RNA m6A methylation fine-tunes the jasmonic acid biosynthesis pathway to modulate plant aging.

N 6-methyladenosine (m6A) is a context-dependent mRNA modification, yet how its dynamics integrate with endogenous signaling to shape plant development remains unclear. Here, we provide further evidence for HIZ2 as an essential component of the m6A writer complex in Arabidopsis, which methylates and destabilizes mRNAs decoding multiple enzymes acting in sequential steps of jasmonic acid (JA) biosynthesis, thereby ensuring JA homeostasis required for timely leaf senescence. HIZ2 interacts with and is indispensable for maintaining the abundance of key m6A writer complex components, including MTA

Role of Nitric Oxide in Postharvest Senescence of Fruits.

Nitric oxide (NO) acts as a gaseous signalling molecule and is considered to be a key regulator in the postharvest storage of fruits. Postharvest senescence is one of the most serious threats affecting the usage and economic value of fruits. Most recent studies have found that exogenous NO application can effectively improve the quality and prolong the shelf life of fruit postharvest by inhibiting postharvest diseases and alleviating chilling injury. Understanding the roles of NO is essential to elucidating how NO activates the appropriate set of responses to postharvest senescence. Here, we c

The H2S-responsive transcription factor ERF.D3 regulates tomato abscisic acid metabolism, leaf senescence, and fruit ripening.

Hydrogen sulfide (H2S) is a signaling molecule that regulates plant senescence. In this study, we found that H2S delays dark-induced senescence in tomato (Solanum lycopersicum) leaves. Transcriptome and reverse transcription quantitative PCR (RT-qPCR) analyses revealed an ethylene response factor ERF.D3 is quickly induced by H2S. H2S also persulfidated ERF.D3 at amino acid residues C115 and C118. CRISPR/Cas9-mediated gene editing, and gene overexpression analyses showed that ERF.D3 negatively regulates leaf senescence and fruit ripening. Abscisic acid (ABA) levels were reduced by ERF.D3 overex

Temporal Dynamics of Viral Load and Genomic Diversity of Plantago Asiatica Mosaic Virus During Long-Term Infection in a Herbaceous Perennial.

Plant positive-strand RNA viruses exhibit high mutation rates that drive genetic diversity and adaptation to different hosts. While herbaceous perennials potentially serve as reservoirs for viral diversification through persistent infections, the mechanisms underlying this process remain largely unexplored. We investigated the temporal dynamics of viral load and genomic diversity of Plantago asiatica mosaic virus (PlAMV) during long-term infection in its natural herbaceous perennial host, Plantago asiatica. Mechanical inoculation of P. asiatica with PlAMV isolate Vi resulted in systemic infect

Targeting oxidative stress, senescence, and neurodegeneration: anti-aging perspectives of Urtica dioica and Centella asiatica.

The aging process is essentially a result of a network of biological events including oxidative stress, chronic inflammation, cellular senescence, mitochondrial dysfunction, and neurodegeneration. In the search for effective multi-target anti-aging strategies, there is a growing trend towards the use of plant compounds with strong bioactivities. Among numerous medicinal plants, Urtica dioica and Centella asiatica are two of the most extensively studied for their antioxidant, anti-inflammatory, neuroprotective, and skin-regenerative effects. This paper is a comprehensive review of the phytochem

Chlorogenic acid ameliorates ovarian aging in laying chickens by restoring lipid metabolism and steroidogenesis via the ERK/PPARα signaling pathway.

BACKGROUND: Ovarian aging is characterized by progressive impairments of follicular development and reproductive performance. Increasing evidence indicates that metabolic dysregulation plays a central role in the decline of ovarian function. Chlorogenic acid (CGA), a widely distributed plant-derived polyphenol, is known for its antioxidant, anti-inflammatory, and metabolic properties. However, its potential role in ovarian aging and the underlying mechanisms remain unexplored. METHODS: D-galactose-induced granulosa cell (GC) senescence model and naturally aging laying chickens were used for e

Mutation Types of CYP71P1 Cause Different Phenotypes of Mosaic Spot Lesion and Premature Leaf Senescence in Rice.

