Preconception paternal alcohol exposure exerts sex-specific effects on offspring growth and long-term metabolic programming.

BACKGROUND: Although clinical data support an association between paternal alcohol use and deficits in child neurocognitive development, the relationship between paternal drinking and alcohol-induced growth phenotypes remains challenging to define. Using an established mouse model of chronic exposure, previous work by our group has linked preconception paternal alcohol use to sex-specific patterns of fetal growth restriction and placental dysfunction. The aim of the present study was to investigate the long-term impact of chronic preconception paternal alcohol use on offspring growth and metab

Paternal anabolic-androgenic steroid exposure promotes autism-like behavior in adult mouse offspring of both sexes.

Paternal environmental factors before conception and during sperm development may influence the offspring's health later in life. This study aimed to investigate whether paternal exposure to anabolic-androgenic steroids (AAS) before conception predisposes mouse offspring to autism spectrum disorder (ASD)-like behavior. For this purpose, male Swiss mice were randomly divided into two groups: the control group received peanut oil, while the treated group was administered testosterone propionate (7.5 mg/kg, s.c.) twice a week for five weeks. After this period, these males were mated, and their of

Paternal lead exposure induces transgenerational oxidative stress and down-regulation of antioxidant genes in Drosophila melanogaster.

Lead (Pb) is one of the most prevalent environmental toxicants and is of great concern due to its adverse effects. Despite lead's oxidative properties, studies on the transgenerational effects of paternal Pb exposure remain inadequate. This study aimed to evaluate these effects on oxidative stress and the expression of specific genes using the D. melanogaster model. Male fruit flies were fed a normal diet supplemented with lead acetate as the Pb source for 20 consecutive days. The Pb-exposed males were mated with unexposed females, producing F1 offspring. F1 flies were fed a normal diet, and t

Paternal and Maternal Exposures to Per- and Polyfluoroalkyl Substances and Child Behavioral Difficulties: A Parental Comparison Study.

Per- and polyfluoroalkyl substances (PFAS) are widespread environmental pollutants with documented developmental toxicity. Prior research of prenatal PFAS exposure and offspring neurodevelopment did not consider the possible influence from paternal exposure. Using the INUENDO cohort, we studied 334 father-mother-singleton triads enrolled from antenatal clinics in Greenland, Poland, and Ukraine. We measured five PFAS in parental serum samples collected around the 31 weeks of gestation. We assessed child behavioral difficulties at ages 5-9 years by the parent-rated Strength and Difficulties Ques

Chronic paternal alcohol exposures induce dose-dependent changes in offspring craniofacial shape and symmetry.

Although dose-response analyses are a fundamental tool in developmental toxicology, few studies have examined the impacts of toxicant dose on the non-genetic paternal inheritance of offspring disease and dysgenesis. In this study, we used geometric morphometric analyses to examine the impacts of different levels of preconception paternal alcohol exposure on offspring craniofacial shape and symmetry in a mouse model. Procrustes ANOVA followed by canonical variant analysis of geometric facial relationships revealed that Low-, Medium-, and High-dose treatments each induced distinct changes in cra

"Cocktail" of environmental chemicals and early reproductive outcomes of IVF: The insight from paternal and maternal exposure.

BACKGROUND: Humans are exposed to various chemicals, including organophosphate esters (OPEs), phthalates (PAEs), and phenols. The effects on early reproductive outcomes of in vitro fertilization (IVF) remain unclear. METHODS: We recruited 192 women and 157 men who underwent IVF treatment. A total of forty-nine urinary chemicals were detected, including six OPEs, fifteen PAEs, six parabens, two chlorophenols, nine bisphenols, five benzophenones, and six synthetic phenolic antioxidants. We examined the individual and joint effects of parental chemical exposure on early reproductive outcomes. R

Paternal Exposure to Immunosuppressive and/or Biologic Agents and Birth Outcomes in Patients With Immune-Mediated Inflammatory Diseases.

BACKGROUND & AIMS: We conducted a retrospective cohort study to inform the safety of exposure to immunosuppressive and/or biologic agents around conception in expectant fathers with immune-mediated inflammatory diseases (IMIDs) on birth outcomes. METHODS: Using a deidentified administrative claims database (OptumLabs Data Warehouse), we identified 7453 expectant fathers with IMIDs (inflammatory bowel diseases, rheumatoid arthritis, psoriasis/psoriatic arthritis, and ankylosing spondylitis) linked to newborns with periconception medication exposure between 38 and 60 weeks before the newborn bi

Therapeutic exposures and pubertal testicular dysfunction are associated with adulthood milestones and paternity after childhood cancer.

BACKGROUND: Childhood cancer therapy may cause long-term effects. This cross-sectional study evaluated adulthood milestones in male childhood cancer survivors (CCS). METHODS: The study population comprised 252 male CCS with 6 to 42 years of survival diagnosed at the Children's Hospital in Helsinki (1964-2000) at the age of 0 to 17 years. Sex-, age-, and area of residence-matched population controls were randomly selected from the Finnish national registries. Data on moving away from the parental home, marital status, offspring, and adoption in CCS were compared with the population controls. W

Fetal exposure to maternal and paternal smoking and the risks of wheezing in preschool children: the Generation R Study.

