Chemical Profiling of Polar Lipids and the Polyphenolic Fraction of Commercial Italian Phaseolus Seeds by UHPLC-HRMS and Biological Evaluation.

The common bean (Phaseolus vulgaris L.) is one of the oldest food crops in the world. In this study, the ultra-high-performance liquid chromatography high-resolution mass spectrometry (UHPLC-MS/MS) technique was used to characterize the polar lipid composition and polyphenolic fraction of five bean varieties commonly consumed in Italy: Cannellino (PVCA), Controne (PVCO), Borlotti (PVBO), Stregoni (PVST), and Vellutina (PVVE). Lipid content represents a minor fraction of the whole metabolome in dry beans, and little is known about their polar lipids, which could be potentially bioactive compone

Root environment is a key determinant of fungal entomopathogen endophytism following seed treatment in the common bean, Phaseolus vulgaris.

The common bean is the most important food legume in the world. We examined the potential of the fungal entomopathogens Beauveria bassiana and Metarhizium anisopliae applied as seed treatments for their endophytic establishment in the common bean. Endophytic colonization in sterile sand:peat averaged ca. 40% higher for fungus treatments and ca. six times higher for volunteer fungi (other fungal endophytes naturally occurring in our samples), relative to sterile vermiculite. Colonization by B. bassiana and M. anisopliae was least variable in sterile vermiculite and most variable in sterile soil

Differential effects of type and quantity of leaf damage on growth, reproduction and defence of lima bean (Phaseolus lunatus L.).

Folivores are major plant antagonists in most terrestrial ecosystems. However, the quantitative effects of leaf area loss on multiple interacting plant traits are still little understood. We sought to contribute to filling this lack of understanding by applying different types of leaf area removal (complete leaflets versus leaflet parts) and degrees of leaf damage (0, 33 and 66%) to lima bean (Phaseolus lunatus) plants. We quantified various growth and fitness parameters including above- and belowground biomass as well as the production of reproductive structures (fruits, seeds). In addition,

Effect of an aqueous extract of Phaseolus vulgaris on plasma insulin and hepatic key enzymes of glucose metabolism in experimental diabetes.

Oral administration of 200 mg/kg of aqueous extract of Phaseolus vulgaris pods (PPEt) to diabetic animals for 45 days resulted in a significant decrease in blood glucose, glycosylated haemoglobin and significant increase in total haemoglobin and plasma insulin. Similarly oral administration of PPEt to normal animals resulted in a significant hypoglycemic effect. The activities of hepatic hexokinase, glucose 6-phosphatase, fructose-1,6-bisphosphatase and glucose-6-phosphate dehydrogenase, a lipogenic enzyme, were measured in the liver of normal and experimental animals. The activities of the li

Freeze-Drying Enhances Functional Properties and Sensory Acceptability of Cranberry Bean (Phaseolus vulgaris L.) Flour in Gluten-Free Cupcakes.

While legume flours can improve nutritional quality of gluten-free baked products, the influence of drying methods on precooked cranberry bean (Phaseolus vulgaris L.) flour and its baking performance remain underexplored. This study compared oven-drying (OD) and freeze-drying (FD) effects on the physicochemical and nutritional properties of cranberry bean flour (CBF) and evaluated their impact on cupcake quality. Functional properties, proximate composition, mineral content, and total phenolic compounds of CBF, along with color, moisture content, texture, storage behavior, and sensory attribut

ACC Deaminase Producing Bacteria With Multifarious Plant Growth Promoting Traits Alleviates Salinity Stress in French Bean (Phaseolus vulgaris) Plants.

Plant growth promoting rhizobacteria (PGPR) with 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity has the potential to promote plant growth and development under adverse environmental conditions. In the present study, rhizobacterial strains were isolated from Garlic (Allium sativum) rhizosphere and were screened in vitro ACC deaminase activity in DF salt minimal media supplemented with 3 mM ACC. Out of six isolates, two could degrade ACC into α-ketobutyrate, exhibiting ACC deaminase activity producing more than ∼1500 nmol of α-ketobutyrate mg protein-1 h-1, and assessed for other

Projections from fetal neocortical transplants placed in the frontal neocortex of newborn rats. A Phaseolus vulgaris-leucoagglutinin tracing study.

