Excitation-Contraction Coupling in Ureteric Smooth Muscle: Mechanisms Driving Ureteric Peristalsis.

The ureter acts as a functional syncytium and is controlled by a propagating plateau-type action potential (AP) which gives rise to a wave of contraction (ureteral peristalsis) via a process called excitation-contraction (E-C)coupling. The second messenger Ca2+ activates Ca2+/calmodulin-dependent myosin light chain kinase-dependent phosphorylation of 20-kDa regulatory light chains of myosin which leads to ureteric contraction. Ca2+ entry from the extracellular space via voltage-gated L-type Ca2+ channels (VGCCs) provides the major source of activator Ca2+, responsible for generation of both th

Gastric Peristalsis Control by Mono Situ Electrical Stimulation: a Preliminary Study.

BACKGROUND: This study was initiated 3 years ago when antral gastric stimulation was first used successfully to reduce free feeding in swine. METHODS: Three swine weighing 45 kg each were implanted with one subserosal bipolar electrode, positioned in the antrum, close to the pylorus, at the anterior side of the lesser gastric curvature. RESULTS: During 4 hours of kethamine anesthesia we paced the stomach by various patterns of electrical stimulation and obtained both forward and backward peristalsis, as well as gastric peresis. CONCLUSION: Variations in antral electrical stimulation produce ch

Ureteric drainage and peristalsis after stenting studied using colour Doppler ultrasound.

OBJECTIVE: To study peristalsis and drainage of the stented ureter under normal physiological conditions. PATIENTS AND METHODS: Fifteen non-obstructed patients with ureteric stents and 15 control subjects (i.e. 45 unstented ureters and 15 stented ureters) had ureteric jets and flow analysed using colour Doppler ultrasound (CDU). Cross-sectional and longitudinal studies were performed. RESULTS: Unstented ureters had forceful ureteric jets at a frequency of 0.5-4 jets/min per ureter. Asymmetry of frequency within subjects was common (1-3 jets/min). Some patent stented ureters had no recordable

Identification of superstalsis, a motility program distinct from peristalsis in the Drosophila midgut.

Enteric organs display the intrinsic ability to generate slow-wave potentials, rhythmic electrochemical events regulating gastrointestinal motility. In mammals, slow waves arise from the pacemaker activity of interstitial cells of Cajal, which, by recruiting visceral muscles, ultimately lead to peristalsis, a movement pattern conserved across species. Here, we identify a motility program in Drosophila distinct from peristalsis that generates strong whip-like midgut contractions. This activity, here named superstalsis, is associated with specific physiological conditions and involves longitudin

The use of transvaginal ultrasound alters physiologic uterine peristalsis in gynecologic participants.

OBJECTIVE: To study whether transvaginal ultrasound (TVUS) affected the uterine peristalsis (UP) patterns in nonpregnant participants. DESIGN: Institutional review board-approved, prospective observational cohort study. The noninvasive UP imaging (UPI) system uses electrode patches placed on the patient's skin just above the pubic bone and on the low back to quantify the 3-dimensional electrical activation pattern during UP by calculating peristalsis frequency, duration, magnitude, and activation ratio. A 20-minute UPI scan was completed without TVUS followed by a 10-minute UPI scan acquired

Diagnosis System for Swallowing and Peristalsis Function for Artificial Tongue and Esophagus Development.

With the progress of surgical technology, the survival rate after resection of esophageal and tongue carcinomas has improved. However, the surgical protocol for esophageal and tongue surgery is complex, and surgery for elderly esophageal and tongue carcinoma patients with cardiopulmonary dysfunction is difficult. Using an artificial tongue and esophagus will be helpful for patients. However, peristalsis of foods depends on food size, taste, and viscosity. This study developed and evaluated a new diagnosis machine for drinking and peristalsis motion. Before clinical evaluation, animal experimen

Influence of prucalopride on esophageal secondary peristalsis in reflux patients with ineffective motility.

BACKGROUND AND AIM: Ineffective esophageal motility (IEM) is associated with gastroesophageal reflux disease. Secondary peristalsis contributes to esophageal clearance. Prucalopride promotes secondary peristalsis by stimulating 5-hydroxytrypatamine 4 receptors in the esophagus. We aimed to determine whether prucalopride would augment secondary peristalsis in gastroesophageal reflux disease patients with IEM. METHODS: After a baseline recording of primary peristalsis, secondary peristalsis was stimulated by slow and rapid mid-esophageal injections of air in 15 patients with IEM. Two separate s

Effect of galanin and galanin antagonists on peristalsis in esophageal smooth muscle in the opossum.

