Therapeutic potency of marine collagen/pectin scaffolds - Fabrication, characterization and evaluation.

Skin is an important vital organ that must be given proper care and protection from external damage and harmful microbes. If injured, it must be treated with an ideal wound dressing material with potent hemostatic and non-toxic properties. In the present study, fish collagen (FC) was extracted from the fins and tails of Black pomfret (Parastromateus niger). The isolated fish collagen was homogenized with pectin (P) and freeze dried to obtain fish collagen/pectin (FC/P) scaffolds. Scanning electron microscopic analysis showed the porous nature of scaffolds with intermittent holes. UV-Visible an

Crosslinked and PEGylated Pectin Chitosan nanoparticles for delivery of Phytic acid to colon.

Polysaccharide-based nanoparticles (NPs) such as pectin/ chitosan (PN/CN) had always been of greatest interest because of their excellent solubility, biocompatibility, and higher suitability for oral drug delivery. This study employed blending-crosslinking of polymers (PN&CN) followed by emulsification-solvent evaporation to prepare and compare two sets of PEGylated NPs to deliver phytic acid (IP6) to colon orally as it has potential to manage colon cancer but fails to reach colon when ingested in pure form. The first set was crosslinked with Glutaraldehyde (GE) (GE*PN-CN-NPs) while the second

Pectin-Alginate Does Not Further Enhance Exogenous Carbohydrate Oxidation in Running.

PURPOSE: Maximizing carbohydrate availability is important for many endurance events. Combining pectin and sodium alginate with ingested maltodextrin-fructose (MAL + FRU + PEC + ALG) has been suggested to enhance carbohydrate delivery via hydrogel formation, but the influence on exogenous carbohydrate oxidation remains unknown. The primary aim of this study was to assess the effects of MAL + FRU + PEC + ALG on exogenous carbohydrate oxidation during exercise compared with a maltodextrin-fructose mixture (MAL + FRU). MAL + FRU has been well established to increase exogenous carbohydrate oxidati

Functional Mechanics of a Pectin-Based Pleural Sealant after Lung Injury.

Pleural injury and associated air leaks are a major influence on patient morbidity and healthcare costs after lung surgery. Pectin, a plant-derived heteropolysaccharide, has recently demonstrated potential as an adhesive binding to the glycocalyx of visceral mesothelium. Since bioadhesion is a process likely involving the interpenetration of the pectin-based polymer with the glycocalyx, we predicted that the pectin-based polymer may also be an effective sealant for pleural injury. To explore the potential role of an equal (weight%) mixture of high-methoxyl pectin and carboxymethylcellulose as

A pectin-honey hydrogel prevents postoperative intraperitoneal adhesions in a rat model.

BACKGROUND: Adhesions are a common postoperative surgical complication. Liquid honey has been used intraperitoneally to reduce the incidence of these adhesions. However, solid barriers are considered more effective than liquids in decreasing postoperative intra-abdominal adhesion formation; therefore, a new pectin-honey hydrogel (PHH) was produced and its effectiveness was evaluated in a rat cecal abrasion model. Standardized cecal/peritoneal abrasion was performed through laparotomy in 48 adult Sprague-Dawley rats to induce peritoneal adhesion formation. Rats were randomly assigned to a contr

Electrostatic Self-Assembled Chitosan-Pectin Nano- and Microparticles for Insulin Delivery.

A polyelectrolyte complex system of chitosan-pectin nano- and microparticles was developed to encapsulate the hormone insulin. The aim of this work was to obtain small particles for oral insulin delivery without chemical crosslinkers based on natural and biodegradable polysaccharides. The nano- and microparticles were developed using chitosans (with different degrees of acetylation: 15.0% and 28.8%) and pectin solutions at various charge ratios (n⁺/n- given by the chitosan/pectin mass ratio) and total charge. Nano- and microparticles were characterized regarding particle size, zeta potential,

Efficacy of a pectin-lecithin complex for treatment and prevention of gastric ulcers in horses.

The objective of this study was to evaluate the effect of a commercial feed supplement containing pectin-lecithin on squamous mucosa ulceration in horses exposed to an experimental ulceration model. Five mares were treated while five mares were controls for this crossover, blinded study. The mares were fed concentrates and hay and were stabled with a two-hour turn out per day for a period of four weeks. The pectin-lecithin complex was fed for the duration of the study on the treated group. At the end of a four-week period, all mares underwent a seven-day alternating feed deprivation (week 5).

Oral sustained delivery of ambroxol from in situ-gelling pectin formulations.

Gels formed in situ following oral administration of dilute aqueous solutions of pectin (1.0 and 1.5%, w/v) to rats were evaluated as vehicles for the sustained release of the expectorant drug ambroxol hydrochloride. The solutions contained calcium ions in complexed form, which on release in the acidic environment of the stomach caused gelation of the pectin. In vitro studies demonstrated diffusion-controlled release of ambroxol from the gels over a period of 6 h. A bioavailability of ambroxol of approximately 64% of that of a commercially available formulation could be achieved from gels cont

Cellulose and pectin alter intestinal beta-glucuronidase (EC 3.2.1.31) in the rat.