Fetal rat neocortex grafted into lesion cavities made in the newborn rat neocortex can exchange multiple axonal connections with the host brain. Most previous studies demonstrating efferent transplant-to-host brain connections have used fluorescent retrograde tracers injected into the host brain (Castro et al. 1985, 1987; Floeter and Jones 1984; O'Leary and Stanfield 1989). Other studies have used anterograde axonal tracing with either tritium-labelled amino acids impregnating the transplant and its efferents (Floeter and Jones 1985) or horseradish peroxidase injected into the transplants (Cha

Effects of Elevated CO2 on Bean Pod Mottle Virus Infection in Both Incompatible and Compatible Interactions With Phaseolus vulgaris L.

Plant viruses cause significant crop losses, a situation that could worsen due to anthropogenic activities driving global climate change, one factor of which is the increase in atmospheric CO2 concentration. This study assessed the impact of elevated CO2 concentration (eCO2, 1000 vs. 400 ppm) on two genotypes of common bean (Phaseolus vulgaris L.), one susceptible and one resistant, infected with bean pod mottle virus (BPMV, Comovirus siliquae). For both genotypes, we found that plant growth, development and physiology were not enhanced under eCO2 enrichment in healthy plants, at the stage of

Minerals and chosen heavy metals retention in immature common bean (Phaseolus vulgaris L.) seeds depending on the method of preservation.

BACKGROUND: Legumes are a good source of protein, and are also abundant in carbohydrates, B-group vita-mins, dietary fibre and mineral compounds. MATERIAL AND METHODS: This work evaluates the retention of ash, eleven minerals and two heavy metals in products obtained from two common bean cultivars harvested before reaching full maturity, with a dry matter content of about 40%. Analyses were conducted on raw, blanched and cooked seeds and three products prepared for consumption after 12-month storage: two frozen and one canned (sterilized). The former comprised two types of frozen product: one

Sargassum latifolium-mediated Se/CuO/MgO/ZnO nanocomposite enhances salt tolerance in Phaseolus vulgaris and exhibits antibacterial and antioxidant activities.

Macroalgae represents a powerful, renewable bioplatform for generating high-performance nanomaterials that bridge sustainability with advanced biotechnological applications. However, crop production and multi-drug resistance microbes are considered the main challenge worldwide. Also, the incorporation of macroalgae metabolites to generate multimetallic nanocomposite, as a new active compound, remains unexplored. In this work, the brown macroalga Sargassum latifolium was exploited as a robust biofactory for the green fabrication of multifunctional active tetrametallic Se/CuO/MgO/ZnO (TSCMZ) nan

Genetic variation and historical breeding patterns in common bean (Phaseolus vulgaris L.) affect fermentation patterns by the human gut microbiome.

Common beans, which contain diverse bioactive molecules, have not been systematically studied for their variation in how they affect the human gut microbiome. We measured taxonomic shifts and metabolite production of three human gut microbiomes cultured with 299 common bean cultivars under conditions that mimic the nutrient availability of the human colon. Common bean population structure (landrace and market class) had significant effects on microbiota diversity, composition, and metabolite production. Genome-wide association analysis identified seven multiple effect loci (MEL) where genetic

Genetic differentiation of a southern Africa tepary bean (Phaseolus acutifolius A Gray) germplasm collection using high-density DArTseq SNP markers.

Genetic resources of tepary bean (Phaseolus acutifolius A. Gray) germplasm collections are not well characterized due to a lack of dedicated genomic resources. There is a need to assemble genomic resources specific to tepary bean for germplasm characterization, heterotic grouping, and breeding. Therefore, the objectives of this study were to deduce the genetic groups in tepary bean germplasm collection using high-density Diversity Array Technology (DArT) based single nucleotide polymorphism (SNP) markers and select contrasting genotypes for breeding. Seventy-eight tepary bean accessions were g

Taxonomical, functional, and cytopathological characterization of Bacillus spp. from Lake Magadi, Kenya, against Rhizoctonia solani Kühn in Phaseolus vulgaris L.

