Divergent immune dynamics in European perch (Perca fluviatilis): Population-specific responses of peritoneal cells to Aeromonas hydrophila.

Fish populations provide valuable genetic resources for sustainable breeding programs. For species like the European perch (Perca fluviatilis), which are early in domestication, systematic characterization is key to developing strong breeding lines. This pilot study assessed the immune response to the bacterium Aeromonas hydrophila in six different perch populations. Bacterial injection triggered two distinct patterns of immune reactions. Fish originating from Finland (FIN) and Slovakia (SVK-LM and SVK-RV) exhibited a rapid immune response, with a dramatic influx of cells, including 4 million

PCR Detection and Phylogenetic Analysis of Megalocytivirus Isolates in Farmed Giant Sea Perch Lates calcarifer in Southern Taiwan.

The Megalocytivirus genus includes three genotypes, red sea bream iridovirus (RSIV), infectious spleen and kidney necrosis virus (ISKNV), and turbot reddish body iridovirus (TRBIV), and has caused mass mortalities in various marine and freshwater fish species in East and Southeast Asia. Of the three genotypes, TRBIV-like megalocytivirus is not included in the World Organization for Animal Health (OIE)-reportable virus list because of its geographic restriction and narrow host range. In 2017, 39 cases of suspected iridovirus infection were isolated from fingerlings of giant sea perch (Lates cal

Hepatic oxidative stress and metal subcellular partitioning are affected by selenium exposure in wild yellow perch (Perca flavescens).

Yellow perch (Perca flavescens) collected from 11 lakes in the Canadian mining regions of Sudbury (Ontario) and Rouyn-Noranda (Quebec) display wide ranges in the concentrations of cadmium (Cd), nickel (Ni), selenium (Se), and thallium (Tl) in their livers. To determine if these trace elements, as well as copper (Cu) and zinc (Zn), are causing oxidative stress in these fish, we measured three biochemical indicators (glutathione (GSH), glutathione disulfide (GSSG) and thiobarbituric acid-reactive substances (TBARS)) in their livers. We observed that 44% of the yellow perch that we collected were

Host-parasite relationships as determinants of heavy metal concentrations in perch (Perca fluviatilis) and its intestinal parasite infection.

The concentrations of As, Cd, Cr, Cu, Hg, Mn, Ni, Pb, and Zn and their bioconcentration factors (BCFs) were determined in two intestinal parasites, an acanthocephalan, Acanthocephalus lucii, a tapeworm, Proteocephalus percae, present in the same host, the European perch (Perca fluviatilis, L.), in the heavily polluted Ružín reservoir in eastern Slovakia. The bioaccumulation of heavy metals in the fish organs and parasites was studied for acanthocephalan and tapeworm monoinfections or mixed infections by the two parasites and for the size of their parasitic infrapopulations. Bioconcentration fa

Morphometric and metabolic indicators of metal stress in wild yellow perch (Perca flavescens) from Sudbury, Ontario: a review.

Eighteen lakes studied near Sudbury and across Northeastern Ontario (Canada) over a five-year period provided a wide contamination gradient of cadmium (Cd), copper (Cu) and other metals such as nickel (Ni) and zinc (Zn). All were inhabited by yellow perch (Perca flavescens), which was sometimes the only species present. Liver Cd and Cu concentrations were monitored in these lakes, in some cases for several consecutive years and for multiple seasons. This data suggests that yellow perch from clean to mildly-contaminated environments loosely control their hepatic Cu concentrations between 7 and

Integrated Physiological and Transcriptomic Analyses Suggest Key Adaptive Mechanisms of European Perch (Perca fluviatilis) to Acute Heat Stress.

The European perch (Perca fluviatilis) is highly susceptible to heat stress, limiting its sustainable aquaculture. While single-organ thermal responses are partially understood, the systemic, multi-organ cooperative survival mechanisms under acute heat stress remain poorly characterized. To elucidate the underlying tolerance mechanisms and provide genetic markers for breeding, this study investigated the multi-organ responses of European perch (n = 90; body length: 13.15 ± 1.75 cm; body weight: 30.54 ± 7.17 g) transferred from 24 °C to an acute heat stress challenge (31 °C) at an increasing ra

White perch health relative to urbanization and habitat degradation in Chesapeake Bay tributaries. II. Hepatic and splenic macrophage aggregates.

