A new species of terrestrial planarian of the genus Microplana (Platyhelminthes: Turbellaria; Tricladida: Continenticola) from Yorkshire, United Kingdom; with a discussion of Microplana humicola Vejdovsky 1890.

Two mostly white terrestrial planarians, about 7 mm long, found in a garden in Yorkshire, UK, are described. They have a single pair of eyes and both specimens are fully mature, with a single pair of ovaries, several testes, a conical penis papilla and a genito-intestinal duct, characters of the genus Microplana, but differ in color and size from other species of the genus and are described as Microplana edwardsi sp. nov.

Failure to classically condition G. dorotocephala and S. mediterranea flatworms using historical and contemporary light-shock paradigms.

Planarian flatworms represent one of the most evolutionarily informative nervous systems for an account of ancient bilaterian brains. Likewise, the unparalleled regenerative ability of planarians makes possible certain investigations of neural development, memory, and behavior that are simply impossible with other model organisms. Despite these facts, learning and memory are today underexplored in planarians, likely due in part to the controversial legacy of 20th-century experiments on the transfer of memories between individual flatworms. Here, we attempted to replicate and extend the classic

'Death and axes': unexpected Ca²⁺ entry phenologs predict new anti-schistosomal agents.

Schistosomiasis is a parasitic flatworm disease that infects 200 million people worldwide. The drug praziquantel (PZQ) is the mainstay therapy but the target of this drug remains ambiguous. While PZQ paralyses and kills parasitic schistosomes, in free-living planarians PZQ caused an unusual axis duplication during regeneration to yield two-headed animals. Here, we show that PZQ activation of a neuronal Ca²⁺ channel modulates opposing dopaminergic and serotonergic pathways to regulate 'head' structure formation. Surprisingly, compounds with efficacy for either bioaminergic network in planarians

Drift of positional identity drives reproductive aging in a long-lived regenerative animal.

Most animals experience irreversibly declining health with advancing age, in part due to limitations in cell turnover and the accumulation of damage. The highly regenerative planarian Schmidtea mediterranea has abundant pluripotent stem cells that drive continuous cell turnover, yet it experiences an age-related loss of fertility, which can be restored through regeneration. We find that the source of planarian age-related infertility lies in the female reproductive system, accompanied by the formation of posterior ectopic ovaries and disrupted accessory reproductive structures, which are resto

DRUG ACTION IN GALVANOTROPIC RESPONSES.

Under strychninization involving reversal of reciprocal inhibition of the circular and longitudinal muscles, planarians and earthworms show reversed, i.e., anodal, galvanotropic responses, which neither nicotine nor atropine induce (in planarians). The results strengthen the conclusion (Crozier, 1926-27) that in causing reversal of inhibition strychnine acts primarily on central synapses or on homologous elements. Like other arthropods, on the other hand, Asellus is little affected by strychnine; no reversal occurs. Caffeine and atropine are even less effective. Nicotine evokes abnormal postur

Stem cells and fluid flow drive cyst formation in an invertebrate excretory organ.

Cystic kidney diseases (CKDs) affect millions of people worldwide. The defining pathological features are fluid-filled cysts developing from nephric tubules due to defective flow sensing, cell proliferation and differentiation. The underlying molecular mechanisms, however, remain poorly understood, and the derived excretory systems of established invertebrate models (Caenorhabditis elegans and Drosophila melanogaster) are unsuitable to model CKDs. Systematic structure/function comparisons revealed that the combination of ultrafiltration and flow-associated filtrate modification that is central

The adaptive evolution divergence of triosephosphate isomerases between parasitic and free-living flatworms and the discovery of a potential universal target against flatworm parasites.

Triosephosphate isomerase (TIM) is an important drug target or vaccine candidate for pathogenetic organisms such as schistosomes. Parasitic and free-living flatworms shared their last common ancestor but diverged from each other for adapting to parasitic and free-living lives afterwards, respectively. Therefore, adaptive evolution divergence must have occurred between them. Here, for the first time, TIMs were identified from three free-living planarian flatworms, namely Dugesia japonica, Dugesia ryukyuensis, and Schmidtea mediterranea. When these were compared with parasitic flatworms and othe

Ca²⁺ channels and praziquantel: a view from the free world.

Targeting the cellular Ca(2+) channels and pumps that underpin parasite Ca(2+) homeostasis may realize novel antihelmintic agents. Indeed, the antischistosomal drug praziquantel (PZQ) is a key clinical agent that has been proposed to work in this manner. Heterologous expression data has implicated an action of PZQ on voltage-operated Ca(2+) channels, although the relevant in vivo target of this drug has remained undefined over three decades of clinical use. The purpose of this review is to bring new perspective to this issue by discussing the potential utility of free-living planarian flatworm

Behavioural and neurochemical mechanisms of learning and memory in Planaria.

Planaria, a freshwater flatworm with unique regenerative abilities, has emerged as a valuable model organism for studying learning and memory. A growing body of evidence shows that planarians can recover their learning behaviour even after decapitation and regeneration. Behavioural paradigms, including classical, operant, and threat-related conditioning responses, show the capacity of planaria in learning and expressing both appetitive and aversive memories. Evidence at the molecular level, further confirms the presence of principal neurotransmitters, including glutamate, gamma-aminobutyric ac

The protection of invertebrates, fish, and vascular plants against inorganic mercury poisoning by sulfur and selenium derivatives.

