Activation of the Phenylpropanoid pathway in Nicotiana tabacum improves the performance of the whitefly Bemisia tabaci via reduced jasmonate signaling.

BACKGROUND: Phloem-feeding insects can manipulate plant-induced resistance and are able to suppress effective jasmonic acid/ethylene (JA/ET) defenses by the induction of inefficient salicylic acid (SA) based responses. As a result, activation of the phenylpropanoid biosynthesis pathway in transgenic plants is anticipated to cause complex interactions between phloem-feeding insects and their host plants due to predicted contradiction between two defense forces: the toxicity of various phenylpropanoids and the accumulation of SA via a branch of the activated pathway. METHODOLOGY/PRINCIPAL FINDI

Tomato sucrose transporter 2 facilitates Pseudomonas syringae proliferation via sugar efflux and repressing salicylic acid biosynthesis.

Bacterial pathogens require carbohydrates from host plants to support their proliferation, in which sugar transporters play critical roles. Sucrose transporters (SUTs) execute phloem loading, while their functions in the plant-pathogen interaction are largely unknown. Here, we pinpoint the role of SlSUT2 during Pseudomonas syringae pv tomato (Pst) DC3000 infection of tomato plants. SlSUT2 transcription is positively correlated with bacterial proliferation during Pst DC3000 infection. Knocking out SlSUT2 impairs plant growth and enhances resistance to the pathogen. The slsut2 mutant has reduced

Functional ratios diverge across climates but converge within environments in 55 woody canopy plants.

Functional coordination among leaf, xylem, phloem and ray parenchyma governs the carbon-water economy of woody plants, yet the allocation principles and climatic drivers remain poorly explored. We hypothesized that species at lower altitudes and/or latitudes invest more in xylem and phloem per unit leaf area to compensate for higher transpirational losses, whereas species from higher altitudes and/or latitudes allocate proportionally more to ray parenchyma to enhance storage. Using canopy cranes across four Chinese forests, we quantified how leaf area, branch tissue cross-sectional areas, cons

Immunolabeling Sieve Element Cell Walls with the LM26 Antibody.

Sieve elements in the phloem transport carbon and small molecules, such as RNA and phytohormones, throughout the plant body. Understanding the physical dimensions of sieve elements and phloem tissue is thus crucial for predicting how much carbon can be moved at any given time. Quantification of sieve element diameters and areas has previously been performed using transmission electron microscopy, scanning electron microscopy, and light microscopy, but sieve element identification is difficult because the phloem is a heterogeneous tissue. The recently identified LM26 antibody labels a pectin in

PtoHsfB1 regulates growth and salt response by affecting ABA biosynthesis in Populus tomentosa.

Poplar is an important commercial timber species and a model organism for forest molecular biology. Here, we identified PtoHsfB1, a subgroup B heat shock transcription factor from Populus tomentosa that is homologous to AtHsfB1 and PtrHsfB1, and contains the conserved repression motif '-LFGV-'. To elucidate its function, transgenic poplars overexpressing PtoHsfB1 were generated. Overexpression significantly enhanced growth rate and biomass accumulation. Histological analyses revealed increased cambial cell layers and enlarged phloem caps, indicating a positive role in cambial activity and phlo

Stationary sieve element proteins.

Vascular plants use the phloem to move sugars and other molecules from source leaves to sink organs such as roots and fruits. Within the phloem, enucleate sieve elements provide the low-resistance pipe system that enable bulk flow of sap. In this review, we provide an overview of the highly specific protein machinery that localize to mature sieve elements without entering the phloem translocation stream. Generally, the proteins either maintain the flow, protect the sieve element against pathogens or transmit system wide signals. A notable exception is found in poppy, where part of the opium bi

Transcriptional evidence for cross talk between JA and ET or SA during root-knot nematode invasion in tomato.

studies have demonstrated that jasmonic acid (JA) reduces root-knot nematode (RKN) infections in tomato plants. RKN invasion is sensed by roots, and root-derived JA signaling activates systemic defense responses, though this is poorly understood. Here, we investigate variations in the RKN-induced transcriptome in scion phloem between two tomato plant grafts: CM/CM ( Lycopersicum esculentum Mill. cv. Castlemart) and CM/ spr2 (a JA-deficient mutant). A total of 8,716 genes were differentially expressed in the scion phloem of the plants with JA-deficient rootstock via RNA sequencing. Among these

Water-carbon coordination in Taxodium distichum but not T. ascendens  under prolonged off-season submergence in the Three Gorges Reservoir riparian zone.

