Multiple interspecific hybridization and microsatellite mutations provide clonal diversity in the parthenogenetic rock lizard Darevskia armeniaca.

BACKGROUND: The parthenogenetic Caucasian rock lizard Darevskia armeniaca, like most other parthenogenetic vertebrate species, originated through interspecific hybridization between the closely related sexual Darevskia mixta and Darevskia valentini. Darevskia armeniaca was shown to consist of one widespread allozyme clone and a few rare ones, but notwithstanding the origin of clonal diversity remains unclear. We conduct genomic analysis of D. armeniaca and its parental sexual species using microsatellite and SNP markers to identify the origin of parthenogenetic clonal lineages. RESULTS: Four

Wolbachia: endosymbiont of onchocercid nematodes and their vectors.

BACKGROUND: Wolbachia is an obligate intracellular maternally transmitted, gram-negative bacterium which forms a spectrum of endosymbiotic relationships from parasitism to obligatory mutualism in a wide range of arthropods and onchocercid nematodes, respectively. In arthropods Wolbachia produces reproductive manipulations such as male killing, feminization, parthenogenesis and cytoplasmic incompatibility for its propagation and provides an additional fitness benefit for the host to protect against pathogens, whilst in onchocercid nematodes, apart from the mutual metabolic dependence, this bact

Sexual characteristics and spermatogenesis in males of the parthenogenetic gecko Lepidodactylus lugubris (Reptilia, Gekkonidae).

Obligately parthenogenetic lizards usually are all-female populations of hybrids producing diploid oocytes by premeiotic endomitosis and quasi-normal meiosis. In an all-female strain of the gekkonid lizard Lepidodactylus lugubris several phenotypic males arose spontaneously. The sexual characteristics of these males were studied using light and electron microscopy and compared with normal males of the bisexual genus Lygodactylus. Emphasis was layed on morphology of seminiferous tubules, occurrence of spermatogenic stages and ultrastructure of spermatozoa. The phenotypic males possessed preanal

TRIPLOID IN PARTHENOGENETIC SPECIES OF THE TEIID LIZARD, GENUS CNEMIDOPHORUS.

Mitotic figures from the bone marrow of three parthenogenetic species of Cnemidophorus (whiptail lizards) exhibit chromosome numbers in the triploid range. The species are C. velox with 68 chromosomes, C. tesselatus with 69, and C. exsanguis with 70. A fourth parthenogenetic species, C. perplexus, has 46 chromosomes, the same number as the bisexual subspecies C. tigris septentrionalis.

Severe Fanconi Anemia phenotypes in Fancd2 depletion mice.

Fanconi anemia (FA) is a genetic disorder characterized by congenital malfunction, bone marrow failure and hypersensitivity to DNA damage. FANCD2 protein play the central role in FA pathway. To study the in vivo role of FANCD2, we generated and characterized a new Fancd2 knockout mouse strain with 7bp deletion in Fancd2 gene 5' terminus using Crispr-Cas9 in congenic C57BL/6J background. This Fancd2-/- mice displayed similar but overall more severe manifestation than the previous ES cell targeted Fancd2 model. These features include increased embryonic and postnatal lethality rate, higher incid

Widespread prevalence of wolbachia in laboratory stocks and the implications for Drosophila research.

Wolbachia is an intracellular microbe harbored by a wide variety of arthropods (including Drosophila) and filarial nematodes. Employing several different strategies including male killing, induced parthenogenesis, cytoplasmic incompatibility, and feminization, and acting by as-yet-unknown mechanisms, Wolbachia alters host reproduction to increase its representation within a population. Wolbachia is closely associated with gametic incompatibility but also interacts with Drosophila in other, little understood ways. We report here significant and widespread infection of Wolbachia within laborator

Discovery of a new Wolbachia supergroup in cave spider species and the lateral transfer of phage WO among distant hosts.

