The genetic and biochemical basis of perchlorate-dependent H2S oxidation (PSOX) was investigated in the dissimilatory perchlorate-reducing microorganism (DPRM) Azospira suillum PS (PS). Previously, it was shown that all known DPRMs innately oxidize H2S, producing elemental sulfur (So). Although the process involving PSOX is thermodynamically favorable (ΔG°' = -206 kJ ⋅ mol-1 H2S), the underlying biochemical and genetic mechanisms are currently unknown. Interestingly, H2S is preferentially utilized over physiological electron donors such as lactate or acetate although no growth benefit is obtai

Perchlorates are strong oxidants widely employed in military and civilian energetic materials and recently have been scrutinized as persistent environmental pollutants. The perchlorate anion, ClO(4)(-), is a well-known and potent competitive inhibitor of iodide transport by the sodium iodide symporter (NIS) expressed in the basolateral membranes of thyroid follicular cells (thyrocytes). Iodide uptake by thyroid follicular cells is rapid and reproducible. The competitive radiotransporter assay in this study shows promise as a rapid and convenient method to assay for ClO(4)(-) in water samples a

Perchlorate, an environmental contaminant, is known to disturb the hypothalamus-pituitary-thyroid (HPT) axis by blocking iodide accumulation in the thyroid. Iodide deficiency can lead to hypothyroidism and goiter in rats. The objective of the study was to characterize the pharmacokinetics of perchlorate in male Sprague-Dawley rats relative to inhibition of thyroidal radiolabeled iodide uptake and onset of up-regulation of the HPT axis. Radiolabeled perchlorate (3.3 mg/kg (36)ClO(-)(4)) was excreted in urine (99.5% over a 48-h period). (36)ClO(-)(4) is rapidly distributed into tissues with pref

A modified method for the diagnosis of the Pendred Syndrome in children by the Perchlorate discharge test using 123I is described. The older child, who has the Pendred Syndrome and the obligatory hearing deficit, frequently has neither a goitre nor hypothyroidism, but other investigations (bone growth, scars and function tests) can also show changes. However a more certain diagnosis of this disorder in children is possible by the Perchlorate discharge test using 123I. The test using 131I is no longer justified in the face of more recent radiobiological knowledge.

Catalyzing ammonium perchlorate (AP) thermal decomposition is essential for improving the energy release of composite solid propellants. Single-atom catalysts (SACs) show great potential in AP catalysis, but systematic research on their regulation of AP decomposition and propellant combustion remains insufficient. Herein, four carbon black-supported transition metal single-atom catalysts (M-SACs, M = Fe, Co, Ni, Cu) were prepared via a coordination adsorption-pyrolysis strategy, and their atomic dispersion and coordination structure were systematically characterized. Thermal tests demonstrate

Several exposure limits for perchlorate have been developed based on an early key event, inhibition of radioactive iodide uptake (RAIU) by the thyroid. These assessments have used a variety of definitions of the point of departure. The current assessment revisited the modeling for inhibition of RAIU, using state of the science methods. Bayesian hierarchical modeling was used to account for the repeated measures on the same individuals in the key dataset, and the underlying Beta distribution used for the modeling correctly reflected the bounding of RAIU between 0 and 1. We defined the BMR as a

In the case of a nuclear power plant accident, repetitive/prolonged radioiodine release may occur. Radioiodine accumulates in the thyroid and by irradiation enhances the risk of cancer. Large doses of non-radioactive iodine may protect the thyroid by inhibiting radioiodine uptake into the gland (iodine blockade). Protection is based on a competition at the active carrier site in the cellular membrane and the Wolff-Chaikoff effect, the latter being, however, only transient (24-48 h). Perchlorate may alternatively provide protection by a carrier competition mechanism only. Perchlorate has, howev

INTRODUCTION: Incompleteness of epidemiological databases is a major drawback when it comes to analyzing data. We conceived an epidemiological study to assess the association between newborn thyroid function and the exposure to perchlorates found in the tap water of the mother's home. Only 9% of newborn's exposure to perchlorate was known. The aim of our study was to design, test and evaluate an original method for imputing perchlorate exposure of newborns based on their maternity of birth.
METHODS: In a first database, an exhaustive collection of newborn's thyroid function measured during a

OBJECTIVE: To determine, on the basis of iodide uptake inhibition (IUI), whether associations between urinary concentrations of IUI agents (perchlorate, nitrate, and thiocyanate), as total perchlorate equivalent concentration (PEC), and serum thyroid parameters suggest functional thyroid abnormalities. Additional thyroid hormone measures were released to augment the National Health and Examination Survey (NHANES) 2001 to 2002 data set, which we used in this study.

