Recombinant expression and affinity purification of snake venom gland parvalbumin in Escherichia coli.

Parvalbumins (PV) are small, acidic, water soluble and calcium-binding proteins generally present in muscular and nervous tissues. In the present study, we identified and characterized a cDNA clone encoding PV, named AplPV, from a snake (Agkistrodon piscivorus leucostoma) venom gland cDNA library. AplPV belongs to EF-hand proteins with six alpha-helices constituting three EF-hand domains. The deduced amino acid sequence of AplPV is 91% and 68% identical to the previously characterized PVs of Boa constrictor and Cyprinus carpio, respectively. The full-length cDNA was subcloned into the expressi

Altered parvalbumin-positive neuron distribution in basal ganglia of individuals with Tourette syndrome.

Tourette syndrome (TS) is a childhood neuropsychiatric disorder characterized by motor and vocal tics. Imaging studies found alterations in caudate (Cd) and putamen volumes. To investigate possible alterations in cell populations, postmortem basal ganglia tissue from individuals with TS and normal controls was analyzed by using unbiased stereological techniques. A markedly higher total neuron number was found in the globus pallidus pars interna (GPi) of TS. In contrast, a lower neuron number and density was observed in the globus pallidus pars externa and in the Cd. An increased number and pro

Postnatal developmental expression of calbindin, calretinin and parvalbumin in mouse main olfactory bulb.

The distribution of calbindin, calretinin and parvalbumin during the development of the mouse main olfactory bulb (MOB) was studied using immunohistochemistry techniques. The results are as follows: (1) calbindin-immunoreactive profiles were mainly located in the glomerular layer, and few large calbindin-immunoreactive cells were found in the subependymal layer of postnatal day 10 (P10) to postnatal day 40 (P40) mice; (2) no calbindin was detected in the mitral cell layer at any stage; (3) calretinin-immunoreactive profiles were present in all layers of the main olfactory bulb at all stages, e

Age-related changes of parvalbumin immunoreactive neurons in the rat main olfactory bulb.

Parvalbumin (PV) is found in the olfactory system, including the main olfactory bulb, and is thought to be one of the neuroactive substances in olfaction. Changes in PV immunoreactivity in the olfactory system during aging have not been examined. We investigated such changes in the main olfactory bulb (MOB) of the rat at postnatal month 1 (PM 1), PM 3, PM 6, PM 12 and PM 24. PV-IR neurons were almost completely restricted to the external plexiform layer. At PM 1 there were only a few PV-IR neurons; at PM 3, the number of PV-IR neurons was at its greatest but they were not well developed morpho

Vitamin-D-dependent calcium-binding-protein and parvalbumin occur in bones and teeth.

Vit-D-Dependent calcium binding protein and parvalbumin have both been detected in ameloblasts and calcified cartilage by immunohistochemical techniques. These two Ca2+ binding proteins may play a crucial role in the local accumulation of Ca2+ ions during the process of mineralization. The mechanisms underlying the deposition of inorganic substances in bone and teeth during physiologic calcification are still the object of intense debate [1, 2]. The hypotheses concerning the factors controlling the initiation of mineralization can be subdivided into three large categories: enzymatic (or non-en

A distinct lineage pathway drives parvalbumin chandelier cell fate in human interneuron reprogramming.

Direct lineage reprogramming of glial cells to induced neurons has the potential for restoring brain circuits and function in neuronal disorders and states. We introduce three-dimensional (3D) human glia reprogramming into neurons with a GABAergic interneuron phenotype using stem cell-derived human glia. Single-nucleus RNA sequencing of the converted cells demonstrates distinct neuronal clusters within 2 weeks, including a parvalbumin (PV) cluster with high neuronal maturity and features of chandelier interneurons. A lineage trajectory analysis of the glia-to-neuron conversion reveals a distin

Environmental Enrichment Preserves Retrosplenial Parvalbumin Density and Cognitive Function in Female 5xFAD Mice.

