Testosterone stimulates perineuronal nets development around parvalbumin cells in the adult canary brain in parallel with song crystallization.

Perineuronal nets (PNN) of the extracellular matrix are dense aggregations of chondroitin-sulfate proteoglycans that usually surround fast-spiking parvalbumin-expressing inhibitory interneurons (PV). The development of PNN around PV appears specifically at the end of sensitive periods of visual learning and limits the synaptic plasticity in the visual cortex of mammals. Seasonal songbirds display a high level of adult neuroplasticity associated with vocal learning, which is regulated by fluctuations of circulating testosterone concentrations. Seasonal changes in testosterone concentrations and

Tuina therapy alleviates neuropathic pain by modulating the CX3CL1/CX3CR1 axis to inhibit degradation of the perineuronal nets.

INTRODUCTION: Tuina, a traditional Chinese non-pharmacological therapy, is effective in treating neuropathic pain (NP). However, its underlying molecular mechanisms remain elusive. The degradation of perineuronal nets (PNNs) via the CX3CL1/CX3CR1 axis is a key driver of central sensitization. This study investigates whether Tuina exerts analgesia by modulating this specific neuro-immune axis to preserve PNNs integrity in a rat model of chronic compression of the dorsal root ganglion (CCD). METHODS: Sprague-Dawley rats were subjected to CCD surgery to establish the NP model. To validate the ro

Differential impacts of germline and adult aggrecan knockout in PV+ neurons on perineuronal nets and PV+ neuronal function.

Perineuronal nets (PNNs) are a condensed form of extracellular matrix primarily found around parvalbumin-expressing (PV+) interneurons. The postnatal maturation of PV+ neurons is accompanied with the formation of PNNs and reduced plasticity. Alterations in PNN and PV+ neuron function have been described for mental disorders such as schizophrenia and autism. The formation of PNNs is highly dependent on aggrecan, a proteoglycan encoded by the ACAN gene, but it remains unknown if it is produced by the PV+ neurons themselves. Thus, we established a knockout (KO) mouse model (ACANflx/PVcre) and an

Perineuronal nets in the rat medial prefrontal cortex alter hippocampal-prefrontal oscillations and reshape cocaine self-administration memories.

The medial prefrontal cortex (mPFC) is a major contributor to relapse to cocaine in humans and to reinstatement behavior in rodent models of cocaine use disorder. Output from the mPFC is modulated by parvalbumin (PV)-containing fast-spiking interneurons, the majority of which are surrounded by perineuronal nets (PNNs). Here we tested whether chondroitinase ABC (ABC)- mediated removal of PNNs prevented the acquisition or reconsolidation of a cocaine self-administration memory. ABC injections into the dorsal mPFC prior to training attenuated the acquisition of cocaine self-administration. Also,

Possible Involvement of Perineuronal Nets in Anti-Depressant Effects of Electroacupuncture in Chronic-Stress-Induced Depression in Rats.

Acupuncture can alleviate depression-like behaviors. However, the neural mechanisms behind the anti-depressive effect remain unknown. Perineuronal net (PNN) abnormalities have been reported in multiple psychiatric disorders. This study investigated the modulation and neural mechanism of PNNs in the anti-depressant process of electroacupuncture (EA) at Baihui (GV20) and Yintang (GV29) points. A rat depression model was induced by chronic unpredicted mild stress (CUMS). The results revealed that CUMS, applied for four weeks, specifically reduces PNNs around parvalbumin (PV). In addition, EA and

Schizophrenia-associated glycoprotein Adamtsl3 regulates perineuronal net formation, maintenance, and adult cortical plasticity.

Perineuronal nets (PNNs) regulate the maturation of parvalbumin-positive (PV+) interneurons during postnatal critical periods and continue to support their function thereafter. Their disruption is implicated in neuropsychiatric disorders such as schizophrenia, yet key endogenous PNN regulators remain unknown. Here, we identify the schizophrenia-associated glycoprotein Adamtsl3 as a PV+ cell-autonomous regulator of PNN integrity. Using mouse genetics, morphological and biochemical analyses, we demonstrate that Adamtsl3 deletion, both in early postnatal and adult stages, results in PNN deficits.

Perineuronal Net and Inhibitory Synapse Remodeling on Striatal Fast-Spiking Interneurons by Chronic Alcohol Exposure.