Wolbachia are widespread intracellular bacteria infecting the major classes of arthropods and some filarial nematodes. In arthropods, Wolbachia have evolved various intriguing reproductive manipulations, including cytoplasmic incompatibility, parthenogenesis, feminization, and male killing. Sixteen supergroups of Wolbachia have been identified, named A-Q (except G). Though Wolbachia present great diversity in arthropods, spiders, especially cave spiders, are still a poorly surveyed group of Wolbachia hosts. Here, we report a novel Wolbachia supergroup from nine Telema cave spiders (Araneae: Te

Optimizing oocyte electroporation for genetic modification of porcine embryos: Evaluation of the parthenogenetic activation.

In reproductive biology, understanding the effects of novel techniques on early embryo development is of paramount importance. To date, the effects of electrical activation on oocytes prior to in vitro fertilization (IVF) are not well understood. The aim of this study was to investigate the effects of oocyte electroporation prior to IVF on embryo development and to differentiate between true embryos and parthenotes by using a TPCN2 knock-out (KO) male to evaluate the presence of the KO allele in the resulting blastocysts. The study consisted of three experiments. The first one examined oocyte

The causative agents of fascioliasis in animals and humans: Parthenogenetic Fasciola in Asia and other regions.

Parthenogenetic Fasciola is the causative agent of fascioliasis in animals and humans and is widely distributed in Asian countries, such as Japan, South Korea, China, Vietnam, Thailand, the Philippines, Myanmar, Bangladesh, Nepal, and India. Parthenogenetic Fasciola geographically originated from central and eastern China, where it exists between the habitats of Fasciola hepatica and Fasciola gigantica; it likely appeared thousands of years ago following hybridization between F. hepatica and F. gigantica. Parthenogenetic Fasciola consists of diploids and triploids that possess nuclear genome o

Viable Triploid Honey Bees (Apis mellifera capensis) Are Reliably Produced in the Progeny of CO2 Narcotised Queens.

The haplodiploid system of sex determination of Hymenoptera acts as an exaptation for species to evolve novel forms of asexual reproduction including thelytoky (clonal offspring of the mother). During normal reproduction in Hymenoptera, three of the four products of meiosis that are present in newly-laid eggs are lost as polar bodies, while the remaining pronucleus either develops as a haploid male or fuses with a sperm nucleus to produce a diploid zygote. In contrast, in thelytokous reproduction, which is uncommon but taxonomically widespread, two of the four products of meiosis fuse, as if o

Effect of melatonin treatment on the developmental potential of parthenogenetic and somatic cell nuclear-transferred porcine oocytes in vitro.

Melatonin secreted from the mammalian pineal gland is a free-radical scavenger that protects tissues from cell damage. The present study examined the effects of addition of melatonin to the culture medium on the developmental potential of parthenogenetic and somatic cell nuclear-transferred (SCNT) porcine oocytes. Supplementation of the maturation medium with melatonin did not increase the maturation rate, the proportion of oocytes that cleaved and developed into blastocysts after parthenogenetic activation, or the blastocyst cell number compared to controls. When 10-7 M melatonin was added to

The molecular through ecological genetics of abnormal abdomen in Drosophila mercatorum. I. Basic genetics.

The abnormal abdomen syndrome (aa) in Drosophila mercatorum is characterized by the persistence of juvenilized cuticle on the adult abdomen. The aa phenotype is shown to depend on at least two X-linked genetic elements that are about one map unit apart near the centromeric end of the X chromosome. These two genetic elements are necessary for aa expression; one behaves as a dominant element and the other as a recessive. Overlaying these genetic studies upon molecular work reported elsewhere, it is argued that the dominant element is the presence of a 5 kb insertion in a majority of the X-linked

Full-term development after transplantation of parthenogenetic embryonic nuclei into fertilized mouse eggs.

Diploid parthenogenetically activated oocytes were obtained after gonadotropin-induced ovulation of virgin females of the LT/Sv (LT) inbred mouse strain. These oocytes cleave spontaneously and develop into blastocysts which implant in the uterus but die within a few days. We examined the developmental potential of nuclei from parthenogenetic embryos after transplantation into fertilized eggs. The inner cell mass (ICM) and trophectoderm (TE) of LT parthenogenetic blastocysts were mechanically isolated and dissociated into single cells. Their nuclei were then injected into fertilized C57BL/6J eg

Chondrodystrophy: an inherited lethal condition in turkey embryos.

