Increased skin permeability for lipophilic molecules.

Treatment of the epidermis with surfactants can markedly increase the transport of polar molecules but only marginally increases the transport of nonpolar (lipophilic) molecules. Thus, other vehicle systems are needed to increase the transport of lipophilic molecules. One method to accomplish this increased transport is to add small quantities of polar lipids to a base vehicle containing propylene glycol. The transport of nonpolar materials such as salicylic acid can be increased by an order of magnitude by the addition of small amounts of fatty acids or alcohols to a formulation. The effect o

Study on the evolution mechanism of permeability and displacement efficiency in CO2-ECBM in low-permeability coal seam.

Unmineable coal seams and residual coal resources in closed mines provide ideal carriers for CO₂ sequestration. However, the evolution mechanisms of permeability and displacement efficiency during CO₂-ECBM in low-permeability coal seams remain unclear, which restricts the engineering application efficiency of this technology. In this study, the No. 15 coal seam of the Lutaishan Coal Mine was taken as the research object. A fully coupled thermal-hydraulic-mechanical model of CO₂-ECBM was constructed and validated using COMSOL Multiphysics software, and a quantitative evaluation method for key i

Influence of permeability enhancers on the paracellular permeability of metformin hydrochloride and furosemide across Caco-2 cells.

Permeability enhancers can affect absorption of paracellularly transported drugs. This study aims to evaluate effects of permeability enhancers (chitosan, methyl-β -cyclodextrin, sodium caprate, sodium lauryl sulfate, etc.) on the permeability of paracellularly absorbed furosemide and metformin hydrochloride. Methyl thiazole tetrazolium bromide test was carried out to determine the drug concentrations in permeability study. Trans-epithelial electrical resistance (TEER) values determined to assess the integrity of tight junctions. Permeability enhancers were applied at different concentrations

Soluble CD40 ligand directly alters glomerular permeability and may act as a circulating permeability factor in FSGS.

CD40/CD40 ligand (CD40L) dyad, a co-stimulatory bi-molecular complex involved in the adaptive immune response, has also potent pro-inflammatory actions in haematopoietic and non-haematopoietic cells. We describe here a novel role for soluble CD40L (sCD40L) as modifier of glomerular permselectivity directly acting on glomerular epithelial cells (GECs). We found that stimulation of CD40, constitutively expressed on GEC cell membrane, by the sCD40L rapidly induced redistribution and loss of nephrin in GECs, and increased albumin permeability in isolated rat glomeruli. Hydrogen peroxide constricts rat arteries by activating Na+-permeable and Ca2+-permeable cation channels.

Oxidative stress is associated with many cardiovascular diseases, such as hypertension and arteriosclerosis. Oxidative stress reportedly activates the L-type voltage-gated calcium channel (VDCCL) and elevates [Ca2+]i in many cells. However, how oxidative stress activates VDCCL under clinical setting and the consequence for arteries are unclear. Here, we examined the hypothesis that hydrogen peroxide (H2O2) regulates membrane potential (Em) by altering Na+ influx through cation channels, which consequently activates VDCCL to induce vasoconstriction in rat mesenteric arteries. To measure the ton

Predicting the extent of metabolism using in vitro permeability rate measurements and in silico permeability rate predictions.

The Biopharmaceutics Drug Disposition Classification System (BDDCS) can be utilized to predict drug disposition, including interactions with other drugs and transporter or metabolizing enzyme effects based on the extent of metabolism and solubility of a drug. However, defining the extent of metabolism relies upon clinical data. Drugs exhibiting high passive intestinal permeability rates are extensively metabolized. Therefore, we aimed to determine if in vitro measures of permeability rate or in silico permeability rate predictions could predict the extent of metabolism, to determine a referenc

Improved Method for Measuring the Permeability of Nanoporous Material and its Application to Shale Matrix with Ultra-Low Permeability.

Nanoporous materials have a wide range of applications in clean energy and environmental research. The permeability of nanoporous materials is low, which affects the fluid transport behavior inside the nanopores and thus also affects the performance of technologies based on such materials. For example, during the development of shale gas resources, the permeability of the shale matrix is normally lower than 10-3 mD and has an important influence on rock parameters. It is challenging to measure small pressure changes accurately under high pressure. Although the pressure decay method provides an

Model-based evaluation of connexin hemichannel permeability.

Connexin (Cx) hemichannels form intercellular gap junction channels but can also function independently. These large-pore channels are permeable not only to ions but also to small signalling molecules and metabolites. This functional property is relevant to many physiological processes and can be altered by various biochemical factors or disease-causing mutations. In this study, we present a methodology for quantifying and comparing the permeabilities of hemichannels formed by different Cx isoforms using a combination of fluorescence imaging, electrophysiological recordings and mathematical mo

A novel cell permeability assay for macromolecules.

