Developing Single-Atomic Manganese Nanozymes for Synergistic Mild Photothermal/Multienzymatic Therapy.

Synergistic mild photothermal/nanozyme therapy with outstanding hyperthermia performance and excellent multienzyme properties is highly needed for osteosarcoma treatment. Herein, we have developed efficient single-atom nanozymes (SANs) consisting of Mn sites atomically dispersed on nitrogen-doped carbon nanosheets (denoted as Mn-SANs) for synergistic mild photothermal/multienzymatic therapy against osteosarcoma. Benefiting from their black N-doped carbon nanosheet matrices, Mn-SANs showed an excellent NIR-II-triggered photothermal effect. On the other hand, Mn-SANs with atomically dispersed Mn

Topically applied liposome-in-hydrogels for systematically targeted tumor photothermal therapy.

Transdermal drug delivery for local or systemic therapy provides a potential anticancer modality with a high patient compliance. However, the drug delivery efficiency across the skin is highly challenging due to the physiological barriers, which limit the desired therapeutic effects. In this study, we prepared liposome-in-hydrogels containing a tumor targeting photosensitizer IR780 (IR780/lipo/gels) for tumor photothermal therapy (PTT). The formulation effectively delivered IR780 to subcutaneous tumor and deep metastatic sites, while the hydrogels were applied on the skin overlying the tumor o

Multifunctional metal rattle-type nanocarriers for MRI-guided photothermal cancer therapy.

In the past decade, numerous species of nanomaterials have been developed for biomedical application, especially cancer therapy. Realizing visualized therapy is highly promising now because of the potential of accurate, localized treatment. In this work, we first synthesized metal nanorattles (MNRs), which utilized porous gold shells to carry multiple MR imaging contrast agents, superparamagnetic iron oxide nanoparticles (SPIONs), inside. A fragile wormpore-like silica layer was manipulated to encapsulate 8 nm oleylamine SPIONs and mediate the in situ growth of porous gold shell, and it was fi

Multifunctional ICG-SB@Lip-ZA Nanosystem Focuses on Remodeling the Inflammatory-Immunosuppressive Microenvironment After Photothermal Therapy to Potentiate Cancer Photothermal Immunotherapy.

Achieving full eradication of residual tumors post photothermal therapy (PTT) hinges on the immune system's activation and response. Nevertheless, the resultant local inflammation attracts a significant influx of aberrant immune cells and fibroblasts, such as tumor-associated macrophages (TAMs) and cancer-associated fibroblasts (CAFs), following tumor PTT. This phenomenon exacerbates immune evasion and the persistence of residual tumor cells, culminating in tumor recurrence and advancement. To tackle this challenge, a combined therapeutic approach utilizing multifunctional ICG-SB@Lip-ZA nanosy

Tubelike Gold Sphere-Attapulgite Nanocomposites with a High Photothermal Conversion Ability in the Near-Infrared Region for Enhanced Cancer Photothermal Therapy.

Near-infrared (NIR)-induced photothermal therapy (PTT) is now considered to be a promising and highly efficient method for tumor therapy. Photothermal agents play a crucial role in PTT, and they are required to possess the ability to harvest NIR light and transform the photon energy into heat energy. This work reports a facile method to synthesize a new PTT agent, which is based on the electrostatic binding of the Au nanospheres (Au NSs, ∼15 nm) to the surface of a nanometer-sized mineral, attapulgite, to form tubelike Au-attapulgite nanocomposites. These nanocomposites consist of numerous Au

Multimodal antibacterial agents based on photodynamic therapy and photothermal therapy: principles, design strategies and applications.

Antimicrobial resistance (AMR) and bacterial biofilms pose a dire global health threat. To combat localized infections (e.g., chronic wounds, implant-associated infections), multimodal strategies combining photodynamic therapy (PDT) and photothermal therapy (PTT)-collectively referred to as photoactivated therapies (PT)-have emerged as promising antibiotic-sparing photoactivated approaches. This review systematically surveys multimodal platforms that integrate PDT/PTT with chemodynamic therapy (CDT), sonodynamic therapy (SDT), gas therapy, antibiotics, and immunotherapy. We highlight the ratio

10Boron-doped carbon nanoparticles as delivery platforms for boron neutron capture therapy and photothermal therapy.

The current clinical application of the boron drug boronophenylalanine (BPA) in BNCT faces various issues owing to its low boron loading (approximately 5%), which limits its therapeutic efficacy. Therefore, the development of boron drugs with higher boron contents is essential. Enhancing the boron content in boron drug materials is the focus of this study. Two-dimensional (2D) boron nitride-doped nano graphene (BNNG) with a high boron content of 24.97% ± 1.14% w/w was synthesized via the chemical vapor deposition (CVD) method. The size of BNNG was controlled through gradient density centrifuga

Tumor-targeted near-infrared/ultraviolet-triggered photothermal/gas therapy nanoplatform for effective cancer synergistic therapy.

The integration of photothermal therapy (PTT) and gas therapy (GT) on a nanoplatform shows great potential in cancer treatment. In this paper, a tumor-targeted near-infrared/ultraviolet (NIR/UV) triggered PTT/GT synergistic therapeutic nanoplatform, PB-CD-PLL(NF)-FA, was designed based on Prussian blue (PB) nanoparticles, 5-chloro-2-nitrobenzotrifluoro (NF)-grafted polylysine (PLL(NF)), and folic acid (FA). PB serves as a core to load PLL(NF) through host-guest interaction and can further modify FA. PB-CD-PLL(NF)-FA can be enriched in tumor tissues by passive targeting with enhanced permeabili

MRI-guided dual-responsive anti-tumor nanostructures for synergistic chemo-photothermal therapy and chemodynamic therapy.

