Hygromycin A Treatment of Borrelia burgdorferi-Infected Peromyscus leucopus Suggests Potential as a Reservoir-Targeted Antibiotic.

Lyme disease spirochetes are maintained in natural reservoirs before spilling over into human populations. Targeting these reservoirs with vaccinations or antibiotics could impact the Borrelia burgdorferi enzootic cycle and reduce the risk of human Lyme disease. In this work we report that the narrow-spectrum antibiotic hygromycin A is sufficient to disrupt B burgdorferi transmission from the main eastern US reservoir, Peromyscus leucopus, to ticks. Additionally, hygromycin A-containing baits can clear B burgdorferi from P leucopus. These studies lay the foundation for the use of hygromycin A

A systematic review of the distribution and prevalence of viruses detected in the Peromyscus maniculatus species complex (Rodentia: Cricetidae).

The North American Deermouse, Peromyscus maniculatus, is one of the most widespread and abundant mammals on the continent. It is of public health interest as a known host of several viruses that are transmissible to humans and can cause illness, including the acute respiratory disease Hantavirus pulmonary syndrome (HPS). However, recent taxonomic studies indicate that P. maniculatus is a complex of multiple species, raising questions about how to identify and interpret three decades of hantavirus monitoring data. We conducted a systematic review investigating the prevalence and spatial distrib

Effects of partial cage dividers on animal welfare and social behavior in California mice (Peromyscus californicus).

California mice (Peromyscus californicus) are an important model species for studying social behaviors that are not readily expressed in standard laboratory mouse and rat lines. Under natural conditions, this species is highly territorial, and both males and females exhibit high levels of aggression in laboratory conditions. Like other rodent species, fighting among cage-mates can create animal welfare problems for California mice. Here we tested whether the use of partial cage dividers could reduce the impact of cage-mate fighting in California mice. Partial cage dividers were made of polycar

Broad diversity of host responses of the white-footed mouse Peromyscus leucopus to Borrelia infection and antigens.

Peromyscus leucopus, the white-footed mouse, is one of the more abundant mammals of North America and is a major reservoir host for at least five tickborne diseases of humans, including Lyme disease and a newly-recognized form of relapsing fever. In comparison to Mus musculus, which is not a natural reservoir for any of these infections, there has been little research on experimental infections in P. leucopus. With the aim of further characterizing the diversity of phenotypes of host responses, we studied a selection of quantitative traits in colony-bred and -reared outbred P. leucopus adults

Effects of Trichinella spiralis on survival, total mass, and organ mass of oldfield mice (Peromyscus polionotus).

Trichinella spiralis is a parasitic nematode that infects many mammals, including humans. Hosts may experience significant physiological changes or may die because of acute inflammatory immune responses toward the parasite. In this study, oldfield mice (Peromyscus polionotus) were used as a new experimental host for T. spiralis. Males of P. polionotus were infected with increasing doses of T. spiralis to determine the effect infection had on survival, mass change, total mass, and relative organ masses. Total juvenile worm burden increased in an asymptotic fashion with infective dose. Large dos

Effects of some factors on the susceptibility of Peromyscus leucopus to infestation by larvae of cuterebra fontinella (Diptera: Cuterebridae).

Two levels of 3 factors, (1) host age, (2) host dietary state in regard to vitamin A, and (3) previous infestation history, were tested individually and in combinations for their effects on the susceptibility of Peromyscus leucopus to infestation by larvae of the rodent bot fly, Cuterebra fontinella. Previous infestation was the only significant single factor. Previously uninfested (Pu) mice usually were more susceptible to infestation than previously infested (Pi) mice. Interactions of age and state of vitamin A had the following effects: 1) Young Pu mice that had been denied vitamin A were m

Integrated control of juvenile Ixodes scapularis parasitizing Peromyscus leucopus in residential settings in Connecticut, United States.