Groups of rats were given a fibre-free diet containing none or one of the three fibre components: pectin, cellulose or galactomannan. After feeding for 16 weeks, total protein level and beta-glucuronidase (EC 3.2.1.31) activity in the contents and mucosa of jejunum and ileum, and in the contents only of the caecum, were determined. The pectin supplement reduced protein concentration in jejunal contents while cellulose reduced protein concentration in the ileal and caecal contents. beta-Glucuronidase activity of caecal contents was significantly reduced in both the pectin- and cellulose-fed gro

Preparation and characterization of zein-caseinate-pectin complex nanoparticles for encapsulation of curcumin: pectin extracted by high-speed shearing from passion fruit (Passiflora edulis f. flavicarpa) peel.

BACKGROUND: Pectin extracted by high-speed shearing from passion fruit peel (HSSP) is a potentially excellent wall material for encapsulating curcumin, which has multiple advantages over pectin prepared by heated water extraction. HSSP was used to fabricate complex nanoparticles of zein-sodium caseinate-pectin for encapsulation of curcumin in this study. The influence of heating on the physicochemical properties of the composite nanoparticles was also investigated, as well as the effect of composite nanoparticles on the encapsulation efficiency, antioxidant activity and release characteristics

Ca2+ and degree of methylation on pectin-proanthocyanidin interactions: Structural, thermal, and rheological insights.

Pectins and proanthocyanidins (PA) are known to undergo extensive intermolecular interactions. Nevertheless, the distinct interaction patterns between PA and pectins of varying methylation levels and the regulatory function of endogenous calcium have not been systematically clarified. This study was therefore conducted to reveal the combined effects of the degree of methylation and calcium on their interaction and related physicochemical properties. The non-covalent binding and covalent grafting strength between pectin and PA are higher for low-methoxyl pectin. Benefiting from its high content

Pectin as cancer targeting nano-vehicle and therapeutic - Physicochemical-biological perspectives: A review.

Pectin is cytotoxic against colorectal/pancreatic/liver/bladder/prostate/ovarian/breast cancers. It induces apoptosis, antioxidant, anti-inflammatory, and immunoregulatory properties and mitigate the metastatic and angiogenic tendency of cancer involving MAPK and NFkβ signalling pathways. It has been found to act as a targeting ligand binding to cancer cells that overexpress asialoglycoprotein receptors and galectin adhesive molecules as a function of its galactan moiety. Its cancer targeting specificity can be further raised via incorporating additional targeting ligand (eg. folate). Nanopart

Influence of plasticizer type on the structure and drug release characteristics of LM-pectin hydrogels.

This study investigated the effects of different plasticizers-castor oil (CO), polyvinylpyrrolidone (PVP), and polyethylene glycol (PEG) with varying molecular weights (MWs)-on the structure and drug-release performance of low-methoxyl pectin (LM-pectin) hydrogels. Theophylline was used as a model drug to evaluate release behavior under physiologically relevant conditions. The incorporation of plasticizers modulated polymer-polymer interactions, swelling behavior, and thermal properties, thereby affecting drug-release kinetics. CO, a hydrophobic triglyceride, created microdomain-induced diffus

Comparative study on the mechanisms of citrus pectin and polygalacturonic acid in alleviating food allergies.

Pectin has a complex structure, and different structures lead to different biological functions. Citrus pectin (PC) has been widely proven to possess immunomodulatory activity. This study compared and analyzed the anti-allergic activities of PC and polygalacturonic acid (PGA) based on intestinal microbiota, metabolomics, and molecular docking. The results showed that dietary interventions with PC and PGA significantly relieved allergic symptoms, intestinal damage, and the contents of specific antibodies IgE and IgG1. Among them, PGA exhibited a significant suppressive effect on histamine and a

Crystal Structure of the Multidomain Pectin Methylesterase PmeC5 from Butyrivibrio fibrisolvens D1T.

Pectin is a dynamic and complex polysaccharide that forms a substantial proportion of the primary plant cell wall and middle lamella of forage ingested by grazing ruminants. Pectin methylesterases (PMEs) are enzymes that belongs to the carbohydrate esterase family 8 (CE8) and catalyze the demethylesterification of pectin, a key polysaccharide in cell walls. Here we present the crystal structure of the catalytic domain of PmeC5 that is associated with a gene from Butyrivibrio fibrisolvens D1T that encodes a large secreted pectinesterase family protein (2089 aa) determined to a resolution of 1.3

Modified citrus pectin protects aortic dissection development involving macrophage pyroptosis.

BACKGROUND: Uncontrolled aortic aneurysms can progress to aortic dissection (AD), a severe vascular disorder characterized by hematoma formation in the aortic wall, with inflammation playing a crucial role. Galectin-3 (Gal-3), a 26-kDa lectin, regulates many aspects of inflammatory cell behavior. Inhibition of Gal-3 ameliorates diabetic neuroinflammation and cardiomyopathy. Modified citrus pectin (MCP) is a PH-modified dietetic supplement produced from citrus pectin. This study investigates the therapeutic potential of MCP, which has a known affinity for Gal-3, in AD. METHODS: A murine model

Oral Administration of Apple Pectin Solution Improves Atopic Dermatitis in a Mouse Model.

The development of atopic dermatitis (AD) involves multiple factors. Three such factors are particularly important in AD onset: immune abnormalities, skin barrier dysfunction, and itching. Many studies report that an imbalance between helper T (Th)1 and Th2 cells causes AD. Apple pectin, a prebiotic, has preventative effects in other allergic diseases (e.g., bronchial asthma and AD), but its potential benefits in AD are unclear. In this study, we investigated the effect of oral apple pectin administration on skin inflammation in an AD mouse model and examined changes in T cells involved in AD.
