) increased discharge frequency. Two days after terminating a one week treatment with flurazepam (FZP), both diazepam and beta CCM showed decreased ability to alter SNpr neuronal activity. Neither residual FZP nor down-regulation of benzodiazepine receptors can account for these results. In contrast, behavioral testing revealed no change in the ability of i.

Exofocal alterations in opioidergic receptor densities following focal cerebral ischemia in the mouse.

In previous studies of our group, we have reported differential alterations in opioidergic receptor subtypes densities in infarcted and periinfarcted brain tissue following middle cerebral artery occlusion (MCAO) in mice. Other studies have also described subcortical alterations consecutive to focal cortical ischemia. For a better understanding of ischemic processes in exofocal areas, we have investigated the evolution of opioidergic receptors following focal cortical ischemia through the quantification of relative binding densities, B(max) and K(d) values for the mu, delta, and kappa subtypes

The neuronal mechanism underlying parkinsonism and dyskinesia, and differential roles of the putamen and caudate nucleus.

The basal ganglia were manipulated pharmacologically by using GABA antagonists and also 1-methyl-14-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in rats, cats, and monkeys, and neuronal activities were recorded from the basal ganglia. It was concluded that (a) in parkinsonism, neuronal activities of the caudate nucleus decreased, those of the pars reticulata of the substantia nigra and internal segment of the globus pallidus increased, and those of the ventromedial nucleus of the thalamus depressed markedly; (b) in dyskinesia, periodical discharges of the caudate-putamen complex were the primary e

Dopamine D4 receptor stimulation prevents nigrostriatal dopamine pathway activation by morphine: relevance for drug addiction.

Morphine is one of the most effective drugs used for pain management, but it is also highly addictive. Morphine elicits acute and long-term adaptive changes at cellular and molecular level in the brain, which play a critical role in the development of tolerance, dependence and addiction. Previous studies indicated that the dopamine D4 receptor (D4 R) activation counteracts morphine-induced adaptive changes of the μ opioid receptor (MOR) signaling in the striosomes of the caudate putamen (CPu), as well as the induction of several Fos family transcription factors. Thus, it has been suggested tha

Nigra-Subthalamic Dopaminergic Circuitry Modulates and Represents Distinct Pain Modality in Physiological and Pain States in Mice.

Dopaminergic (DA) neurons in the substantia nigra pars compacta (SNc) degenerate in Parkinson's disease (PD). Although pain is a common non-motor symptom in PD, it remains unclear whether and how degeneration of SNc DA neurons contributes to hyperalgesia. In the present study, we revealed a nigro-subthalamic DA circuit, composed of a subset of SNc DA neurons, the SNc DA projection to the subthalamic nucleus (STN), and the downstream STN neurons. These components regulate mechanical, but not thermal, pain threshold on the contralateral side, exhibiting distinct responses to mechanical and therm

Regional changes in 2-deoxyglucose uptake associated with neuroleptic-induced tardive dyskinesia in the Cebus monkey.

The neural mechanisms that mediate a primate model of tardive dyskinesia have been investigated using the 2-deoxyglucose (2-DG) uptake technique. Three groups of Cebus monkeys were used. Some of the animals received long-term neuroleptic treatment. These animals were allotted to one of two groups depending on whether they developed tardive dyskinesia or not. A third group of animals served as untreated controls. The neuroleptic-treated dyskinetic animals showed reduced uptake of 2-DG in the medial segment of the globus pallidus and in the ventral anterior (VA) and ventral lateral (VL) nuclei o

Intra-nigra infusion of AMPA attenuates dopamine-dependent rotation in the rat.

It has previously been shown that glutamatergic overactivity of the subthalamic nucleus (STN) is involved in hypokinetic movement disorders such as Parkinson's disease. Conversely, it has been hypothesized that hyperkinetic behavioral syndromes may be associated with reduced glutamatergic transmission of the STN to its target areas, the substantial nigra pars reticulata (SNR) and the globus pallidus pars interna (GPI). In the present experiment, apomorphine injected systemically into unilaterally dopamine-denervated rats induced the hyperkinetic syndrome of contralateral rotation. The involvem

The role of the anterior pretectal nucleus in pain modulation: A comprehensive review.