BACKGROUND: Alcohol use disorder is characterized by persistent drinking in the face of negative consequences. Such inflexible drinking requires dorsolateral striatum fast-spiking interneurons, which comprise approximately 1% of all striatal neurons. How chronic ethanol exposure affects fast-spiking interneuron physiology is poorly understood. METHODS: We exposed adult male and female mice to chronic intermittent ethanol and examined synaptic transmission onto dorsolateral striatum fast-spiking interneurons. RESULTS: We discovered that chronic ethanol exposure induced a dramatic loss of GABA

Perineuronal Net and Inhibitory Synapse Remodeling on Striatal Fast-spiking Interneurons by Chronic Alcohol Exposure.

Alcohol use disorder is characterized by persistent drinking in the face of negative consequences. Such inflexible drinking requires dorsolateral striatum fast-spiking interneurons, which comprise roughly 1% of all striatal neurons. How chronic ethanol exposure affects fast-spiking interneuron physiology is poorly understood. We discover in mice that chronic ethanol exposure induced a dramatic loss of GABAergic, but not glutamatergic, synapses onto dorsolateral striatum fast-spiking interneuron somata and proximal dendrites where perineuronal nets, a subdivision of the extracellular matrix, ar

Do Perineuronal Net Elements Contribute to Pathophysiology of Spinal Muscular Atrophy? In Vitro and Transcriptomics Insights.

Spinal muscular atrophy (SMA) is one of the most common childhood onset neurodegenerative disorders in global health whereby novel biomarkers and therapeutic targets are sorely needed. SMA is an autosomal recessive genetic disorder resulting in degeneration of α-motor neurons in the brain stem and spinal cord that leads to mortality in infants worldwide. In majority of the patients, SMA is caused by homozygous deletion of the SMN1 gene. The clinical spectrum of the SMA displays, however, large person-to-person variations where the underlying mechanisms are poorly understood. We report in this

Running to remember: The effects of exercise on perineuronal nets, microglia, and hippocampal angiogenesis in female and male mice.

Exercise is accepted as a positive health behaviour; however, the mechanisms of exercise on neuroprotection and cognitive health are not completely understood. The purpose of this study was to explore the neurobiological benefits of chronic treadmill exercise in female and male mice through its role in microglial content and morphology, cerebral vascularization, and perineuronal net (PNN) expression. We further examined how these neurobiological changes relate to spatial memory outcomes. Adult mice were assigned to a sedentary or treadmill exercise group for eight weeks. During the final week,

Running-induced neurogenesis reduces CA1 perineuronal net density without substantial temporal delay.

Aerobic exercise has many effects on brain function, particularly at the hippocampus. Exercise has been shown to increase the rate of adult neurogenesis within the dentate gyrus and decrease the density of perineuronal nets in area CA1. The relationship between the rate of neurogenesis and the density of perineuronal nets in CA1 is robust; however, these studies only ever examined these effects across longer time scales, with running manipulations of 4 weeks or longer. With such long periods of manipulation, the precise temporal nature of the relationship between running-induced neurogenesis a

Prenatal stress history modifies adolescent stress effects on adult social behavior and basolateral amygdala GABAergic neurons with perineuronal nets.

Developmental stress is a well-established risk factor for mental health disorders, yet the neural mechanisms underlying these outcomes remain incompletely understood. Inhibitory brain networks, particularly within the amygdala, are disrupted by stress and implicated in stress-related psychopathologies. Using a rodent model, the current study investigated the isolated and combined effects of prenatal and adolescent stress on adult social interactions and GABAergic neurons surrounded by perineuronal nets (PNNs) in the basolateral amygdala (BLA). Male and female rats were exposed to chronic vari

Regrouping reverses social impairments and prefrontal parvalbumin-perineuronal nets alterations induced by post-weaning social isolation in male mice.

Social isolation during early development can lead to abnormal adult behavior. These behavioral deficits are often associated with disruptions of perineuronal nets (PNNs), a condensed form of the brain's extracellular matrix. PNNs regulate neuroplasticity by enwrapping parvalbumin-expressing (PV) interneurons, and abnormal maturation of PNN in early life has been associated with behavioral deficits reminiscent of those following isolation. Past studies demonstrate that social regrouping following early isolation can improve behavioral deficits, but the underlying mechanisms of this recovery ar

Age-Dependent Sex Differences in Perineuronal Nets in an APP Mouse Model of Alzheimer's Disease Are Brain Region-Specific.

