The effects of chronic oral vasodilator therapy were studied in a group of patients with refractory congestive heart failure. Fifteen patients were treated acutely with intravenous sodium nitroprusside and sublingual isosorbide dinitrate. After continuous therapy with nitroprusside and isosorbide dinitrate for up to 72 hours the patients were then placed on isosorbide dinitrate and oral phenoxybenzamine. Hemodynamic responses to nitroprusside, isosorbide dinitrate, and phenoxybenzamine with isosorbide dinitrate were determined. After a mean follow-up of seven months, nine patients who were rec

Toxin purified from venom of the scorpion Leiurus quinquestriatus was used to release the norepinephrine from adrenergic nerve terminals in isolated pancreatic islets perifused in vitro. Addition of toxin (10 mug./ml) to the perifusion medium caused a sixfold increase in release of norepinephrine in the presence or absence of 3 X 10(-5) M phenoxybenzamine. During 20 minutes of stimulation with toxin, the pancreatic islets released an average of 15 pg. of norepinephrine per islet, which represented 20 per cent of the normal content of norepinephrine in islets. Insulin secretory rates in respons

Adrenalectomies for canine adrenal tumours are associated with peri-operative morbidity and mortality. Objectives of this study included assessing the prognostic value of tumour- or surgery-related variables in predicting peri-operative mortality and overall survival in dogs undergoing adrenalectomies for primary adrenal tumours as well as pre-treatment with phenoxybenzamine on survival to discharge with pheochromocytomas specifically. A multi-institutional retrospective cohort study was performed across nine institutions. Electronic medical record searches identified 302 dogs which met the in

Prazosin, phenoxybenzamine and clozapine, but not sulpiride, abolished the increase in flexor reflex activity induced by clonidine and they accelerated the noradrenaline utilization in the brain of rats. These findings indicate that the first three drugs block central alpha-adrenoreceptors. The alpha-methyltyrosine-induced disappearance of dopamine in the corpus striatum and the limbic system was decelerated by prazosin and phenoxybenzamine, was accelerated by sulpiride and was not significantly changed by clozapine. Prazosin and phenoxybenzamine almost completely reversed the sulpiride-induce

This prospective clinical study evaluates the possible beneficial effects of increased phenoxybenzamine dosage in the preoperative treatment of patients with pheochromocytoma. For this purpose total blood volume (TBV) prior to and after treatment with phenoxybenzamine and hemodynamic changes during surgery were determined in two groups of patients: group I (n = 12) received a mean dosage of 140 mg, group II (n = 12) 270 mg/day. The mean TBV in group I showed no changes after treatment with phenoxybenzamine, while the TBV in group II increased by 5.6 ml/kg body weight, corresponding to an incre

We examined in a double blind fashion and placebo controlled the effects of some alpha and beta adrenergic receptor agonists and antagonists on the recovery of finger skin temperature 12 min after finger cooling (5 min waterbath for both hands) in twelve patients with Raynaud's syndrome. A favourable effect was established on phenoxybenzamine 20 mg as compared to placebo. A significant but rather small effect on orciprenaline 10 mg. The beta-agonists prenalterol (10 mg) and terbutaline (5 mg) did not influence the recovery of finger skin temperature. The beneficial effect of phenoxybenzamine 2

OBJECTIVE: To evaluate the therapeutic potential of sympatholytic agents on premature ejaculation in an animal model, using monitoring of rat seminal vesicle pressure change in response to electrical stimulation of the lesser splanchnic nerve.
MATERIALS AND METHODS: Male Wistar rats (aged 12-14 weeks) were injected intra-arterially with sympatholytic agents (phenoxybenzamine, prazosin, WB-4101, chloroethylclomidine, yohimbine and RX 821002) at various concentrations 10 min before electrical stimulation of the lesser splanchnic nerve. The change in phasic tension (triangle upmmHg) of the semin

The effects of phenoxybenzamine an alpha-adrenergic blocker, propranolol a beta-adrenergic blocker, and diethyldithiocarbamate a noradrenaline synthesis inhibitor, on the LH increase induced by naloxone an opiate antagonist, was investigated in 25 day old female rats. Pretreatment with phenoxybenzamine or diethyldithiocarbamate suppressed the LH increase induced by naloxone, whereas pretreatment with propranolol had no significant effects on the naloxone-induced LH release. These results suggest that naloxone-induced increase in LH release is mediated via a noradrenergic mechanism.

The effects of chlorpromazine, pentoxifylline, terbutaline, allopurinol, phenoxybenzamine, naftidrofuryl, hydralazine and trimetazidine were investigated on caudally based dorsal flaps. The study was performed on 108 rats, divided in 9 groups of 12 animals each, one of which served as a control group. All treated groups showed a significantly greater survival of the flap than the control group. Comparisons among different groups showed better outcome in those receiving trimetazidine and hydralazine, followed by those receiving naftidrofuryl and phenoxybenzamine.