Prolonged winter submergence induced by reservoir impoundment poses severe physiological challenges to riparian trees, yet post-submergence recovery dynamics remain poorly understood. In this study, we investigated hydraulic function, xylem anatomy and non-structural carbohydrate (NSC) dynamics in two congeneric species, Taxodium ascendens and T. distichum, following a winter impoundment in the Three Gorges Reservoir. During the initial post-submergence exposure phase, we comparatively analysed stem xylem micro-anatomy, percentage loss of conductivity (PLC) and NSC allocation across the xylem

Salicylic acid transport in Ricinus communis involves a pH-dependent carrier system in addition to diffusion.

Despite its important functions in plant physiology and defense, the membrane transport mechanism of salicylic acid (SA) is poorly documented due to the general assumption that SA is taken up by plant cells via the ion trap mechanism. Using Ricinus communis seedlings and modeling tools (ACD LogD and Vega ZZ softwares), we show that phloem accumulation of SA and hydroxylated analogs is completely uncorrelated with the physicochemical parameters suitable for diffusion (number of hydrogen bond donors, polar surface area, and, especially, LogD values at apoplastic pHs and Delta LogD between apopla

Role of magnesium in carbon partitioning and alleviating photooxidative damage.

Magnesium (Mg) deficiency exerts a major influence on the partitioning of dry matter and carbohydrates between shoots and roots. One of the very early reactions of plants to Mg deficiency stress is the marked increase in the shoot-to-root dry weight ratio, which is associated with a massive accumulation of carbohydrates in source leaves, especially of sucrose and starch. These higher concentrations of carbohydrates in Mg-deficient leaves together with the accompanying increase in shoot-to-root dry weight ratio are indicative of a severe impairment in phloem export of photoassimilates from sour

Temperature-Responsive Bottlebrush Polymers Deliver a Stress-Regulating Agent In Vivo for Prolonged Plant Heat Stress Mitigation.

Anticipated increases in the frequency and intensity of extreme temperatures will damage crops. Methods that efficiently deliver stress-regulating agents to crops can mitigate these effects. Here, we describe high aspect ratio polymer bottlebrushes for temperature-controlled agent delivery in plants. The foliar-applied bottlebrush polymers had near complete uptake into the leaf and resided in both the apoplastic regions of the leaf mesophyll and in cells surrounding the vasculature. Elevated temperature enhanced the in vivo release of spermidine (a stress-regulating agent) from the bottlebrush

Low-affinity cation transporter (OsLCT1) regulates cadmium transport into rice grains.

Accumulation of cadmium (Cd) in rice (Oryza sativa L.) grains poses a potential health problem, especially in Asia. Most Cd in rice grains accumulates through phloem transport, but the molecular mechanism of this transport has not been revealed. In this study, we identified a rice Cd transporter, OsLCT1, involved in Cd transport to the grains. OsLCT1-GFP was localized at the plasma membrane in plant cells, and OsLCT1 showed Cd efflux activity in yeast. In rice plants, strong OsLCT1 expression was observed in leaf blades and nodes during the reproductive stage. In the uppermost node, OsLCT1 tra

Post-véraison irrigation frequency, rather than amount, better preserves vascular function and delays late-season dehydration in Cabernet Sauvignon grapes.