Lesion mimic mutants (LMMs) are ideal materials for studying programmed cell death and defense response in plants. Here we report investigations on two LMMs (msl-1 and msl-2) from the indica rice cultivar JG30 treated by ethyl methyl sulfone. Both of the mutants showed similar mosaic spot lesions at seedling stage, but they displayed different phenotypes along with development of the plants. At tillering stage, larger orange spots appeared on leaves of msl-2, while only small reddish-brown spots exhibit on leaves of msl-1. At heading stage, the msl-2 plants were completely dead, while the msl-

Dual roles of pear EARLY FLOWERING 4 -like genes in regulating flowering and leaf senescence.

BACKGROUND: Flowering is a critical agronomic trait in fruit tree cultivation, essential for sexual reproduction and fruit yield. Circadian clock system, governing processes such as flowering, growth, and hormone signaling, plays a key role in plant adaptability. While some clock-related genes influencing pear flowering have been studied, the role of the PbELF4 (EARLY FLOWERING 4) family remains largely unexplored. RESULTS: In this study, we identified five ELF4 homologous genes within the pear (Pyrus bretschneideri) genome. Phylogenetic analysis delineated two distinct groups within the PbEL

Formononetin attenuates cellular senescence through a FOS-mediated mechanism.

Cellular senescence is characterized by cell cycle arrest accompanied by mitochondrial dysfunction. Despite extensive efforts to overcome this stable state of growth arrest, effective strategies for treating cellular senescence have not been identified. In this study, we screened plant-derived secondary metabolites and confirmed that formononetin is a substance that potently induces the proliferation of senescent fibroblasts. Formononetin-induced proliferation was associated with the suppression of expression of key cell cycle inhibitors, including p53 and Rb-1. This proliferative response was

Monoterpenoids from the Roots of Liquidambar formosana (Formosan Sweet Gum) Exhibit Senomorphic Activity Against Cellular Senescence.

BACKGROUND/OBJECTIVES: Cellular senescence is a hallmark of aging that contributes to tissue dysfunction and age-related diseases. This process is characterized by the activation of the cyclin-dependent kinase inhibitor p16INK4A and the secretion of pro-inflammatory factors collectively known as the senescence-associated secretory phenotype (SASP). In this study, we used human lung-derived cells, including A549 and IMR90 fibroblasts, to identify bioactive compounds from the roots of Liquidambar formosana that suppress p16INK4A activity and attenuate SASP expression. METHODS: Bioactivity-guide

Do red and yellow autumn leaves make use of different photoprotective strategies during autumn senescence?

Our goal was to determine whether anthocyanin-producing species (red) use different photoprotective strategies to cope with excess light during fall senescence compared with non-anthocyanin-producing species (yellow). In a previous study, we found that a yellow species retained the photoprotective PsbS protein in late autumn, while a red species did not. Specifically, we tested the hypothesis that red species make less use of zeaxanthin and PsbS-mediated thermal dissipation, as they rely on anthocyanins for photoprotection. We monitored four red (Acer ginnala, Rhus typhnia, Parenthocissus quin

Re-Appearing Legacy Effect in Timing of Autumnal Leaf Senescence and Compensation Growth After Severe Drought in Fagus sylvatica L.

Drought events are expected to induce both immediate and delayed effects on tree functioning, yet the persistence and dynamics of these legacy effects remain poorly understood. In an experimental set-up, European beech (Fagus sylvatica L.) saplings were subjected to two consecutive water-withholding treatments during the 2022 growing season, resulting in five drought-severity categories. We monitored these categories over the following three years to assess drought legacy effects on phenology and growth. In the first post-drought year, bud burst proceeded more slowly in all drought categories,

AtMPK1/AtMPK2 Interact with AtVQ25 to Enhance Its Protein Stability and Coordinately Regulate Salicylic Acid-Mediated Leaf Senescence.

Leaf senescence is a pivotal developmental program in plants, precisely regulated by diverse signals including salicylic acid (SA). The Arabidopsis VQ protein AtVQ25 has been previously characterized as a positive regulator of SA-mediated leaf senescence, functioning through interaction with AtWRKY53 to relieve the transcriptional self-repression of AtWRKY53 at its own promoter. However, the upstream regulatory mechanisms governing AtVQ25 itself remain elusive. In this study, a yeast library screening was performed, and the mitogen-activated protein kinases AtMPK1 and AtMPK2 were identified as

Fisetin as a senotherapeutic agent: Evidence and perspectives for age-related diseases.

