Pseudomonas aeruginosa is a major opportunistic pathogen implicated in a wide range of infections, including chronic respiratory infections, burn wound infections, urinary tract infections, and device-associated infections. Its intrinsic and acquired resistance mechanisms, particularly its capacity for biofilm formation, pose serious challenges to conventional antibiotic therapy. With the continued rise of multidrug-resistant and pan-drug-resistant strains, the need for alternative therapeutic strategies has become increasingly urgent. Phages, viruses that specifically recognize and lyse bacte

Antibiotic resistance continues to rise, necessitating alternative strategies. Bacteriophages have emerged as promising natural antibacterial agents, offering a targeted approach to combating bacterial infections. Combining bacteriophages with nanoparticles presents a novel approach that could enhance antibacterial potency while reducing the risk of resistance. While phage/antibiotic cocktails are widely explored to enhance antibacterial efficacy and prevent resistance, research on phage/nanoparticle combinations remains limited. We explore the synergy between green tea extract-capped silver n

UNLABELLED: Methicillin-resistant Staphylococcus aureus (MRSA) causes biofilm-related medical device infections. Phage-antibiotic combinations offer potential therapy due to proven in vitro antibiofilm efficacy. We evaluated phage-antibiotic synergy against biofilms using modified checkerboard and 24-h time-kill assays. Humanized-simulated daptomycin (DAP) (10, 8, and 6 mg/kg q24h) and ceftaroline (CPT) (600 mg q12h) were combined with Intesti13, Sb-1, and Romulus phages (tMOI 1, q12h). Assays were conducted in 168-h biofilm reactor models against DAP non-susceptible (DNS) vancomycin intermedi

Persistent antibiotic use results in the rise of antimicrobial resistance with limited or no choice for multidrug-resistant (MDR) and extensively drug resistant (XDR) bacteria. This necessitates a need for alternative therapy to effectively combat clinical pathogens that are resistant to last resort antibiotics. The study investigates hospital sewage as a potential source of bacteriophages to control resistant bacterial pathogens. Eighty-one samples were screened for phages against selected clinical pathogens. Totally, 10 phages were isolated against A. baumannii, 5 phages against K. pneumonia

Acute pancreatitis (AP) is a common clinical inflammatory condition whose severity is closely associated with both local pancreatic injury and systemic complications. Research indicates that gut microbiota dysbiosis and subsequent bacterial translocation can exacerbate disease progression via the gut-pancreas axis, representing a critical factor influencing patient outcomes. Although antibiotics remain a primary approach for infection control, their extensive use has led to increasingly prominent issues of bacterial resistance, and their broad-spectrum nature may further disrupt microbial home

BACKGROUND: Lower respiratory tract infections (LRTIs) pose a significant threat to global health, causing more than 2 million deaths worldwide. This menace is intensified by the alarming increase in drug resistance, which limits the availability of effective antibiotics for bacterial respiratory infections. Consequently, there is an urgent demand for alternative therapeutic options. Phage therapy (PT) has re-emerged as a promising therapeutic approach and as an adjunct to antibiotic treatment.
OBJECTIVE: This systematic review synthesises the application of PT for LRTIs in humans, providing

In the face of rising antimicrobial resistance, bacteriophage therapy, also known as phage therapy, is seeing a resurgence as a potential treatment for bacterial infections including urinary tract infection (UTI). Primarily caused by uropathogenic Escherichia coli, the 400 million UTI cases annually are major global healthcare burdens and a primary cause of antibiotic prescriptions in the outpatient setting. Phage therapy has several potential advantages over antibiotics including the ability to disrupt bacterial biofilms and synergize with antimicrobial treatments with minimal side effects or

Solutions for bone and joint infection (BJI) are needed where conventional treatments are inadequate. Bacteriophages (phages) are naturally occurring viruses that infect bacteria and have been harnessed for refractory bone and joint infections (BJI) in many case reports. Here we examine the safety and efficacy of English-language published cases of BJI since 2010 with phage therapy. From 33 reported cases of BJI treated with phage therapy, 29 (87%) achieved microbiological or clinical success, 2 (5.9%) relapsed with the same organisms, and 2 (5.9%) with a different organism. Of these 4 relapse

