Subicular spatial codes arise from predictive mapping.

The successor representation has emerged as a powerful model for understanding mammalian navigation and memory; explaining the spatial coding properties of hippocampal place cells and entorhinal grid cells. However, the diverse spatial responses of subicular neurons, the primary output of the hippocampus, have eluded a unified account. Here, we demonstrate that incorporating rodent behavioural biases into the successor representation successfully reproduces the heterogeneous activity patterns of subicular neurons. This framework accounts for the emergence of boundary and corner cells-neuronal

CA3 transiently modulates spatial representation in CA1.

Neuronal representations of the world are dynamic. A striking example is the rapid remapping of the hippocampal spatial code, which occurs even when the environment and behavior remain unchanged. CA3 input to CA1 has been shown to exert a key role in triggering synaptic plasticity in place-encoding CA1 cells - a phenomenon which could provide the cellular foundations for the remapping of the hippocampal code for space. However, how CA3 input directly contributes to place field formation and remapping of the place code in CA1 remains incompletely understood. By combining longitudinal two-photon

Adaptive modulation of theta sweeps in the brain's navigation circuit.

Efficient navigation requires prioritizing information from behaviorally relevant locations. In rats, such selective sampling may arise from theta-paced sweeps in grid and place cell populations, which scan nearby space in a left-right alternating pattern coordinated by parasubicular direction signals. This alternation promotes uniform spatial coverage during exploration, but whether sweeps can be flexibly tuned to moment-to-moment demands remains unknown. Using large-scale Neuropixels recordings in freely behaving rats, we show that sweeps and direction signals are rapidly and dynamically mod

Tactile modulation of hippocampal place fields.

Neural correlates of spatial representation can be found in the activity of the hippocampal place cells. These neurons are characterized by firing whenever the animal is located in a particular area of the space, the place field. Place fields are modulated by sensory cues, such as visual, auditory, or olfactory cues, being the influence of visual inputs the most thoroughly studied. Tactile information gathered by the whiskers has a prominent representation in the rat cerebral cortex. However, the influence of whisker-detected tactile cues on place fields remains an open question. Here we studi

Trading Place for Space: Increasing Location Resolution Reduces Contextual Capacity in Hippocampal Codes.

Many animals learn cognitive maps of their environment - a simultaneous representation of context, experience, and position. Place cells in the hippocampus, named for their explicit encoding of position, are believed to be a neural substrate of these maps, with place cell "remapping" explaining how this system can represent different contexts. Briefly, place cells alter their firing properties, or "remap", in response to changes in experiential or sensory cues. Substantial sensory changes, produced, e.g., by moving between environments, cause large subpopulations of place cells to change their

A neural code for time and space in the human brain.

Time and space are primary dimensions of human experience. Separate lines of investigation have identified neural correlates of time and space, yet little is known about how these representations converge during self-guided experience. Here, 10 subjects with intracranially implanted microelectrodes play a timed, virtual navigation game featuring object search and retrieval tasks separated by fixed delays. Time cells and place cells activate in parallel during timed navigation intervals, whereas a separate time cell sequence spans inter-task delays. The prevalence, firing rates, and behavioral

Distributed neural codes of the 3D position in the marmoset frontal cortex and hippocampus.

The neural encoding of the animal's position is essential for spatial navigation and cognition, yet the three-dimensional (3D) positioning system in the primate brain remains poorly understood. By combining wireless multi-channel recordings from common marmosets with a free-forging task in the 3D space, we identified 42% and 24% neurons in the frontal cortex (FC) and hippocampus (HPC) as place cells, respectively. Place cells in FC and HPC encoded the 3D position with similar spatial information, sparsity and stability, and exhibited stronger functional connections than non-place cells. Notabl

[Visualization of the functional neural circuit breakdown process in a mouse model of Alzheimer's disease and application to the development of new therapies].

Alzheimer's disease (AD) patients cause cognitive impairment in time and space as early symptoms, in which the hippocampus plays an essential role. In the hippocampus, place cells play an important role in processing spatial information. The hippocampus recognizes space and guides creatures to their destinations using these place cells. In AD patients, the hippocampal neuronal network that place cells form could malfunction with age. However, it has remained unknown how the disruption of the neural circuits progresses at a single-cell resolution in AD. Conventional methods such as electrophysi

A stable hippocampal representation of a space requires its direct experience.

In humans and other mammals, the hippocampus is critical for episodic memory, the autobiographical record of events, including where and when they happen. When one records from hippocampal pyramidal neurons in awake, behaving rodents, their most obvious firing correlate is the animal's position within a particular environment, earning them the name "place cells." When an animal explores a novel environment, its pyramidal neurons form their spatial receptive fields over a matter of minutes and are generally stable thereafter. This experience-dependent stabilization of place fields is therefore

Place and behavioral modulation of hippocampal neurons during immobility.

Behavioral state fluctuations profoundly impact episodic memory processing. To explore the underlying mechanisms, we recorded CA1 place cells in head-fixed male mice and focused on awake immobility to capture spontaneous behavioral state fluctuations by facial motion, pupillometry, and local field potential (LFP) analysis. We found that during awake immobility, the duration of spontaneous whisker-pad motion events correlated with ongoing levels of arousal and modulated both the frequency and the power of theta oscillations. CA1 place cells continued to encode location during immobility, with t

Focal lesions of human hippocampal CA1 neurons in transient global amnesia impair place memory.