Chondrodystrophy was found to occur as an embryonic lethal in a special line of turkeys. Inheritance of the condition was studied in embryos produced parthenogenetically from virgin dams and also in embryos from the same dams in bisexual matings. Expression was similar to other forms of the disorder encountered in various avian species. Micromelia and brachycephaly were recognized in affected embryos after 9 days while lethality occurred mainly after 16 days of incubation. A few affected embryos expressed modified phenotypes and these were also nonviable. Modified phenotypes were not observed

Chromatin Immunoprecipitation Assays of Histone Modifications in Daphnia magna.

Daphnia magna is a widely distributed freshwater planktonic crustacean that has been extensively used as a model organism in ecotoxicological studies, due to its sensitivity to environmental pollutants. It has recently received enough attention as a model organism to study transgenerational epigenetic mechanisms, owing to its ability to reproduce via parthenogenesis. This unique ability of D. magna to generate clonal offspring makes it an ideal model organism to study how environmental cues-including environmental pollutants-can affect the epigenome of future generations. This is important sin

Host-associated populations in the lettuce root aphid, Pemphigus bursarius (L.).

Pemphigus bursarius is a host-alternating aphid in which annual rounds of sexual reproduction on its primary host, Populus nigra, are interspersed with parthenogenesis on a range of secondary hosts. Evidence was sought for the existence of genetically distinct populations, associated with different secondary hosts, in P. bursarius. Microsatellite markers revealed that genetically distinct populations were present on three different secondary host species. Microsatellites were also used, in conjunction with mitochondrial DNA sequence variation, to investigate the relationships between aphids on

MADS-BOX PROTEIN3 acts cooperatively with AGAMOUS-like6 and MADS-BOX PROTEIN22 to suppress parthenocarpy in tomato.

Ovule fertilization initiates fruit set, the transition from a quiescent ovary to a growing ovary (fruit), yet how ovary growth is arrested before fertilization remains poorly understood. Previously, we have shown that the ovaries of the tomato (Solanum lycopersicum) AGAMOUS-like6 (SlAGL6) loss-of-function mutant (slagl6CR-sg1) develop normally until anthesis but nevertheless can set fruits independently of fertilization (parthenocarpy). Here, we demonstrate that SlAGL6 suppresses parthenocarpy specifically from the ovules, as reintroducing SlAGL6 into slagl6CR-sg1 using an ovule-specific prom

The miR166-SlHB15A regulatory module controls ovule development and parthenocarpic fruit set under adverse temperatures in tomato.

Fruit set is inhibited by adverse temperatures, with consequences on yield. We isolated a tomato mutant producing fruits under non-permissive hot temperatures and identified the causal gene as SlHB15A, belonging to class III homeodomain leucine-zipper transcription factors. SlHB15A loss-of-function mutants display aberrant ovule development that mimics transcriptional changes occurring in fertilized ovules and leads to parthenocarpic fruit set under optimal and non-permissive temperatures, in field and greenhouse conditions. Under cold growing conditions, SlHB15A is subjected to conditional ha

Increase in concentration of patch test allergen reduces patch test occlusion time to 12 hours without affecting patch test reactivity in patients with parthenium dermatitis.

BACKGROUND: Patch testing is the standard method to diagnose contact allergy. Patches are applied for 48 hours, which is inconvenient to patients in tropical weather. Therefore, we evaluated different patch test occlusion times with increased concentrations of an allergen to determine if occlusion time can be reduced without compromising patch test reactivity. METHODS: Patch test positive patients with parthenium dermatitis were enrolled and patch tested using five different concentrations (10%, 4%, 2%, 1%, and 0.5%) of parthenium extract. The patches were applied in triplicate. The first set