Descending pain modulation involves multiple encephalic sites and pathways that range from the cerebral cortex to the spinal cord. Behavioral studies conducted in the 1980s revealed that electrical stimulation of the pretectal area causes antinociception dissociation from aversive responses. Anatomical and physiological studies identified the anterior pretectal nucleus and its descending projections to several midbrain, pontine, and medullary structures. The anterior pretectal nucleus is morphologically divided into a dorsal part that contains a dense neuron population (pars compacta) and a ve

Time- and dose-dependent effects of the serotonergic agent quipazine on regional cerebral metabolism in rats.

The time course and the relation to dose of regional cerebral metabolic rates for glucose (rCMRglc) were measured in awake male adult Fischer-344 rats after administration of quipazine, a serotonin 5-HT2-3 receptor agonist. rCMRglc was determined, using the quantitative autoradiographic [14C]deoxyglucose technique, in 92 brain regions at 30, 60, 90 and 120 min after quipazine 20 mg/kg i.p. and at 60 min after quipazine 5 mg/kg i.p. Peak metabolic effects were observed 60 min after quipazine 20 mg/kg i.p. when rCMRglc was significantly elevated in 27 (29%) brain regions (mean rise 17%). Quipazi

Environmental risk factors and Parkinson's disease: selective degeneration of nigral dopaminergic neurons caused by the herbicide paraquat.

Environmental toxicants and, in particular, pesticides have been implicated as risk factors in Parkinson's disease (PD). The purpose of this study was to determine if selective nigrostriatal degeneration could be reproduced by systemic exposure of mice to the widely used herbicide paraquat. Repeated intraperitoneal paraquat injections killed dopaminergic neurons in the substantia nigra (SN) pars compacta, as assessed by stereological counting of tyrosine hydroxylase (TH)-immunoreactive and Nissl-stained neurons. This cell loss was dose- and age-dependent. Several lines of evidence indicated se

Locating the Sex Determining Region of Linkage Group 12 of Guppy (Poecilia reticulata).

Despite over 100 years of study, the location of the fully sex-linked region of the guppy (Poecilia reticulata) carrying the male-determining locus, and the regions where the XY pair recombine, remain unclear. Previous population genomics studies to determine these regions used small samples from recently bottlenecked captive populations, which increase the false positive rate of associations between individuals' sexes and SNPs. Using new data from multiple natural populations, we show that a recently proposed candidate for this species' male-determining gene is probably not completely sex-lin

Evidence for a glutamatergic projection from the zona incerta to the basal ganglia of rats.

This study explores the organisation and neurochemical nature of the projections from the zona incerta (ZI) to the basal ganglia. Sprague-Dawley rats were anaesthetised with ketamine (100 mg/kg) and Rompun (10 mg/kg), and injections of cholera toxin subunit B were made into each of the following nuclei: the ZI, the substantia nigra (SN), the pedunculopontine tegmental nucleus (PpT), and the entopeduncular nucleus (Ep). Brains were aldehyde fixed, sectioned, and processed using standard methods. Tracer-labelled sections were then doubly labelled with antibodies to glutamate (Glu), nitric oxide

Anatomical and cellular localization of melatonin MT1 and MT2 receptors in the adult rat brain.

The involvement of melatonin in mammalian brain pathophysiology has received growing interest, but information about the anatomical distribution of its two G-protein-coupled receptors, MT1 and MT2 , remains elusive. In this study, using specific antibodies, we examined the precise distribution of both melatonin receptors immunoreactivity across the adult rat brain using light, confocal, and electron microscopy. Our results demonstrate a selective MT1 and MT2 localization on neuronal cell bodies and dendrites in numerous regions of the rat telencephalon, diencephalon, and mesencephalon. Confoca