Protection of organisms against mercury (Hg) poisoning is most commonly associated with the antagonistic effects of selenium (Se)-compounds against mercury alkyls in higher animals. This study shows that there is no consistent difference among Periodic Group VIA derivatives including S(IV), S(II) organic, Se (IV), Se (II) organic, possibly Se VI, as well and Te (IV) in their ability to protect against mercury poisoning. The organisms used in assays were: Coleus explants (leaf abscission); turnip (germination); pea (growth inhibition and Hg uptake); a planarian (regeneration); the brineshrimp (

Danionella cerebrum.

Model organisms are bargains with biology. Squid gave neurophysiology an oversized cable. Arabidopsis gave plant biology a weed that grew too fast to ignore. Planarians gave regeneration biology a body that could be cut into questions. Danionella cerebrum offers a different bargain still: a sexually mature vertebrate whose brain and body remain tiny, transparent and accessible.

Effect of Donepezil, Tacrine, Galantamine and Rivastigmine on Acetylcholinesterase Inhibition in Dugesia tigrina.

Dugesia tigrina is a non-parasitic platyhelminth, which has been recently utilized in pharmacological models, regarding the nervous system, as it presents a wide sensitivity to drugs. Our trials aimed to propose a model for an in vivo screening of substances with inhibitory activity of the enzyme acetylcholinesterase. Trials were performed with four drugs commercialized in Brazil: donepezil, tacrine, galantamine and rivastigmine, utilized in the control of Alzheimer's disease, to inhibit the activity of acetylcholinesterase. We tested five concentrations of the drugs, with an exposure of 24 h,

PHRED-1 is a divergent neurexin-1 homolog that organizes muscle fibers and patterns organs during regeneration.

Regeneration of body parts requires the replacement of multiple cell types. To dissect this complex process, we utilized planarian flatworms that are capable of regenerating any tissue after amputation. An RNAi screen for genes involved in regeneration of the pharynx identified a novel gene, Pharynx regeneration defective-1 (PHRED-1) as essential for normal pharynx regeneration. PHRED-1 is a predicted transmembrane protein containing EGF, Laminin G, and WD40 domains, is expressed in muscle, and has predicted homologs restricted to other lophotrochozoan species. Knockdown of PHRED-1 causes abno

The brain: a concept in flux.

One of the most important aspects of the scientific endeavour is the definition of specific concepts as precisely as possible. However, it is also important not to lose sight of two facts: (i) we divide the study of nature into manageable parts in order to better understand it owing to our limited cognitive capacities and (ii) definitions are inherently arbitrary and heavily influenced by cultural norms, language, the current political climate, and even personal preferences, among many other factors. As a consequence of these facts, clear-cut definitions, despite their evident importance, are

Ethanol exposure induces a delay in the reacquisition of function during head regeneration in Schmidtea mediterranea.

Prenatal exposure to ethanol affects neurodevelopmental processes, leading to a variety of physical and cognitive impairments collectively termed Fetal Alcohol Spectrum Disorders (FASD). The molecular level ethanol-induced alterations that underlie FASD are poorly understood and are difficult to study in mammals. Ethanol exposure has been shown to affect regulation and differentiation of embryonic stem cells in vitro, suggesting that in vivo effects such as FASD could arise from similar alterations of stem cells. In this study, we hypothesize that ethanol exposure affects head regeneration and

Mephedrone ("bath salt") pharmacology: insights from invertebrates.

Psychoactive bath salts (also called meph, drone, meow meow, m-CAT, bounce, bubbles, mad cow, etc.) contain a substance called mephedrone (4-methylcathinone) that may share psychostimulant properties with amphetamine and cocaine. However, there are only limited studies of the neuropharmacological profile of mephedrone. The present study used an established invertebrate (planarian) assay to test the hypothesis that acute and repeated mephedrone exposure produces psychostimulant-like behavioral effects. Acute mephedrone administration (50-1000 μM) produced stereotyped movements that were attenua

Regeneration of the flatworm Prosthiostomum siphunculus (Polycladida, Platyhelminthes).

Fueled by the discovery of head regeneration in triclads (planarians) two and a half centuries ago, flatworms have been the focus of regeneration research. But not all flatworms can regenerate equally well and to obtain a better picture of the characteristics and evolution of regeneration in flatworms other than planarians, the regeneration capacity and stem cell dynamics during regeneration in the flatworm order Polycladida are studied. Here, we show that as long as the brain remained at least partially intact, the polyclad Prosthiostomum siphunculus was able to regenerate submarginal eyes, c

Explosive cytotoxicity of ruptoblasts bridges hormone surveillance and immune defense.

Current understanding of cytotoxic immunity is shaped by hematopoietic-derived cells-T cells, natural killer cells, and neutrophils. Here, we identify "ruptoblasts," a previously unknown cytotoxic glandular cell type in regenerative planarian flatworms. Ruptoblasts undergo an explosive cell death, "ruptosis," triggered by activin, a multifunctional hormone acting as an inflammatory cytokine. Excessive activin-induced through protein injection, genetic chimerism, or bacterial infection-initiates ruptosis, discharging potent diffusible cytotoxic agents capable of eliminating nearby cells, bacter

LPS‑induced impairment of neuroregeneration is alleviated by fluoxetine in Dugesia japonica.

Freshwater planarians (Dugesia japonica) are excellent models for neural regeneration due to their neoblast-dependent regenerative capacity. This study explored the regulatory effects of fluoxetine (FLX) on lipopolysaccharide (LPS)-triggered neuroinflammation as well as cyclosporin A (CsA)-mediated mitophagy inhibition during the regeneration of Dugesia japonica. Planarians were exposed to LPS, FLX, and CsA alone or in combination for 10 days, with assessments of regeneration, locomotion, antioxidant function, DNA damage, neoblast proliferation, apoptosis, and PINK1/Parkin pathway activity.