Lyme disease continues to be the most common vector-borne disease in the United States with an estimated 330,000 human cases annually. In the eastern United States, the blacklegged tick, Ixodes scapularis, is the primary vector of the Lyme disease spirochete, Borrelia burgdorferi, and the white-footed mouse, Peromyscus leucopus, is a primary reservoir host. In four residential neighborhoods in Connecticut over three years, we tested the effectiveness of different low-toxicity integrated tick management approaches to control larval and nymphal I. scapularis parasitizing P. leucopus. Combination

A method to distinguish morphologically similar Peromyscus species using extracellular RNA and high-resolution melt analysis.

A method applying high-resolution melt (HRM) analysis to PCR products copied and amplified from extracellular RNA (exRNA) has been developed to distinguish two morphologically similar Peromyscus species: Peromyscus leucopus and Peromyscus maniculatus. P. leucopus is considered the primary reservoir host of Borrelia burgdorferi, the causative agent for Lyme disease in North America. In northern Minnesota the habitat ranges of P. leucopus overlaps with that of P. maniculatus. Serum samples from live mice of both species were collected from cheek bleeds, total extracellular RNA (exRNA) was extrac

Pathogenesis of experimental vesicular stomatitis virus (New Jersey serotype) infection in the deer mouse (Peromyscus maniculatus).

The pathogenesis of vesicular stomatitis virus (VSV) infection has not been investigated previously in native New World rodents that may have a role in the epidemiology of the disease. In the present study, 45 juvenile and 80 adult deer mice (Peromyscus maniculatus) were inoculated intranasally with VSV New Jersey serotype (VSV-NJ) and examined sequentially over a 7-day period. Virus was detected by means of immunohistochemistry and in situ hybridization in all tissues containing histologic lesions. Viral antigen and mRNA were observed initially in olfactory epithelium neurons, followed by olf

Bot fly (Cuterebra fontinella fontinella) parasitism of cotton mice (Peromyscus gossypinus) on St. Catherines Island, Georgia.

From February 1993 through October 1994, 382 cotton mice (Peromyscus gossypinus) were live-trapped and examined for bot fly (Cuterebra fontinella fontinella) larvae. Thirty-nine (10%) of the mice were parasitized by a total of 44 bot fly larvae. All bots were situated in the inguinal or lower abdominal region of the mice. Monthly data strongly suggested that this bot is bivoltine at this latitude with distinct summer and winter population peaks. Prevalence of parasitism was highest in June 1994 when 56% of 16 mice examined were infested with a mean intensity of 1.3 bots. Adult flies were succe

Borrelia burgdorferi (Spirochaetales: Spirochaetaceae) Infection Prevalence and Host Associations of Ticks Found on Peromyscus spp. in Maryland.

Lyme disease, caused by Borrelia burgdorferi sensu stricto and most commonly transmitted by Ixodes scapularis Say (Ixodida: Ixodidae), is the most common tick-borne disease in Maryland. Because B. burgdorferi s.s. is maintained in enzootic cycles among wild mice (Peromyscus spp) and Ixodes spp ticks, differing patterns of parasitism of ticks on mice could impact the infection prevalence with B. burgdorferi. We determined the infection prevalence of Peromyscus spp as well as questing and partially engorged nymphal ticks collected at six sites on private land in five counties in Maryland from Ma

Vertical Transmission: A Vector-Independent Transmission Pathway of Babesia microti in the Natural Reservoir Host Peromyscus leucopus.

BACKGROUND: Babesia microti, a malaria-like pathogen, is increasing in mammal and human populations in endemic areas and is unlikely to be the sole result of horizontal pathogen transmission. METHODS: Peromyscus leucopus mice, natural reservoir hosts, were infected via Ixodes scapularis nymphs. Infected parental females (n = 6) produced F1 offspring (n = 36) that were screened for B. microti using quantitative PCR. Xenodiagnostic larvae were fed on infected offspring to determine horizontal transmission and pathogen viability. Fifty engorged larvae were screened; the rest were allowed to molt

Liver microbiome of Peromyscus leucopus, a key reservoir host species for emerging infectious diseases in North America.