Exposure to perchlorate or other thyrotoxic compounds can cause hypothyroidism in most vertebrates, and this may affect levels of endogenous antioxidants and cause oxidative stress. Arsenic also induces oxidative stress in animals by modifying the antioxidant capacity and may alter the thyroid homeostasis. Therefore, hypothyroidism may affect the toxicity of arsenate. In order to test this hypothesis, zebrafish (Danio rerio) were made hypothyroid by exposure to perchlorate, and toxicity of arsenate in hypothyroid and euthyroid fish was compared. The endpoints were LC50 and thyroid histopatholo

In voltage clamped frog skeletal muscle fibres 0.2 mM tetracaine strongly suppresses Ca(2+) release. After this treatment Ca(2+) release flux lacks its characteristic initial peak and the remaining steady component is strongly reduced when compared with the control condition. We studied the effect of two agonists of Ca(2+) release on these tetracaine treated fibres. 8 mM ClO(4)(-) added after tetracaine potentiated release flux from 0.11 +/- 0.03 mM s(-1) to 0.34 +/- 0.07 mM s(-1) (n = 6) although without recovery of the peak at any test voltage. The voltage dependence of the increased release

We set up a modified technetium-99m (Tc-99m) pertechnetate/Tc-99m MIBI (Tc-MIBI) subtraction scintigraphy for parathyroid imaging by introducing the use of potassium perchlorate (KCLO4). Initially, the effect of KCLO4 on technetium thyroid wash-out was evaluated in five healthy volunteers: 40-minute dynamic studies of the thyroid were obtained 20 minutes after the injection of technetium 150 MBq (4 mCi), both in baseline conditions and after the oral administration of 400 mg KCLO4. After an average latency time of 10.5 minutes, KCLO4 administration resulted in fast and relevant technetium thyr

Antibiotic-resistant Staphylococcus aureus (S. aureus) has severely threatened public health, colonizing approximately 20% of the human population. Thus, targeting S. aureus virulence factors appears to be a promising strategy for infection control. In this study, DP treatment significantly reduced sortase activity, inhibiting S. aureus adhesion and invasion. Computational biology and site-directed mutagenesis confirmed that DP bound to sortase via P30 and T105 residues, which occupied the active center of sortase and inhibited its activity. Additionally, treatment with DP significantly reduce

This study explored relationship between concentrations of perchlorate, nitrate, and thiocyanate and serum liver function markers using data from 3366 adults in the 2013-2018 National Health and Nutrition Examination Survey (NHANES) of the United States. Generalized linear model (GLM), restricted cubic spline (RCS) regression model, and quartile g-computation (Qgcomp) regression model were used to assess the relationship. The median concentrations of perchlorate, nitrate, and thiocyanate in urine were 2.33, 42,900, and 1060 ng/mL. The median concentrations for serum liver function indicators w

Perchlorate is an endocrine-disrupting compound (EDC) used for its oxidative properties and has been detected in many industrialized countries in drinking water, soil, foods, and breast milk. Perchlorate competitively inhibits the uptake of iodide into the thyroid, thus reducing thyroid hormone synthesis. Perchlorate causes steatosis in the liver, alters gonadal development, and increases tubule size in kidneys in some fish models but not others. Much less is known of perchlorate's effects on mammals with respect to these organs. We examined the morphological effects of environmentally relevan

Extensive carbonate precipitation has occurred on Mars. To gain insight into the carbonation mechanisms and formation processes under ancient Martian aqueous conditions, we examine the precipitation of carbonates resulting from atmospheric carbon fixation, focusing on interactions between various brines and silicate and perchlorate solutions in alkaline environments. The micro-scale morphology and composition of the resulting precipitates are analysed using ESEM micrographs, EDX chemical compositional analysis, X-ray diffraction, and micro-Raman spectroscopy. Our findings indicate a significan

Ion exchange chromatography-tandem mass spectrometry (IEC-MS/MS) has recently become the preferred method for detecting ionic substances in tea. In this study, an IEC-MS/MS method was developed for the rapid determination of chlorate and perchlorate residues in tea samples. The optimal sample extraction process, pretreatment column, and chromatographic and mass spectrometric conditions were systematically investigated. In the optimal process, the tea samples were ultrasonically extracted with methanol-water (13∶7, v/v), and a PRiME HLB SPE column was used to purify the sample extract. An AceCh

Perchlorate, a widespread environmental contaminant originating from various industrial applications, agricultural practices, and natural sources, poses potential risks to ecosystems and human health. While previous studies have highlighted its influence on the thyroid endocrine system and its impact on gonadal maturation, reproduction, and sex hormone synthesis, the specific interplay between thyroid and steroid hormones, in this context, remains largely unexplored. Therefore, this study was undertaken to investigate the adverse effects and underlying mechanisms triggered by exposure to sodiu