Macrophage aggregate (MA) abundance in fish is a useful general biomarker of contaminant exposures and environmental stress. Hepatic and splenic MAs were evaluated in semi-anadromous white perch Morone americana (Gmelin, 1789) from the urbanized Severn River (S) and the more rural Choptank River (C), Chesapeake Bay. Fish were collected from different sites in the annual migratory circuit in each river that corresponded to active spawning in late winter-early spring, summer regenerating, autumn developing, and winter spawning-capable phases. An age-associated progressive increase in the total v

Effects of dietary carbohydrate to lipid ratios on growth, biochemical indicators, lipid metabolism, and appetite in Chinese perch (Siniperca chuatsi).

An 8-week feeding trial was conducted to evaluate the effects of dietary carbohydrate to lipid (CHO:L) ratios on growth performance, body composition, serum biochemical indexes, lipid metabolism, and gene expression of central appetite regulating factors in Chinese perch (Siniperca chuatsi) (mean initial weight: 12.86 ± 0.10 g). Five isonitrogenous and isoenergetic diets (fish meal, casein as main protein sources) were formulated to contain different graded CHO:L ratio diets ranging from 0.12, 0.86, 1.71, 3.29, and 7.19. Each diet was assigned to triplicate groups of 18 experimental fish. Our

First cytogenetic characterization of the southern pygmy perch Nannoperca australis (Teleostei: Percichthyidae): Karyotype, heterochromatin and repetitive DNA organization.

Chromosome organization and repetitive DNA distribution are important components of genome evolution in teleost fishes but remain poorly characterized in many species of ecological and conservation relevance. We performed a detailed cytogenetic analysis of the southern pygmy perch, Nannoperca australis, a small freshwater species widely used in studies of population structure and evolutionary ecology in southeastern Australia. Mitotic chromosomes were examined using Giemsa staining, C-banding, CMA3 fluorochrome staining, fluorescence in situ hybridization (FISH) with 5S and 28S rDNA probes, te

Combined use of red ginger extract and a cooling perch improves thermoregulation and performance of broilers in tropical semi-open housing.

This study evaluated the combined effects of red ginger extract and a cooling perch system on thermoregulation and performance of broiler chickens under chronic heat stress in tropical semi-open housing. A total of 198 broiler chicks were allocated to 18 floor pens in a completely randomized design with three treatments: control, red ginger supplementation, and red ginger combined with a cooling perch. Birds were exposed to daily ambient temperatures exceeding the thermoneutral range. The combined treatment significantly reduced cloacal temperature, water intake, and the heterophil/lymphocyte

Standardized Cryopreservation Protocols for European Perch (Perca fluviatilis) and Pikeperch (Sander lucioperca) Semen Enabling Hatchery-Scale Fertilization.

Cryopreservation of fish semen ensures the year-round availability of gametes from species with seasonal spawning, supports genetic resource banking, and underpins selective breeding programs. However, percid species such as European perch and pikeperch pose specific challenges, notably the spontaneous activation of sperm motility in seminal plasma, which can reduce sperm motility reserves during cryopreservation. Building on systematic studies, here we present standardized protocols for semen collection, sperm quality evaluation, extender preparation, freezing, thawing, and post-thaw handling

Salmonella Salamae and S. Waycross isolated from Nile perch in Lake Victoria show limited human pathogenic potential.

Non-enterica subspecies of Salmonella enterica are rarely associated with human infections. Paradoxically, food safety legislations consider the entire genus Salmonella as pathogenic to humans. Globally, large amounts of seafoods are rejected and wasted due to findings of Salmonella. To inform better food safety decisions, we investigated the pathogenicity of Salmonella Salamae 42:r- and Salmonella Waycross isolated from Nile perch from Lake Victoria. Genome-wide analysis revealed absence of significant virulence determinants including on key Salmonella pathogenicity islands in both serovars.

High prevalence of biliary neoplasia in white perch Morone americana: potential roles of bile duct parasites and environmental contaminants.