The influence of cardiovascular changes on ventilation has been demonstrated in adult animals and humans (Jones, French, Weissman & Wasserman, 1981; Wasserman, Whipp & Castagna 1974). It has been suggested that neonatal hypoxic ventilatory depression may be related to some of the hemodynamic changes that occur during hypoxia (Brown & Lawson, 1988; Darnall, 1985; Suguihara, Bancalari, Bancalari, Hehre & Gerhardt, 1986). To test the possible relationship between the cardiovascular and ventilatory response to hypoxia in the newborn, eleven sedated spontaneously breathing piglets (age: 5.9 +/- 1.6

In an attempt to improve the selection of patients with neurogenic bladder dysfunction likely to benefit from phenoxybenzamine therapy, the effect of intravenous phentolamine (Regitine) on the urinary flow rate was determined in normal and abnormal subjects. A nomogram relating mean flow rate to volume voided was constructed to aid in the interpretation of results. In 16 patients with neurogenic bladder dysfunction it was found that a response of more than 0.8 nomogram units indicates a likelihood of clinical improvement with phenoxybenzamine therapy. The clinical applications of this test are

The use of amyl nitrite and phenoxybenzamine in the treatment of acute cyanide poisoning was evaluated. Sixty anesthetized beagle dogs were injected i.v. with sodium cyanide (2.5 mg/kg) and were followed for changes in the heart rate, electrocardiogram, respiration, blood pressure and methemoglobin concentration. Twenty control dogs died within 5 to 7 min, showing severe bradycardia, a sharp drop in arterial blood pressure, and respiratory paralysis. Pretreatment with phenoxybenzamine (0.5 mg/kg) prevented these changes in 8 of 10 dogs; however, this drug was ineffective if given after the cya

The influence of noradrenergic mechanisms on the effects of D- and L-amphetamine on lordosis behavior and stereotype activity was studied in ovariectomized estrogen + progesterone treated rats. Phenoxybenzamine, phentolamine and prazosin, which all block alpha-adrenergic receptors potentiated the inhibitory effect of D-amphetamine on lordosis behavior. Phenoxybenzamine and prazosin also potentiated the effect of L-amphetamine, while phentolamine did not increase the effect. No augmented effect of D- or L-amphetamine was obtained on stereotype activity after pretreatment with phenoxybenzamine o

THE VENTILATORY RESPONSE TO HYPOXIA WAS STUDIED IN TWO GROUPS OF SUBJECTS WITH ABNORMAL SYMPATHETIC NERVOUS CONTROL: (a) human subjects with familial dysautonomia (Riley-Day syndrome), and (b) unanesthetized goats treated with an alpha-adrenergic blocking agent (phenoxybenzamine). The ventilatory response to hypoxia was evaluated in two ways: (a) from the slope of the relationship between ventilation and alveolar P(Co2) ([unk]V(E)-P(ACo2) slope) during the rebreathing of hypoxic and hyperoxic gases, and (b) from the change in ventilation produced when hypoxia was abruptly relieved. The ventila

Four patients with pheochromocytoma underwent pre-surgical treatment with phenoxybenzamine and volemic expansion. The postural hypotension was evaluated by daily monitoring of the arterial pressure and heart rate in clino orthostatism. The patients underwent surgery and hemodynamic monitoring by Swan-Ganz catheter when postural hypotension was at the lowest level. Hemodynamic data were: elevated cardiac output, normal systemic vascular resistance and filling pressure. These results confirm that the orthostatic hypotension reduction is a valid indication to evaluate the effectiveness of the tre

Evidence has been obtained suggesting that cathinone-induced analgesia depends upon stimulation of alpha-adrenoceptors, followed by release of opioid peptides and by activation of serotonergic pathways. This hypothesis is supported by the following. (1) Cathinone potentiated morphine analgesia and the whole effect was antagonized by naloxone whereas onto the cathinone potentiation was counteracted by phenoxybenzamine. (2) Bestatin potentiated cathinone-induced analgesia and this effect was sensitive to both naloxone and phenoxybenzamine blockade. (3) The analgesic effect of cathinone + bestati

The role of beta 1- and beta 2-adrenoceptors in the modification of Na, K, and Cl transport in submandibular main duct of rat perfused with bicarbonate saline solution was studied with direct sympathetic nerve stimulation (4 V, 5 ms, 20 Hz) in the presence of specific adrenergic antagonists. Nerve stimulation in the presence of phenoxybenzamine (3 mg/kg b. wt., i.p.) and butoxamine (3 mg/kg b. w.t., i.p.) enhanced Na (44%) and Cl (35%) absorption but inhibited K (24%) secretion. On the other hand, the nerve-evoked response in the presence of phenoxybenzamine and metoprolol (3 mg/kg b. wt., i.p

A 20-year-old woman with a four-week history of dysphagia, weight loss of four kilograms and unspecific abdominal pain was admitted because of sudden haematemesis. The physical examination showed a patient with a prominent kyphoskoliosis. The patient reported of having a situs inversus abdominalis and a tethered cord syndrome. Bladder function disorders were present since childhood. Upper endoscopy demonstrated a 4 cm large, exophytically growing necrotic tumour of the oesophagus. The CT scan showed a space occupying tumour of the oesophagus and metastases in a size of 1.5 cm in both lungs. Fu

This study assessed the role of vascular and central alpha-adrenoceptors in the regional cerebral blood flow response to moderate hypoxia. Studies were conducted in 21 rabbits using radioactive microspheres under normoxic and hypoxic (10% O2 in N2) conditions. Animals were divided into three groups and administered either saline, N-methyl chlorpromazine, or phenoxybenzamine. During normoxia, there were regional differences in cerebral blood flow distribution in the saline- and N-methyl chlorpromazine-treated rabbits which were eliminated by phenoxybenzamine. In control, hypoxia significantly i

The efficacy and side effects of the combined administration of propranolol and phenoxybenzamine were examined in 19 patients with moderate and moderately severe essential hypertension. By titrating the dosage of both drugs against pulse rate and blood pressure response, propranolol was given between 80 and 160 mg. and phenoxybenzamine between 20 and 50 mg. per day in divided doses. There was a substantial reduction in both systolic and diastolic blood pressure in both recumbent and upright positions without orthostatic hypotension. Normal blood pressure (140/90 mm. Hg or less) or near normal