Microbiome studies generally focus on the gut microbiome, which is composed of a large proportion of commensal bacteria. Here we propose a first analysis of the liver microbiome using next generation sequencing as a tool to detect potentially pathogenic strains. We used Peromyscus leucopus, the main reservoir host species of Lyme disease in eastern North America, as a model and sequenced V5-V6 regions of the 16S gene from 18 populations in southern Quebec (Canada). The Lactobacillus genus was found to dominate the liver microbiome. We also detected a large proportion of individuals infected by

Peromyscus maniculatus, a possible reservoir host of Borrelia garinii from the Gannet Islands off Newfoundland and Labrador.

Thirty-five deer mice, Peromyscus maniculatus, were trapped on Gannet Cluster 2 (GC-2), one of a group of islands numbered by convention in the Gannet Island Archipelago, and examined for ectoparasites. One species each of Acari (Ixodes uriae) and Siphonaptera (Orchopeas leucopus) were recovered. Samples of mice favored males to females (3.4∶1). Twenty-nine percent (10) of the mice were free of ectoparasites. Males were more heavily parasitized than females when both parasites were considered. No ticks were recovered from the female mice, while the males that were parasitized carried adult Ixo

Type 2 diabetes mellitus, hyperlipidemia, and extremity lesions in California mice (Peromyscus californicus) fed commercial mouse diets.

BACKGROUND AND PURPOSE: We characterized abnormalities of carbohydrate and lipid metabolism and determined whether those metabolic abnormalities are associated with extremity lesions in California mice (Peromyscus californicus). METHODS: Blood samples were evaluated for glucose, cholesterol, triglyceride, and insulin concentrations. Necropsy and histologic evaluation were done on selected mice, including staining pancreatic sections for insulin. Physical examinations also were performed. RESULTS: California mice were found to have Type 2 diabetes mellitus (T2DM). Sections of pancreas from di

Pathogenesis of Trypanosoma brucei infection in deer mice (Peromyscus maniculatus): hematologic, erythrocyte biochemical, and iron metabolic aspects.

Trypanosoma brucei EATRO 110 infection in deer mice (Peromyscus maniculatus) produced anemia in 15 of 42 mice between postinoculation days 14 and 70. The infected anemic (IA) mice had significantly higher reticulocyte counts (P less than 0.025), spleen (P less than 0.001) and liver (P less than 0.005) weights, and higher parasitemia than did infected nonanemic (INA) mice. gamma-Globulin concentrations of infected mice were markedly increased, and values for INA mice were 10% higher than values for IA mice. The effects of diet composition on body fat and hepatic steatosis in an animal (Peromyscus californicus) model of the metabolic syndrome.

The objective of this research was to determine body composition, total fat content, fat distribution, and serum leptin concentration in hyperlipidemic (high responder, HR) and normolipidemic (low responder, LR) California mice (Peromyscus californicus). In our initial experiments, we sought to determine whether differences in regional fat storage were associated with hyperlipidemia in this species. To further characterize the hepatic steatosis in the mice, we performed 2 additional experiments by using a diet containing 45% of energy as fat. The body fat content of mice fed a low fat-diet (12

Phenotypic plasticity of reproductive traits in response to food availability and photoperiod in white-footed mice (Peromyscus leucopus).

Although most temperate-zone mammals are seasonal breeders, many populations display variation in winter reproductive phenotype. For most mammals, the primary environmental cues regulating reproductive status are food availability and photoperiod, and these two factors can interact in their effect. Low food availability is primarily thought to suppress reproduction by reducing body mass and thereby forcing energy allocations to survival alone. However, because most small mammals rely on an increase in food intake rather than stored nutrients for reproduction, we hypothesized that food availabi

Exposure of nontarget white-footed mice (Peromyscus leucopus) to second-generation anticoagulant rodenticides in an urban context.

The pathways of nontarget exposure to anticoagulant rodenticides (ARs) are poorly understood and have yet to be examined in Ontario, Canada. We sought to determine if second-generation anticoagulant rodenticide (SGAR) baiting practices used by pest management professionals targeting commensal rodents may be causing rodenticide spillover into nontarget rodents in urban wildland areas, which could act as a vector of ARs to predators. We used snap traps to capture white-footed mice (Peromyscus leucopus) in urban wildland areas of Toronto and Vaughan, Ontario, near ongoing rodenticide baiting prog