The rate of cognitive decline in Alzheimer's disease (AD) varies considerably from person to person. Numerous epidemiological studies point to the protective effects of cognitive, social, and physical enrichment as potential mediators of cognitive decline in AD; however, there is much debate as to the mechanism underlying these protective effects. The retrosplenial cortex (RSC) is one of the earliest brain regions with impaired functions during AD pathogenesis, and its activity is affected by cognitive, social, and physical stimulation, making it a particularly interesting region to investigat

Cortical Layer-Specific Remodelling of Parvalbumin and Perineuronal Net Networks in Alcohol Use Disorder.

BACKGROUND: Alcohol use disorder (AUD) is a chronically relapsing condition marked by a pathological shift in behaviour, where excessive motivational drive predominates over cognitive control. Brodmann area 6 (BA6) is a key cortical region that integrates cognitive control with motor output and striatal circuits. Cellular alterations in the BA6 can shift from flexible, goal-directed planning to habitual, compulsive behaviours. METHOD: Here, we examined cellular changes in the human post-mortem cortex from AUD cases (n = 9) and age-matched controls (n = 10). The density of parvalbumin (PV) neu

The Role of Anterior Insular Parvalbumin GABAergic Interneurons in Impulsivity-Related and Alcohol-Seeking Behaviors.

Adolescent impulsivity is linked to increased alcohol seeking and altered activation of anterior insular PV interneurons. Chemogenetic modulation of these neurons regulates inhibitory control and alcohol-related behaviors, highlighting their role in vulnerability to alcohol use disorder.

Parvalbumin-positive neurons in the medial septum participate in the formation of hippocampal-dependent spatial memory.

GABAergic medial septal neurons play a key role in regulating hippocampus-dependent spatial memory, but the underlying mechanisms are not yet fully understood. In male mice, we establish an object-place recognition impairment model using an acute sleep deprivation protocol. Here we show that parvalbumin-positive neurons in the medial septum regulate object-location memory specificity during encoding by modulating hippocampal place cell population activity. Using in vivo electrophysiology combined with optogenetics, we characterize the role of medial septal parvalbumin-positive neurons in objec

Microglial galectin-3 disrupts parvalbumin interneurons and hippocampal synchrony, driving cognitive deficits.

Sepsis-associated encephalopathy (SAE), a devastating neurological complication of systemic inflammation, affects approximately 70% of patients with sepsis. It not only increases mortality but also leaves survivors with persistent cognitive deficits. However, the mechanisms underlying SAE progression remain incompletely understood. Here, using a lipopolysaccharide (LPS)-induced mouse model of SAE, we identify microglial galectin-3 (Gal-3) as a central pathogenic mediator driving systemic inflammation-induced cognitive impairment. Mechanistically, systemic LPS challenge robustly upregulates mic

Proteomic characterization of cellular responses to the fish allergen β-parvalbumin using ZenoSWATH-MS.

β-Parvalbumin (β-PRVB) is a calcium-binding protein and the major allergenic macromolecule responsible for most IgE-mediated fish allergy reactions. Despite its clinical relevance, the molecular mechanisms underlying interactions between β-PRVB and host cellular systems remain incompletely understood. In this study, high-resolution quantitative proteomics using data-independent acquisition ZenoSWATH-MS was applied to investigate cellular responses associated with exposure to β-PRVB in a controlled experimental model. Parvalbumin 5 potentially regulates Fsh cells but not Lh cells in the pituitary of ricefield eels†.

Parvalbumins (Pvalb) are members of the EF-hand protein superfamily which have been shown to play important roles in the nervous system and muscle. In the present study, the expression and potential roles of parvalbumin 5 (Pvalb5), a member of Parvalbumin subfamily, was characterized in a teleost the ricefield eel. Reverse transcription-polymerase chain reaction (RT-PCR) analysis revealed a very restricted tissue distribution of pvalb5 mRNA, with dominant expression in the pituitary and eye of both female and male ricefield eels. Specific antisera against ricefield eel Pvalb5 were generated, a

Impaired parvalbumin interneurons in the retrosplenial cortex as the cause of sex-dependent vulnerability in Alzheimer's disease.

