Pahhph-5: a mouse mutant deficient in phenylalanine hydroxylase.

Mutant mice exhibiting heritable hyperphenylalaninemia have been isolated after ethylnitrosourea mutagenesis of the germ line. We describe one mutant pedigree in which phenylalanine hydroxylase activity is severely deficient in homozygotes and reduced in heterozygotes while other biochemical components of phenylalanine catabolism are normal. In homozygotes, injection of phenylalanine causes severe hyperphenylalaninemia and urinary excretion of phenylketones but not hypertyrosinemia. Severe chronic hyperphenylalaninemia can be produced when mutant homozygotes are given phenylalanine in their dr

Detection of hepatic phenylalanine 4-hydroxylase in classical phenylketonuria.

An extract from a liver biopsy of a patient with classical phenylketonuria contains phenylalanine 4-hydroxylase (EC 1.14.3.1) with a specific activity 0.27% of the normal activity of the hydroxylase. This enzyme from a patient with phenylketonuria is a structurally altered form of phenylalanine 4-hydroxylase that probably results from a mutation in the gene coding for the hydroxylase.

Performance of the Egoo test for phenylalanine measurement in females with phenylketonuria.

BACKGROUND: Accurate measurement of phenylalanine (Phe) is a requirement for the diagnosis and management of individuals with Phenylketonuria (PKU). The current methods for quantifying Phe, plasma amino acids, and dried blood spots (DBS) have several analytical limitations and are not suitable for self-monitoring. This study evaluated the validity of a point-of-care testing (POCT) device, the Egoo Phe Test, in a sample of females with PKU who participated in a five-day camp intervention. METHODS: Blood samples were collected from 20 participants (median age: 18 years; IQR: 16, 28.3) on the fi

Phenylalanine free infant formula in the dietary management of phenylketonuria.

BACKGROUND: Phenylalanine-free infant formula is an essential source of safe protein in a phenylalanine restricted diet, but its efficacy is rarely studied. We report a multicentre, open, longitudinal, prospective intervention study on a phenylalanine-free infant formula (PKU Start: Vitaflo International Ltd.). RESULTS: This was a 2-part study: part I (28 days short term evaluation) and part II (12 months extension). Data was collected on infant blood phenylalanine concentrations, dietary intake, growth, and gastrointestinal tolerance. Ten infants (n = 8 males, 80%), with a median age of 14 w

[The mutant spectrum of phenylalanine hydroxylase gene in Northern Chinese].

OBJECTIVE: To understand the mutant spectrum of phenylalanine hydroxylase (PAH) gene in Northern Chinese. METHODS: All the exons and flaking introns of PAH gene were detected by PCR-single strand conformation polymorphism (PCR/SSCP) and sequencing in 230 patients with phenylketonuria (PKU). RESULTS: (1) A total of 75 different mutations were detected in 435 out of 460 mutant alleles (94.6%). Among them 3 mutations (S251-R252>SfsX89, Y387D and A389G) have not been reported previously. The mutations, R243Q, EX6-96A>G, R111X, R413P and Y356X, were the prevalent mutations with relative frequenci

Mutations in the regulatory domain of phenylalanine hydroxylase and response to tetrahydrobiopterin.

Tetrahydrobiopterin (BH4) is a co-factor that enhances the activity of other enzymes, and this co-factor level is found to be affected in phenylketonuria (PKU), an amino acid metabolism disorder. The present study was aimed at understanding the effect of BH4 on mutations in the regulatory domain of phenylalanine hydroxylase (PAH). Among 14 patients, 5 patients were classical PKU, 3 were atypical PKU, and 6 were mild PKU. All of these patients had at least one mutation in the regulatory domain. Patients were given 10 mg/kg BH4, and the response of blood phenylalanine (Phe) levels was monitored

A potential role for phenylalanine hydroxylase in mosquito immune responses.

In mosquitoes the melanotic encapsulation immune response is an important resistance mechanism against filarial worms and malaria parasites. The rate limiting substrate for melanin production is tyrosine that is hydroxylated by phenoloxidase (PO) to produce 3, 4-dihydroxyphenylalanine. The single pathway for endogenous production of tyrosine is by hydroxylation of phenylalanine by phenylalanine hydroxylase (PAH). In this study we describe a potential role for PAH in melanotic immune responses in the yellow fever mosquito, Aedes aegypti. A 1.6 kb A. aegypti PAH cDNA, encoding a 51 kDa protein,

[Phenotypic expression of 12 mutations of the phenylalanine hydroxylase gene].

BACKGROUND: Several mutations in the human phenylalanine hydroxylase (PAH) gene have been described and it may be interesting to tentatively correlate mutant genotypes and clinical phenotypes of phenylketonuria (PKU). METHODS: Twelve mutations were searched for using classical techniques of molecular biology in a total of 126 patients. 3 phenotypes were arbitrarily defined: typical PKU, atypical PKU or Mediterranean form, and persistent mild hyperphenylalaninemia. The patients were classified according to the residual in vivo PAH activity. RESULTS: Mutations were found in 64 patients. One mu

The effects of high phenylalanine concentration on chick embryonic development.

Cells from a particular portion of the cranial neural crest (cardiac neural crest) migrate from the neural fold into pharyngeal arches 3, 4 and 6, where they provide the support for the endothelium of the aortic arch arteries, and by migration into the outflow tract become involved in septation of the truncus arteriosus. Ablation of the premigratory cardiac neural crest results in persistent truncus arteriosus and other defects reminiscent of the DiGeorge syndrome in man. Removal of a small area of the cardiac neural crest causes a spectrum of heart defects classified together as dextraposed a

[Diagnosis of deficiency in cofactor of phenylalanine hydroxylase: a metabolic emergency].