A decline in common bean production and the ineffectiveness of synthetic chemical products in managing plant pathogens has led to exploiting Kenyan soda lakes as an alternative search for biocontrol agents. This study aimed to identify phylogenetically Bacillus spp. from Lake Magadi and their antagonistic activity against Rhizoctonia solani under in vitro and in vivo conditions. The 16 S ribosomal RNA (rRNA) subunit sequences of six bacterial strains isolated from Lake Magadi showed diversity similar to the Bacillus genus; Bacillus velezensis, Bacillus subtilis, and Bacillus pumilus. In vitro,

Variation in cyanogenic glycosides across populations of wild lima beans (Phaseolus lunatus) has no apparent effect on bruchid beetle performance.

Cyanogenic glycosides (CNGs) act as feeding or oviposition deterrents and are toxic after enzymatic hydrolysis, thus negatively affecting herbivore performance. While most studies on CNGs focus on leaf herbivores, here we examined seeds from natural populations of Phaseolus lunatus in Mexico. The predominant CNGs, linamarin and lotaustralin, were quantified for each population by using ultra-high pressure liquid chromatography-mass spectrometry. We also examined whether there was a correlation between the concentration of CNGs and the performance of the Mexican bean beetle, Zabrotes subfasciat

Effect of salicylic acid and structurally related compounds in the accumulation of phytoalexins in cotyledons of common bean (Phaseolus vulgaris L.) cultivars.

In the present work, isoflavonoid phytoalexin production in response to the application of salicylic acid in cotyledons of four common bean (Phaseolus vulgaris) cultivars (SA) was evaluated. The time-course and dose-response profiles of the induction process were established by quantifying the isoflavonoids by HPLC. Cotyledons of anthracnose-resistant cultivars induced by SA produced substantially higher phytoalexin contents as compared to the susceptible ones. In addition, maximum levels of phytoalexins (50-100 fold increases) were reached between 96 and 144 h, and when a concentration of SA

Quantitative variability of direct chemical defense in primary and secondary leaves of lima bean (Phaseolus lunatus) and consequences for a natural herbivore.

Ontogenetic variability in chemical plant defenses against herbivores is a common phenomenon, but the effects of this variability on herbivore-plant interactions are little understood. In a previous study on lima bean (Phaseolus lunatus), we found a trade-off between cyanogenesis, a direct defense, and the release of herbivore-induced volatile organic compounds (VOCs; mainly functioning as an indirect defense). Moreover, the expression of these two defenses could change during plant ontogeny. The present study aimed at elucidating whether such ontogenetic changes in plant defense can affect he

Drought, abscisic acid and transpiration rate effects on the regulation of PIP aquaporin gene expression and abundance in Phaseolus vulgaris plants.

BACKGROUND AND AIMS: Drought causes a decline of root hydraulic conductance, which aside from embolisms, is governed ultimately by aquaporins. Multiple factors probably regulate aquaporin expression, abundance and activity in leaf and root tissues during drought; among these are the leaf transpiration rate, leaf water status, abscisic acid (ABA) and soil water content. Here a study is made of how these factors could influence the response of aquaporin to drought. METHODS: Three plasma membrane intrinsic proteins (PIPs) or aquaporins were cloned from Phaseolus vulgaris plants and their express

The effect of external Ca2+ and Ca(2+)-channel modulators on red-light-induced swelling of protoplasts of Phaseolus radiatus L.

Red-light-induced swelling of the protoplasts isolated from hypocotyl of etiolated mung bean (Phaseolus radiatus L.) was observed only when Ca2+ ions were present in the medium. The optimal CaCl2 concentration was 250 microM. Swelling response declined when Ca2+ was supplied into the medium after red light irradiation. The Ca(2+)-chelator EGTA eliminated the red-light-induced swelling and 45Ca2+ accumulation in the protoplasts. In contrast, A23187, a Ca(2+)-ionophore, could mimic the effect of red light in darkness. These results indicate that Ca2+ may play a role in light signal transduction.