Galanin, a neuropeptide that is widely distributed in the esophageal nerves, is known to exert a neuromodulatory action in the gut. These studies examined the effect of galanin and galanin antagonists on esophageal peristalsis in anesthetized opossums in vivo. Intraluminal esophageal pressures were recorded at 1, 3, 5, 7, and 9 cm above the lower esophageal sphincter. Esophageal peristaltic contractions were induced by swallow and short- (1-s) and long-train (10-s) vagal stimulation (VS). Galanin (1 nmol/kg) inhibited the amplitude of swallow-induced peristaltic contractions and increased peri

Lidocaine inhibition of esophageal peristalsis and lower esophageal sphincter pressure in baboons.

Intravenous lidocaine was infused at 0.82 ml/min in a concentration of 1.2 mg/ml (2.3 mg/kg) for 120 min in awake chair-restrained baboons (Papio anubis), and measurements of esophageal peristalsis and LES pressure were compared before and after lidocaine or control infusions. Lidocaine produced a progressive and significant (P less than 0.05) decrease in amplitude in the peristaltic wave in the smooth muscle portion of the distal esophagus during the 120-min infusion. Lower esophageal sphincter pressure was similarly significantly lower than control after the infusion of lidocaine (P less tha

Roles of adrenergic drugs in the renal pelvic pacemaker control of ureteral peristalsis.

Experiments were performed to explore the influence of noradrenaline and isoproterenol on the ureteral peristalsis in the canine pelviureteral system in vitro. The rhythmic changes of intrapelvic pressure no longer became manifest, and simultaneously recorded electromyograms revealed disappearance of peristalsis at sites distal to the upper region of the renal pelvis following application of isoproterenol, while the pacemaker at the pelvicalyceal border continued to discharge at an unaltered rate. It is concluded from our findings that both noradrenaline and isoproterenol act upon conduction o

[Pharmacological analysis of the inhibition of peristalsis of the isolated guinea-pig ileum by tetramethylammonium].

Tetramethylammonium in small concentrations contracted the longitudinal muscle of the guinea-pig isolated ileum, but had no effect on the peristaltic reflex. However, in high concentrations, it contracted the longitudinal muscle and blocked the peristaltic reflex. Acetylcholine, methacholine, carbachol, eserine and neostigmine antagonized the peristaltic block previously produced by tetramethylammonium. On the other hand, propionylcholine, butyrylcholine, nicotine, pilocarpine, arecoline, histamine and 5-hydroxytryptamine did not remove the peristaltic block. From these experiments, it is conc

Ureteric peristalsis studies in loin pain and haematuria syndrome: another diagnostic disappointment.

Although not a cause of progressive renal damage, loin pain and haematuria syndrome is nevertheless associated with significant morbidity. The management of pain often presents a formidable problem to urologists, physicians and general practitioners. An earlier study noted hold-up of urine in the renal pelvis and implicated this in the pathogenesis of pain. In this study we have failed to demonstrate an excess incidence of disordered urinary peristalsis in loin pain and haematuria syndrome.

Bolus pressure and bolus mismatch in patients with dysphagia and preserved esophageal peristalsis.

Dysphagia is common in patients with preserved contraction phase of esophageal peristalsis, such as those diagnosed with functional dysphagia (FD) or esophagogastric junction outflow obstruction (EGJOO); it amounts to ≥50% patients undergoing high-resolution manometry impedance (HRMZ) study. We aimed to characterize the distension phase of esophageal peristalsis (bolus domain) and identify abnormalities that may contribute to dysphagia in these patients. HRMZ recordings from 35 healthy controls, 35 patients with FD, and 35 patients with EGJOO were analyzed. Distension-contraction plots were us

Primary Peristalsis Is the Dominant Mechanism of Refluxate Clearance Following Gastroesophageal Reflux.

BACKGROUND: Refluxate volume and pH drop following gastroesophageal reflux are mostly cleared by peristalsis. We evaluated the roles of primary volume clearing peristaltic wave (VCPW), secondary VCPW, post-reflux swallow-induced peristaltic wave (PSPW), and late primary peristaltic wave (LPPW) in refluxate clearance. METHODS: We retrospectively analyzed pH-impedance studies performed off therapy in 40 patients with typical esophageal symptoms. Mechanism of refluxate clearance was evaluated for each reflux episode (primary VCPW vs. secondary VCPW vs. none), as well as presence of PSPW, LPPW wh
