Human Gut Commensal Membrane Vesicles Modulate Inflammation by Generating M2-like Macrophages and Myeloid-Derived Suppressor Cells.

Immunomodulatory commensal bacteria modify host immunity through delivery of regulatory microbial-derived products to host cells. Extracellular membrane vesicles (MVs) secreted from symbiont commensals represent one such transport mechanism. How MVs exert their anti-inflammatory effects or whether their tolerance-inducing potential can be used for therapeutic purposes remains poorly defined. In this study, we show that MVs isolated from the human lactic acid commensal bacteria Pediococcus pentosaceus suppressed Ag-specific humoral and cellular responses. MV treatment of bone marrow-derived mac

Lactic acid bacteria derived postbiotic preparations disrupt biofilm architecture of mastitis-related pathogens in vitro.

Rising antimicrobial resistance necessitates alternative therapeutic approaches for bovine mastitis, especially strategies targeting biofilm-related infections. This study explores the antibiofilm effects of LAB-derived postbiotic preparations (neutralized cell-free supernatants, nCFS) from Pediococcus pentosaceus and Lactococcus lactis, isolated from local sources as potential complementary or adjuncts to conventional antibiotic therapy. Antimicrobial activity was assessed using the agar well diffusion method, while antibiofilm efficacy was determined via the microtiter plate assay. Biofilm e

Lactic acid bacteria decarboxylates hydroxycinnamic acids under acidic environments.

Some lactic acid bacteria (LAB) enzymatically decarboxylate hydroxycinnamic acids (HCAs) as a detoxification mechanism, yielding 4-vinylphenols. These compounds can in turn react with anthocyanins to form more stable chromophores, called pyranoanthocyanins, which are potential candidates as food colorants. However, LAB growth conditions differ from the ones needed for pyranoanthocyanin production. This study aimed to determine if Lactiplantibacillus plantarum, Enterococcus mundtii and Pediococcus pentosaceus strains could perform decarboxylation under acidic conditions that favor pyranoanthocy

Recovery of vancomycin-resistant gram-positive cocci from pediatric liver transplant recipients.

Between November 1988 and October 1989, 49 first-time pediatric liver transplant recipients at the Children's Hospital of Pittsburgh were prospectively monitored for the presence of stool colonization and the development of disease caused by vancomycin-resistant gram-positive cocci (VRGPC). Quantitative stool culturing was done on a weekly basis, and cultures were planted onto a selective medium for VRGPC. Isolates for which the MIC was greater than or equal to 8 were considered resistant to vancomycin. Patients were monitored clinically for the development of infection, and their charts were

From gluten structure to immunogenicity: Investigating the effects of lactic acid bacteria and yeast co-fermentation on wheat allergenicity in steamed buns.

The aim of this study was to explore the reduction of allergenic proteins in sourdough by selecting lactic acid bacteria (LAB) to promote protein hydrolysis while maintaining the techno-functional properties of steamed buns. Here, we measured the protein hydrolysis activity of 37 LAB strains and identified three strains with significant protein hydrolysis effects (L. sanfranciscensis Fs5, L. sanfranciscensis Ls5, and P. pentosaceus P.p7). Co-fermentation of LAB and yeast led to the depolymerization of gluten macromolecules and a more severe decrease in immunoreactivity (46.93 % ∼ 78.51 %). Qua

Commensal-derived acetylcholine enhances mucosal immune education.

The microbiota produces thousands of potentially bioactive small molecules1-3. High-throughput bioactivity screens of in vitro commensal cultures have exposed microbiota metabolites that shape host physiology by activating diverse G-protein-coupled receptors (GPCRs)4-7. However, owing to technical limitations, the GPCRome-wide bioactivities of in vivo metabolomes, which result from complex diet-microorganism-host interactions, remain unclear. Here we used a multiplexed GPCR screening technology to assess GPCRome-wide bioactivities of 100 commensal strains grown in vivo in monoassociated germ-f

A synthetic probiotic engineered for colorectal cancer therapy modulates gut microbiota.

BACKGROUND: Successful chemoprevention or chemotherapy is achieved through targeted delivery of prophylactic agents during initial phases of carcinogenesis or therapeutic agents to malignant tumors. Bacteria can be used as anticancer agents, but efforts to utilize attenuated pathogenic bacteria suffer from the risk of toxicity or infection. Lactic acid bacteria are safe to eat and often confer health benefits, making them ideal candidates for live vehicles engineered to deliver anticancer drugs. RESULTS: In this study, we developed an effective bacterial drug delivery system for colorectal ca

Comprehensive insights into the mechanism of flavor formation driven via inoculation with mixed starter cultures in dry-fermented tilapia sausages: Integration of macrogenomics, volatilomics, and lipidomics.

To investigate the mechanisms of flavor formation in dry-fermented tilapia sausages, the volatiles, bacterial community, and lipid composition during fermentation were analyzed using gas chromatography-ion mobility spectrometry, 16S high throughput sequencing, and ultra-performance liquid chromatography-mass spectrometer. Pediococcus pentosaceus, Staphylococcus xylosus, and Staphylococcus carnosus became dominant bacteria during the fermentation. A total of 66 volatiles and 293 lipids (48 differential lipids) were identified. PC and PE content decreased. Aldehyde and 1-octen-3-ol content decre

Commensal Bacteria Maintain a Qa-1 b -restricted Unconventional CD8 + T Population in Gut Epithelium.