We report on two cases of children suffering from biopterin synthetase deficiency. Both were treated with the same treatment schedule with biopterin and neurotransmitters: 6-hydroxytryptophan and dihydrophenylalanine (DOPA). The only difference between the two cases is the time of diagnosis and therefore of treatment. The child who was treated early has a normal neurologic development. The other one has been treated since he was 7 months old and is mentally deficient (DQ = 0.60). This older child also suffers from dystonia probably secondary to Levodopa treatment. The authors emphasize the unc

Is phenylalanine requirement in infants and children related to protein intake?

Two groups of children with phenylketonuria (PKU) received protein at two different levels. The protein source was a protein hydrolysate, devoid of phenylalanine, and intact protein from milk, vegetables and fruit. One group (RDA group) was given protein at a level based on the recommendations of the (US) Food and Nutrition Board (1974, 1980). The other group (FAO group) was given protein at the level of intake corresponding to the Joint FAO/WHO ad hoc Expert Committee (1973) safe levels of intake of egg or milk protein. The children were monitored very closely for several years. From an earli

[Heterozygote detection in phenylketonuria by means of an oral load of phenylalanine].

The testing of heterozygosity for PKU by mean of an oral load of phenylalanine is a widely used method. In this paper, the need of a genetic counselling for two adults, with family story of PKU, lead us to reinvestigate criteria of heterozygote recognition. With all variables of the phenylalanine tolerance test, overlapping and mis classification occured. Probability density functions plotting, as proposed by GOLD, illustrated well the genetic risk of a patient. However, in practice, the heterozygote likelihood ratio determination seemed to be the best way to avoid possibly erroneous categoric

Effect of phenylalanine and its metabolites on the metabolism of leucocytes and lymphocytes.

1. The pathogenesis of the mental retardation in phenylketonuria remains obscure. Leucocytes have proved of value in the study of other inborn errors of metabolism. The lymphocyte is a suitable model cell for the study of mammalian metabolism, because of its ability to divide in vitro in response to various stimuli. 2. We have examined the effects of phenylalanine, phenylpyruvate, phenyl-lactate and phenylacetate on the human leucocyte and the resting and phytohaemagglutinin-stimulated rabbit lymphocyte. 3. Phenylpyruvate and phenyl-lactate reduced acetate incorporation into leucocyte lipid by

Distribution and metabolism of phenylalanine and tyrosine during tualraemia in the rat.

Tularaemia in rats causes a doubling of the serum phenylalanine/tyrosine ratio and a more than twofold increase in muscle phenylalanine which cannot be accounted for by decreased hepatic phenylalanine hydroxylase (EC 1.14.16.1) activity, but appear to result from a generalized movement of amino acids from muscle to liver during infection.

Enzymes involved in phenylalanine metabolism in the human foetus and child.

The activities of phenylalanine p-hydroxylase, tyrosine: alpha-ketoglutarate and phenylalanine: pyruvate aminotransferases, and aromatic alpha-ketoacid reductase have been measured in liver biopsies obtained from children with a series of disorders other than phenylketonuria and from human foetuses aged 8.5 to 24 weeks. The importance of such a study in relation to the management of infants with hyperphenylalaninaemia and pregnant phenylketonuric women is discussed

Metabolism of phenylalanine in mice homozygous for the gene 'dilute lethal'.

Mice homozygous for d(l) have been suggested as models for phenylketonuria. We found: (1) the concentration of phenylalanine in the blood was normal at all ages examined; (2) phenylalanine hydroxylase activity in the liver in vitro equalled that in unaffected littermates; (3) the apparent K(m) values for phenylalanine and cofactor respectively in d(l)/d(l) mice were the same as in their normal littermates; (4) inhibition of the overall reaction by the particulate fraction, excess of substrate, excess of cofactor or phenylpyruvic acid showed no difference between d(l)/d(l) mice and their unaffe

Nanopore-enabled time-resolved monitoring of catecholamine-related phenylalanine metabolism.

Catecholamines, including dopamine, noradrenaline (also known as norepinephrine) and adrenaline (also known as epinephrine), are essential regulators of neural and endocrine function. Although they are synthesized through a branched phenylalanine metabolic pathway, direct and dynamic monitoring of the enzymatic processes governing their production has remained challenging. Here we reconstitute the complete catecholamine biosynthetic pathway from phenylalanine to adrenaline in vitro using purified enzymes and cofactors, and couple this system with nanopore-based single-molecule sensing. DNAJC12 Stabilizes Phenylalanine Hydroxylase and Facilitates Its Substrate-Dependent Activation.

Phenylalanine hydroxylase (PAH) is a tetrahydrobiopterin (BH4)-dependent enzyme that converts L-phenylalanine (L-Phe) to L-tyrosine. PAH dysfunction leads to the accumulation of L-Phe in the blood (hyperphenylalaninemia; HPA), which may reach neurotoxic levels, resulting in phenylketonuria (PKU). PKU is associated with pathogenic variants in PAH, mostly causing misfolding and instability, leading to decreased levels of PAH protein and activity. Recently, variants in the J-domain protein DNAJC12 have also been associated with HPA in patients, demonstrating the importance of protein homeostasis

L-Phenylalanine promotes liver steatosis by inhibiting BNIP3-mediated mitophagy.

BACKGROUND: L-Phenylalanine (L-Phe) levels are elevated in patients with metabolic dysfunction-associated steatotic liver disease (MASLD). However, whether L-Phe induces liver steatosis and the underlying mechanism remain unknown. This study aimed to investigate the mechanism through which L-Phe promotes liver steatosis. METHODS: We utilized human data from the UK Biobank and SPECT-China studies. Plasma/serum samples were collected for metabolomic testing to measure L-Phe levels. A rat model with L-Phe in the drinking water was established to investigate changes in hepatic lipid metabolism. I