Alzheimer's disease (AD) is the most common form of dementia, which disproportionately affects women. AD symptoms include progressive memory loss associated with amyloid-β (Aβ) plaques and dismantled synaptic mechanisms. Perineuronal nets (PNNs) are important components of the extracellular matrix with a critical role in synaptic stabilisation and have been shown to be influenced by microglia, which enter an activated state during AD. This study aimed to investigate whether sex differences affected the density of PNNs alongside the labelling of microglia and Aβ plaques density. We performed neu

Identification of Functional Cell Groups in the Abducens Nucleus of Monkey and Human by Perineuronal Nets and Choline Acetyltransferase Immunolabeling.

The abducens nucleus (nVI) contains several functional cell groups: motoneurons of the singly-innervated twitch muscle fibers (SIF) and those of the multiply-innervated muscle fibers (MIF) of the lateral rectus muscle (LR), internuclear neurons (INTs) projecting to the contralateral oculomotor nucleus (nIII) and paramedian tract-neurons (PMT) that receive input from premotor neurons of the oculomotor system and project to the floccular region. In monkey, these cell populations can be delineated by their chemical signature. For correlative clinico-pathological studies the identification of the

Cannabidiol attenuates behavioral impairments and deficits in parvalbumin interneurons and their perineuronal nets induced by adolescent stress in adult mice.

Adolescence is a critical period for the development of neural circuits, and exposure to stressors during this stage constitutes a risk factor for psychiatric disorders. Previous studies have shown that adolescent stress in rodents leads to behavioral alterations and functional loss of parvalbumin-containing GABAergic (PV+) interneurons and their perineuronal nets (PNNs) in the medial prefrontal cortex (mPFC) and ventral hippocampus (vHip). Although less explored, somatostatin-expressing (SST+) interneurons may also be impaired. Given the reported anti-stress effects of cannabidiol (CBD) in ad

Activating systemic inflammation with lipopolysaccharide drives microglia-mediated degradation of perineuronal nets after cervical spinal cord injury in female rats.

Functional gains from rehabilitative motor training after spinal cord injury (SCI) depend on neuroplasticity, which naturally declines over time. We previously demonstrated that combining the pro-inflammatory compound lipopolysaccharide (LPS) with motor training reawakens neuroplasticity and enhances its therapeutic efficacy in chronic cervical SCI rats, though the underlying mechanisms remained unclear. Here, we investigated whether LPS-induced inflammation facilitates neuroplasticity by degrading perineuronal nets (PNNs), extracellular structures known to restrict recovery after SCI. Adult f

Perineuronal nets in cerebellar nuclei neurons orchestrate social behaviour via regulation of neuronal activity in circuits innervated by the cerebellum.

Certain types of neurons in the central nervous system are wrapped in extracellular matrix protein complexes named perineuronal nets (PNNs). While it is known that PNNs regulate neuronal activity by modulating synaptic plasticity, their pathophysiological role in psychiatric disorders has not been sufficiently clarified. Here, we demonstrate that the expression of PNNs in autism-spectrum-disorder (ASD)-associated mice, valproic acid-injected mice and chromodomain helicase DNA-binding protein 8 (Chd8) gene haploinsufficient mice was decreased in cerebellar nuclei neurons. Enhanced expression of parvalbumin and perineuronal nets in the medial prefrontal cortex after extended-access cocaine self-administration in rats.

The medial prefrontal cortex (mPFC) drives cocaine-seeking behaviour in rodent models of cocaine use disorder. Parvalbumin (PV)-containing GABAergic interneurons powerfully control the output of the mPFC, yet few studies have focused on how these neurons modulate cocaine-seeking behaviour. Most PV neurons are surrounded by perineuronal nets (PNNs), which regulate the firing of PV neurons. We examined staining intensity and number of PV and PNNs after long-access (6 h/day) cocaine self-administration in rats followed by either 8-10 days extinction ± cue-induced reinstatement or short-term (1-2

The absence of brain-specific link protein Bral2 in perineuronal nets hampers auditory temporal resolution and neural adaptation in mice.

Brain-specific link protein Bral2 represents a substantial component of perineuronal nets (PNNs) enwrapping neurons in the central nervous system. To elucidate the role of Bral2 in auditory signal processing, the hearing function in knockout Bral2(-/-) (KO) mice was investigated using behavioral and electrophysiological methods and compared with wild type Bral2(+/+) (WT) mice. The amplitudes of the acoustic startle reflex (ASR) and the efficiency of the prepulse inhibition of ASR (PPI of ASR), produced by prepulse noise stimulus or gap in continuous noise, was similar in 2-week-old WT and KO m