BACKGROUND: Many cell permeabilisation methods to mediate internalisation of various molecules to mammalian or bacterial cells have been developed. However, no size-specific permeability assay suitable for both cell types exists. RESULTS: We report the use of intrinsically biotinylated cell components as the target for reporter molecules for assessing permeabilisation. Due to its well-described biotin binding activity, we developed an assay using Streptavidin (SAv) as a molecular weight marker for assessing eukaryotic and prokaryotic cell internalisation, using flow cytometry as a readout. GM1-Binding Conjugates To Improve Intestinal Permeability.

Drugs and proteins with poor intestinal permeability have a limited oral bioavailability. To remediate this problem, a receptor-mediated endocytosis and transcytosis approach was explored. Indeed, the nontoxic β subunit of cholera toxin (CTB) can cross the intestinal barrier by binding to receptor GM1. In this study, we explored the use of GM1-binding peptides and CTB as potential covalent carriers of poorly permeable molecules. GM1-binding peptides (G23, P3) and CTB were conjugated to poorly permeable fluorescent probes such as fluorescein isothiocyanate (FITC) and albumin-FITC using triethyl

Aroma permeability of hydroxypropyl maize starch films.

In this study, the role of water and the impact of the glass to rubber transition on aroma mass transport through hydroxypropyl maize starch films were followed. The permeability of four aroma compounds (ethanol, pyrazine, menthone, and decanone) was monitored by atmospheric pressure chemical ionization mass spectroscopy. The increase in water content within the film promoted greater water diffusion and film flexibility and resulted in enhanced transport of the four aroma compounds through the film. At low water contents in the glassy state, the permeability of ethanol was much higher than tho

pH regulated anion permeability of aquaporin-6.

The kidney is a model organ for transport physiology (Nielsen 1996). AQPs are well-characterized in mammalian kidneys, where they facilitate transepithelial water reabsorption. Most renal AQPs are expressed either in proximal tubule cells or in collecting duct principal cells, which are known as sites for water reabsorption. AQP1 is present in both apical and basolateral membranes of proximal tubules, and in descending limbs of Henle's loop where 70% of filtrated water is isoosmotically reabsorbed (King and Agre 1996). AQP2 is expressed in principal cells of the collecting duct; in response to

Ultrasonic sonoporation can enhance the prostate permeability.

As possible existence of the blood-prostate barrier and decreased prostate permeability due to inflammation, it is difficult to form an effective drug concentration in prostate tissues, which influences medication efficacy. This is one of principal reasons why it is difficult to treat prostate diseases including prostatitis etc. How to increase the permeability of drugs into prostate is inevitable to become an issue concerned by clinical research. Ultrasonic sonoporation may increase permeability of cells and tissues, so we propose a hypothesis that sonoporation induced by ultrasonic cavitatio

Insulin alters the permeability of sheep pleura.

AIM: Insulin promotes ion transportation across epithelia, mainly kidneys, leading to water and electrolyte abnormalities, possibly causing 'insulin oedema syndrome', which rarely presents as pleural effusion. Direct stimulation of sheep pleura by insulin and the possible electrophysiology mechanisms involved were investigated. MATERIAL AND METHODS: Sheep visceral and parietal pleural specimens were mounted between Ussing chambers. Insulin solutions (10 (-9) to 10 (-5) M), L-NAME, Nitroprussid sodium, amiloride and ouabain were used. Trans-mesothelial Resistance was determined. Immunohistoche

The mitochondrial permeability transition in neurologic disease.

Mitochondria, being the principal source of cellular energy, are vital for cell life. Yet, ironically, they are also major mediators of cell death, either by necrosis or apoptosis. One means by which these adverse effects occur is through the mitochondrial permeability transition (mPT) whereby the inner mitochondrial membrane suddenly becomes excessively permeable to ions and other solutes, resulting in a collapse of the inner membrane potential, ultimately leading to energy failure and cell necrosis. The mPT may also bring about the release of various factors known to cause apoptotic cell dea

Effects of hypoxia on corneal epithelial permeability.

PURPOSE: Epithelial permeability to fluorescein (Pdc) increases with closed-eye soft contact lens wear. Possible mechanisms responsible for this change include corneal hypoxia (and acidosis) during overnight wear and interference of the lens with normal tear exchange. In this study we used a local environmental chamber to examine the effects of hypoxia on Pdc. METHODS: After baseline slit-lamp examination, autofluorescence, and central corneal thickness measurements, 35 subjects were fitted with a pair of airtight swimming goggles. One eye was exposed to a humidified gas mixture containing 95