Fisetin, a flavonoid naturally occurring in plants, fruits, and vegetables, has recently gained attention for its potential role as a senotherapeutic agent for the treatment of age-related chronic diseases. Senotherapeutics target senescent cells, which accumulate with age and disease, in both circulating immune cell populations and solid organs and tissues. Senescent cells contribute to development of many chronic diseases, primarily by eliciting systemic chronic inflammation through their senescence-associated secretory phenotype. Here, we explore whether fisetin as a senotherapeutic can eli

Hydrogen sulfide: A versatile gaseous molecule in plants.

Hydrogen sulfide (H2S) is a gasotransmitter and signaling molecule associated with seed germination, plant growth, organogenesis, photosynthesis, stomatal conductance, senescence, and post-harvesting. H2S is produced in plants via both enzymatic and non-enzymatic pathways in different subcellular compartments. Exogenous application of H2S facilitates versatile metabolic processes and antioxidant machinery in plants under normal and environmental stresses. This compound interacts with phytohormones like auxins, abscisic acid, gibberellins, ethylene, jasmonic acid, and salicylic acid. Furthermor

Functions of Melatonin during Postharvest of Horticultural Crops.

Melatonin, a tryptophan-derived molecule, is endogenously generated in animal, plant, fungal and prokaryotic cells. Given its antioxidant properties, it is involved in a myriad of signaling functions associated with various aspects of plant growth and development. In higher plants, melatonin (Mel) interacts with plant regulators such as phytohormones, as well as reactive oxygen and nitrogen species including hydrogen peroxide (H2O2), nitric oxide (NO) and hydrogen sulfide (H2S). It shows great potential as a biotechnological tool to alleviate biotic and abiotic stress, to delay senescence and

Relationship between DNA methylation and ethylene in plants: A review.

DNA methylation, as a central mechanism of epigenetic regulation, plays a key role in coordinating plant growth and stress responses. Simultaneously, the gaseous phytohormone ethylene is also a crucial regulator in these processes. Both form a metabolic competition through S-adenosyl-L-methionine (SAM) and mutually regulate each other at the level of gene expression. Studies demonstrate that DNA methylation functions to either enhance or block ethylene synthesis and ethylene signal transduction, thereby regulating a spectrum of developmental processes including seed development, root growth, f

Adapting Crops to Rising Temperatures: Understanding Heat Stress and Plant Resilience Mechanisms.

Global temperature rise has become a critical challenge to agricultural sustainability, severely affecting crop growth, productivity, and survival. Human-induced climate change and greenhouse gas emissions cause heat stress, disrupting plant metabolism and physiology at all developmental stages from germination to harvest. Elevated temperatures during germination impair water uptake, enzyme activity, and energy metabolism, leading to poor or uneven seedling emergence. At key phases such as flowering and grain filling, heat stress limits photosynthesis and transpiration by inducing stomatal clo

Solanum nigrum L.-derived nanovesicles as novel nanotherapeutics suppressing prostate cancer progression via senescence-based antitumor activity.

Current therapies for prostate cancer are limited by toxicity and acquired resistance, motivating development of biocompatible nanotherapeutics. Here, Solanum nigrum L.-derived nanovesicles (SDNVs) were isolated and characterized, showing a mean diameter of 93.4 ± 0.2 nm and a particle concentration of (4.2 ± 0.4) × 1011/mL. SDNV uptake, antitumor activity, and safety were assessed in PC-3 and RWPE-1 cells, while efficacy and biodistribution were further evaluated in a subcutaneous PC-3 xenograft mouse model following oral administration. SDNVs were readily internalized by PC-3 cells and suppr

Love Me Not Meter: A Sensor Device for Detecting Petal Detachment Forces in Arabidopsis thaliana.

Abscission is the developmentally controlled loss of plant organs, providing diverse functions such as shedding senescent or diseased leaves, dispersing seeds, or dropping ripened fruit. This process is defined by the hydrolytic breakdown of the middle lamella in the abscission zone, allowing for organ detachment. While the model plant Arabidopsis thaliana does not undergo leaf abscission, it does have a predictable progression of floral organ abscission. To study abscission zone physical integrity in Arabidopsis, a breakstrength meter for Arabidopsis was developed to reliably measure the peta