Klebsiella pneumoniae (K. pneumoniae) is a WHO priority pathogen, with multidrug-resistant (MDR) and carbapenem-resistant (CRKP) strains causing severe, often untreatable urinary tract infections (UTIs). The therapeutic impasse has reignited interest in bacteriophage (phage) therapy, a precision antibacterial approach that uses viruses to selectively lyse bacterial pathogens. The potential of phage therapy for MDR K. pneumoniae UTIs is critically evaluated in this review. We summarize the underlying biological rationale, concentrating on phage-mediated lysis and biofilm disruption. We then exa

Phage therapy (PT) shows promising potential in managing biofilm infections, which include refractory orthopedic infections. We report the case of a 13-year-old girl who developed chronic polymicrobial biofilm infection of a pelvic bone allograft after Ewing's sarcoma resection surgery. Chronic infection by Clostridium hathewayi, Proteus mirabilis and Finegoldia magna was worsened by methicillin-susceptible Staphylococcus aureus exhibiting an inducible Macrolides-Lincosamides-Streptogramin B resistance phenotype (iMLSB). After failure of conventional conservative treatment, combination of in s

Antimicrobial resistance in multidrug-resistant Escherichia coli has severely compromised the efficacy of conventional antibiotics, particularly in hospital-acquired infections. Bacteriophage therapy offers a targeted approach, yet the rapid emergence of phage-resistant mutants during monotherapy limits its durability. However, phage cocktail therapy can overcome this issue. In this study, we isolated and characterized six novel lytic bacteriophages from hospital wastewater that specifically infect MDR E. coli clinical isolates. All six phages exhibited strictly lytic lifestyles, and genomic a

The emergence of multidrug-resistant Acinetobacter baumannii (MDR-AB) has raised concerns regarding the lack of effective treatment options, prompting interest in phage therapy. In this study, two phages, designated vB_MZM_2AB-P and vB_MZM_4AB-P, were isolated and characterized for their lytic activity against MDR-AB strains. vB_MZM_2AB-P demonstrated considerable stability, retaining activity at 70 °C and pH 11.00, with over 37.10% survival rate after three months of storage. It displayed a 10-min latent period and a burst size of 39.72 PFU/cell. vB_MZM_4AB-P was stable at 50 °C and pH 7.00,

Bacteriophages (phages) are the viral predators of bacteria. Their ability to infect and kill bacterial strains has been harnessed for use in clinical practice for more than a century, but their development stalled for decades in many regions. Now, in the face of increasingly complex bacterial infections that fail conventional therapeutic options, and the growing global threat of antibiotic resistance, interest in phage therapy has resurged. Data evaluating the impact of phage therapy via clinical trials are steadily increasing, but remain sparse. Yet, an ever-growing number of case reports an

Enterotoxigenic Escherichia coli (ETEC) is a major causative agent of animal diarrhea, severely constraining the sustainable development of animal husbandry and endangering food safety and human health. Driven by the worsening antibiotic resistance crisis and the policy implementation of antibiotic reduction and replacement, phages have attracted extensive attention as novel antibacterial candidates. With prominent strengths including rapid bactericidal action, strict host specificity, no disturbance to intestinal microflora, and no drug residues, phages exhibit promising prospects for antibac

Phage therapy is a promising approach against multidrug-resistant infections, yet systemic administration can lead to incomplete cures. We investigated the distribution, immune responses, and efficacy of the therapeutic phage KPP10 delivered via intranasal or intraperitoneal (IP) routes in murine Pseudomonas aeruginosa lung infection models. Intranasal pre-treatment achieved markedly higher localization of KPP10 in the lungs and bronchoalveolar compartment compared to IP delivery. Intranasal administration elicited minimal systemic antibody responses, whereas IP injection triggered significant