Alzheimer's disease is a debilitating neurodegenerative disorder with no cure and few treatment options. In early stages of Alzheimer's disease, impaired metabolism and functional connectivity of the retrosplenial cortex strongly predict future cognitive impairments. Therefore, understanding Alzheimer's disease-related deficits in the retrosplenial cortex is critical for understanding the origins of cognitive impairment and identifying early treatment targets. Using the 5xFAD mouse model, we discovered early, sex-dependent alterations in parvalbumin-interneuron transcriptomic profiles. This co

Prefrontal and Hippocampal Parvalbumin Interneurons in Animal Models for Schizophrenia: A Systematic Review and Meta-analysis.

BACKGROUND: Consistent with postmortem findings in patients, most animal models for schizophrenia (SCZ) present abnormal levels of parvalbumin (PV), a marker of fast-spiking GABAergic interneurons, in the prefrontal cortex (PFC) and hippocampus (HIP). However, there are discrepancies in the literature. PV reductions lead to a functional loss of PV interneurons, which is proposed to underly SCZ symptoms. Given its complex etiology, different categories of animal models have been developed to study SCZ, which may distinctly impact PV levels in rodent brain areas. STUDY DESIGN: We performed a qu

Electrical Coupling between Parvalbumin Basket Cells is Reduced after Experimental Status Epilepticus.

Acquired epilepsies, characterized by abnormal increase in hypersynchronous network activity, can be precipitated by various factors including brain injuries which cause neuronal loss and increases in network excitability. Electrical coupling between neurons, mediated by gap junctions, has been shown to enhance synchronous neuronal activity and promote excitotoxic neurodegeneration. Consequently, neuronal gap junctional coupling has been proposed to contribute to development of epilepsy. Parvalbumin expressing interneurons (PV-INs), noted for their roles in powerful perisomatic inhibition and

CTCF in parvalbumin-expressing neurons regulates motor, anxiety and social behavior and neuronal identity.

CCCTC-binding factor (CTCF) is a regulator of chromatin organization and has direct effects on gene transcription. Mutations in CTCF have been identified in individuals with neurodevelopmental conditions. There are wide range of behaviors associated with these mutations, including intellectual disabilities, changes in temperament, and autism. Previous mice-model studies have identified roles for CTCF in excitatory neurons in specific behaviors, particularly in regards to learning and memory. However, the role of CTCF in inhibitory neurons is less well defined. In the current study, specific kn

Seizures, ataxia and parvalbumin-expressing interneurons respond to selenium supply in Selenop-deficient mice.

Mice with constitutive disruption of the Selenop gene have been key to delineate the importance of selenoproteins in neurobiology. However, the phenotype of this mouse model is exquisitely dependent on selenium supply and timing of selenium supplementation. Combining biochemical, histological, and behavioral methods, we tested the hypothesis that parvalbumin-expressing interneurons in the primary somatosensory cortex and hippocampus depend on dietary selenium availability in Selenop-/- mice. Selenop-deficient mice kept on adequate selenium diet (0.15 mg/kg, i.e. the recommended dietary allowan

BACKGROUND: Clinical cross-reactivity between bony fish, cartilaginous fish, frog, and chicken muscle has previously been demonstrated in fish-allergic patients. In indicative studies, two reports of anaphylaxis following the consumption of crocodile meat and IgE-cross-binding were linked to the major fish allergen parvalbumin (PV). This study investigates IgE-binding proteins in crocodile meat with a focus on PV and their clinical relevance. METHODS: Proteins were extracted from muscle tissue of crocodile, three bony fish, and two cartilaginous fish. A cohort of fish-allergic pediatric patie

CSF parvalbumin levels reflect interneuron loss linked with cortical pathology in multiple sclerosis.

INTRODUCTION AND METHODS: In order to verify whether parvalbumin (PVALB), a protein specifically expressed by GABAergic interneurons, could be a MS-specific marker of grey matter neurodegeneration, we performed neuropathology/molecular analysis of PVALB expression in motor cortex of 40 post-mortem progressive MS cases, with/without meningeal inflammation, and 10 control cases, in combination with cerebrospinal fluid (CSF) assessment. Analysis of CSF PVALB and neurofilaments (Nf-L) levels combined with physical/cognitive/3TMRI assessment was performed in 110 naïve MS patients and in 32 controls
