Integrated metabolome-SNP-gut microbiome analysis deciphers the molecular basis of EHP resistance in Penaeus vannamei.

Enterocytozoon hepatopenaei (EHP) has emerged as a highly virulent pathogen in shrimp aquaculture, yet the molecular determinants governing host resistance remain incompletely characterized. This study employed an integrated multi-omics strategy (metabolome, transcriptome, and microbiome) to elucidate the molecular basis of EHP resistance in Penaeus vannamei. Comparative metabolomic profiling of EHP-resistant (R4029) and susceptible (S4104) family lines identified 1614 metabolites, primarily organic acids (35.07 %) and lipids (21.56 %). Orthogonal partial least squares discriminant analysis (O

Properties of Pacific white shrimp (Litopenaeus vannamei) collagen and its degradation by endogenous proteinases during cold storage.

Many factors are responsible for the diminished quality of shrimp during cold storage, while the role of collagen has rarely been studied. This study therefore investigated the relationship between collagen degradation and changes of textural properties of Pacific white shrimp, and its hydrolysis by endogenous proteinases. The textural properties of shrimp decreased gradually along with disruption of shrimp muscle tissues, and the chewiness property of shrimp muscle showed a linear relationship with collagen contents in muscle during 6-day-storage at 4 °C. Pepsin-solubilized collagen in shrimp

Effect of phytic acid combined with lactic acid on color and texture deterioration of ready-to-eat shrimps during storage.

To retard the deterioration of texture and color of ready-to-eat (RTE) shrimps during storage, phytic acid (PA) and lactic acid (LA) were used to soak the shrimp prior to cooking. The factors affecting texture (water holding capacity, protein oxidation and degradation, and microstructure) and color (Maillard reaction and lipid oxidation) were determined separately. The free radical and copper ion content were also determined in order to investigate the regulation mechanism of phytic acid and lactic acid on the texture and color of RTE shrimps. It was found that the inhibitor-treated RTE shrimp

Microbiological Investigation of Persistent Mortalities in Litopenaeus vannamei Grown in Low Saline Waters in India.

Abstract Microbial diseases are a serious hindrance to successful shrimp aquaculture. The Pacific white shrimp Litopenaeus vannamei is an exotic species recently introduced in India to supplement the failing aquaculture of the Asian tiger shrimp Penaeus monodon due to viral diseases. However, after a brief initial success, the aquaculture of L. vannamei is also experiencing serious problems due to microbial diseases. In this study, we conducted a microbiological investigation into the problem of persistent mortalities in selected L. vannamei farms on the southeastern coast of India. The infect

[Development of rapid detection of infectious hypodermal and hematopoietic necrosis virus by loop-mediated isothermal amplification].

Loop-mediated isothermal amplification (LAMP) assay is a novel method of gene amplification with high specificity, sensitivity and rapidity, which can be applied for disease diagnosis in shrimp aquaculture. The method is performed under isothermal conditions with a set of four specially designed primers that recognize six distinct sequences of the target. In the present study, according to the conservative regions of non-structural protein gene NS1, a set of four specific primers were designed, and a rapid detection of IHHNV was established by LAMP assay. The parameters of reaction time and te

[Bacterial conversion of sulfur-and phosphorous-compounds and microbial diversity in sediments from a near-shore marine-cultural region].

The H2S-producing bacteria and the phosphorous-cycling bacteria in sediments from near-shore marine-cultural region were investigated. Results indicate that the bacterial H2S production in aerobic condition is the dominating process to produce H2S in the sediment of cultural pond. The total counts of H2S-producing bacteria utilizing cysteine and Na2S2O3 were 1.6 x 10(6) and 4.35 x 10(3) cells g(-1) respectively. The counts of sulfate-reducing bacteria in the sediments were very little, only 2.5 x 10(1) g(-1). Further results show that the bacterial counts of decomposing lecithin and secreting

Genomic characterization and phage cocktail suppression of a multidrug-resistant Vibrio sinaloensis isolate from diseased shrimp.

UNLABELLED: Vibrio sinaloensis has been reported in association with disease events in aquaculture, yet its genomic features and non-antibiotic suppression strategies remain largely unexplored. Here, we report genomic characterization of a multidrug-resistant V. sinaloensis strain, ZZ006, isolated from diseased shrimp and evaluate phage-mediated suppression of this strain in vitro. The ZZ006 genome harbors multiple putative virulence-associated secretion systems and antimicrobial resistance genes, suggesting chromosomal carriage of traits potentially relevant to host association and antimicrob

Designing sustained-release sodium carriers: Oil-modified starch gels enhance the texture and delay sodium release in low-sodium shrimp protein gels.