Intestinal intraepithelial lymphocytes (IELs) are characterized by an unusual phenotype and developmental pathway, yet their specific ligands and functions remain largely unknown. Here by analysis of QFL T cells, a population of CD8 + T cells critical for monitoring the MHC I antigen processing pathway, we established that unconventional Qa-1 b -restricted CD8 + T cells are abundant in intestinal epithelium. We found that QFL T cells showed a Qa-1 b -dependent unconventional phenotype in the spleen and small intestine of naïve wild-type mice. The splenic QFL T cells showed innate-like function

Anti-bacterial and anti-biofilm activity of probiotic bacteria against oral pathogens.

In this study, three lactic acid bacteria (LAB), isolated from barley, traditional dried meat and fermented olive were characterized and tested for their anti-bacterial and anti-biofilm activities against oral bacteria. Our results revealed that the tested LAB were γ-hemolytic and were susceptible to four antibiotics. All the strains were resistant to low pH, bile salt, pepsin and pancreatin. Furthermore, FB2 displayed a high aut-oaggregative phenotype (99.54%) while FF2 exhibited the best co-aggregation rate. Concerning the microbial adhesion to solvent, FB2 was the most hydrophobic strain (d

Rapid monitoring of sourdough starter and contaminants using a dual-lateral flow immunochromatographic assay.

Sourdough maintenance via daily backslopping faces the persistent risk of starter lactic acid bacteria (LAB) being displaced by competing contaminants. Here, we developed a dual-lateral flow immunochromatographic assay (LFIA) system to simultaneously monitor starter health (Fructilactobacillus sanfranciscensis; Fsanf-LFIA) and contaminant presence (Pediococcus acidilactici; Ped-LFIA). Fsanf-LFIA and Ped-LFIA both exhibited detection limits of 5 log cells/test. Fsanf-LFIA was highly specific to certain strains of F. sanfranciscensis, whereas Ped-LFIA demonstrated a broader detection spectrum, c

Insights into the conversion of odor compounds by a novel microbial agent during swine manure storage: a multi-omics integration of microbiome, metabolome and genome.

Deep-pit manure storage in intensive swine farming has emerged as a major source of malodorous emissions, posing significant environmental and public health concerns. This study developed a targeted microbial agent by systematic screening of malodor-degrading microorganisms (Alcaligenes faecalis A1, Bacillus velezensis B6, Pediococcus pentosaceus L4 and Meyerozyma guilliermondii Y7), and multi-omics approach was used to elucidate the mechanism of odorous compounds conversion during manure storage after inoculation with microbial agent. The agent significantly reduced emissions of NH3 (42.22%)

Anti-inflammatory effect of lignans from flaxseed after fermentation by lactiplantibacillus plantarum SCB0151 in vitro.

Lignan, a beneficial constituent of Flaxseed (Linum usitatissimum L.) showed great interest in researchers because of its multiple functional properties. Nonetheless, a challenge arises due to the glycosidic structure of lignans, which the gut epithelium cannot readily absorb. Therefore, we screened 18 strains of Lactiplantibacillus plantarum, Lacticaseibacillus casei, Lactobacillus acidophilus, Lacticaseibacillus rhamnosus, Pediococcus pentosaceus, Pediococcus acidilactici, and Enterococcus durans to remove glycosides from flaxseed lignan extract enzymatically. Characteristics of the microstructure and the key components of white kidney bean sourdough bread induced by mixed-strain fermentation and its influence on gut microbiota.

In this study, the effect of mixed-strain fermentation using Kluyveromyces marxianus with either Lactobacillus plantarum or Pediococcus pentosaceus on the physiochemical and nutritional properties of white kidney bean flour sourdough was investigated. The results indicated that mixed-strain fermentation reduced the anti-nutritional factors produced from the white kidney bean flour, especially in the sourdough fermented by L. plantarum and K. marxianus (WKS-LK) compared to that by P. pentosaceus and K. marxianus (WKS-JK). Meanwhile, the content of lactic acid and acetic acid and the proportion

The predominant lactic acid bacteria and yeasts involved in the spontaneous fermentation of millet during the production of the traditional porridge Hausa koko in Ghana.

Spontaneous fermentation of cereals like millet involves a diverse population of microbes from various sources, including raw materials, processing equipment, fermenting receptacles, and the environment. Here, we present data on the predominant microbial species and their succession at each stage of the Hausa koko production process from five regions of Ghana. The isolates were enumerated using selective media, purified, and phenotypically characterised. The LAB isolates were further characterised by 16S rRNA Sanger sequencing, typed using (GTG)5 repetitive-PCR, and whole genome sequencing, wh

Microbial community structure, functional potential, probiotic signatures, and MAG reconstruction of fermented bamboo shoots from Northeast India.

Fermented bamboo shoot (FBS) products are widely consumed traditional foods across the Northeast region (NER) of India, yet their microbiome structure, functional capacity, biosynthetic potential, and safety attributes remain insufficiently explored. Here, comparative shotgun metagenomics of ten traditional FBS products from six NER states was used to address these gaps integrating previously generated metagenomic data from Tripura with newly generated datasets from Manipur, Meghalaya, Arunachal Pradesh, Nagaland, and Sikkim thereby bringing the total number of samples to 24.