A critical role in place learning has been attributed to place cells within the cornu ammonis 1 (CA1) sector of the hippocampus in rodents. The role of CA1 cells in the human hippocampus with regard to place learning remains elusive. Using a virtual Morris water maze, we investigated patients with acute transient global amnesia (TGA), a rare self-limiting dysfunction of the hippocampal system. Fourteen individuals with selective and focal lesions in the CA1 sector of the hippocampus showed a profound impairment in place learning. The size of the lesions and the duration of the TGA correlated w

Field repetition and local mapping in the hippocampus and the medial entorhinal cortex.

Hippocampal place cells support spatial cognition and are thought to form the neural substrate of a global "cognitive map." A widely held view is that parts of the hippocampus also underlie the ability to separate patterns or to provide different neural codes for distinct environments. However, a number of studies have shown that in environments composed of multiple, repeating compartments, place cells and other spatially modulated neurons show the same activity in each local area. This repetition of firing fields may reflect pattern completion and may make it difficult for animals to distingu

Integrated decoding of local and prospective spatial representations for future decision prediction.

Objective.Decoding future spatial decisions from the brain's cognitive map represents a critical step toward cognitive brain-computer interfaces (BCIs) and closed-loop neuromodulation for neurological disorders. However, accurately predicting future paths from hippocampal neural dynamics remains challenging, partly because previous studies have largely overlooked the temporal evolution of early decision-making phases.Approach.We recorded hippocampal CA1 ensemble activity from rats performing a continuous spatial decision task in a modified T-maze. By segmenting the decision process into starti

Mild Traumatic Brain Injury Decreases Spatial Information Content and Reduces Place Field Stability of Hippocampal CA1 Neurons.

Both clinical and experimental studies have reported that mild traumatic brain injury (mTBI) can result in cognitive impairments in the absence of overt brain damage. Whether these impairments result from neuronal dysfunction/altered plasticity is an area that has received limited attention. In this study, we recorded activity of neurons in the cornu Ammonis (CA)1 subfield of the hippocampus in sham and mild lateral fluid percussion injured (mFPI) rats while these animals were performing an object location task. Electrophysiology results showed that the number of excitatory neurons encoding sp

A model of episodic memory: mental time travel along encoded trajectories using grid cells.

The definition of episodic memory includes the concept of mental time travel: the ability to re-experience a previously experienced trajectory through continuous dimensions of space and time, and to recall specific events or stimuli along this trajectory. Lesions of the hippocampus and entorhinal cortex impair human episodic memory function and impair rat performance in tasks that could be solved by retrieval of trajectories. Recent physiological data suggests a novel model for encoding and retrieval of trajectories, and for associating specific stimuli with specific positions along the trajec

Unique deficits in place coding across subfields of the hippocampus in a mouse model of temporal lobe epilepsy.

OBJECTIVE: Memory problems are comorbid with temporal lobe epilepsy (TLE). Animal models of TLE reveal impairments in spatial firing fields of hippocampal place cells, providing a potential neural substrate for memory problems. Each subfield of the hippocampus carries out unique aspects of spatial memory, yet little is known about how individual subfields are perturbed. Here, we investigated the spatial coding properties of the three major subfields of the hippocampus. METHODS: Single unit recordings were made from CA1, CA3, and the dentate gyrus (DG) of mice (n = 10, 6 male [M]/4 female [F])

Disrupted hippocampal replay is associated with reduced offline map stabilization in an Alzheimer's mouse model.

Alzheimer's disease is characterized by progressive memory decline associated with hippocampal degeneration. However, the specific physiological mechanisms underlying hippocampal dysfunction in the disease remain poorly understood-improved knowledge may aid diagnosis and identify new avenues for therapeutic intervention. We investigated how disruptions in hippocampal reactivations relate to place cell stability and spatial memory deficits in an Alzheimer's mouse model. Using the App knockin mouse model NL-G-F, we conducted simultaneous behavioral and electrophysiological recordings in a radial

Dual-mode dynamics in hippocampal populations enable rapid and precise spatial coding.

Hippocampal spatial representations emerge within minutes of first exposure to novel environments, challenging models based solely on gradual synaptic plasticity. Here, we show that CA1 spatial coding is organized according to a dual-mode architecture comprising a pre-organized backbone and an experience-dependent component. Backbone cells expressing immediately emerging place fields form a stable spatial scaffold and dominate positional decoding from the first traversal, whereas experience-dependent cells bearing gradually emerging fields are progressively incorporated to refine spatial repre

Lateralization of CA1 assemblies in the absence of CA3 input.

In the hippocampal circuit CA3 input plays a critical role in the organization of CA1 population activity, both during learning and sleep. While integrated spatial representations have been observed across the two hemispheres of CA1, these regions lack direct connectivity and thus the circuitry responsible remains largely unexplored. Here we investigate the role of CA3 in organizing bilateral CA1 activity by blocking synaptic transmission at CA3 terminals through the inducible transgenic expression of tetanus toxin. Although the properties of single place cells in CA1 were comparable bilateral