This study developed an oil-modified starch gel (OMSGs) as delivery system to modulate texture and control sodium release in low-sodium shrimp myofibrillar protein (SMP) gels. OMSGs prepared with 1% oil and 100 mg/mL starch formed a dense network with high water-holding capacity and gel strength. OMSGs containing varying NaCl (5-20%), denoted OMSGs-Na, were blended with SMP at a 10:1 ratio to obtain OMSGs-Na-SMP composites. Among these, the OMSGs-Na (15%, equivalent to 1.19% NaCl in SMP) exhibited uniform and dense microstructure, effectively delaying sodium ion release during simulated oral d

Multidimensional strategy for discovering saltiness-enhancing peptides in shrimp heads integrating ultra-high pressure hydrolysis and machine learning.

This study aims to develop a comprehensive strategy to investigate whether the integration of ultra-high pressure (UHP)-assisted enzymatic hydrolysis with machine learning and molecular docking can effectively identify salty peptides (SPs) from Litopenaeus vannamei heads. UHP treatment (400 MPa, 30 min) significantly enhanced proteolysis, yielding more low-molecular-weight peptides (<3 kDa, 66.44 %) than conventional methods. Peptidomics identified 211 differentially expressed peptides enriched in hydrophobic/acidic residues (e.g., Asp, Glu) linked to saltiness. A machine learning model integr

Crustacean cardioactive peptide (CCAP) negatively regulates sand-diving behaviour in kuruma shrimp, Penaeus japonicus.

Kuruma shrimp (Penaeus japonicus) exhibits sand-diving behaviour. The genetic control mechanism of this behaviour is still unclear, although previous single-cell transcriptome sequencing suggests that crustacean cardioactive peptide (CCAP) may be involved. In this study, we cloned the CCAP precursor cDNA (Pj-CCAP) in kuruma shrimp. It was predicted that the gene can be translated into four related peptides and one mature peptide (PFCNAFTGC-NH2), and is highly similar to crustaceans and insects. The predicted Pj-CCAP itself is highly conserved in other crustaceans. RT-qPCR and in situ hybridiza

MicroRNA sequencing analysis reveals immune responses in hepatopancreas of Fenneropenaeus penicillatus under white spot syndrome virus infection.

White Spot Disease is one of the most harmful diseases of the red tail shrimp, which can cause devastating economic losses due to the highest mortality up to 100% within a few days. MicroRNAs (miRNAs) are large class of small noncoding RNAs with the ability to post-transcriptionally repress the translation of target mRNAs. MiRNAs are considered to have a significant role in the innate immune response of crustaceans, particularly in relation to antiviral defense mechanisms. Numerous crustacean miRNAs have been verified to be required in host immune defense against viral infection, however, till

Polystyrene nanoplastics induce lipid metabolism disorder and alter fatty acid composition in the hepatopancreas of Pacific whiteleg shrimp (Litopenaeus vannamei).

The impact of nanoplastics (NPs) on environmental pollution and aquatic organisms has gradually attracted attention, but there are relatively few reports of the effects of NPs on the lipid metabolism of crustaceans. In this study, we exposed Pacific whiteleg shrimp (Litopenaeus vannamei) to different concentrations of polystyrene NPs (0, 0.1, 1, 5, and 10 mg/L) for 28 days. We then evaluated the effects of NP exposure on metabolite content, histology, lipid metabolism-related enzyme activity, and gene expression. Our results showed that with increasing NPs concentrations and exposure time, (1)

Functionality of probiotics on the resistance capacity of shrimp against white spot syndrome virus (WSSV).

Shrimp aquaculture is currently regarded as a significant commercial and food production sector due to its growing importance as a source of human-consumable protein, As shrimp farming has become more intensive, disease outbreaks have become more common, necessitating the overuse of antimicrobial drugs, which has had a number of unintended consequences. The white spot syndrome virus (WSSV) is now recognized as one of the world's most pervasive and potentially fatal diseases affecting shrimp. However, there is currently no cure to prevent the disease's uncontrolled incidence and spread. Probiot
