Comprehensive chemical profiling of Vitex agnus-castus L. leaves: analytical validation of bioactive lipids, phytosterols, and nutritional elements.

This study provides a targeted bioactive lipid and nutritional profile of the under-investigated leaves of Vitex agnus-castus L. An absolute methanolic extract of shade-dried leaves was analysed using gas chromatography/mass spectrometry (GC/MS) after derivatization, alongside essential mineral quantification via atomic absorption spectroscopy (AAS) and proximate analysis. Rigorous data validation excluded analytical artefacts. GC/MS tentatively identified ten validated bioactive compounds, predominantly glycerol (45.21%, a lipid hydrolysis artefact), oleic acid (9.89%), and palmitic acid (9.1

Size-Tailored Nanospaces in Hierarchical Magnetic Core-Shell Microspheres for Lipase Immobilization and Phytosterol Ester Synthesis.

A size-matched magnetic core-shell platform (Fe3O4@MnO2@mSiO2) was constructed for immobilizing Candida rugosa lipase (CRL). The flower-like MnO2 interlayer provides chemical shielding, while mesoporous channels were expanded to 6.45 nm to geometrically match CRL (approximately 5 nm). Driven by interfacial activation, the support exhibited a selective purification effect, achieving a high activity-based immobilization yield (85.2%) and protein loading (143.1 mg g-1). The immobilized specific activity reached 31.4 U mg-1 (1.57-fold higher than the free enzyme) with 88.2% recovered activity. Cru

A Case of Peripheral Precocious Puberty May Be Caused by a Diet Containing Phytosterols in a 20-Month-Old Boy.

INTRODUCTION: Precocious puberty in boys generally remains an etiology. In addition to the causes of endogenous hormone changes, endocrine-disrupting chemicals of exogenous substances may interfere with children's pubertal development. CASE PRESENTATION: A 20-month-old boy presented with peripheral precocious puberty may be due to a phytosterol-containing diet. The patient came to see a doctor because of acne, hairiness, increased penis size, and coarse voice. Genital examination revealed a Tanner stage of 2 for pubic hair and a stretched penile length of 5 cm, which disagreed with the prepub

Fat type in phytosterol products influence their cholesterol-lowering potential: A systematic review and meta-analysis of RCTs.

The most common form of phytosterol (PS) fortified foods are fat spreads and dairy products. The predominant fats used are soybean/sunflower (SS) or rapeseed/canola (RC) oils and animal fat (D) in dairy products. This review aimed to investigate whether the carrier fat is a determinant of the hypocholesterolaemic effects of PS fortified foods. Databases were searched using relevant keywords and published RCTs from 1990 investigating the effects of dietary PS intervention (≥1.5g per day) on total cholesterol and LDL-C were included. After methodological quality assessment and data extraction, a

[Clinical study of a phytosterol extract of Prunus arborea and 3 amino acids: glycine, alanine and glutamic acid].

We have carried out a therapeutic study on "Tebetane compuesto", by applying it to one hundred patients, whose pathologies, prostatic adenoma, prostatitis and postadenomectomy sequelas, come within the field of action of the product, as shown by other authors. The particular feature of "Tebetane compuesto" is that it is a non-hormonal product, which contributes to the fact that it may be used in high doses without producing harmful effects on the patient. We have found no cases of intolerance or side effects, which we feel makes it superior to other similar products. To be specific, in the cas

A hierarchical systems metabolic engineering framework for modular construction of Mycolicibacterium neoaurumcell factories for efficient phytosterol bioconversion.

Androst-4-ene-3,17-dione (AD), Androst-1,4-diene-3,17-dione (ADD), and 9-hydroxy-4-androstene-3,17-dione (9-OHAD) are valuable C19 steroid intermediates derived from low-cost phytosterol bioconversionby Mycolicibacterium neoaurum. However, its slow growth rate, limited sterol uptake and complex metabolic networks restrict efficient and selective steroid biosynthesis. Here, we established a hierarchical systems metabolic engineering framework that sequentially integrates chassis evolution, metabolic flux rewiring, pathway optimization, cofactor regeneration, and transporter engineering for modu

Incorporation of liposomal phytosterols into human cells in culture: a potential in vitro model for investigating pathological effects of phytosterolemia.

A potential in vitro cell culture model was developed for studies concerning the pathological effect of phytosterolemia in which liposomal phytosterols were incorporated into human skin fibroblasts and hepatoblastoma (HepG2) cells. After incubation with phytosterols, fibroblasts and HepG2 cells contained a significant amount (20-27%) of phytosterols (campesterol and beta-sitosterol). Phytosterol accumulation caused a significant reduction in the cholesterol content of cells. Labeled sitosterol and cholesterol showed similar uptake with lower esterification of sitosterol when compared to choles

Blood phytosterols in relation to cardiovascular diseases and mediating effects of blood lipids and hematological traits: a Mendelian randomization analysis.

BACKGROUND: Short-term clinical trials have shown the cholesterol-lowering potentials of phytosterols, but their impacts on cardiovascular disease (CVD) remain controversial. This study used the Mendelian randomization (MR) to investigate the relationships between genetic predisposition to blood sitosterol concentration and 11 CVD endpoints, along with the potential mediating effects of blood lipids and hematological traits. METHODS: Random-effect inverse-variance weighted method was used as the main analysis of MR. Genetic instruments of sitosterol (seven SNPs, F = 253, and R2 = 15.4 %) were

Effect of hydrophilic phytosterol glycosyl derivatives on lipid digestion and cholesterol precipitation during in vitro dynamic gastrointestinal digestion.

Hydrophilic phytosterol derivatives are synthetic analogs of phytosterols conjugated with hydrophilic moieties, designed to enhance water solubility and bioaccessibility compared to free phytosterols. The interactive effect of these derivatives on lipid digestion and cholesterol precipitation remains unclear. Building on our prior synthesis of a homologous series of hydrophilic phytosterol glycosyl derivatives, this study employed an in vitro dynamic human stomach-intestine system IV (DHSI-IV) to elucidate the effect of free phytosterols and phytosterol glycosyl derivatives on these two closel

Mechanisms of deodorizing rapeseed oil with ethanol steam at a low temperature: A focus on free fatty acids, tocopherols, and phytosterols.

Previous studies have demonstrated that deodorization of oil with ethanol steam at a low-temperature (L-ESD) can achieve the same effect in removing harmful substances and preserving micronutrients while better inhibiting the formation of new harmful substances compared to deodorization with water steam at a high-temperature. To elucidate the mechanism of ESD, the kinetics and thermodynamics of ethanol steam deodorized distillate were studied in comparison with those of water steam deodorized distillate. Results revealed their deodorization kinetics and thermodynamic models, and further indica