Late-season dehydration (LSD) reduces grape yield and quality in semi-arid regions, yet the roles of post-véraison irrigation amount (IA) and frequency (IF) in regulating berry vascular function remain unclear. This study investigated how IA and IF influence berry physicochemical properties, phenolic composition, and vascular function in Vitis vinifera L. cv. Cabernet Sauvignon. Irrigation frequency exerted a stronger influence than amount on berry composition, phenolic accumulation, and vascular continuity. Significant four-way interactions among developmental stage, irrigation amount, IF, an

Sieve Element Plastids: An Elusive Organelle.

Vascular plants use the phloem to transport photoassimilates via sieve elements, which are cells with modified anatomy to accommodate long distance transport. Mature sieve elements lack a majority of cell organelles to reduce friction in the tube system, but some organelles like the endoplasmic reticulum, mitochondria, phloem-proteins and sieve element plastids remain. Most of the functions of sieve element organelles are poorly characterized. This leads to the fact that sieve element plastids are often omitted from textbooks as a plastid type, although they apparently derive from proplastids.

Synthesis, Antifungal Activity, and Plant Systemic Mobility of Amino Acid Conjugates of a Thiasporine A-Derived Phenylthiazole Scaffold.

To develop fungicidal candidates with both high antifungal activity and phloem mobility, a total of 34 2-[2-(trifluoromethoxy)phenyl]thiazole-4-carbonyl amino acid and amino acid ester conjugates were designed and synthesized. Their structures were characterized by 1H NMR, 13C NMR, and HRMS. Their in vitro antifungal activities against five phytopathogenic fungi and their phloem and xylem mobility in castor bean seedlings were subsequently evaluated. The results showed that the amino acid conjugates 9a-9p generally exhibited higher antifungal activity than the corresponding amino acid ester co

Modulating the spatiotemporal dynamics of starch metabolism in postharvest sweetpotato using high voltage alternating electric field.

Although postharvest starch degradation was extensively studied, its initiation site and distribution patterns remain unclear. This study investigated the spatiotemporal dynamics of starch metabolism in sweetpotato tubers along vertical (top, middle, bottom) and radial directions (phloem, outer xylem, inner xylem) under 4 kV·m-1 high voltage alternating electric field (HVAEF) treatment stored at 13 °C for 8 weeks. Results illustrated that starch degradation was first detected in the middle region during early storage, and extended to the bottom and top, whereas HVAEF's inhibitory effect concen

Evolution of jasmonate and salicylate signal crosstalk.

The evolution of land plants approximately 470 million years ago created a new adaptive zone for natural enemies (attackers) of plants. In response to attack, plants evolved highly effective, inducible defense systems. Two plant hormones modulating inducible defenses are salicylic acid (SA) and jasmonic acid (JA). Current thinking is that SA induces resistance against biotrophic pathogens and some phloem feeding insects and JA induces resistance against necrotrophic pathogens, some phloem feeding insects and chewing herbivores. Signaling crosstalk between SA and JA commonly manifests as a reci

ABCG9, ABCG11 and ABCG14 ABC transporters are required for vascular development in Arabidopsis.

In order to obtain insights into the regulatory pathways controlling phloem development, we characterized three genes encoding membrane proteins from the G sub-family of ABC transporters (ABCG9, ABCG11 and ABCG14), whose expression in the phloem has been confirmed. Mutations in the genes encoding these dimerizing 'half transporters' are semi-dominant and result in vascular patterning defects in cotyledons and the floral stem. Co-immunoprecipitation and bimolecular fluorescence complementation experiments demonstrated that these proteins dimerize, either by flexible pairing (ABCG11 and ABCG9) o

Near-infrared spectroscopy as a high-throughput phenotyping method for fusiform rust resistance in loblolly pine.

Fusiform rust, caused by the pathogen Cronartium quercuum (Berk.) Miyabe ex Shirai f. sp. fusiforme, is the most important disease of loblolly pine (Pinus taeda L.) in the U.S., causing millions of dollars in damage each year. Using resistant genotypes has proven a successful strategy to limit the disease, but resistance selection still relies on visual inspection for symptoms, which can lead to misclassification due to human error and the presence of 'escaped susceptibles' (i.e., susceptible individuals with no visible symptoms due to either an extended asymptomatic phase of the disease or th