BACKGROUND: Previous studies have suggested that fetal smoke exposure is associated with increased risks of wheezing during childhood. The underlying pathways are unknown. We examined the associations of parental smoking during pregnancy with wheezing in preschool children and whether these associations are explained by postnatal smoke exposure or small for gestational age at birth. METHODS: This study was embedded in a population-based prospective cohort study. Parental smoking was prospectively assessed by questionnaires. Wheezing was reported at 1 to 4 years. Small for gestational age at b

Prenatal exposure to maternal and paternal smoking on attention deficit hyperactivity disorders symptoms and diagnosis in offspring.

The study examined the effect of maternal and paternal smoking during pregnancy on the child's inattention and hyperactivity/impulsivity symptoms, and the risk for attention deficit hyperactivity disorder (ADHD) and oppositional defiant disorder (ODD). Generalized estimating equations, incorporating data from multiple informants (parents and teachers), was used to evaluate levels of ADHD as a function of parental smoking. The risk for ADHD, ODD, and comorbid ADHD and ODD was evaluated using polytomous logistic regression. We found that maternal, but not paternal, smoking was significantly asso

Paternal Nicotine/Ethanol/Caffeine Mixed Exposure Induces Offspring Rat Dysplasia and Its Potential "GC-IGF1" Programming Mechanism.

Clinical and animal studies suggest that paternal exposure to adverse environments (bad living habits and chronic stress, etc.) has profound impacts on offspring development; however, the mechanism of paternal disease has not been clarified. In this study, a meta-analysis was first performed to suggest that paternal exposure to nicotine, ethanol, or caffeine is a high-risk factor for adverse pregnancy outcomes. Next, we created a rat model of paternal nicotine/ethanol/caffeine mixed exposure (PME), whereby male Wistar rats were exposed to nicotine (0.1 mg/kg/d), ethanol (0.5 g/kg/d), and caffe

Paternal exposure to recreational drugs before conception and its effect on live-born offspring: A scoping review.

BACKGROUND: Men of reproductive age increasingly use recreational drugs. While many of these substances may reduce the quantity and quality of sperm, less is known about the effects of these exposures on their offspring. We performed a scoping review to summarize the available literature and identify areas for future research on the outcome of live-born offspring of fathers who were exposed to recreational drugs before conception. METHODS: A systematic search was conducted of the Medline, EMBASE, and Web of Science databases, which included keywords for the following substances: cannabis-rela

Childhood cancer and paternal exposure to ionizing radiation: preliminary findings from the Oxford Survey of Childhood Cancers.

Paternal occupational data already collected as part of the Oxford Survey of Childhood Cancers have been reviewed. Information on occupations during or before the relevant pregnancy was sought for 15,279 children dying from cancer in England, Wales, and Scotland in the period 1953-81, and for an equal number of matched controls. Estimates were made for paternal exposure to human-made external ionizing radiation in the six months before conception of the survey child--as judged from job histories and dates of birth. Assessments were also made for potential exposure to unsealed sources of radion

Paternal exposure to low-dose N-nitrosodimethylamine (NDMA) induces intergenerational alterations in the prostatic microenvironment of rats.

NDMA is a nitrosamine recognized as an environmental contaminant of water and food, unintentionally formed during industrial processes and water treatment. Due to its mobility in aquatic environments, persistence, and carcinogenic potential, NDMA has emerged as a toxicological and public health concern. This study evaluated the effects of low-dose exposure (7.2 ng/kg/day) on the prostate of male Wistar rats across two generations within the Paternal Origins of Health and Disease (POHaD) framework. F0 males were exposed via gavage from postnatal day (PND) 60 to 104. In F1, offspring were evalua

Paternal and maternal exposures to per- and polyfluoroalkyl substances (PFAS) and birth outcomes: a multi-country cohort study.

BACKGROUND: Maternal prenatal exposures to per- and polyfluoroalkyl substances (PFAS) have been linked to adverse birth outcomes. However, few investigations have considered paternal PFAS exposure. We estimated the parent-specific associations of prenatal PFAS exposures with adverse birth outcomes. METHODS: This study included 498 couples from the INUENDO cohort recruited at antenatal care visits in Greenland, Poland, and Ukraine during 2002-2004. We measured five major types of PFAS in parental serum during pregnancy. We analyzed three birth outcomes ascertained from medical records, includi

Influence of the AT1 Receptor Antagonists Telmisartan and Losartan on Reproduction and Offspring After Paternal Exposure to Ionizing Radiation.

This study evaluated the repercussions of paternal exposure to radiation on reproduction and offspring in rats, as well as whether treatment with the angiotensin II type 1 (AT1) receptor antagonists telmisartan and losartan has a mitigating effect. Rats were randomly divided into 6 groups: control, radiation, telmisartan, losartan, radiation + telmisartan, and radiation + losartan. A single 5 Gy dose of radiation was administered directly into the scrotum, followed by treatment with telmisartan (12 mg/kg/d) or losartan (34 mg/kg/2 times per day) for 60 days in the groups receiving these medica

Paternal lineage effects of prenatal ethanol exposure: Offspring neuroanatomy and behavior in a multigenerational mouse model of FASD.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) encompass a range of phenotypes in offspring exposed to ethanol via maternal consumption during pregnancy. In a series of studies, our laboratory identified deleterious effects of prenatal ethanol exposure (PrEE) in our FASD mouse model. The first filial generation (F1) of PrEE offspring exhibited abnormal neocortical gene expression, ectopic intraneocortical connectivity, altered neuroanatomy, and disrupted behavior. Our results suggest that PrEE can induce transgenerational transmission of the abnormal phenotypes, potentially via epigenetic