Recent surveys of white perch Morone americana from Chesapeake Bay, USA, revealed a high prevalence of hepatic and biliary lesions, including neoplasia, and bile duct parasites. Here, we describe lesions in the liver and gallbladder and evaluate for statistical associations among lesions, parasites, and biomarkers of chemical exposure in fish from 2 tributaries of Chesapeake Bay. Fish were collected from an estuarine site in the Choptank River (n = 122, ages 3-11), a tributary with extensive agriculture within the watershed, and the Severn River (n = 131, ages 2-16), a tributary with extensive

Cumulative effects of municipal effluent and parasite infection in yellow perch: A field study using high-throughput RNA-sequencing.

Multiple metabolic, immune and reproductive effects have been reported in fish residing in effluent-impacted sites. Natural stressors such as parasites also have been shown to impact the responses of organisms to chronic exposure to municipal effluent in the St. Lawrence River (Quebec, Canada). In order to comprehensively evaluate the cumulative impacts of anthropogenic and natural stressors on the health of yellow perch, differential mRNA transcription profiles were examined in juvenile females collected from effluent-impacted and upstream sites with low or high infection levels of the larval

Seasonal development of the coccidian parasite Goussia bayae and hepatobiliary histopathology in white perch Morone americana from Chesapeake Bay.

The coccidium Goussia bayae infects the gallbladder and bile ducts of white perch Morone americana from Chesapeake Bay, USA. Seasonal changes in coccidian infections were analyzed from bile specimens of 1588 fish from the Choptank River during 2016-2018 using wet mount preparations with a Sedgwick-Rafter counting chamber. Histopathology of the gallbladder and liver was analyzed from a subset (n = 480) of these fish. Maximum parasite prevalence (100%) and intensities in the gallbladder occurred during the fish spawning season in March and April. Asynchronous coccidian development and prevalence

Evolutionary change driven by metal exposure as revealed by coding SNP genome scan in wild yellow perch (Perca flavescens).

Pollution can drive rapid evolutionary change in wild populations. This study targets functional polymorphisms of chronically metal-contaminated wild yellow perch (Perca flavescens). A de novo transcriptome scan contrasted subsets of individuals from clean (n = 16) and contaminated (n = 16) lakes to identify 87 candidate annotated coding SNPs. Candidate genotypes and liver [metal] were obtained in 10 populations (n = 1,052) and a genome scan distinguished outliers: one nuclear (cyclin G1 gene) and two mitochondrial (cytochrome b and NADH dehydrogenase subunit 2 genes) also displaying allelic c

Impact of toxic cyanobacterial blooms on Eurasian perch (Perca fluviatilis): experimental study and in situ observations in a peri-alpine lake.

Due to the importance of young-of-the-year (YOY) perch in the peri-alpine regions where they are consumed, the microcystin (MC) contamination of YOY perch was analysed both in field (Lake Bourget, France) and experimentally using force-feeding protocols with pure MCs. In-situ, schools of YOY perch present in the epilimnion of the lake were never found in direct contact with the P. rubescens blooms that were present in the metalimnion. However, MCs were detected in the muscles and liver of the fish and were thus assumed to reach YOY perch through dietary routes, particularly via the consumption

Co-assembly of GABA rho subunits with the GABA(A) receptor gamma(2) subunit cloned from white perch retina.

Although it is well established that GABA(C) receptors are comprised in part of GABA rho subunits, the exact subunit composition of neuronal GABA(C) receptors is yet to be determined. A detailed comparison of GABA(C)-mediated neuronal responses elicited from retinal neurons with those obtained from receptors formed by GABA rho subunits revealed a number of significant differences both in the kinetics and the pharmacology of the responses. Our previous studies indicated that the human GABA(A) receptor gamma(2) subunit could co-assemble with one (rho(1B)) of the white perch GABA rho subunits to

Targeting HSP90ab1: curcumin and silybin inhibit NNV infection in sea perch (Lateolabrax japonicus) via receptor blockade and immune enhancement.

Nervous necrosis virus (NNV), a member of the family Nodaviridae, is a devastating pathogen in global aquaculture, yet effective preventive and therapeutic strategies remain limited. This study aimed to identify natural compounds that inhibit NNV infection by targeting the host receptor Lateolabrax japonicus heat shock protein 90 alpha family class B1 (LjHSP90ab1). Herein, we identified curcumin and silybin as potent inhibitors of LjHSP90ab1. Molecular docking predicted that curcumin and silybin bind to the N-terminal and C-terminal domains of LjHSP90ab1, respectively, and fluorescence spectro