Image-guided stimulus-responsive theranostics are beneficial for identifying malignant lesions and integrating multiple cell-killing mechanisms to enhance tumor cell clearance. Herein, an intelligent dual-responsive nanostructure (HSPMH-DOX) was developed for magnetic resonance imaging (MRI)-guided synergistic chemo-photothermal therapy (PTT) and chemodynamic therapy (CDT). The core-shell nanostructure was synthesized by layering polydopamine (PDA), manganese oxide (MnO2), and hyaluronic acid (HA) onto drug-loaded hollow mesoporous silica nanoparticles (HS). The constructed nanoagent has both

Photothermal-responsive Biomimetic Xenon Nanobubbles for Spatiotemporally Targeted Therapy of Ischemic Stroke.

Ischemic stroke (IS) involves persistent oxidative stress, exacerbated inflammation, and neuronal apoptosis. However, the therapeutic efficacy of current neuroprotective agents is severely restricted by the blood-brain barrier, which hinders their effective delivery to ischemic brain tissue. Although xenon (Xe) exhibits well-documented neuroprotective properties, its rapid diffusion and short in vivo retention limit its ability to match the evolving pathology of stroke. Therefore, developing Xe-based delivery systems with enhanced lesion targeting and controllable release is essential to fully

Hydrophilic porphyrin copolymers combined with autophagy inhibitors enhance tumor photothermal/photodynamic therapy.

Synergistic photodynamic and photothermal therapy (PDT/PTT) holds immense promise for cancer treatment, yet its efficacy is often compromised by limited photosensitizer performance, glutathione (GSH)-mediated ROS quenching, and tumor-protective autophagy. Herein, we developed a GSH-responsive hydrophilic porphyrin copolymer (PPS) that covalently integrates porphyrin into the polymer backbone, ensuring high loading capacity and minimal carrier-related toxicity. PPS exhibits robust absorption around 800 nm, enabling simultaneous PDT and PTT under a single NIR laser. By incorporating the autophag

Enhancing photothermal therapy effectiveness via tartrazine-induced optical clearing of biological tissues.

Photothermal therapies offer a powerful strategy for noninvasive treatment of cancer and other pathological conditions by converting light energy into localized heat to ablate diseased tissues or induce local release of antigens. However, the efficacy and precision of these therapies are severely restricted by optical scattering in biological tissues, which limits light penetration and spatial accuracy in vivo. In this work, we leverage the food coloring dye tartrazine-a strongly absorbing molecule in UV-VIS range ([Formula: see text]250 nm and [Formula: see text]430 nm) and with minimal absor

Supramolecular peptide hydrogel-mediated synergistic antibiotic-photothermal therapy for potent antibacterial applications.

Bacterial infection usually exacerbates inflammation and is one of the important factors impeding wound healing. At present, dressings for treating wound infections often include antibiotics, silver ions, fibers, etc., but their therapeutic efficacy is still limited. To enhance antibiotic effectiveness and overcome drug resistance, developing efficient drug delivery systems is imperative. Herein, we rationally designed a three-component peptide hydrogel Nap-Phe-Phe-Thr-Asp-Asp-Tyr (NapFFTDDY) co-encapsulating the photosensitizer indocyanine green (ICG) and ciprofloxacin (Cip) to establish a no

Perspectives on photothermal therapy to enhance osteogenesis-angiogenesis coupling in bone repair.

Recent attention has focused on bone's highly vascularized characteristics to improve bone defect repair, an interplay referred to as osteogenesis-angiogenesis coupling. Regarding this point, it is promising to utilize photothermal therapy (PTT) to enhance the coupling, with increasing research in the field. However, the application models, mechanisms, and therapeutic efficacy remain underexplored. Previous reviews have mainly addressed the general osteogenic applications of PTT. This review instead focuses on how PTT regulates the spatiotemporal coordination of osteogenesis and angiogenesis d

Oxygen-Independent Supramolecular Nanoplatform for Photothermal-Thermodynamic Therapy Against Periodontal Anaerobic Infections.

Radical-based nanotherapeutics hold great promise for the treatment of periodontitis. However, the hypoxic/anaerobic microenvironment within periodontal pockets significantly restricts the antibacterial efficacy and clinical translatability of oxygen-dependent radical therapy. By conjugating gold nanorods (AuNRs) with carboxylatopillar[5]arene (CP5), followed by the loading of 2,2'-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH) via host-guest recognition interactions, we develop an oxygen-independent radical delivery nanoplatform (AIPH⊂CP5-AuNR) to overcome the abovementioned barr

Biofilm-Binding Phages Enhance Biofilm Eradication by Synergistic Photothermal and Photodynamic Therapy.

Biofilms, a major cause of chronic bacterial infections, present significant treatment challenges due to their protective extracellular matrix that shields bacteria from both antibiotics and host immune defenses. To treat biofilms more effectively, here we discovered a biofilm-binding peptide from a phage library and verified that it selectively bound the polysaccharides on the biofilm. We then engineered M13 phage into trifunctional nanofibers displaying the biofilm-binding peptide at the tip and carrying gold nanoparticles (AuNPs, as photothermal agents) and tetrakis(4-carboxyphenyl) porphyr
